Mouse lethality assays (MLA) remain the gold standard for evaluating antivenom potency despite ethical concerns regarding animal welfare. This study assessed whether reducing observation periods from 48 to 24 h in MLA for determining median lethal dose (LD50) and median effective dose (ED50) of Bothrops jararaca and Crotalus durissus terrificus venoms and antivenoms maintains scientific validity while improving animal welfare. Through retrospective analysis of 518 quality control assays conducted between 2009-2023 at INCQS, including 334 potency assays and 27 median lethal dose assays for B. j

Plant compounds that inhibit snake venom activities are relevant and can provide active molecules to counteract snake venom effects. Numerous studies on snake viperid venoms found that metalloproteinases play a significant role in the pathophysiology of hemorrhage that occurs on envenomation. Preclinical studies using vitro and in vivo protocols investigated natural compounds and viperid snake venoms, evaluating the enzymatic, procoagulant, hemorrhagic, edematogenic, myotoxic, and lethal activities. Many studies focused on Bothrops venoms and ascribed that angiorrhexis and hemorrhage resulted

African cobra (Naja spp.) venom contains toxins dominated by proteins and peptides with inter- and intra-specific variations. There are several FDA-approved drugs from snake venom toxins from other regions, including South America and Asia. Profiling the proteomes of medically important African cobra venoms from different locations will aid in developing more effective anticancer agents. The venoms of spitting cobras (Naja pallida and Naja nigricincta woodi) and non-spitting cobras of the Uraeus subgenus (Naja anchietae, Naja annulifera, and Naja nivea) were fractionated by reverse phase-high

Venoms have traditionally been studied from a proteomic and/or transcriptomic perspective, often overlooking the true genetic complexity underlying venom production. The recent surge in genome-based venom research (sometimes called "venomics") has proven to be instrumental in deepening our understanding of venom evolution at the molecular level, particularly through the identification and mapping of toxin-coding loci across the broader chromosomal architecture. Although venomous snakes are a model system in venom research, the number of high-quality reference genomes in the group remains limit

Snake venoms contain a variety of different components, most of which are protein and peptide toxins. Although certain metal ions are known to be integral parts of several important venom enzymatic toxins, the elemental content of snake venoms has received scant attention. X-ray fluorescence (XRF) spectroscopy is proving to be a rapid, high-throughput method for analyzing venoms and is revealing several interesting compositional trends never before seen in venoms. Using XRF, we have analyzed the dominant elements found in snake venoms extracted from two species of rattlesnakes, Crotalus polyst

Strophanthus sarmentosus is recognised for various ethnomedicinal applications, including treatment after snakebites. However, only limited scientific evidence exists on its antivenomous capabilities. This study investigates the efficacy of methanol and ethylacetate extracts from S. sarmentosus leaves and roots against Echis ocellatus venom. A non-toxic range for the extracts was determined in rats, and assays were performed to test their anti-hemorrhagic and anti-hemolytic activity as well as their influence on venom-induced blood clotting. In all of these experiments, the extracts demonstrat

Acetylcholinesterase (AChE) is present in the venom of Elapid snakes, except the Mambas. Although the physiological role of AChE in snake venom is uncertain, it may contribute to acute neuromuscular paralysis following envenomation. With the aim of repurposing clinically used drugs for snakebite envenoming, this study investigated the inhibition of AChE from snake species of Southern Africa. Hemachatus haemachatus and Naja nivea venom presented with high AChE activity, which was inhibited by tacrine, galantamine and donepezil with IC50 values ranging from 0.49 to 3.73 μM.

The genus Crotalus comprises venomous pit vipers widely distributed discontinuously from southern Canada to northern Argentina. A dichotomy in Crotalus venom is observed, divided in type I and type II, and these different types could be correlated with geographic distribution and ontogeny of these snakes. C. d. ruruima inhabits Roraima state, Northen of Brazil, close to the Equatorial line and to North American Rattlesnakes. Brazilian crotalic antivenom is produced by the mixture of C. d. terrificus and C. d. collilineatus venoms, not including other subspecies. The knowledge of snake venom va

Pinpointing the emergence of toxicological evolutionary novelties can be challenging. In American pit vipers, anticoagulant venoms are the paradigm, with a notable exception being the genus Bothrops, which are typically procoagulant. A recent study found that the basal Bothrops (B. pictus) is anticoagulant, raising two competing hypotheses: ancestral Bothrops were anticoagulant with procoagulant venom evolving later, or ancestral Bothrops were procoagulant with anticoagulant venom in B. pictus being a derived trait. To help resolve this, we tested venoms of the sister genus Bothrocophias for p

Snake genomes attract significant attention from multiple disciplines, including medicine, drug bioprospection, and evolutionary biology. However, genomic research within the Viperidae family has mostly focused on the subfamily Crotalinae, while the true vipers (Viperinae) have largely been overlooked. European vipers (Vipera) have been the subject of extensive research due to their phylogeographic and ecological diversification, as well as their venoms. Nevertheless, phylogeography and systematics in this genus have primarily relied on biased information from mitochondrial genes, which fail t

Crotalus atrox venoms show high levels of metalloproteinase activity and generally display low levels of geographic variation, but proteolytic activity is known to increase moving northeast across the distribution in the United States, inversely to venom toxicity. We tested the venoms of five individuals from the northeastern distributional limit in Arkansas and found the highest activities and abundances of snake-venom metalloproteinases compared to other assayed populations. We discuss the ecological implications of the heightened lytic activity in venoms from Arkansas rattlesnakes.

The composition of Australian snake venoms is the least well-known of any continent. We characterised the venom proteome of the southern death adder Acanthophis antarcticus-one of the world's most morphologically and ecologically divergent elapids. Using a combined bottom-up proteomic and venom gland transcriptomic approach employing reverse-phase chromatographic and gel electrophoretic fractionation strategies in the bottom-up proteomic workflow, we characterised 92.8% of the venom, comprising twelve different toxin identification hits belonging to seven toxin families. The most abundant prot

Species recovery efforts may require establishment of ex situ populations to supply augmentation or reintroduction projects. The Eastern Massasauga (Sistrurus catenatus) is a small rattlesnake with an ex situ breeding program maintained by a consortium of conservation breeding centres. Small population size in human care has restricted the supply of animals for in situ conservation, thus, the development of improved ex situ breeding protocols was recommended. The objectives of our retrospective study were to (1) describe and compare reproductive traits between two groups of Massasauga litters

We report a clinical case of severe and prolonged hypofibrinogenemia caused by envenomation from Protobothrops cornutus, an pit viper species found mainly in Vietnam and parts of southern China. A 60-year-old woman was bitten while hiking in Cuc Phuong national forest of Ninh Binh province, Vietnam. Although WHO guidelines discourage antivenom administration for minor coagulation abnormalities, in this case, the decision was made due to a fibrinogen level <1.0 g/l and the patient's risk factors, including age (60 years) and untreated arterial hypertension, increasing the risk of spontaneous ce

Snakebite envenomation remains a life-threatening public health challenge in developing regions of the world. In India, Russell's viper (Daboia russelii) is responsible for the highest number of snakebite-inflicted mortality and morbidity. Diverse toxins have been reported in D. russelii venoms. However, the precise role of these toxins and whether they function independently, additively or synergistically remains unclear. To bridge this knowledge gap, we resolved D. russelii venom into distinct fractions containing major toxins and subjected them to various in vitro and in vivo experiments. O

INTRODUCTION: Crotalidae polyvalent immune Fab (ovine) is indicated for the treatment of Agkistrodon contortrix (copperhead) envenomation. Medical toxicologists have used lower Crotalidae polyvalent immune Fab (ovine) antivenom doses than those recommended in the prescribing information for the treatment of copperhead envenomation.
METHODS: We conducted a single-center retrospective chart review of patients with copperhead envenomation seen by our medical toxicology consult service between January 2001 and June 2023. Patients with dry bites, envenomation by other snakes, no antivenom treatmen

Many vertebrates have evolved resistance to snake venom as a result of coevolutionary chemical arms races. In Australian skinks (family Scincidae), who often encounter venomous elapid snakes, the frequency, diversity, and molecular basis of venom resistance have been unexplored. This study investigated the evolution of neurotoxin resistance in Australian skinks, focusing on mutations in the muscle nicotinic acetylcholine receptor (nAChR) α1 subunit's orthosteric site that prevent pathophysiological binding by α-neurotoxins. We sampled a broad taxonomic range of Australian skinks and sequenced

Australian elapid snakes possess potent procoagulant venoms, capable of inducing severe venom-induced consumption coagulopathy (VICC) in snakebite victims through rapid activation of the coagulation cascade by converting the FVII and prothrombin zymogens into their active forms. These venoms fall into two mechanistic categories: FXa-only venoms, which hijack host factor Va, and FXa:FVa venoms, containing a complete venom-derived prothrombinase complex. While previous studies have largely focused on human plasma, the ecological and evolutionary drivers behind prey-selective venom efficacy remai

INTRODUCTION: Copperheads and cottonmouths are responsible for most snake envenomations in Louisiana. While the United States Food and Drug Administration has approved both Crotalidae polyvalent immune Fab (FabAV) and Crotalidae immune F(ab')2 (Fab2AV) for Agkistrodon envenomations, data is limited comparing their efficacies for this indication.
METHODS: This is a retrospective study comparing FabAV to Fab2AV in the treatment of suspected Agkistrodon envenomations in Louisiana between April 2017 and October 2024. Cases identifying rattlesnakes were excluded. The primary outcome was the need f
