Toxicology and seafood toxins: domoic acid.

Marine and terrestrial food sources are susceptible to contamination by various industrial chemicals and microbial pathogens. Both types of hazard are amenable to regulatory assessment using a single toxicology data base, along with some knowledge of contaminant levels and consumption figures for food. On the other hand, regulatory problems persist with acutely toxic naturally occurring phycotoxins, which may accumulate unpredictably to toxic levels in seafood. However, a scarce supply of pure toxin often precludes the availability of acceptable toxicology studies describing their biological e

Antimicrobials in shrimp aquaculture in the United States: regulatory status and safety concerns.

The consumption of seafood, especially shrimp, increases yearly in the U.S. The U.S. is the second largest importer of shrimp in the world, consuming more than 11% of the total world production. Aquaculture is becoming an increasingly important source of the world's shrimp, currently accounting for approximately 30% of the world's supply. Unfortunately, in this era of international trade deficits, U.S. production of aquacultured shrimp is insignificant (< 0.1%) compared with world production. As shrimp aquaculture expands in the U.S., so does the use of intensive farming techniques. Shrimp aqu

Domoic Acid and Amnesic Shellfish Poisoning - A Review.

A new type of seafood toxicity, called amnesic shellfish poisoning, was described from 107 human cases after individuals consumed mussels containing domoic acid harvested from Prince Edward Island, Canada, in 1987. Most of these cases experienced gastroenteritis, and many older persons or others with underlying chronic illnesses developed neurologic symptoms including memory loss. Standard treatment procedures for the neurologic condition were not effective and three patients died. Domoic acid is a known neurototoxin, and it is believed that in these cases enough toxin was absorbed through the

Eutrophication, marine biotoxins, human health.

Eutrophication phenomena in marine coastal waters can today be explained on the basis of natural or anthropogenic causes. Undesirable effects and also sanitary problems in both types of eutrophication are often produced, but they may differ greatly in frequency and significance. Some algal biotoxins can affect both marine animals and man, whilst others affect man alone. From data currently available it appears that the sanitary state of man can be affected through the digestive, respiratory and cutaneous apparatus. Four main dinoflagellate biointoxications are now recognized: paralytic shellfi

Metal, mutagenicity, and biochemical studies on bivalve molluscs from Spanish coasts.

Three species of marine bivalve molluscs (Chamelea gallina, Ruditapes decussatus, and Crassostrea gigas) have been studied in order to evaluate the levels of pollution on the South Atlantic Spanish littoral. Several transition metals (Cu, As, Cd, Sn, Hg, Pb) were determined as a general index of total contamination. Animals from putative contaminated areas exhibited higher metal contents than those from cleaner waters. C. gigas showed 5-20-fold higher total metal content than the other two species. The mutagenicity of ethanolic extracts was assayed by using both the His reversion and the Ara f

Carcinogen adducts as an indicator for the public health risks of consuming carcinogen-exposed fish and shellfish.

A large variety of environmental carcinogens are metabolically activated to electrophilic metabolites that can bind to nucleic acids and protein, forming covalent adducts. The formation of DNA-carcinogen adducts is thought to be a necessary step in the action of most carcinogens. Recently, a variety of new fluorescence, immunochemical, and radioactive-postlabeling procedures have been developed that allow the sensitive measurement of DNA-carcinogen adducts in organisms exposed to environmental carcinogens. In some cases, similar procedures have been developed for protein-carcinogen adducts. In

Transfer constants for blood-brain barrier permeation of the neuroexcitatory shellfish toxin, domoic acid.

The cause of the toxic mussel poisoning episode in 1987 was traced to a plankton-produced excitotoxin, domoic acid. Experiments were undertaken to quantitate the degree to which blood-borne domoic acid can permeate the microvasculature to enter the brain. Pentobarbital-anesthetized, adult rats received an i.v. injection of 3H-domoic acid which was permitted to circulate for 3-60 min. Transfer constants (Ki) describing blood-to-brain diffusion of tracer were calculated from analysis of the relationship between brain vs plasma radioactivity with time. Mean values (mL.g-1.s-1 X 10(6] for permeati

Cellular retention, toxicity and carcinogenic potential of seafood arsenic. I. Lack of cytotoxicity and transforming activity of arsenobetaine in the BALB/3T3 cell line.

Cytotoxicity, morphological neoplastic transformation, intracellular retention and metabolic behaviour have been investigated in BALB/3T3 Cl A 31-1-1 cells for arsenobetaine, the main form of arsenic in certain seafoods, in comparison to inorganic sodium arsenite. In order to avoid false results, particular attention was paid to the purity, checking for the presence of any trace amounts of inorganic arsenic as well as methylated contaminants in the chemically synthesized arsenobetaine. Cytotoxicity and morphological transformation assays gave obvious positive results for sodium arsenite at a d

Light and scanning electron microscopic studies on effects of marine algal toxins toward freshly prepared hepatocytes.

Mussels exposed to dinoflagellates may represent a human health risk due to accumulation of a variety of algal toxins. In several parts of the world, algal toxins leading to diarrhea (diarrhetic shellfish poisons, DSP) are found in mussels for extended periods of the year. Routine monitoring of these toxins involves ip injections in mice. Chemical analytical methods have been developed for only some of the toxins in question, namely, those giving diarrhea. Other toxins in the DSP complex are not easily detected by analytical methods. In this report we show that freshly prepared hepatocytes fro

The toxin tetramine from the "edible" whelk Neptunea antiqua.

The chemical nature of extracts of the marine gastropod Neptunea antiqua collected in the North Sea has been investigated. Amino acid analysis and spectroscopical studies (FAB MS, 1H NMR, 13C NMR and IR) on fractions purified by Biogel P-2 and Amberlite IR 120 column chromatography allowed the identification of 19 components. It was established that the water-soluble toxin responsible for poisonings, following ingestion of this snail, is tetramine (the tetramethylammonium ion present as an unknown salt). Contrary to what occurs in other Neptunea species (N. arthritica and N. intersculpta), tet

Clinical applications of fish oils.

Fish oil supplements are currently being nationally advertised, and many physicians are being queried about their clinical utility. Epidemiologic studies reveal a low incidence of cardiovascular disease in people, such as the Eskimos, who eat large amounts of seafood. Cardiovascular health may be improved because fish and fish oil supplements lower plasma lipid levels (especially triglycerides), inhibit platelet aggregation, and may decrease blood pressure and viscosity and increase high-density lipoprotein (HDL) levels. Preliminary observations also suggest a potential future role for fish oi

Determination of "heavy" organotin pollution of water and shellfish by a modified hydride atomic absorption procedure.

Tin speciation in aquatic environment is very complex. To the natural SnIV and methylated compounds, human activities add mainly butylated, octylated, phenylated or even methylated derivatives. The most environmentally significant, due to their high toxicity and direct introduction in water through biocidal use, are the trisubstituted ones. Several sophisticated speciation procedures have been proposed, they are not susceptible of common use. We propose a simple and fast procedure allowing routine global distinction of "heavy" tin species that are most susceptible of exerting harmful effects.

Methylmercury in fish: a review of residue levels, fish consumption and regulatory action in the United States.

The dangers associated with the consumption of large amounts of methylmercury in fish are well recognized, and there is some evidence to suggest that methylmercury may be the cause of subtle neurological impairments when ingested at even low to moderate levels, particularly the prenatal and early childhood periods. This concern has prompted a continuing assessment of the risk of methylmercury toxicity among fish consumers in the United States as well as other countries. The toxicokinetics of methylmercury in humans are reviewed and used to estimate body burdens associated with toxic effects. T

Impact of effects of acid precipitation on toxicity of metals.

Acid precipitation may increase human exposure to several potentially toxic metals by increasing metal concentrations in major pathways to man, particularly food and water, and in some instances by enhancing the conversion of metal species to more toxic forms. Human exposures to methylmercury are almost entirely by way of consumption of fish and seafood. In some countries, intakes by this route may approach the levels that can give rise to adverse health effects for population groups with a high consumption of these food items. A possible increase in methylmercury concentrations in fish from l

Formation of gizzard erosion-inducing substance in heated casein-histidine mixture.

Formation of a toxic substance inducing gizzard erosion (GE) was examined by using mainly a casein-histidine mixture as a simple model of fish meal. When a casein-histidine mixture was heated at 135 degrees C for three or five hours or at 160 degrees C for one hour, it induced severe GE. Changes in pH of histidine solution seemed to have little effect on the production of the toxic substance. Addition of a heated mixture of 30 mg of histidine and 150 g of casein (per kg of diet) failed to induce GE. A mixture of 750 mg of histidine and 150 g of casein was enough to produce a maximal toxicity.

Effect of dietary protein on the response of rainbow trout (Salmo gairdneri) to aflatoxin B1.

Diets containing either 49.5% or 32% casein or fish protein concentrate (FPC) were fed to young rainbow trout (Salmo gairdneri) for 12 months. Five levels [0, 2, 6, 18, and 54 parts per billion (ppb)] of aflatoxin B1 (AFB1) were given in each of four different diets. A 30-fish sample was taken at 6, 9, and 12 months to determine the influence of diet on the carcinogenicity of AFB1. Both levels of casein produced similar hepatoma incidences at each level of AFB1. The diet high in FPC produced more tumours than did the casein diets at 2, 6, and 18 ppb AFB1, whereas fish fed the diet low in FPC h

Current status of PCB toxicity to mink, and effect on their reproduction.

Experiments were conducted from 1968 to 1974 to investigate reproductive complications and mortality in mink fed Great Lakes coho salmon and to ascertain the effects of polychlorinated biphenyls (PCB's) on this fur bearer. The results of mink feeding trials indicated that coho salmon, as such, were not responsible for the loss of reproduction in the adult, or the kit mortality. Mink diets that contained other species of Great Lakes fish caused similar reproductive complications, but to a lesser degree. Rancidity, mercury poisoning and chlorinated hydrocarbon pesticide contamination of the fish

[Clinical comments on poisoning caused by marine animals in the Marquesas].

Intoxications due to sea animals in the Marquesas seem to be relatively frequent in this archipelago. Fish consumption under rarely realized favorable conditions does not induce intoxications. Although statistics are hazardous in Polynesia, the phenomena seem to progress from East to West, and to be cyclic, with a regression phase at the end of 1973. Two complementary syndromes have been isolated: -- one is primary, neuro-sensory, and is prevalent in the case of toxic crustaceans, molluscs or echinoderms consumption. Hemorragic signs are sometimes observed in the case of consumption of fish pr

Dimethylnitrosamine--formation in fish meal and toxic effects in pigs.

The production of fish meal from fresh and from formaldehyde- and sodium benzoate-preserved catches results in the formation of considerable dimethylnitrosamine (DMNA). This is in contrast to the prevailing opinion that only nitrite preservatives can be incriminated in producing this toxin. With capelin fish preserved in a mixture of 920 g of sodium benzoate and 920 ml of formaldehyde/hectoliter for 11 days, 7.5 mgs of DMNA/kg fish meal were produced. Pigs exposed to different daily levels of DMNA, of commercial source, proved to be rather resistant to intoxication compared with other domestic

A serum enzyme increasing effect of non-toxic herring meal fed to sheep.

In a series of experiments it was demonstrated that highly increased activities of aspartate aminotransferase (= GOT), total lactate dehydrogenase (LDH), and α-hydroxybutyrate dehydrogenase (HBD) occurred in serum of sheep on herring meal feeding. The alanine aminotransferase (A1AT = GPT) level remained unchanged. The enzyme increase was apparently not related to the liver toxic agent dimethylnitrosamine (DMNA) occasionally occurring in lethal doses in meals produced from raw materials preserved with excesses- of nitrite. Histological changes of the liver or other tissues were never detected i
