Accumulation and depuration of ciguatoxins in green-lipped mussels fed toxic Gambierdiscus polynesiensis: Evidence from controlled feeding and transcriptomic analysis.

Ciguatera poisoning (CP) was traditionally associated with fish consumption, but other marine invertebrates such as gastropods and bivalves can also serve as non-traditional vectors. In this study, ciguatoxins (CTXs) accumulation, tissue distribution, depuration kinetics, and associated transcriptomic responses were studied using controlled feeding trials on green-lipped mussels (Perna canaliculus), an important seafood species, fed Gambierdiscus polynesiensis. In two experiments, mussels accumulated CTXs rapidly, peaking at concentrations of 5.14 µg kg-1 (day 19, Experiment 1) and 4.57 µg kg-

The occurrence of paralytic shellfish toxins in invertebrates, forage fishes and predatory fishes across southern Alaska.

In Alaska, paralytic shellfish poisoning (PSP) is associated with the toxic dinoflagellate Alexandrium catenella, which blooms annually along the coastline from the Canadian border to the Beaufort Sea. While it has long been recognized that these blooms cause annual outbreaks of shellfish toxicity each spring and summer, little is known about other pathways for PSP toxins in the marine food web. Here, we present data from a four-year study to characterize the occurrence of PSTs in marine non-bivalve invertebrates, forage fishes and predatory fishes across the breadth of southern Alaska. Sample

Transcriptomic comparison unveils saxitoxin biosynthesis genes in the marine dinoflagellate Gymnodinium catenatum.

The marine dinoflagellate Gymnodinium catenatum is known to produce saxitoxins (STXs) that are responsible for paralytic shellfish poisoning (PSP); however, the genes involved in STX synthesis are insufficiently understood. In the present study, we determined transcriptome sequences of toxic G. catenatum isolated from Korea (Gc-KR) and compared them with G. catenatum strains reported in other regions such as Spain (Gc-SP) and the United States (Gc-US). Toxin analysis showed that the Korean strain produced the toxins at 6.0 ± 1.9 STXs eq fmol/cell. Comparative transcriptomics of the three strai

Marine Biotoxins in Crustaceans and Fish-A Review.

In recent years, there has been an increase in the consumption of seafood such as shellfish and crustaceans due to their pleasant taste and nutritional value. Fish are also a crucial part of a healthy, balanced diet. However, the consumption of these products may cause food poisoning through marine biotoxins. In recent years, several legal acts have been published by the European Commission to regulate toxin limits and describe their reference analysis methods. Commission Regulation (EC) No. 853/2004 established the maximum contents of marine biotoxins only in bivalve mollusks. Although other

Differences in Marine Toxin Poisonings Reported to US Poison Centers After Pandemic Restrictions.

This study investigated whether marine toxin poisonings reported to U.S. Poison Centers changed during the height of the pandemic period (April 2020 to December 2021). The National Poison Data System was queried for single-substance human exposure calls between 1 January 2000 and 31 March 2022 pertaining to ichthyosarcotoxins. Incidence rate ratios for exposure calls were calculated using mixed-effects negative binomial regression. Call counts were aggregated by year and regressed on a binary indicator for occurrence during pandemic restrictions. During the peak pandemic period, exposure calls

Estimating Network Adequacy of Autism Service Providers in Affordable Care Act Markets.

Castrate-Resistant Prostate Cancer (CRPC) is characterized by persistent androgen receptor-driven tumor growth in the apparent absence of systemic androgens. Current evidence suggests that CRPC cells can produce their own androgens from endogenous sterol precursors that act in an intracrine manner to stimulate tumor growth. The mechanisms by which CRPC cells become steroidogenic during tumor progression are not well defined. Herein we describe a novel link between the elevated cholesterol phenotype of CRPC and the TERE1 tumor suppressor protein, a prenyltransferase that synthesizes vitamin K-2

Spatiotemporal transitions in Pseudo-nitzschia species assemblages and domoic acid along the Alaska coast.

The toxic diatom genus Pseudo-nitzschia is distributed from equatorial to polar regions and is comprised of >57 species, some capable of producing the neurotoxin domoic acid (DA). In the Pacific Arctic Region spanning the Bering, Chukchi, and Beaufort seas, DA is recognized as an emerging human and ecosystem health threat, yet little is known about the composition and distribution of Pseudo-nitzschia species in these waters. This investigation characterized Pseudo-nitzschia assemblages in samples collected in 2018 during summer (August) and fall (October-November) surveys as part of the Distri

Concentrations of Tetrodotoxin (TTX) and Its Analogue 4,9-Anhydro TTX in Different Tissues of the Silver-Cheeked Pufferfish (Lagocephalus sceleratus, Gmelin, 1789) Caught in the South-Eastern Mediterranean Sea, Lebanon.

Pufferfishes are among the best-known marine organisms that accumulate marine biotoxins such as Tetrodotoxin (TTX). In the Mediterranean Sea, the silver-cheeked toadfish Lagocephalus sceleratus is the most reported TTX-bearer, causing many fatal and non-fatal cases. In Lebanon, no previous studies have measured TTX levels although the possibility of TTX-poisoning is high since L. sceleratus is caught in different sizes and can be mistaken with other small fishes. Hence, this study reports TTX and its analogue 4,9-anhydro TTX in L. sceleratus collected from Lebanese waters in the Eastern Medite

Toxic Effects and Tumor Promotion Activity of Marine Phytoplankton Toxins: A Review.

Phytoplankton are photosynthetic microorganisms in aquatic environments that produce many bioactive substances. However, some of them are toxic to aquatic organisms via filter-feeding and are even poisonous to humans through the food chain. Human poisoning from these substances and their serious long-term consequences have resulted in several health threats, including cancer, skin disorders, and other diseases, which have been frequently documented. Seafood poisoning disorders triggered by phytoplankton toxins include paralytic shellfish poisoning (PSP), neurotoxic shellfish poisoning (NSP), a

Potent allelopathy and non-PSTs, non-spirolides toxicity of the dinoflagellate Alexandrium leei to phytoplankton, finfish and zooplankton observed from laboratory bioassays.

The dinoflagellate genus Alexandrium has been well known for causing paralytic shellfish poisoning (PSP) worldwide. Several non-PSP toxin-producing species, however, have shown to exhibit fish-killing toxicity. Here, we report the allelopathic activity of Alexandrium leei from Malaysia to other algal species, and its toxicity to finfish and zooplankton, via laboratory bioassays. Thirteen microalgal species that co-cultured with Al. leei revealed large variability in the allelopathic effects of Al. leei on the test algae, with the growth inhibition rates ranging from 0 to 100%. The negative all

Harmful algal blooms and their effects in coastal seas of Northern Europe.

Harmful algal blooms (HAB) are recurrent phenomena in northern Europe along the coasts of the Baltic Sea, Kattegat-Skagerrak, eastern North Sea, Norwegian Sea and the Barents Sea. These HABs have caused occasional massive losses for the aquaculture industry and have chronically affected socioeconomic interests in several ways. This status review gives an overview of historical HAB events and summarises reports to the Harmful Algae Event Database from 1986 to the end of year 2019 and observations made in long term monitoring programmes of potentially harmful phytoplankton and of phycotoxins in

Deeper insight into Gambierdiscus polynesiensis toxin production relies on specific optimization of high-performance liquid chromatography-high resolution mass spectrometry.

Ciguatera food poisoning affects consumer health and fisheries' economies worldwide in tropical zones, and specifically in the Pacific area. The wide variety of ciguatoxins bio-accumulated in fish or shellfish responsible for this neurological illness are produced by marine dinoflagellates of the genus Gambierdiscus and bio-transformed through the food web. The evaluation of the contents of ciguatoxins in strains of Gambierdiscus relies on the availability of standards and on the development of sensitive and specific tools to detect them. There is a need for sensitive methods for the analysis

New evidence of pectenotoxins in farmed bivalve molluscs from Sardinia (Italy).

Several planktonic dinoflagellates can produce lipophilic phycotoxins that represent a significant threat to public health as well as to shellfish and fish farming. Poisoning related to some of these toxins is categorised as diarrhetic shellfish poisoning. We analysed 975 shellfish samples from Tortoli in the central-eastern region of Sardinia (Italy) from January 2016 to March 2020, to investigate the prevalence of different lipophilic marine biotoxins in mollusc bivalves. The results highlighted the predominant presence of toxins belonging to the okadaic acid group in all samples with toxin

Spectroscopic ellipsometry methods for brevetoxin detection.

The spectroscopic ellipsometry (SE), and attenuated internal reflection spectroscopic ellipsometry (TIRE) are promising methods in label-free biosensing applications. An ellipsometer running under surface plasmon resonance (SPR) conditions has unique advantages over other SPR-based methods in terms of sensitivity and real-time/label-free measurement capability. In this study, both SE and TIRE-based brevetoxin B (BTX) sensors were developed using two anti-BTX aptamers reported before. A new aptamer sequence was also derived from these two antiBTX aptamers using predictive modeling tools and an

Emerging harmful algal bloom species over the last four decades in China.

The first recorded micro-algae bloom in Chinese coastal waters dates back to 1933 and was caused by a mixture of Noctiluca scintillans and Skeletonema costatum sensu lato along the Zhejiang coast (the East China Sea). While well-documented harmful algal blooms (HABs) appeared to be extremely scarce from the 1950s to 1990, both the frequency and intensity have been reportedly increasing since 1990. Among them, the fish-killing HABs, mainly caused by Karenia mikimotoi, Karlodinium digitatum, Karlodinium veneficum, Margalefidinium polykrikoides, and Heterocapsa spp., have intensified. Karenia mik

Toxic effects, mechanisms, and ecological impacts of harmful algal blooms in China.

Over the last 30 years, harmful algal blooms (HABs) have occurred frequently in the coastal waters of China, resulting in financial losses of over 5.9 billion yuan (about 0.87 billion US dollars) due to massive fish and shellfish mortalities and negative impacts on tourism. To better understand HABs in China, herein we summarized bloom events with massive fish/shellfish mortalities and/or economic losses. Our results suggest that the diversity of HAB species has increased over the last 30 years, with the main causative species shifting from the raphidophyte Chattonella marina and dinoflagellat
