Morphological and molecular identification of Kalicephalus sp. in the Yucatan Neotropical rattlesnake Crotalus tzabcan in Yucatan, Mexico.

Female nematodes were collected from the intestines of a road-killed Yucatan Neotropical rattlesnake, Crotalus tzabcan, in Yucatan, Mexico. Morphometric analysis of the nematode specimens allowed their identification to the genus Kalicephalus. However, the absence of male specimens prevented species-level identification. Notably, second- and third-stage larvae were observed in the reproductive tract of some Kalicephalus females. Additionally, the internal transcribed spacer (ITS) region, the cytochrome c oxidase subunit 1 (COI) gene, and the 28S rRNA gene (28S) were amplified and sequenced. Pa

A chromosome-level genome assembly and annotation for the beauty snake Elaphe taeniura.

The genus Elaphe Fitzinger, a large species clade within Colubridae, comprises 18 non-venomous snake species. Among them, Elaphe taeniura serves as a representative species due to its wide distribution and strong adaptability. However, genomic studies on this group remain limited. Here, we present a chromosome-level, high-quality reference genome for this species. The genome is assembled by integrating high-accuracy PacBio HiFi long-read sequencing and Hi-C chromatin conformation capture technologies and comprehensively annotated with the aid of transcriptomic data. The genome of E. taeniura s

The Golden Lancehead Genome Reveals Distinct Selective Processes Acting on Venom Genes of an Island Endemic Snake.

High-quality genomic resources are important for accurate assessments of adaptive evolution in rapidly evolving island endemic species. The golden lancehead (Bothrops insularis) is a critically endangered venomous species endemic to the Queimada Grande island located on the southeast coast of Brazil, with no reference genome available. Here, we present a high-quality near chromosome-level and well-annotated genome for the golden lancehead. A macrosyntenic analysis using genomes from other viper species revealed that microchromosomes present higher rearrangements consisting of fission and fusio

Reference Plasma Proteome Map of Crotalus durissus terrificus.

Snakebite envenoming remains a major public health concern worldwide, while the maintenance of captive snakes is essential for conservation, venom production, and biomedical research. However, little is known about the plasma proteome of snakes, a gap that limits physiological assessments and the development of species-specific reference protein profile. This study aimed to characterize the plasma proteome of healthy adult Crotalus durissus terrificus snakes to establish a molecular baseline that supports biological evaluation and future diagnostic research. Blood samples were collected from 1

Pharmacological inhibition of myosin reduces the cytotoxic effect of a Lys49 PLA2-like myotoxin on human and rat cardiac muscle.

Viperid snake venoms contain a group of cytotoxic proteins called Lys49 phospholipase A2 (PLA2)-like myotoxins. These proteins lack enzymatic activity, but can still disrupt the sarcolemma and induce cell death. On skeletal and cardiac muscle, PLA2-like myotoxins induce substantial alterations to the sequence of events in the excitation-contraction coupling, including increased Ca²⁺ influx and consequent hypercontraction. This study aimed to quantify the increase in force development induced by a Lys49 PLA2-like myotoxin and to assess whether this effect contributes to the overall cytotoxicity

The Venom Proteome and Immunorecognition Profile of Clinically Important Echis carinatus sochureki from Northwestern India Underscores the Need for Regionally Specific Antivenoms.

The saw-scaled viper Echis carinatus, one of the "Big Four" causes of snakebites in India, is found from Sri Lanka to eastern Iraq. To investigate clinical reports regarding the limited efficacy of Indian polyvalent antivenom (IPAV) against envenomation in Echis carinatus sochureki (ECS) in northwestern India, we obtained 22 snakes from three locations in Rajasthan and identified 148-174 toxin isoforms belonging to 21-25 toxin families in their venom using a bottom-up proteomics approach. All samples showed a high abundance of snake venom metalloproteinases (SVMPs), particularly SVMP class III

Integrating epidemiological evidence and immunodiagnostic development for early diagnosis of Bothrops spp. envenomation in Brazil.

Snakebite envenoming remains a major public health issue in Brazil, with Bothrops genus responsible for most cases. To support public health planning, we analyzed epidemiological data from Paraná State and reported, for the first time, a species-level identification of snakes responsible for accidents in this region. The results revealed that Bothrops jararaca accounted for over 85% of cases, followed by Bothrops jararacussu, reinforcing their epidemiological and clinical relevance. Based on this evidence, two monoclonal antibodies were produced by hybridoma technology for application in a dia

The hills are alive with the sound of venom: Clinical implications of intraspecific variations in Factor Va mediated activations of Factor VII and Factor X in Swiss populations of Vipera aspis (Asp Viper).

This study investigated the individual variation in the procoagulant toxicity of Vipera aspis venoms collected from multiple Swiss localities, in addition to the neutralisation capacity of three currently available antivenoms. Functional coagulation assays revealed quantifiable geographic differences in venom potency and clotting kinetics. Mechanistic analyses demonstrated that V. aspis venoms exert their procoagulant effects primarily through Factor Va-dependent action of the coagulation cascade, leading to accelerated thrombin generation and clot formation. These findings highlight the compl

First characterization of the venom of the endemic coral snake Micrurus camilae (Serpentes: Elapidae) from Colombia: Proteome, toxic activities, immunorecognition, and neutralization by antivenoms.

Thirty-one species of Micrurus (coral snakes) are distributed in Colombia. However, functional and proteomic analyses of their venoms have only been performed on six of them. Micrurus camilae is endemic to Colombia, and no information exists regarding its venom. The proteome of M. camilae venom, its biochemical and toxic activities, immunorecognition, and neutralization by commercial equine antivenoms and an experimental one prepared in rabbits are here reported. In addition, the phylogenetic position of M. camilae within the genus was explored. The venom was characterized by RP-HPLC, SDS-PAGE

Cross-continental venom narratives: proteo-transcriptomic insights into Hydrophis cyanocinctus of India and China.

Hydrophis cyanocinctus, annulated sea snake, is a widely distributed marine elapid whose venom composition and gene-expression architecture remain poorly characterized across its geographic range. Here, we present the first high-resolution proteo-transcriptomic profile of venom and venom gland of this species from India. This research underscores RNA-seq comparison of Indian and Chinese H. cyanocinctus venom gland gene expression to elucidate population-level variation in venom gland machinery, gene diversity, and other regulatory biology. Coiling chirality? Wild eastern diamondback rattlesnakes (Crotalus adamanteus) do not exhibit clear lateralized coiling behaviour.

Behavioural lateralization has been recorded in various groups, from lower vertebrates to humans. Handedness, or the preferred use of one hand over the other, as observed in humans, is thought to be related to brain structure and function. Snakes have been recorded to potentially display such sided behaviours. The asymmetrical internal anatomy of snakes may provide a basis for coiling handedness in the form of clockwise versus anti-clockwise coiling to protect vital organs or provide an advantageous strike to capture prey. The few studies on behavioural lateralization in snakes have documented

Survey of Synanthropic Spiders in Ireland Reveals Expansion and Dominance of the Invasive Noble False Widow Steatoda nobilis in Urban Habitats (Araneae: Theridiidae).

Rapid urbanisation has led several spider species to adapt to synanthropic microhabitats and establish large populations outside of their native ranges. In Ireland, the establishment and widespread distribution of the Noble false widow spider Steatoda nobilis (Araneae: Theridiidae) (Thorell, 1875) has raised questions about its impact on native spider populations across the country. Through an extensive field survey of six urban centres over an 11-month period, we sought to establish population demographics for S. nobilis and other synanthropic spiders in Ireland for the first time. We surveye

A country-wide survey of knowledge and practice regarding prevention, snake identification and pre-hospital management of children with snakebites amongst Public Health Midwives in Sri Lanka.

BACKGROUND: Snake envenoming remains a major public health problem in Sri Lanka, disproportionately affecting rural children. Timely and appropriate pre-hospital management is crucial to reducing morbidity and mortality. Public Health Midwives (PHMs), as frontline community health workers, are strategically positioned to contribute to snakebite prevention and early management. However, their knowledge and confidence regarding paediatric snakebite care have not been systematically evaluated. METHODOLOGY/PRINCIPAL FINDINGS: A cross-sectional, country-wide survey was conducted among PHMs across

Structural and pharmacological analysis of a PLA2-like toxin in complex with the sPLA2 inhibitor AZD2716: Comparisons to varespladib.

Phospholipase A2 (PLA2) and PLA2-like toxins are key contributors to the local myonecrotic effects induced by Bothrops snake venoms and represent important targets for inhibition. Synthetic molecules such as varespladib (LY315920) and AZD2716 were originally developed as inhibitors of human group IIA PLA2, with varespladib emerging as a promising candidate for repurposing against venom toxins. Here, we investigated a snake venom Lys49-PLA2-like toxin and another potent sPLA2 inhibitor, AZD2716, using ex vivo neuromuscular blockade assays, microscale thermophoresis, crystallographic, and bioinf

Ruthenium Compounds Differentially Inhibit Group IA and IIA Snake Venom Phospholipase A2 Anticoagulant Activity.

Neurotoxicity caused by snake venom phospholipase A2 (PLA2) activity derived from coral snakes (e.g., Micrurus tener, Micrurus fulvius, group IA PLA2) and some rattlesnakes (e.g., Crotalus scutulatus, group IIA PLA2) is medically significant. Of interest, the catalytic site of PLA2 also binds to activated clotting factor X, causing anticoagulation. Given that ruthenium (Ru)-containing compounds have been demonstrated to inactivate hemotoxic venoms in a solvent-dependent manner (e.g., 0.9% NaCl, phosphate-buffered saline), we wished to determine if RuCl3 would cause solvent-dependent inhibition

Inappropriate antivenom use in Crotalus snakebite treatment reports: A national data-based analysis (2014-2024).

Snakebite envenoming by Crotalus snakes is a relevant public health problem in Brazil. From 2015 to 2024, 26,021 cases were reported in DataSUS, with 78,329 antivenom vials administered. However, only 27,257 vials had the specific antivenom type recorded, indicating 51,072 undocumented vials. In addition, the database records the administration of antivenoms that are not typically indicated for crotalic envenoming. Because these data originate from a secondary surveillance system, such discrepancies may reflect documentation gaps, data entry errors, or potential inconsistencies in antivenom se

De Novo Assembly and Characterization of Venom Gland Transcriptome for Rhabdophis lateralis.

Rhabdophis lateralis is a snake species within the family Natricidae, which is widely distributed across mainland China, Russia, and Korea. Although this species was once thought to be non-venomous, there are quite a few cases demonstrating its bite could be fatal. In this study, we performed de novo assembly and analysis of the transcriptome data from the Duvernoy's gland of R. lateralis, aiming to characterize its venom transcriptome and reveal the molecular basis of its toxicity. Among 6196 annotated transcripts, 77 were identified as potential toxin transcripts belonging to 26 toxin famili
