Snakebite envenoming (SBE) is a critical yet neglected public health issue in ASEAN countries, including Malaysia. Antivenom immunotherapy is the primary treatment for SBE, functioning by neutralising toxic venom components through antibodies raised from immunised animals. Current antivenoms come with potential reduced efficacy due to regional venom variability. The aim of this systematic review is to consolidate and compare the venom proteome composition from Malaysian venomous snakes (elapids and viperids) sampled from Malaysia, with their counterparts sampled across other Southeast Asia (SE

Snakebite envenoming is a neglected tropical disease that causes death and disability. Lysine 49 phospholipase A2-like (Lys49 PLA2-like) proteins are abundant toxins in many viperid venoms and play a significant role in their toxicity. They induce skeletal muscle necrosis by disrupting the plasma membrane through a mechanism independent of phospholipid hydrolysis. X-ray structures of Bothrops asper Lys49 PLA2-like proteins were analysed for sequence, folding and quaternary structure, and compared with dimer geometries of Lys49 PLA2s from other viper species in the PDB. Sequence and folding wer

The noble false widow spider (Steatoda nobilis), a rapidly spreading member of the Theridiidae family, has gained attention for its increasing presence near human habitats and its medical significance due to envenomation reports. Recent studies have revealed that its venom contains α-latrotoxins, toxins also found in Latrodectus (black widows), responsible for latrodectism symptoms. Despite this growing interest, little is known about the lipidome and metabolome of S. nobilis, which could offer insights into its ecological role, dietary metabolism, and chemical communication. In this study, we

Envenomation by viperid snakes causes acute muscle tissue injury (myonecrosis). An important group of myotoxic components comprises catalytically-inactive Lys49 phospholipase A2 homologs, which disrupt the integrity of the plasma membrane of skeletal muscle fibers through a mechanism that does not involve phospholipid hydrolysis. However, it remains unknown whether other mechanisms are involved in the cytotoxic action of these myotoxins. In this work, isolated calcium release channels (ryanodine receptor, RyR1) incorporated into an artificial lipid bilayer were used to study the action of the

Snakebites are classified as a neglected tropical disease due to its global impact. The genus Bothrops is the main responsible for envenoming in Brazil, with symptoms that are both local and systemic. Snake venoms are subject to intraspecific variation related to several internal and external factors, including ontogeny. Bothrops erythromelas are small snakes with generalist feeding habits and the ontogenetic variation in its venom has not yet been fully elucidated. Thus, the present work aimed to biologically and biochemically characterize the venom of B. erythromelas snakes from neonates to

The spectacled cobra (Naja naja) and monocled cobra (Naja kaouthia), widespread venomous snakes in South and Southeast Asia, occur in diverse habitats and cause neurotoxic envenoming. Despite reported venom variability of these two cobras across their range, no comparative study has been conducted from the interconnected but distinct habitats of Bangladesh. Using venomics and antivenomics, we analysed 26 individual venom samples of N. kaouthia and 17 of N. naja from Bangladesh across age groups and locations, respectively. Significant interspecific and intraspecific venom variability was obser

Snakebite envenoming causes more than 130,000 deaths and more than 400,000 disabilities per year and has been classified as a priority Neglected Tropical Disease by the World Health Organization (WHO). While antivenom therapy remains the mainstay of snakebite treatment, small molecule therapeutics (SMTs) have been proposed as potential adjuncts to antivenom, particularly as oral treatment in the prehospital setting. Several SMTs have demonstrated efficacy in preclinical models of snakebite envenoming, with varespladib, a secreted phospholipase A2 (sPLA2) inhibitor, being granted orphan drug st

Snake venoms have surged as model systems in evolutionary biology thanks to the dynamic diversification and accelerated evolution of many toxin families. Among these, phospholipase A2 (PLA2) constitute a prime example as they are ubiquitous across the venomous snake radiation and have evolved a wide variety of pathophysiological activities. This is especially true in vipers (family Viperidae), one of the most successful and medically significant venomous snake lineages worldwide. In this study, we gathered publicly available sequences of viper venom PLA2s to recreate the molecular phylogeny an

Mamba (Dendroaspis species) snakebites are critical medical emergencies across sub-Saharan Africa. Envenomings can result in the rapid onset of complex neurotoxic symptoms, often leading to high rates of mortality without timely intervention with antivenom. The ancestral state of mambas is the green coloured, forest dwelling type, with the tan/grey coloured, savannah dwelling D. polylepis (Black Mamba) representing a derived state both ecologically and morphologically. However, it has not been tested whether these changes are paralleled by changes in venom biochemistry or if there are differen

The puff adder (Bitis arietans) is a clinically relevant viper species found throughout Africa, and it is responsible for a greater incidence of health-related envenomations than all other snake species on the continent combined. Unresolved skeletal muscle damage is a common consequence of B. arietans envenomation that can result in long-term morbidity and even death. Antivenom treatment can mitigate the systemic effects of the venom but offers little protection against local tissue damage. Identifying the mechanisms through which B. arietans venom induces tissue damage and impedes skeletal mu

Snakes of the genus Bothrops are responsible for most snakebites in Latin America, frequently associated with hemostatic disorders. Among venom components, C-type lectin-like proteins (CTLPs) stand out for their ability to interact with platelet receptors. In this study, jararaca GPIb-BP, a CTLP from Bothrops jararaca venom was purified and further characterized. In vivo assays demonstrated that the protein induced significant and dose-dependent thrombocytopenia in mice, with the most pronounced effect at 167.35 μg/kg (s.c.), which caused partial recovery of platelet counts at 24 h and complet

Snake venoms have evolved over millions of years to subdue prey and deter predators rapidly. The potency and amount of venom differ both within and across species, and are primarily influenced by their ecology and environment. Understanding venom yields in clinically relevant species is essential for refining treatment approaches for snakebite. Despite this, venom yields have seldom been documented, especially in snakes of the Indian subcontinent. To address this knowledge gap, we analysed venom yields from 338 specimens of the clinically most important "big four" Indian snakes-Naja naja, Dabo

BACKGROUND: Snakebite envenomings (SBE) are an important and neglected health issue due to their frequency and potential for severe clinical outcomes. Envenomations can cause local and systemic complications, depending on the snake species, amount of venom injected, comorbidities, timing and use of antivenom, and access to health care. Systemic effects may be fatal or lead to permanent sequelae, including strokes resulting from venom-induced vascular and tissue damage. The objective of this study is to investigate the main clinical and epidemiological characteristics of individuals who develop

This study characterizes all medically diagnosed bites and stings in active component service members (ACSMs) from snakes, venomous fish, other venomous marine animals, arthropods, and insects identified through an evaluation of medical data from the Defense Medical Surveillance System (DMSS). Incident trends were determined from 2008 through 2023, and incidence rates (IRs) and incidence rate ratios (IRRs) were calculated. In total, there were 42,552 venomous bite and sting medical encounters among 39,201 ACSMs, resulting in an IR of 19.3 cases per 10,000 person-years (p-yrs) during the survei

BACKGROUND: Urban environments are increasingly recognized as important habitats for wildlife, yet information on how reptiles use urban environments remains scarce. The Aesculapian snake (Zamenis longissimus), a non-venomous colubrid widespread in Central Europe, is considered a habitat generalist with a frequent association with human settlements. However, data on its occurrence and ecology in metropolitan areas are limited. To address this gap, we implemented a citizen science initiative in Bratislava, Slovakia, aiming to document the distribution and habitat use of the Aesculapian snake an

BACKGROUND: Antivenom remains the standard and most effective treatment for snakebite envenomation, yet batch-to-batch variability in neutralization potency, even under standardized immunization protocols, continues to present a significant challenge in antivenom production worldwide. This issue is particularly evident in the manufacturing of Deinagkistrodon acutus antivenom in Taiwan, where some hyperimmunized equines fail to achieve the required potency despite identical conditions. Previous studies identified a lower antibody titer against acutolysin A-a key metalloproteinase toxin in D. ac

Although rare, exotic snakebites are a growing medical concern worldwide, partly due to the rising popularity of exotic pet ownership in high-income countries. Envenomation from family Elapidae snakes may cause severe neurological and local symptoms. Timely antivenom administration is crucial, but often complicated by logistical challenges. We report a case of a West African green mamba (Dendroaspis viridis) envenomation in a 59-year-old male in Switzerland who developed progressive weakness and respiratory failure, associated with swelling of a limb. Bothrops pubescens is a clinically significant viperid snake species in Uruguay, accounting for over 60 cases of human envenomation annually, alongside Bothrops alternatus. Formerly classified within the Bothrops neuwiedi complex, recent taxonomic revisions now restrict its distribution to Rio Grande do Sul (Brazil) and Uruguay. This species is not very robust, with a body length ranging from 22 to 92 cm and displays a distinctive dorsolateral pattern consisting of dark brown trapezoidal blotches on each side. The venom of B. pubescens exhibits toxicological properties typical of the Bothrops

Bothrops (B.) ammodytoides is the southernmost viper species in the world and is endemic to Argentina, where it is distributed from the northwest to southern Patagonia. In La Pampa, northern Patagonia where it inhabits, snakebite incidents are recorded. Its venom has toxic potency comparable to that of other Bothrops species found in Argentina. However, its venom and the effectiveness of available antivenoms have not been studied as extensively as those of other Bothrops species. For this reason, some biochemical, immunological and toxicological characteristics of venoms from snakes of this pr

BACKGROUND: Snakebite envenoming is a significant yet often overlooked public health issue. Despite Australia's reputation for venomous snakes, hospitalisation rates for snakebites have remained relatively low due to factors such as low population density, widely available antivenoms, and high-quality healthcare. However, population growth and climate change have increased snake sightings, raising concerns about shifts in snakebite incidence and seasonality. The Australian Venomous Injuries Project (AVIP) analysed hospitalisation data from 2002 to 2020 to assess trends and implications of snak
