Venom composition of the Iberian adder (Vipera seoanei) from northern Portugal reveals limited sex and size variation.

Snake venom is a complex and dynamic ecological trait, with high potential for intraspecific variation. In this study, the venom composition of the Iberian adder (Vipera seoanei) from the Peneda Mountains of northern Portugal was investigated to explore whether venom varies with body size and sex. Venom proteomes from six individuals representing juvenile, subadult, and adult males and females were characterised using LC-MS/MS, while individual variation was assessed from SDS-PAGE profiles of 18 snakes. LC-MS/MS identified 144 proteins, dominated by phospholipases A2 (PLA2), snake venom serine

Purification and Characterization of BfPLA2: Fraction containing Acidic Asp49 Phospholipase A2 Isoforms Isolated from the Snake Bothrops fonsecai.

Among the various proteins found in the venom of snakes belonging to the genus Bothrops, which is responsible for the highest number of snakebite accidents in Brazil, Phospholipase A2 (PLA2) is one of the most important components. The present study demonstrated the isolation of a fraction containing two Asp49 PLA2 isoforms from the endemic southeastern Brazilian snake Bothrops fonsecai, named BfPLA2. The isolation was achieved through two chromatographic techniques: RP-HPLC using a Teknokroma Europa Protein 300 C18 column, followed by anion-exchange chromatography using a HiTrap Q FF column.

Paradoxical viper envenomation: minimal local manifestations with fatal intracranial hemorrhage- an autopsy case report.

Viper envenomation typically presents with prominent local manifestations, including pain, edema, ecchymosis, cellulitis, and tissue necrosis. Severe systemic toxicity in the absence of significant local signs is uncommon. We report an atypical case of a viper envenomation in a 50-year-old male who presented with minimal local manifestations but rapidly developed life-threatening systemic complications. The 20-min whole-blood clotting test was non-clotting on admission. Despite prompt administration of polyvalent anti-snake venom (ASV) and symptomatic management, his condition progressively de

Molecular Recognition and Immunopharmacological Implications of Serratiopeptidase Targeting Snake Venom Bradykinin-Potentiating Peptides.

PURPOSE: Bradykinin-potentiating peptides (BPPs), found in snake venom, are known to modulate cardiovascular functions, but their susceptibility to enzymatic degradation remains poorly understood. METHODS: Models were generated using AlphaFold-Colab for five small BPPs (< 9 amino acids) that did not have experimentally solved structures, and those BPPs which had reliable predictions of structure (pLDDT > 80) were used for docking studies. Protein-peptide docking experiments were done using HADDOCK for studying the interaction between wild-type (WT) and mutant serratiopeptidase with BPPs. Afte

Integrated computational investigation of Glycosmis pentaphylla-derived compounds against major venom proteins of Daboia russelii.

Snakebite envenomation remains a significant neglected tropical disease, particularly in South Asia, where Daboia russelii (Russell's viper) causes substantial morbidity and mortality. Current antivenom therapy is limited by poor efficacy against local tissue damage, high production costs, restricted toxin specificity, and adverse immune reactions, highlighting the need for alternative or adjunct therapeutic strategies. Using an integrated computational and experimental approach, phytocompounds identified from Glycosmis pentaphylla leaf extracts were evaluated as potential inhibitors of major

Preparedness of primary health centres for snakebite management and challenges faced by healthcare providers: a mixed-methods study from a selected district, southern India.

OBJECTIVES: Primary health centres (PHCs) are the first point-of-care in rural settings for snakebite management and are expected to provide early management, including anti-snake venom (ASV) administration. However, evidence on their functional readiness and operational challenges remains limited. This study assessed PHC preparedness for snakebite management and explored challenges faced by healthcare providers in a district of Tamil Nadu, India. METHODS: A convergent mixed-methods study was conducted across 65 PHCs. Quantitative data on preparedness were collected using a structured checkli

Photobiomodulation attenuates BjussuMP-II-induced muscle inflammation: effects on leukocyte recruitment and MIR-223 expression.

Bothrops jararacussu is one of the snake species most frequently involved in snakebite envenomation in Brazil. Snake venom metalloproteinases (SVMPs), which account for approximately 26.2% of its venom composition, play a central role in envenomation by inducing hemorrhage, myonecrosis, and a robust local inflammatory response. Although antivenom effectively neutralizes systemic toxicity, its efficacy against local tissue damage remains limited. In this context, the present study investigated the effects of 945 nm LED photobiomodulation on the inflammatory response induced by BjussuMP-II, a PI

A human-cell recombinant prethrombin-2 platform producing thrombin with enhanced fibrinogen processing for in vitro applications.

Thrombin is a central mediator of haemostasis, immune responses, and inflammation. It arises from prothrombin upon prothrombinase cleavage at R320-I321, which triggers the allosteric maturation typical of chymotrypsin-like serine peptidases. Further processing yields the physiologically dominant α-thrombin (αT), whose competent active site centres on catalytic S525. Despite widespread biomedical and biotechnological use, plasma-derived αT is heterogeneous and risks pathogen or prion transmission, motivating recombinant alternatives. These have largely relied on suboptimal bacterial or non-huma

Rational design of a consensus bothropic metalloproteinase as an immunogen for broadly protective antivenom development.

Antivenoms remain the only specific therapy for snakebite envenoming. In Bothrops venoms, Snake Venom Metalloproteinases (SVMPs) are abundant and highly diverse toxins responsible for the major local and hemotoxic effects induced by envenoming. However, currently available bothropic antivenoms fail to fully neutralize these effects, frequently resulting in permanent and disabling sequelae in affected patients. Here, we describe a rational approach to improve the SVMP neutralization potential of bothropic antivenoms using a consensus toxin. Forty SVMP sequences belonging to PI, P-II, and P-III

Ziyuglycoside I Attenuates Deinagkistrodon acutus Venom-Induced Injury in Renal Tubular Epithelial Cells by Modulating PAR1-NLRP3-IL-1β Signaling.

INTRODUCTION: Sanguisorba officinalis L. (Di Yu) has traditionally been used for snakebite treatment. Ziyuglycoside I, a major bioactive constituent of Di Yu, exhibits anti-inflammatory activity, but its role in snake venom-associated renal injury remains unclear. This study aimed to investigate its potential protective effects against Deinagkistrodon acutus venom (DAV)-induced renal tubular epithelial cell injury in vitro. METHODS: HK-2 cells were exposed to DAV to establish an in vitro renal tubular epithelial injury model. Cellular metabolic activity and intracellular ATP levels were asses

Rotational thromboelastometry guided transfusion in venom-induced consumptive coagulopathy following a haemotoxic snakebite.

A woman in her 30s presented with severe haemotoxic snake envenoming complicated by venom-induced consumptive coagulopathy (VICC) and early multiorgan involvement. Despite receiving adequate polyvalent anti-snake venom and initial transfusions at a referral facility, the patient's coagulopathy persisted. On arrival at our emergency department, conventional coagulation tests (prothrombin time and international normalised ratio) were only mildly abnormal, in contrast to her significant clinical severity. Rotational thromboelastometry (ROTEM) revealed severe hypocoagulability with markedly impair

Intrinsic endonucleolytic activity of snake venom phosphodiesterase.

Snake venoms contain enzymes whose biochemical activities cannot be inferred reliably from sequence or proteomic detection alone. Snake venom phosphodiesterase (svPDE), an extracellular ENPP-family glycoprotein, is generally regarded as a 3'-to-5' exonuclease, although endonucleolytic activity was reported in early preparations. Whether this activity is intrinsic to a molecularly defined svPDE and how DNA structure influences cleavage remained unresolved. Here, nuclease activity in Protobothrops flavoviridis venom was tracked by sequential chromatography, and LC-MS/MS identified svPDE in activ

Core outcome set for intervention research on snakebite envenomation in South Asia.

BACKGROUND: The 2019 WHO strategy to reduce snakebite burden emphasises the need for fostering research on snakebite treatments. A core outcome set (COS) is a consensus minimal list of outcomes that should be measured in research on a particular condition. We aimed to develop a COS for snakebite research in South Asia, the region with the highest burden. METHODS: We used data from a systematic review of outcomes to develop a long list of outcomes which were rated in two rounds of online Delphi survey with healthcare providers, patients and the public, and potential COS users to develop a COS

Lansbermin-I: A Snake Venom Disintegrin With Selective Cytotoxicity and Anti-Adhesive Effects Against Glioblastoma Cells.

Glioblastoma is the most common and aggressive malignant brain tumor, characterized by poor response to current therapies and inevitable recurrence. Novel therapeutic strategies are urgently needed. Lansbermin-I, a disintegrin isolated from the venom of Porthidium lansbergii lansbergii, was purified, sequenced, structurally modeled, and evaluated for its antitumor potential against the human Glioblastoma cell line T98G. Cytotoxicity assays revealed a dose-dependent effect, with Lansbermin-I inducing up to 38.1% cell death at 100 μg/mL, surpassing the activity of temozolomide (34.9%) while exhi

Neddylation Targets and Stabilizes NLRP3 to Augment Inflammasome-Mediated Colitis and Mood Disorder.

The activation of NLRP3 inflammasome contributes to the development of numerous chronic inflammatory diseases, including ulcerative colitis and stress-induced anxiety. Recent studies have revealed that NLRP3 K48-linked ubiquitination by several E3 ligases, including Trim31, leads to degradation. Neddylation is a process highly similar to ubiquitination by covalently conjugating NEDD8 to lysines in specific substrate proteins. Neddylation of substrate proteins alters their subcellular localization, stability, and activity. The role of neddylation in the NLRP3 inflammasome remains elusive. Here,

Comparative analyses of Bothrops fonsecai snake venom: individual variability and sex-based differences.

Snakebite envenomation is a neglected tropical disease affecting thousands of people globally, especially in tropical and subtropical regions. An estimated 81,000 to 138,000 deaths occur annually, with many survivors experiencing permanent disabilities. In Brazil, around 26,000 cases are reported each year, with the genus Bothrops responsible for 83.8 % of them. Bothrops venoms are rich in metalloproteases, serine proteases, phospholipases A2, and L-amino acid oxidases, which can cause hemorrhage, tissue necrosis, coagulation disorders, and hypotension. This study focuses on the biochemical an

Selectivity screening of cytotoxicity evoked by viper venoms and their toxins after nanofractionation.

Cytotoxicity is a major pathological effect that can occur during snakebite envenoming. To better understand the underlying biochemical and molecular mechanisms behind snake venom-induced cytotoxicity, it is essential to use appropriate in vitro tools for bioassaying cytotoxicity evoked by snake venoms. Identifying the toxins causing cytotoxicity is also important in this regard, particularly in the context of developing more effective snakebite treatments. Cytotoxicity induced by venom toxins can result in local pathologies in snakebite victims, which can result in long-term morbidity, and is

An accurate and reproducible method to measure snake venom anticoagulant activity.

INTRODUCTION: Anticoagulant activity is an important clinical effect of human envenoming, seen in Australian black snake (Pseudechis) and cobra (Naja) envenoming. Anticoagulant toxins are not well characterised and anticoagulant assays not standardised. We aimed to develop and validate an accurate and standardised anticoagulant assay for snake venoms. METHODS: We used a turbidimetric assay previously developed to measure procoagulant activity and modified for anticoagulant activity. Cytotoxic and anti-metastatic effects of Pllans-II from Porthidium lansbergii lansbergii venom against prostate cancer cells.

Prostate cancer remains a major health challenge due to therapeutic resistance and the adverse effects of conventional treatments. Snake venom phospholipases A2 have emerged as promising bioactive molecules with selective anti-cancer properties. In this study, we evaluated the cytotoxic and anti-metastatic effects of Pllans-II, an Asp-49 PLA2 purified from Porthidium lansbergii lansbergii venom, on prostate cancer cell lines. Pllans-II exerted a dose-dependent cytotoxic effect on LNCaP, PC-3, and DU-145 cells, with the strongest activity observed in LNCaP cells (IC50 = 100 μg/mL). In contrast,
