Alkylphenolic compounds in edible molluscs of the Adriatic Sea (Italy).

This paper reports the first group of results on alkylphenol (APE) contamination of seafood in the Adriatic Sea, in the framework of a national project on the quality of this Sea (PRISMA 2). Nonylphenol (NP), octylphenol (OP), and their ethoxylates (NPE and OPE) were detected in edible molluscs, either filter feeders or predators (clams, mussels, cuttlefishes, and squids), caught from 15 harbors along the Italian coast in the Adriatic Sea in 1997. NP was the compound found always at levels much higher than the other APEs in all the examined species. It reached the maximum concentration of 696

Heterocyclic amines: human carcinogens in cooked food?

During the frying of meat and fish, genotoxic heterocyclic amines (HCAs) are formed. The dietary exposure to HCAs may be implicated in the aetiology of human cancer, but there may be other factors in our diet that prevent the genotoxic effects of these compounds. Within the project described here, we plan to identify regional and individual cooking habits that affect HCA-levels in our food. These are determined with a validated analytical method and the exposure to HCAs is estimated by dietary assessment. Biomarker analysis will be employed to estimate recent or long-term exposure to HCAs. In

Histamine and biogenic amine production by Morganella morganii isolated from temperature-abused albacore.

Histamine-producing bacteria were isolated from albacore stored at 0, 25, 30, and 37 degrees C. They were screened using Niven's differential medium, and their histamine production was confirmed by high-pressure liquid chromatography analysis. The optimum temperature for growth of histamine-producing bacteria was 25 degrees C. The bacterium producing the highest level of histamine was isolated from fish abused at 25 degrees C. It was identified as Morganella morganii by morphological, cultural, biochemical, and antimicrobial characteristics and by the Vitek microbial identification system. The

Toxic effects, pharmacokinetics and clearance of saxitoxin, a component of paralytic shellfish poison (PSP), in cats.

Saxitoxin (STX) was the first known and most studied toxic component of paralytic shellfish poisoning (PSP). This toxin blocks neuronal transmission by binding to the voltage-gated Na+ channel. Although the toxin's mechanism of action is well known at the molecular level, there are still many unresolved questions about its pharmacokinetics and the PSP intoxication syndrome in mammals. Some of these questions are addressed in the present paper, which describes an experimental design which allowed us to follow the dynamics of STX poisoning in vivo. Adult cats were anaesthetized and permanently c

Aquatic biotoxins: design and implementation of seafood safety monitoring programs.

Naturally occurring toxicants are usually odorless, tasteless, and generally undetectable by any simple chemical test. Various programs have been established that are effective in reducing risks associated with these toxicants in food. These programs include setting regulatory limits, monitoring susceptible commodities for toxin levels, and using decontamination procedures. Bioassays have been used traditionally to monitor suspect products. All traditional bioassays, however, have one common disadvantage, i.e., the lack of specificity for individual toxins. The lack of available reference stan

Delayed evoked potentials in children exposed to methylmercury from seafood.

Methylmercury poisoning may cause constriction of visual fields and deafness, especially if exposure occurs prenatally. However, the risks associated with exposure from contaminated seafood is unclear. We examined 149 children attending first grade in a Madeiran fishing community. As maternal dietary habits were relatively unchanged, current maternal hair concentrations were used as indicator of the child's prenatal exposure to methylmercury (geometric average, 9.64 microg/g [48.2 nmol/g]). After adjustment for age and sex, the mean (+/-SD) latency of peak III of the brainstem auditory evoked

Human health implications associated with mucilage in the northern Adriatic Sea.

Mucilage in the northern Adriatic Sea is well known for its negative impact not only on the ecology of the affected area and on fishing activities but on tourism as well. The microhabitat mucilage creates in the sea can provide favourable conditions for the growth and/or survival of some environmental microorganisms that include human opportunistic pathogens. It also seems to favour the selective development of some marine toxic algae. Finally, mucilage can concentrate chemical contaminants from surrounding waters, hence increasing their bioaccumulation in seafoods. This paper examines the pos

Vitamin A toxicity secondary to excessive intake of yellow-green vegetables, liver and laver.

We report a case of sudden onset of vitamin A poisoning. A 20-year-old Japanese woman had been eating pumpkin and only a very limited amount of other foods on a daily basis for 2 years. She was overly concerned about weight reduction. Aurantiasis cutis and abnormal liver function tests were noted by her family doctor in 1995 when she was 18 years old. At that time, she stopped eating pumpkin. However, she secretly continued an excessive intake of other beta-carotene-rich vegetables, liver and laver for about 2 years. Two and one-half years after being seen by her family physician, she experien

Predicted and observed growth and toxigenesis by Clostridium botulinum type E in vacuum-packaged fishery product challenge tests.

The observed growth and toxigenesis by Clostridium botulinum type E in vacuum-packaged unprocessed, raw pickled and cold-smoked rainbow trout stored at slightly abusive temperatures were compared to predictions generated by two currently available predictive microbiological programs, Food MicroModel and Pathogen Modelling Program. In unprocessed fish there was only a 2 log increase in type E cell count at the time the toxicity first occurred after 2 weeks storage at 8 degrees C. Neither growth or toxin production was observed in raw pickled fish with a NaCl concentration of 6.7% (w/v) during 6

Fate and risk evaluation of persistent organic contaminants and related compounds in Victoria Harbour, Hong Kong.

The Environment Protection Department of Hong Kong has a monitoring program for persistent organic contaminants in sediments of Victoria Harbour, the main harbour of Hong Kong. A fugacity model has been used, based on this sedimentary data, to estimate inputs to the system (probably from sewage, stormwater and industrial discharges) as well as the fate of the contaminants, particularly in terms of the aqueous and biotic concentrations. The risk of deleterious effects on the natural marine system, as well as on the consumers of seafood from the system, was carried out using the estimated aqueou

Dimercaptosuccinic acid (DMSA), a non-toxic, water-soluble treatment for heavy metal toxicity.

Heavy metals are, unfortunately, present in the air, water, and food supply. Cases of severe acute lead, mercury, arsenic, and cadmium poisoning are rare; however, when they do occur an effective, non-toxic treatment is essential. In addition, chronic, low-level exposure to lead in the soil and in residues of lead-based paint, to mercury in the atmosphere, in dental amalgams and in seafood, and to cadmium and arsenic in the environment and in cigarette smoke is much more common than acute exposure. Meso-2,3-dimercaptosuccinic acid (DMSA) is a sulfhydryl-containing, water-soluble, non-toxic, or

Oral toxicity in mice of algal toxins from the diarrheic shellfish toxin (DST) complex and associated toxins.

Mussel samples from four locations along the Norweigian coast were extracted by methods for diarrheic shellfish toxins (DST) and tested by chemical and biological methods, including histopathology. All samples had previously been found to be highly toxic in mice, with symptoms indicating the presence of non-diarrheagenic toxins in the mouse bioassay. Chemical analysis revealed that the DST okadaic acid (OA) and dinophysistoxin-1 (DTX1) were present each one in one sample, but only a minor part of the total toxicity could be attributed to these toxions. In the other two samples, OA and DTX1 wer

Dietary and tissue residue analysis and contaminant intake estimations in rats consuming diets composed of Great Lakes salmon: a multigeneration study.

To further characterize the toxicological risk associated with chemical contaminants in Great Lakes fish, a multigeneration rat reproduction study was designed. Mature chinook salmon (Oncorhynchus tsawytscha), collected during the Fall 1991 spawning runs from Sydenham River, Lake Huron, and Credit River, Lake Ontario, were filleted, lyophilized, and incorporated into standard rat diets at 25% (w/w) or 100% (w/w) of the normal protein compliment [casein, 20% (w/w)]. This resulted in diets composed of 5 or 20% (w/w) lyophilized fish and estimated daily fish intakes by the rats at levels approxim

The role of seafood in foodborne diseases in the United States of America.

In the United States of America, seafood ranked third on the list of products which caused foodborne disease between 1983 and 1992. Outbreaks connected with fish vectors were caused by scombroid, ciguatoxin, bacteria and unknown agents; in shellfish, unknown agents, paralytic shellfish poisoning, Vibrio spp. and other bacteria, followed by hepatitis A virus, were responsible for the outbreaks. At least ten genera of bacterial pathogens have been implicated in seafood-borne diseases. Over the past twenty-five years, bacterial pathogens associated with faecal contamination have represented only

Behavioral effects of prenatal and adult exposure to toxic chemicals found in Lake Ontario fish: two methodological approaches.

Two research approaches are described that were used to determine behavioral changes following a diet of Lake Ontario fish. Approach 1 involved the correlational method, in which human subjects voluntarily ate contaminated Lake Ontario fish. Demographic information, data on the amount of Lake Ontario fish consumed, and control variables were obtained during an interview. Respondents' behavior, as well as the behavior of their children, then was measured. Because subjects were not assigned randomly to eat or not eat Lake Ontario fish, other variables that might have influenced both consumption

A review of the significance of animal food products as potential pathways of human exposures to dioxins.

The polychlorinated dibenzo-p-dioxins and dibenzofurans (dioxins) are groups of compounds with similar chemical and toxicological properties. Carcinogenicity was considered the most serious toxic end point when setting previous regulatory policies, but recent concerns have focused on the possible endocrine-disrupting activities of the dioxins. Toxicity is related to the 2,3,7,8 pattern of chlorine substitution, a pattern that also leads to chemical and metabolic stability. Dioxins are practically insoluble in water and concentrate in lipids of biological systems, leading to low background conc

Dietary mercury intake and human exposure in an Adriatic population.

A study was conducted to examine human exposure to mercury through dietary mercury intake in a population living in an industrially non-polluted area of the Adriatic Sea. The results have shown that approximately 20% of the subjects had a weekly dietary mercury intake above the provisional tolerable weekly intake (PTWI), primarily those consuming fish and other seafood > 6 times/week. The estimated seafood consumption corresponding to a mean intake of PTWI of 300 micrograms total mercury was 1559 g, and 1365 g for a PTWI of 200 micrograms methylmercury. However, the total mercury content in ha

210Po and 210Pb intake by the Portuguese population: the contribution of seafood in the dietary intake of 210Po and 210Pb.

Through analysis of 210Po and 210Pb in the diet, the average ingestion rate for the Portuguese population is estimated at 1.2 and 0.47 Bq d-1 per capita for 210Po and 210Pb, respectively. Detailed analysis of foods indicate that seafood alone contributes up to 70% of the 210Po ingestion rate, whereas cereals, vegetables, and meat altogether contribute 79% of the 210Pb ingestion rate. Consumption of seafood, both in terms of quantities (kg d-1 per person) and preferential consumption of certain marine species, is the cause of the relatively high intake of 210Po and high 210Po:210Pb ratio in the

Cytogenetic monitoring of fishermen with environmental mercury exposure.

Due to high mercury levels in many Mediterranean aquatic organisms, people who live in this area and consume large amounts of seafood are exposed to a toxicological hazard. A group of 51 fishermen exposed to mercury through eating contaminated seafood from the northern Tyrrhenian Sea underwent cytogenetic monitoring. This work is part of a research project consisting of the evaluation of micronuclei (MN), chromosomal aberrations (CA) and sister-chromatid exchanges (SCE) in peripheral blood lymphocytes. Here we present data on mercury levels in blood and on micronucleus frequencies in periphera
