Conserved Enzymatic Peptides in Bitis arietans Venom Revealed by Comparative Proteomics: Implications for Cross-Reactive Antibody Targeting.

Snakebite envenoming remains a critical public health issue, and the molecular variability of venoms limits the cross-species efficacy of conventional antivenoms. Here, we conducted a comparative proteomic analysis of Bitis arietans venom to identify conserved peptide regions derived from enzymatic toxins and evaluate their potential relevance for complementary immunotherapeutic applications. Enzyme-enriched venom fractions were isolated through sequential affinity and ion-exchange chromatography and were subsequently characterized using fluorogenic FRET substrates and inhibitor assays. Omics-guided discovery and functional profiling of Da-29, a bioactive serine protease from Deinagkistrodon acutus venom, with antithrombotic activity.

BACKGROUND: Snake venom serine proteases (SVSPs) are critical toxic components that interfere with the hemostatic balance and serve as a key candidate library for anticoagulant drug development. Despite the diversity of SVSPs in Deinagkistrodon acutus venom, systematic studies of low-abundance SVSPs with thrombin-like activity profiles are limited. Hence, this research aimed to isolate and characterize a bioactive SVSP candidate from D. acutus venom using integrated multi-omics and to assess its antithrombotic effects and bleeding risk. METHODS: PacBio full-length transcriptome sequencing com

Bioinformatics-Driven Design and Evaluation of Recombinant Multi-Epitope Immunogens Derived From Snake Venom Toxins as Potential Antivenom Candidates.

Snakebite envenomation is a major public health concern, particularly in low- and middle-income regions where access to safe and effective antivenoms is limited. Traditional antivenoms, derived from immunization with crude venom, often trigger adverse reactions and lack specificity against key venom components. This study presents a bioinformatics-driven approach to design, construct, and evaluate a novel panel of recombinant multi-epitope toxin-derived immunogens targeting the most clinically significant snake venom toxin families. Five principal toxin families-three-finger toxins (3FT), phos

In vitro-in vivo discord: A preclinical study of AZD2716 and its racemate with comparison to varespladib for the development of snake venom sPLA2 inhibitors.

We evaluated a family of repurposed sPLA2 inhibitors as novel candidate snakebite envenoming therapeutics. Stereospecific (R)-7 AZD2716 and its racemic mixture were compared to varespladib in an in vitro sPLA2 assay against a sample of 26 venoms from medically important snake species from five continents. All compounds demonstrated potent nano-to picomolar IC50 values, comparable to the benchmark inhibitory profile of varespladib. Surprisingly, however, this in vitro efficacy did not translate to survival in an in vivo mouse model under GLP standard conditions at an independent third party lab

Establishing the Kenya National Antivenom Quality Control Laboratory: Preclinical Efficacy Results of Four Antivenoms Against Venoms from the "Big Five" Snake Species in Kenya.

Antivenom administration is currently the only therapy for snakebite envenoming. However, in sub-Saharan Africa, inadequate quality control systems have led to deficits in the availability, accessibility, efficacy and safety of regionally available antivenoms, which, in turn, hinder snakebite treatment and management in the region. To address this impediment to snakebite treatment in Kenya, this study aimed to assess the preclinical neutralising potencies of four different antivenoms previously or currently available in Kenya (SAIMR polyvalent, AFRIVEN, PANAF-PremiumTM and InoserpTM) against k

Scorpion sting: a narrative review and proposed guidelines for contemporary UK armed forces operations.

Scorpions are venomous arthropods with a wide geographical footprint and are endemic throughout all UK Armed Forces operational areas outside of Northern Europe. Scorpion stings are common on military exercises and operations. Here, we present a narrative review summarising epidemiological data pertaining to scorpions in geographical areas of contemporary operational relevance to the UK Armed Forces and suggest evidence-based treatment recommendations suitable for use in a military environment, providing an update to the last detailed review in 2008. The overwhelming majority of stings will re

Inter- and Intraspecific Venom Variation in the Reclusive Rear-Fanged Black-Striped Snakes (Coniophanes).

Our current understanding of snake venom is highly biased towards species known to be medically significant in human envenomations. This vastly under-represents the true evolutionary and ecological breadth of snake venom, with gaps spanning entire clades and unique lifestyles. As a result, many genera of rear-fanged snakes lack well-understood venom profiles despite these taxa composing around 65% of known extant snake species. Methodological challenges associated with venom extraction have long been a key reason responsible for the lack of venom research on this group. Modern advancements in

New Insights into the Bioenergetic and Immunomodulatory Properties of Phospholipases A2 from Bothrops diporus Venom.

Phospholipases A2 (PLA2s) are key mediators of the cytotoxic and inflammatory activities of snake venoms. While PLA2 isoforms from Bothrops diporus venom have been characterized and shown to possess antimetastatic and antiangiogenic properties, their impact on mitochondrial bioenergetics and immune modulation has not yet been investigated. In this study, we examined the bioenergetic and immunomodulatory effects of B. diporus PLA2s using integrated biochemical, metabolic, and multiplex cytokine analyses. In MDA-MB-231 breast cancer cells, pooled PLA2s induced a dose-dependent decrease in MTT-re

A commercial insecticide-induced neurotoxicity and snake venom nerve growth factor-inspired peptides-mediated neuroprotection in Caenorhabditis elegans: Mechanistic, safety, and pharmacokinetic (in vivo imaging) evaluation of peptides.

Agriculture workers are especially at risk from insecticide (INSECT) exposure, which is associated with neurological disorders, such as Parkinson's disease (PD). The present study evaluated the mechanism of neurotoxicity induction in a commercial insect containing chlorpyrifos (50%) and cypermethrin (5%) in Caenorhabditis elegans models, as well as the neuroprotective efficacy of two custom peptides (CPs), HNP (heptadeca-neuropeptide) and TNP (trideca-neuropeptide), derived from snake venom nerve growth factor. CPs significantly mitigated INSECT-induced neurotoxicity by preventing chemosensory

Zinc-dependent prothrombin activator from Rhabdophis tigrinus venom with extended substrate specificity.

Rhabdophis tigrinus, a common venomous snake in Japan, produces venom with potent procoagulant activity through activation of human prothrombin. However, the biochemical properties and classification of its prothrombin activator remain unclear. In this study, we partially purified the prothrombin activator from R. tigrinus venom (provisionally termed "Rhabdarin") and characterized its enzymatic and structural properties. The enzyme was partially purified using gel filtration and hydroxyapatite chromatography, yielding a disulfide-linked homodimer (∼110 kDa non-reducing; ∼55 kDa reducing). Rhab

Neutrophil Inflammasome Activation and Pyroptosis Induced by a Metalloproteinase via HIF-1α and Gasdermin D Pathways.

INTRODUCTION: Bothrops jararacussu venom contains snake venom metalloproteinases (SVMPs) that contribute to inflammation and tissue damage. BjussuMP-II, a PI class SVMP, lacks hemorrhagic activity but retains proteolytic and immunomodulatory properties. METHODS: Here, we uncover a previously unrecognized function of BjussuMP-II in triggering NLRP3 inflammasome activation in human neutrophils, leading to IL-1β release and pyroptosis. Inflammasome activation was investigated through gene expression analysis by quantitative PCR and protein expression by immunoblotting. Caspase-1 activation and g

Comprehensive molecular characterization and comparison of venom proteins and transcripts in three Gloydius species from South Korea.

In Korea, Gloydius brevicaudus, G. ussuriensis, and G. intermedius are the primary venomous species causing human envenomation. Although these species belong to the same genus, their envenomations induce distinct clinical symptoms, and the inconsistent efficacy of antivenoms underscores the need for species-specific therapeutic strategies. However, comprehensive molecular and toxicological profiling of Korean Gloydius species has remained unexplored. In this study, we quantitatively distinguished the venoms of three Gloydius species using two-dimensional gel electrophoresis and MALDI-TOF mass

Specific in vivo detection of V2R-positive metastatic ccRCC using a toxin-based PET radioligand.

BACKGROUND: The diagnosis of metastatic clear cell renal cell carcinoma (mccRCC) remains challenging due to the tumor's molecular heterogeneity, often resulting in low sensitivity and a high false-positive rate. In this study, we introduce and validate a new imaging modality for mccRCC based on the first radioligand targeting the type 2 vasopressin receptor (V2R) suitable for positron emission tomography (PET). V2R is ectopically expressed in mccRCC. This imaging approach utilizes [18F]F-MQ232, a radiolabeled peptide derived from snake venom which exhibits high in vivo selectivity for V2R. ME

Deconstructing cardiovascular and coagulation-related traits links dietary ecology to multi-functional snake venom specificity.

Animal venoms vary greatly in compositional complexity, where complex venoms are hypothesized to be maintained by greater dietary breadth. Beyond explaining venom composition, the dietary breadth hypothesis predicts that these more complex venoms should show greater functional breadth in terms of overall toxicity and by disrupting multiple prey physiological processes. We evaluate these predictions with six distinct physiological assays of cardiac and blood clotting functions and compared the effects of venoms from snake species with a range of taxonomic dietary diversity levels. We compared t

Lansbermin-I, a disintegrin from Porthidium lansbergii lansbergii venom as a potential anticancer prototype: In vitro and in silico effects on cervical cancer cells.

We report the cytotoxic effects of Lansbermin-I, a disintegrin from Porthidium lansbergii lansbergii venom, on HeLa cervical cancer cells but not on leukemic, non-tumorigenic peripheral blood mononuclear cells (PBMCs) or fibroblast lines. Lansbermin-I reduced HeLa cell viability in a concentration-dependent manner, impaired adhesion to fibronectin and collagen, and inhibited migration and clonogenicity. Docking analysis suggested interaction with the RGD domain of integrin α5β1. These results identify Lansbermin-I as a potential antitumor prototype against cervical cancer.

Bispecific single-domain antibody (VHH) fused with human IgG1 Fc with dual specificity effectively neutralize Naja Kaouthia venom.

Naja kaouthia, or the monocled cobra, is one of the most medically important snakes in Thailand, responsible for approximately 17% of snakebite cases. Conventional horse-derived antivenoms are lifesaving, yet they may trigger severe allergic reactions and exhibit batch to batch variability. Nanobodies (VHH) are promising alternatives as recombinant antivenoms having demonstrated the ability to neutralize snake venom both in vitro and in vivo. However, a major challenge in developing them is the diverse and complex composition of snake venoms, which requires therapies capable of targeting multi

Quantifying venom in African snakes: Insights into protein content, yield and body size associations.

Snake venoms are complex mixtures primarily composed of toxic proteins used during prey capture and defence. There is limited knowledge concerning the protein concentration of snake venom and the biases of different protein determination methods. Here, we assess the ability of the Qubit protein assay, bicinchoninic acid (BCA) assay, Bradford assay and NanoDrop spectrometry (A280 with a mass extinction coefficient of one) to accurately quantify protein concentrations of toxins isolated from venom, including three-finger toxins and phospholipase A2. The Bradford assays severely underestimated th
