Quantification of Hydrogen Sulfide in Single Living Cells by Azido-Functionalized Glass Nanopores.

Hydrogen sulfide (H2S) is the third gasotransmitter after nitrogen oxide and carbon monoxide, playing vital roles in physiological controls, such as angiogenesis, vascular homeostasis, thrombosis, inflammation, and remodeling. Monitoring the content level and dynamic variations of H2S within cells will help understand its involvement and relationship in cancer proliferation and may also lead to new cancer therapy options. Herein, we developed a simple and effective approach for ionic rectification sensing detection of H2S in single living cells using p-azidobenzoic acid (PA)-functionalized gla

Formation of dimethyl sulfide (CH3SCH3) and ethanethiol (CH3CH2SH) in interstellar analog ices of methane (CH4) and hydrogen sulfide (H2S).

Hitherto unidentified abiotic formation pathways leading to the organosulfur molecules ethanethiol (C2H5SH), methanethiol (CH3SH) and, dimethyl sulfide (CH3SCH3) were investigated through a series of laboratory simulation experiments. Interstellar analog ices of methane (CH4) and hydrogen sulfide (H2S) were exposed to proxies of galactic cosmic rays (GCRs) in the form of energetic electrons released in the GCR track in interstellar ices simulating typical cold molecular cloud lifetimes of a few 106 to 107 years. During the temperature-programmed desorption phase, the molecules subliming fracti

Harnessing methylotrophic methanogenesis to overcome salinity inhibition and sulfide production in anaerobic digestion.

Saline biomass represents an emerging feedstock for anaerobic digestion (AD), yet methane production is often limited by salinity and competition from sulfate-reducing microorganisms (SRM), leading to hydrogen sulfide formation. Unlike conventional methanogenesis pathway, methylotrophic pathway naturally thrives in saline environments by metabolizing methylated compounds without SRM interference nor saline inhibition. This study compared trimethylamine (TmaCl) and microcrystalline cellulose (MCC) as substrates, with or without molybdate (SRM inhibitor), at 0 and 25 g sea salt/L. Methane yields

Endogenous H₂S promotes HSPA8 sulfhydration to downregulate HIF1α and prevent ferroptosis in septic myocardial injury.

BACKGROUND: Sepsis-induced myocardial injury (SIMI) contributes significantly to morbidity and mortality in sepsis, but its molecular mechanisms are not fully understood. Hydrogen sulfide (H₂S), an endogenous signaling molecule, regulates inflammation, oxidative stress, and cell death in cardiovascular diseases, with protein sulfhydration as a key mechanism. METHODS: We used in vitro and in vivo sepsis models we investigated the protective to examine the effects of H₂S donors (GYY4137 and Allicin) on SIMI. We focused on ferroptosis and the HIF1α/BNIP3 axis, and applied transcriptomic, proteom

Olfactory training selectively modifies cortical responses to odors in healthy adults.

Olfactory training (OT) is an effective method for rehabilitating smell dysfunctions through regular stimulation of the olfactory system. However, the exact mechanism through which OT improves olfaction remains elusive, especially regarding cortical processing of odors. This study aimed to examine the effects of OT on cortical responses to odors in young healthy individuals. Ninety-four participants (Mage = 25.3, SD = 4.5 years; 71 women) were asked to either (1) conduct bi-daily OT with the odors of rose (phenyl ethyl alcohol, PEA), cloves (eugenol), lemon (citronellal), eucalyptus (eucalypto

Architectural modulation of polymeric hydrogen sulfide donors: Structure-responsive materials for controlled therapeutics.

Hydrogen sulfide (H2S), once regarded merely as a malodorous and toxic gas, has emerged as a vital endogenous signaling molecule with multifaceted physiological roles, encompassing neuromodulation, vasorelaxation, cytoprotection, and the regulation of inflammatory responses. Despite this growing recognition of its biological significance, the therapeutic translation of H2S remains impeded by its physicochemical limitations as volatility, poor aqueous solubility, and rapid metabolic clearance. Addressing these challenges has stimulated the development of polymeric hydrogen sulfide (H2S) donors

"Interfacial Activation" High-Fidelity Sensing Strategy: Two-Photon Fluorescence Imaging of Transmembrane H2S Release.

Precise elucidation of dynamic chemical events at the cell membrane interface is vital for understanding fundamental biological processes. However, current fluorescent tools often fail to confine signal generation strictly to the membrane, leading to background interference that compromises imaging fidelity. To address this, we propose an "Interfacial Activation" strategy that transforms the cell membrane from a passive anchoring point to an active signal trigger. This is realized using NACP, an amphiphilic two-photon fluorophore that remains completely nonemissive in aqueous media via aggrega

Hydrogen sulfide in ocular physiology and pathology: molecular Mechanisms, therapeutic Paradoxes, and delivery challenges.

Hydrogen sulfide (H₂S), once known as a toxic gas, is now acknowledged as a fundamental gasotransmitter essential for ocular homeostasis. This review critically examines the paradoxical role of H₂S in the eye, how it acts as both a vital signaling molecule and a potential pathological contributor depending on concentration, cellular context, and disease stage. We explore the compartmentalized synthesis of H₂S via three principal enzymatic pathways, viz., cystathionine-β-synthase (CBS), cystathionine-γ-lyase (CSE), and the 3-mercaptopyruvate sulfurtransferase/cysteine aminotransferase (3MST/CAT

Real-world Study of Three-gas Breath Testing Nationwide and the Association With Symptoms.

GOALS AND BACKGROUND: Hydrogen (H 2 ) breath testing (BT) is used to diagnose small intestinal bacterial overgrowth (SIBO), and methane (CH 4 ) to diagnose constipation. Recently, a third microbially derived gas, hydrogen sulfide (H 2 S), was added to BT and associated with diarrhea. Here, we assess a 3-gas BT nationwide, comparing the results to gastrointestinal (GI) symptoms. STUDY: Consecutive subjects (N=6000) undergoing 3-gas at-home BT were asked to complete a symptom questionnaire. Substrate choice (glucose or lactulose) was at physician's discretion. H 2 , CH 4, and H 2 S levels, and

Investigation of organosulfur molecules in experimental acute pancreatitis: Antioxidant and antiferroptotic actions of ATB-346.

AIMS: Acute pancreatitis (AP), an acute inflammatory disorder of the exocrine pancreas, is a potentially life-threatening disease with no specific treatment. Hydrogen sulfide (H2S) donor organosulfur compounds administered exogenously typically exert anti-inflammatory effects. We aimed to compare the five most widely studied organosulfurs (ATB-346, dimethyl trisulfide, diallyl trisulfide, GYY4137, AP39) as therapeutic agents against experimental AP in mice, select the most effective one and provide insights into its mechanisms of action. RESULTS: Each organosulfur molecule dose-dependently de

Injectable, self-healing and wound microenvironment responsive hydrogel for photothermal, chemodynamic, and hydrogen sulfide gas synergistic therapy of the infected diabetic wound healing.

Bacteria-infected diabetic wounds represent a significant global healthcare concern. Uncontrolled bacterial infection results in excessive production of reactive oxygen species (ROS), sustained inflammatory responses and immunosuppression, which seriously impede healing processes. Conventional antibiotic treatments are often inadequate for addressing these challenges and concurrently pose the risk of promoting bacterial resistance. Here, an injectable, self-healing, and wound microenvironment-responsive hydrogel (HA-PBA/OAP/FeS@PC) with photothermal effects, chemodynamic therapy, and hydrogen

Thiotaurine is a sulfane sulfur donor to the biological thiols glutathione and cysteine.

Persulfide and hydrogen sulfide chemistry is relevant to the prevention of cellular oxidative damage. Thiotaurine is a poorly-studied precursor to the abundant biological molecule taurine and contains a labile sulfane sulfur. Here, reversible reactions of thiotaurine with glutathione or cysteine were investigated to observe formation of persulfides and hydrogen sulfide gas. Raman, Visible, and NMR spectroscopy were employed to characterize the reaction products and observe reaction kinetics. The reactions were verified through a kinetic competition experiment involving hydrogen peroxide. Data

EGCG promotes apoptosis in BT-549 triple-negative breast cancer cells by targeting STAT3.

BACKGROUND: Triple-negative breast cancer (TNBC), defined by the lack of estrogen receptor (ER), progesterone receptor (PR), and human epidermal growth factor receptor 2 (HER2) expression, is an aggressive subtype of breast cancer (BC) with limited therapeutic options. Signal transducer and activator of transcription 3 (STAT3) plays a critical oncogenic role in TNBC by promoting tumor progression and conferring resistance to apoptosis. (-)-epigallocatechin-3-gallate (EGCG), a bioactive green tea polyphenol, has been reported to inhibit STAT3 signaling and exert anti-cancer effects. However, it

Hydrogen sulfide at the intersection of hypoxia and oxidative stress: implications for lung cancer progression and treatment - a narrative review.

Lung cancer remains a leading cause of cancer-related mortality worldwide, with a pathogenesis deeply influenced by the tumor microenvironment. Two central and interrelated factors -hypoxia and oxidative stress - contribute significantly to tumor progression, angiogenesis, metabolic reprogramming, and therapeutic resistance. In recent years, hydrogen sulfide (H2S), traditionally viewed as a toxic gas, has gained recognition as a critical gasotransmitter with a regulatory role in both hypoxic and redox signaling pathways in cancer biology. Endogenously produced by enzymes such as CBS, CSE, and

Effects of High Lithium Concentrations on the Growth, Biomass, Mineral Accumulation, Oxidative Stress, Antioxidant and Gene Expression Response, and DNA Methylation in Sunflower Plants.

This study demonstrates that sunflower plants display integrated, multilevel responses to excessive lithium (Li) exposure. Li concentrations above 5 mM markedly impair germination, growth, and biomass accumulation. Li is preferentially accumulated in the shoots, showing high translocation and bioaccumulation factors, and disrupts mineral nutrient homeostasis, particularly potassium (K) and sodium (Na) uptake, while inducing oxidative stress. Although photosynthetic pigment contents decline, photosynthetic efficiency is largely maintained, except at 10 mM Li. Li treatment enhances superoxide an

Alcohol-Induced Dysregulation of Hydrogen Sulfide Signaling in Alzheimer's Disease-Narrative Mechanistic Synthesis Review.

Alcohol use disorder (AUD) is highly comorbid with psychiatric conditions and is increasingly recognized as a modifiable factor associated with cognitive decline and dementia, including Alzheimer's disease (AD). While epidemiological and experimental studies consistently demonstrate that chronic alcohol exposure exacerbates neurodegenerative vulnerability rather than implying a single dominant causal pathway, accumulating evidence supports a multifactorial and context-dependent framework in which alcohol acts as a disease-modifying stressor that perturbs endogenous adaptive and resilience mech

Hydrogen sulfide promotes wheat immunity against stripe rust through TaATG6c persulfidation.

Hydrogen sulfide (H2S) is a key gaseous signaling molecule involved in plant growth and stress responses, yet its role in wheat resistance to stripe rust remains poorly understood. Here, we show that exogenous H2S enhances resistance of wheat (Triticum aestivum L.) to Puccinia striiformis f. sp. tritici (Pst), the causative agent of stripe rust. Comparative persulfidation proteomics identified the autophagy-related protein TaATG6c as a Pst-responsive H₂S target. Site-specific mass spectrometry and a modified biotin-switch assay demonstrated that Cys177 and Cys180 of TaATG6c undergo H₂S-induced

Causal relationship between hydrogen sulfide and osteoarthritis: A Mendelian randomization analysis.

Hydrogen sulfide (H2S) has been shown to alleviate bone and joint inflammation in osteoarthritis (OA). Exploring the connection between H2S and OA could reveal novel therapeutic avenues for treating this condition. This study utilized H2S as the exposure variable and OA as the outcome within a Mendelian randomization framework. Data for genome-wide association studies related to both exposure and outcome were sourced from the IEU OpenGWAS project. To estimate causal relationships, inverse variance weighted method was employed, along with supplementary analytical techniques (MR-Egger, weighted

AIE fluorescent bio-probe for recognition of exogenous and endogenous H2S signaling molecules, and targeted detection of nonalcoholic fatty liver.

Hydrogen sulfide (H₂S) has attracted attention as a harmful substance causing ecological pollution. However, due to the lack of H2S specific detection tools, the changes in endogenous hydrogen sulfide levels during the pathological progression of chronic liver disease are not fully understood. In this study, a novel aggregation induced emission (AIE) fluorescent bio-probe HBA was developed to facilitate high-selective detection of exogenous and endogenous H2S. HBA showed strong water solubility, high selectivity, low detection limit (0.45 nM), rapid responsiveness, high fluorescence quantum yi
