Why does local edema persist? Immunological mismatch between phospholipase A2 potency and antivenom affinity in Protobothrops mucrosquamatus envenomation.

BACKGROUND: Protobothrops mucrosquamatus is the primary species responsible for snakebite morbidity in Taiwan, typically resulting in severe localized tissue edema. Although freeze-dried hemorrhagic antivenom (FHAV) is the standard treatment, persistent swelling remains a frequent clinical challenge. This study aimed to identify primary edematogenic toxins and evaluate the neutralizing efficacy of FHAV and small-molecule inhibitors (SMIs) to address this therapeutic gap. METHODOLOGY/PRINCIPAL FINDINGS: Multiple linear regression of a retrospective cohort of 50 patients was used to identify pr

Delayed onset of myotoxicity following red-bellied black snake envenoming: a case report.

BACKGROUND: Myotoxicity is an important and recognized effect of red-bellied black snake envenoming that remains poorly understood but can result in severe illness. Prior observational data have shown that myotoxicity develops less frequently and later than other envenoming effects, including systemic symptoms and anticoagulant coagulopathy, which are likely to coexist in an envenomed victim. Current recommendations for managing snake bite victims in Australia include serial clinical and laboratory assessments for envenoming for at least 12 hours post-bite. CASE PRESENTATION: We report the ca

Severe venom-induced consumption coagulopathy, snakebite-associated thrombotic microangiopathy, and local necrosis following Western bush viper (Atheris chlorechis) envenoming in France.

BACKGROUND: Atheris spp. are small African vipers whose bites are rarely reported but can result in significant envenoming. There is no specific antivenom available. We describe a case of systemic A. chlorechis envenoming presenting with venom-induced consumption coagulopathy (VICC), snakebite-associated thrombotic microangiopathy (TMA), and local necrosis. CASE PRESENTATION: A 33-year-old man was bitten on the index finger by a captive A. chlorechis. Within 6 hours, early laboratory abnormalities progressed to VICC with markedly prolonged PT/aPTT, undetectable fibrinogen, elevated fibrin mon

Barietin, a snake venom vascular endothelial growth factor from puff adder Bitis arietans venom, increases the heart rate with minor effect on blood pressure in rats.

Snake venom vascular endothelial growth factors (svVEGFs) are homodimeric proteins comprising two equal subunits bound by two intermolecular disulfide bonds. The main biological effects of svVEGFs are local vasodilatation, increasing of vascular permeability, and hypotension. Barietin, a svVEGF purified from the venom of puff adder Bitis arietans, differs from other representatives of this toxin family by its shortened C-terminal amino acid sequence. No data about barietin effects on hemodynamics are available. Studying those in rats we found that barietin at a dose of 1 mg/kg significantly in

Ziyuglycoside I attenuates coagulopathy induced by Deinagkistrodon acutus venom in human whole blood: a thromboelastography-based study.

INTRODUCTION: Ziyuglycoside I, a hemostatic saponin isolated from Sanguisorba officinalis (Di-Yu), has reported anti-hemorrhagic properties. Deinagkistrodon acutus venom (DAV) is associated with venom-induced consumptive coagulopathy, but its effects on whole-blood coagulation and potential adjunctive interventions remain insufficiently characterized. This study evaluated whether Ziyuglycoside I attenuates DAV-induced coagulation abnormalities using thromboelastography (TEG) and molecular docking. METHODS: Citrated whole blood from healthy donors was exposed to 10, 20, or 30 μg DAV to charact

Hypopituitarism following Russell's viper envenomation in Bangladesh: findings from a prospective longitudinal cohort study.

Envenomation by Russell's viper is a major cause of hemotoxic snakebite in South Asia, associated with venom-induced consumptive coagulopathy (VICC), leading to end-organ damage, including the kidneys, adrenal glands, and pituitary gland. Hypopituitarism is a rare but increasingly recognized delayed complication, reported primarily from South India, Sri Lanka, and Myanmar, but not previously reported from Bangladesh. We conducted a prospective longitudinal cohort study at a tertiary referral hospital in Rajshahi, Bangladesh (2019-2025). A total of 203 male patients with Russell's viper envenom

A new species of Hepatozoon in the common lancehead snake (Bothrops atrox) from the Eastern Amazonia region.

Although venomous snakes from the family Viperidae, such as Bothrops atrox, are recognized for their medical importance due to snakebite accidents, few studies on parasitological aspects have been carried out with them, especially in the Amazonia region. Using morphological and molecular tools, we described a novel haemogregarine species infecting the common lancehead snake B. atrox from Eastern Amazonia, Brazil. Hepatozoon atrocis sp. nov. has mature gamonts that are morphologically distinct from those reported in the literature, which are often compact, with dispersed or encapsulated cytopla

MUSEUM SPECIMENS OF WESTERN DIAMOND-BACKED RATTLESNAKES (VIPERIDAE: CROTALUS ATROX) REVEAL TRENDS IN BODY SIZE, DIET, AND ENDOPARASITES OVER THE LAST 75 YEARS.

Climate projections for the Southwestern United States predict increases in temperature, drought intensity and frequency, and wildfire intensity and frequency. Such climatic change is expected to impact communities and species of the region, including Crotalus atrox (western diamond-backed rattlesnake) and its helminth communities. We hypothesized that C. atrox in New Mexico has experienced measurable changes in body size, diet composition, and helminth communities over the past century. We examined specimens of C. atrox from 3 time periods: 1947-1960 (1950s, n = 34), 1980-1990 (1980s, n = 53)

Exploring the potential therapeutic effects of ultradiluted Crotalus horridus on antibody-dependent enhancement in dengue virus pathogenesis.

DENV remains a global health concern, with ADE playing a major role in severe disease progression. ADE leads to excessive cytokine release, nitric oxide overproduction, and elevated levels of non-structural-1 antigen, contributing to higher vascular dysfunction, thrombocytopenia, and hemorrhagic manifestations. The present study investigated the therapeutic effect of ultradiluted Crotalus horridus on ADE-mediated DENV infection using in vitro system. MTT, colony formation, cytopathic effect observation, ELISA, and qRT-PCR, were performed to assess cytotoxicity, viral pathogenesis, and immunomo

Mechanism of activation of an ancestral TEC kinase by PIP3.

BACKGROUND: The introduction of second-generation antipsychotics (SGAs) over the past 2 decades generated considerable optimism that better antipsychotic treatments for schizophrenia and bipolar disorder were possible. SGAs offer several tolerability benefits over first-generation antipsychotics (FGAs), particularly with respect to extrapyramidal symptoms. However, SGAs can induce serious metabolic dysregulations, especially in drug-naive, first-episode, and child and adolescent populations, with olanzapine and clozapine having the highest propensity to cause these abnormalities. In this conte

Effects of Selective African Spitting Cobra Venoms and Their Three-Finger Toxins on the Modulation of Platelet Function and Blood Clotting.

Venoms of African spitting cobras, including Naja mossambica, N. nigricincta, and N. pallida, are widely recognised for their cytotoxicity. However, their effects on platelets and blood coagulation remain poorly characterised. To address this gap, we characterised their enzymatic profiles and evaluated their effects on haemostasis and cell viability using functional blood-based and cellular assays, respectively. All three venoms markedly impaired haemostatic regulation by affecting intrinsic coagulation, significantly inhibiting platelet function, and inducing haemolysis. They also exerted pot

Pathologic Characterization of Raillietiella orientalis Infection and Comorbidities in Free-Ranging Pygmy Rattlesnakes (Sistrurus miliarius) from Central Florida, USA.

Raillietiella orientalis (Ro) is a parasitic pentastome with a rapidly expanding geographic and herpetofaunal host range in the southeastern USA since its introduction, probably with the invasive Burmese python (Python bivittatus). However, little is known about the health impacts of Ro infection in native USA host species. Pygmy rattlesnakes (Sistrurus miliarius), small pit vipers native to the southeastern USA, were among the first documented naïve hosts of Ro in Florida, USA. Between December 2018 and February 2019, 17 free-ranging S. miliarius from central Florida were evaluated for Ro pen

Insights from the timber rattlesnake (Crotalus horridus) genome for MHC gene architecture and evolution in threatened rattlesnakes.

Conservation of threatened species can benefit from an evaluation of genes in the major histocompatibility complex (MHC), whose loci encode proteins that bind pathogens and are often under strong selection to maintain diversity in immune response to diseases. Despite this gene family's importance to disease resistance, little is known about these genes in reptiles including snakes. To address this issue, we assembled and annotated a highly contiguous genome assembly for the timber rattlesnake (Crotalus horridus), a pit viper which is threatened or endangered in parts of its range, and analyzed

Prospect of using ethnobotanicals to manage snakebites in a cost-effective manner: validating Senegalia mellifera extract's inhibitory potential on Naja nigricincta nigricincta (zebra cobra) venom.

BACKGROUND: Despite Naja nigricincta nigricincta being responsible for most snake envenomation in remote Namibian regions, an effective intervention against its venom remains undiscovered. This study aimed to scientifically validate Namibian folklore claims about Senegalia mellifera extract's efficacy against snake envenomation. METHODS: In vitro assays were conducted to assess the inhibitory potential of S. mellifera stem bark extract on snake venom phospholipase A2 (svPLA2) activity from N. n. nigricincta venom. Venom composition, toxicity and cross-neutralization by PoliVal-ICP antivenom, of Mesoamerican jumping pitvipers genus Metlapilcoatlus (Viperidae: Crotalinae).

BACKGROUND: The genus Metlapilcoatlus was recently erected to include six species of stout venomous snakes, known as the jumping pitvipers, which inhabit mountainous areas of Mesoamerica. This group maintains affinity with Atropoides picadoi, another jumping pitviper with restricted distribution in Costa Rica and Panama. Although the venom of A. picadoi and a couple of Metlapilcoatlus species has previously been characterized, little is known about the interspecific and intraspecific variation of the other species that comprise the genus. In this work, we characterize the venoms of five out of

Genetic variability in snake venom and its implications for antivenom development in sub-Saharan Africa.

Snake venom, a complex mixture of proteins, has attracted human attention for centuries due to its associated mortality, morbidity and other therapeutic properties. In sub-Saharan Africa (SSA), where snakebites pose a significant health risk, understanding the genetic variability of snake venoms is crucial for developing effective antivenoms. The wide geographic distribution of venomous snake species in SSA countries demonstrates the need to develop specific and broad antivenoms. However, the development of broad antivenoms has been hindered by different factors, such as antivenom cross-reacti

Snake venom galactoside-binding lectin from Bothrops jararacussu: Special role in leukocytes activation and function.

Snake venom galactoside-binding lectins (SVgalLs) comprise a group of toxins with the ability to bind specifically, reversibly, and non-covalently to galactose-containing carbohydrates in a Ca2+-dependent manner. Several SVgalLs have been identified and isolated from Bothrops snake venoms, presenting highly similar structures and biological functions. BjcuL is a galactoside binding C-type lectin isolated from the venom of South America Bothrops jararacussu and consists of the most investigated lectin. Previous studies have deeply investigated the participation of BjcuL in physiopathological ev

Enhancing the Bothropic Antivenom through a Reverse Antivenomics Approach.

Antivenoms are the only effective treatment for snakebite envenomation and have saved countless lives over more than a century. Despite their value, antivenoms present risks of adverse reactions. Current formulations contain a fraction of nonspecific antibodies and serum proteins. While new promising candidates emerge as the next generation of antivenoms, it remains clear that animal-derived antivenoms will still play a critical role for years to come. In this study, we improved the bothropic antivenom (BAv), by capturing toxin-specific antibodies through affinity chromatography using immobili
