Preliminary Insights into Geographic Variation in Venom Profiles and Functional Activities of Nigerian Snakes, Bitis arietans and Naja nigricollis.

Snakebite envenoming is a major yet neglected tropical disease in sub-Saharan Africa, where antivenom efficacy is critically limited by intraspecific venom variation shaped by local ecological pressures. Nigeria's sharply contrasting Sudan Savanna (North) and Lowland Rainforest (South) provide an ideal natural system to investigate this variation, yet a comparative analysis of its medically important snakes has been lacking. We conducted an integrated proteomic and functional characterization of venoms from the puff adder (Bitis arietans) and black-necked spitting cobra (Naja nigricollis) coll

Protective effect of scorpion-venom heat-resistant peptide (SVHRSP) against depression associated with Parkinson's disease via suppression of CREB-Akt pathway-mediated inflammation.

BACKGROUND AND PURPOSE: Parkinson's disease (PD), characterized by motor dysfunction and dopaminergic neuron loss in the substantia nigra, is frequently complicated by depression (depression-associated PD, DPD), affecting 40-50% of patients and accelerating disease progression. This study investigated neuroprotective effects in a chronic MPTP-induced mouse model of DPD. EXPERIMENTAL APPROACH: We established a chronic PD model induced by MPTP, with motor deficits assessed via rotarod and gait analysis. Depressive phenotypes were confirmed by tail suspension, sucrose preference and forced swim

Mechano-electrical feedback in transgenic rabbit models of long QT syndrome Type 2 and short QT syndrome Type 1.

AIMS: Electromechanical coupling and mechano-electrical feedback (MEF) are crucial for cardiac function, but their pro-arrhythmic roles in short and long QT syndromes (SQT1 and LQT2) are not fully understood. We aimed to evaluate MEF-induced electrical changes, their arrhythmic impact, and the involvement of stretch-activated channels (SACs) in transgenic rabbit models of SQT1 and LQT2. METHODS AND RESULTS: Patch-clamp and fluorescence imaging were used to analyse action potential duration (APD), Ca²⁺ transients, and contractility in ventricular cardiomyocytes (VCMs) from LQT2, SQT1 and wild-

A Hydrolase-Rich Venom Beyond Neurotoxins: Integrative Functional Proteomic and Immunoreactivity Analyses Reveal Novel Peptides in the Amazonian Scorpion Brotheas amazonicus.

The scorpion family Buthidae, renowned for its neurotoxin-rich venoms, dominates toxinology, while non-buthid venoms remain largely unexplored. Here, we present a comprehensive proteomic and biochemical characterization of the Amazonian chactid scorpion Brotheas amazonicus venom (BamazV), with emphasis on molecular complexity, proteolytic processing, and peptide diversity. Using an integrative venomics approach that combines molecular mass-based fractionation, reversed-phase chromatography, high-resolution mass spectrometry, N-terminal sequencing, and functional and immunological analyses, we

Venom Variation as a Window into the Ecology and Evolution of Snakes.

Snake venoms are complex biochemical systems that function primarily in prey subjugation and defense, yet their composition varies extensively across individuals, populations, species, and environments. This variation provides a powerful framework for investigating ecological and evolutionary processes. Here, we offer a forward-looking synthesis of snake venom diversity that proposes new research directions and highlights how venom variation can illuminate eco-evolutionary dynamics across biological scales. We review evidence for 10 key contexts in which venom variation arises, including withi

Modelling human proteostasis and organelle homeostasis disorders in yeast and its application in drug discovery.

Disruptions in cellular homeostasis and proteostasis are central to many human diseases, yet direct mechanistic investigation in human systems remains constrained by biological complexity and ethical limitations. Therefore, researchers have turned to the use of model systems that allow the more efficient dissection of fundamental cellular processes. The unicellular yeast Saccharomyces cerevisiae has emerged as a powerful eukaryotic model for studying disorders driven by defects in homeostasis and proteostasis. The relevant processes are highly conserved in yeast, enabling precise genetic manip

Discovery of non-opioid peptides that selectively relieve mechanical pain in rodents through inhibition of TRPV4 channels.

Mechanical hyperalgesia is a primary symptom of clinical pain; there remains a therapeutic challenge for severe mechanical and chronic forms of neuropathic pain. The 34-amino-acid neuropeptide GsMTx4, isolated from Tarantula spatulata, was identified as a selective inhibitor of mechanosensitive channels and shown to reduce mechanical hyperalgesia and neuropathic pain in rats. We previously reported two types (type I and type II) of short peptides derived from GsMTx4 that mimicked the inhibitory action of GsMTx4 on a mechanosensitive BK (SAKca) channel. In this study, we investigated whether th

Neuwlectin, a novel C-type lectin from Bothrops neuwiedi venom, inhibits platelet aggregation induced by cancer cells and angiogenesis.

C-type lectins are the major non-enzymatic protein constituents in Viperidae venoms. Neuwlectin, a novel member of the C-type lectin family, was purified to homogeneity from Bothrops neuwiedi venom and characterized as a 32.2 kDa homodimer of 16.2 kDa subunits by MALDI-TOF. In this study, we investigated its effect on platelet aggregation triggered by cancer cells and on angiogenesis. Each subunit contains 135 amino acids with eight cysteine residues and shares high sequence similarity to homologous snake venom Ca2-dependent C-type lectins. Neuwlectin induced agglutination of rabbit and human

The sting that clots: The Factor VII and Factor X activating procoagulant effects of Androctonus scorpion venoms are potentiated by Factor Va as a cofactor.

While scorpion venoms are well-characterised as being potently neurotoxic, their effects upon blood coagulation are understudied. Here, we report novel procoagulant toxicological functions for Androctonus amoreuxi, A. australis, A. bicolor and A. crassicauda venoms. Factor activation tests with A. amoreuxi venom revealed cofactor-dependent activation of Factor VII (FVII) and Factor X (FX) to be the primary zymogen targets, with FX the more potently activated. Activation of both factors was demonstrated to be dependent upon the proteinaceous cofactor Factor Va (FVa) and the biochemical cofactor

Viprutoxin, a snake venom serine proteinase inhibits platelet aggregation and selectively targets A549 cancer cells invitro.

Snake venom serine proteinases (SVSPs) are a well-studied group of toxins commonly found in the venoms of Viperidae, Elapidae and Crotalidae snakes. Serine proteinase toxins present in snake venoms are responsible for a wide variety of pathophysiological consequences in the victim. The problems range from coagulopathy to cytotoxicity. This paper discusses Viprutoxin (VPT), a 27 kDa serine proteinase isolated from Daboia russelli russelli venom (found in eastern India). Viprutoxin fragments obtained after trypsin digestion and Orbitrap High-Resolution Liquid Chromatography (O-HRLCMS) and Liquid

Discovery of Two Novel Scorpion Venom Peptides Activating TRPML2 to Impair ZIKV Internalization.

The endo-lysosomal channel TRPML2 regulates key processes like membrane trafficking and autophagy, which are hijacked by many RNA viruses during endocytic entry. However, the development of TRPML2-targeted therapeutics has been hindered by a notable lack of high-affinity and selective peptide-based activators. Scorpion venom peptides, honed by evolution for exceptional specificity toward diverse membrane ion channels, represent a promising, underexplored natural library for discovering novel pharmacological probes and drug leads. Here, we screened and identified seven candidate peptides intera

Recluse spiders (Araneae: Sicariidae): contesting myths on distribution, bite behavior, and medical risk in Florida.

Recluse spiders are arachnids of potential medical significant due to their necrotic venom. However, in areas populated by the brown recluse, Loxosceles reclusa, bites and necrosis are rare. In Florida, USA, public media reports and medical case studies have propagated three misconceptions about recluse spiders: breeding populations are widespread in Florida, USA; recluse spiders readily bite when encountered; and verified recluse spider bites inevitably produce necrotic wounds. Here, in onsite investigations of 220 Florida properties with alleged infestations of recluse spiders reported to th

Inflammation induced by snake venoms optimizes envenomation.

Snake envenomation activates the immune system through permeability-increasing factors that generate an acute vascular inflammatory response by opening large inflammation-activated pores (IAPs) in the microvasculature. These events facilitate the exudation of plasma from blood into body tissues. However we show that IAPs also allow macromolecules, including venom toxins, to flood directly into the bloodstream, even against an outflow of plasma solutes. Such inflammation-facilitated macromolecular absorption (IFMA) acts together with lymphatic absorption and has a physiological role in removing

Web of Potentials: Neuroactive Components of Spider Venom and Their Emerging Pharmacologic Applications in Neurologic Diseases.

Spider venom has emerged as a promising source of neuroactive compounds with potential applications in the treatment of complex neurological disorders. With over 53,000 described species and more yet to be studied, spiders possess one of the most chemically diverse venoms in the animal kingdom. This diversity has evolved through ecological adaptation, enabling spiders to paralyze and manipulate the nervous systems of a wide range of prey. These same mechanisms, which target ion channels, neurotransmitter receptors, and signaling enzymes, coincide with pathways implicated in human neurologic di

Purified zymogens reveal mechanisms of snake venom metalloproteinase auto-activation.

Snake venoms contain diverse mixtures of toxins that evolved to incapacitate prey, but in humans, they cause extensive pathology following snakebite envenomation. In viper venom, some of the most potent toxins are the haemorrhagic and coagulopathic snake venom metalloproteinases (SVMPs). Because venoms contain an SVMP cocktail and due to their cytotoxicity, SVMP characterisations have been hampered by the lack of purified enzymes. By incorporating their prodomain, which blocks the active SVMP site, we overcame their cytotoxicity and enabled recombinant production of zymogens from all three str

Structural insights into insect-selective sodium channel toxins drive AI-enhanced biopesticide design.

Many voltage-gated sodium channel-targeting animal peptide toxins are renowned for their potency and selectivity against insects. Understanding why these toxins selectively target insect sodium channels over their mammalian counterparts is crucial for developing safer and more effective pest control agents. Here, we present the cryoelectron microscopy (cryo-EM) structures of the insect sodium channel NavPaS bound to two naturally occurring insect-selective toxins, Av3 from the sea anemone and LqhαIT from the scorpion. Both toxins bind to the voltage-sensing domain 4 (VSD4) of NavPaS and disrup
