Paralysis efficiency (PD50) scales linearly with lethality (LD50) in spider venoms.

Historically, venom potencies have been assessed using measures of lethality, such as the median lethal dose (LD50). However, venoms may be selected primarily for their ability to rapidly incapacitate rather than cause mortality, meaning LD50 may not capture the efficacy of venoms in an ecological and evolutionary context. To capture this context, recent studies have adapted measures that assess venoms' ability to rapidly incapacitate, such as the median paralysis dose (PD50). However, while PD50 values are expected to provide a more proximate assessment of ecological variation in venom potenc

Tetracycline reprograms inflammatory and regenerative signaling pathways in human keratinocytes exposed to Loxosceles spider venoms and sphingomyelinases.

INTRODUCTION: Loxosceles spider envenomation, or loxoscelism, constitutes the most severe form of araneism and frequently progresses to dermonecrosis with significant tissue damage. The key venom component, sphingomyelinase D (SMase D), drives both local and systemic effects through its structurally distinct Class I and II isoforms, each differing in toxicity. The current therapies provide limited benefit and, once necrosis is established, interventions are primarily supportive, underscoring the need for more effective pharmacological options. While tetracyclines have emerged as promising modu

Latrophilin-1-Mediated Gαq Signaling, Store-Operated Ca2+ Entry, and CaV2.1 Activation Control Spontaneous Exocytosis at the Mouse Neuromuscular Junction.

Latrophilin 1 (LPHN1/ADGRL1), an adhesion G-protein-coupled receptor (GPCR), is the principal receptor for α-latrotoxin (αLTX), a toxin that triggers massive neurotransmitter release. However, its endogenous signaling mechanism remains elusive. Here, we dissect the LPHN1 signaling pathway at the vertebrate neuromuscular junction, using the pore-deficient αLTX mutant LTXN4C as a selective agonist. Combining electrophysiological recordings from LPHN1 knockout mice with pharmacological inhibitors, calcium imaging, and biochemical assays, we delineate the cascade from receptor activation to sponta

Inhibition of ASIC1a Attenuates Neuronal Pyroptosis and Neuroinflammation Following Traumatic Brain Injury.

BACKGROUND: Traumatic brain injury (TBI) is a neurological disorder that severely affects health and function. Acid-sensing ion channels 1a (ASIC1a), a proton-gated cation channel permeable to Na+ and Ca2+, has been implicated in chronic neurodegeneration after TBI. However, its specific role in post-TBI neuroinflammation remains poorly defined. In this study, we investigated the mechanistic involvement of ASIC1a in neuronal pyroptosis following TBI. METHODS: Adult male C57BL/6 mice were subjected to controlled cortical impact (CCI) modeling using weight-drop models. Expression of ASIC1a was

Molecular arms race classifier for decrypting venom peptide and ion channel interactions.

Animal venoms comprise an astonishing number of peptides, proteins and small molecules. The diversity of venom compounds arises from evolutionary adaptations resulting in both offensive and defensive traits in predators and prey alike. This concept, referred to as "arms race", underpins the specificity and selectivity of venom compounds for certain molecular targets, like ion channels. Ion channels are essential regulators of cellular processes, and their dysfunction or dysregulation facilitates a wide range of diseases. Venom peptides and their derivatives are powerful modulators of ion chann

Spider Venom Peptides as Potential Allosteric Inhibitors of Undecaprenyl Diphosphatase (UppP) from Acinetobacter baumannii: In Silico Identification and Structural Analysis.

The antimicrobial resistance of Acinetobacter baumannii necessitates the development of novel therapeutic strategies targeting essential enzymes such as Undecaprenyl Pyrophosphate Phosphatase (UppP). This study explored spider venom peptides in silico as potential allosteric inhibitors of A. baumannii UppP. A systematic literature review was conducted to select eight α-helical peptides with reported anti-A. baumannii activity, followed by their computational physicochemical characterization. Three-dimensional models of A. baumannii UppP and the candidate peptides were generated, and a putative

Comparative proteomic characterization of web silk toxins between two species of orb-weaving spiders Trichonephila clavipes and Nephilingis cruentata.

The silk strands spun by Nephilinae spiders are known to encompass a diverse array of peptidic and proteinaceous toxins, alongside low-molecular-weight toxins. This study employs a mass spectrometry-based proteomics methodology to comparatively assess the abundance of web silk toxins in two species of orb-weaving spiders, Trichonephila clavipes and Nephilingis cruentata. Results derived from the proteomic analysis revealed the presence of 54 proteinaceous toxin-like compounds on the web of N. cruentata and 63 counterparts in that of T. clavipes. Additionally, a set of 24 toxin-like proteins is

Quantitative analysis of growth and diversification in venom data using database metrics.

Animal venomics is a growing field of research with evolutionary and biotechnological significance. Yet, fundamental questions regarding the origin, diversification, and bioactivity of venoms remain unresolved. Here, we analysed venom tissue-related data curated in Tox-Prot, currently the most comprehensive database for animal venoms, across three snapshots spanning two decades (2005, 2015, 2025). We assessed the taxonomic landscape related to Tox-Prot entries, sequence length distribution, protein family abundances, and habitat-specific venom patterns. Our results consistently show that snake

A Single L17E Mutation Switches the Membrane Disruption Mechanism of the Spider Venom Peptide M-lycotoxin.

Membrane-active peptides (MAPs) disrupt lipid bilayers through mechanisms that depend sensitively on amino acid sequence, yet the molecular determinants governing membrane pore formation remain incompletely understood. The spider-venom peptide M-lycotoxin exhibits strong membrane-disruptive activity, whereas a single substitution of Leu17 with glutamate (L17E) markedly reduces cytotoxicity. Here, we employed microsecond-scale all-atom molecular dynamics simulations to investigate how this mutation alters the membrane disruption pathway. The wild-type (WT) peptide preserves strong amphiphilicit

Piezo1 mediates hypoxia and mineralization in cementoblasts.

Orthodontically induced inflammatory root resorption (OIIRR) is a common complication associated with excessive compressive force during orthodontic treatment, leading to irreversible cementum destruction. However, the cellular and molecular mechanisms underlying this process remain unclear. This study aimed to investigate the role of the mechanosensitive ion channel Piezo1 in regulating cementoblast function under mechanical stress. Cementoblasts (OCCM-30) were subjected to 1 g/cm2 of compressive force, and global gene expression was analyzed via RNA sequencing, revealing 6932 differentially

Epithelial cells fire voltage spikes.

Bioelectric signaling is well characterized in neurons and cardiomyocytes but remains largely unexplored in epithelia. Using multielectrode arrays, we demonstrate that localized laser injury to epithelial monolayers (primary human keratinocytes and MDCK cells) triggers voltage spikes in the range of 4 to 12 per min for over 60 min postinjury. These spikes exhibit depolarization, repolarization, and hyperpolarization phases lasting 1 to 2 s, a timescale three orders of magnitude slower than neuronal action potentials. Spike amplitudes and frequencies detected at 740 μm from the injury site (the

The new vrdg183 toxin from the Colombian spider Pamphobeteus verdolaga inhibits L-type Ca2+ currents through an allosteric mechanism.

BACKGROUND: Spider venoms are enriched in toxins that modulate mammalian voltage-gated ion channels (VGIC) with pharmacological potential. However, the inventory of toxins of American spiders with effects on voltage-gated Ca2+ channels has lagged the number of toxins interacting with other channels. Here, we aimed at identifying new Ca2+ channel-modulating toxins of the Colombian spider Pamphobeteus verdolaga. METHODS: The sequences of five novel short, positively charged, disulfide-bridged peptides, and one more short, non-bridged control peptide, were selected from the transcriptomic databa

Survey of Synanthropic Spiders in Ireland Reveals Expansion and Dominance of the Invasive Noble False Widow Steatoda nobilis in Urban Habitats (Araneae: Theridiidae).

Rapid urbanisation has led several spider species to adapt to synanthropic microhabitats and establish large populations outside of their native ranges. In Ireland, the establishment and widespread distribution of the Noble false widow spider Steatoda nobilis (Araneae: Theridiidae) (Thorell, 1875) has raised questions about its impact on native spider populations across the country. Through an extensive field survey of six urban centres over an 11-month period, we sought to establish population demographics for S. nobilis and other synanthropic spiders in Ireland for the first time. We surveye

Hypertonicity and Piezo1 Activation Enhance Radiosensitivity of B16 Melanoma.

Radiation therapy is widely used for cancer treatment; however, radioresistance remains a major obstacle, particularly in malignant melanoma. In this study, we investigated the effects of hypertonicity and activation of the mechanosensitive ion channel Piezo1 on the radiation response of B16 mouse melanoma cells. Hypertonic conditions significantly impaired radiation-induced DNA damage signaling and/or repair, as evidenced by reduced γH2AX and 53BP1 foci formation, and increased the number of unrepaired DNA damage sites at later time points. Consistently, hypertonicity enhanced radiation-induc
