Potential of Venom-Derived Compounds for the Development of New Antimicrobial Agents.

The emergence of antimicrobial resistance is a significant challenge in global healthcare, necessitating innovative techniques to address multidrug-resistant pathogens. Multidrug-resistant pathogens like Klebsiella pneumoniae, Acinetobacter baumannii, and Pseudomonas aeruginosa pose significant public health threats, as they are increasingly resistant to common antibiotics, leading to more severe and difficult-to-treat infections. These pathogens are part of the ESKAPE group, which includes Enterococcus faecium, Staphylococcus aureus, and Enterobacter species. Animal venoms, derived from a wid

Highlighting the Potential of LyeTx I, a Peptide Derived from the Venom of the Spider Lycosa erythrognatha, as a Potential Prototype for the Development of a New Antimicrobial Against Carbapenem-Resistant Klebsiella pneumoniae.

Background: Carbapenem-resistant Klebsiella pneumoniae (CRKP) is a multidrug-resistant (MDR) gram-negative bacterium frequently involved in hospital-acquired pneumonia. The infection caused by this superbug has spread quickly in health centers worldwide, leading to high mortality rates. Due to this emerging scenario, the World Health Organization has categorized CRKP as the highest-priority species for the development of new compounds. In this context, antimicrobial peptides (AMPs) stand out as prototypes for alternative antimicrobials against superbugs, including CRKP. Objectives: We aimed to

Shotgun sequencing of airborne eDNA achieves rapid assessment of whole biomes, population genetics and genomic variation.

Biodiversity and its associated genetic diversity are being lost at an unprecedented rate. Simultaneously, the distributions of flora, fauna, fungi, microbes and pathogens are rapidly changing. Novel technology can help to capture and record genetic diversity before it is lost and to measure population shifts and pathogen distributions. Here we report the rapid application of shotgun long-read environmental DNA (eDNA) analysis for non-invasive biodiversity, genetic diversity and pathogen assessments from air. We also compared air eDNA with water and soil eDNA. Coupling long-read sequencing wit

Use of spheroids as a model to evaluate the anticancer action of animal venoms and derived molecules: 2010-2024 review.

BACKGROUND: Cancer is one of the leading causes of death worldwide, with incidence rates continuously increasing, thereby posing a major healthcare challenge. Although many oncological drugs fulfill therapeutic requirements, they often show high toxicity due to their limited specificity. To address this problem, there has been a search for natural therapies, including animal venoms that harbor bioactive molecules with therapeutic potential, as well as biological models that facilitate their study. Consequently, three-dimensional culture models, such as spheroids, play a pivotal role in evaluat

Synthesis of U-theraphotoxin-Pv1a_1, an Aedes aegypti larvicidal disulfide bridged peptide from the Colombian tarantula Pamphobeteus verdolaga (Araneae: Theraphosidae).

The attention to the increased resistance of Aedes aegipty to traditional insecticides has been directed to the development of bioinsecticides, such as those produced by insect predators, e.g. spiders. Here we present the solid-phase synthesis of native U-theraphotoxin-Pv1a_1 (n-Pv1a_1) from Pamphobeteus verdolaga, an active (by contact) insecticidal peptide against A. aegipty. U-theraphotoxin-Pv1a_1 sequence was gathered from venom proteomics and venom gland transcriptomics of Pamphobeteus verdolaga, and synthesized by solid phase using the Fmoc strategy followed by dimethyl sulfoxide promote

Beyond venomous fangs: Uloboridae spiders have lost their venom but not their toxicity.

BACKGROUND: Venom, one of nature's most potent secretions, has played a crucial role in the evolutionary success of many animal groups, including spiders. However, Uloboridae spiders appear to lack venom and capture their prey, unlike venomous spiders, by extensive silk-wrapping and regurgitation of digestive fluids onto the entire prey package. A prevailing hypothesis posits that toxins may have been reallocated from the venom to alternative secretions, like silk or digestive fluids. Yet, whether uloborids have retained venom toxins and the mechanisms underlying prey immobilisation remain unr

Intranasal Absorption Enhancement of Antidiabetic Therapeuticals by the Functional Peptide Segment of Latroeggtoxin-VI.

Latroeggtoxin-VI (LETX-VI) is an active peptide found from the eggs of the spider Latrodectus tredecimguttatus. Previous studies demonstrated that LETX-VI can penetrate the plasma membrane of secretory PC12 cells and its 17-residue C-terminal sequence is the functional peptide segment (FPS), suggesting that the FPS may act as a vector for drug transmembrane delivery. In the present proof-of-concept study, the ability and efficiency of FPS to transmembrane delivery of antidiabetic therapeuticals were preliminarily evaluated. The FPS was covalently fused with a glucagon-like peptide-1 analogue (

Screening inhibitory peptides against sphingomyelinase D from the livestock tick Rhipicephalus microplus using phage display technology.

The cattle tick Rhipicephalus microplus, which is distributed worldwide, causes significant economic loss to the livestock industry. A sphingomyelinase (SMase) D-like protein was previously identified in R. microplus saliva (RmSMase) and characterized in terms of its cofactors, optimum pH, and products generated from sphingomyelin hydrolysis. SMases are present in the venom of spiders from Loxosceles spp. and in the digestive system of non-venomous spiders from Uloborus spp. and are secreted by pathogenic bacteria. In this study A peptide library was screened using Phage Display technology and

Proteolytic stabilization of a spider venom peptide results in an orally active bioinsecticide.

BACKGROUND: The toxin peptide U1-AGTX-Ta1b from the Hobo spider, Eratigena agrestis (Walckenaer, 1802), was studied to determine its potential to serve as a bioinsecticide. RESULTS: U1-AGTX-Ta1b has insecticidal potencies similar to commercial insecticides when injected directly into insect hemolymph but lacks activity when ingested by lepidopterans due to trypsin-like gut proteases. Alanine scanning identified an arginine and lysine rich patch on the peptide's surface that is critical for bioactivity. Targeted stability studies on these basic residues identified a single site, R9, to be the

GsMTx4-loaded GelMA promotes tendon regeneration and suppresses heterotopic ossification via the Apelin signaling pathway.

Tendon injuries pose significant challenges in both athletes and the general population, often leading to prolonged healing, impaired functionality, and increased risk of re-injury. Current treatment options are limited and often yield unfavorable outcomes. Given that tendons are highly mechanosensitive tissues, recent studies highlight the crucial role of mechanotransduction in tissue repair. Piezo1, a mechanosensitive ion channel, has been recognized as a crucial factor attributing to many pathological processes in various tissues, but its specific role in tendon healing has not been previou

Tensile stress promotes the chondrogenic ability of condylar cartilage stem/progenitor cells in the temporomandibular joint via the Piezo1-Ca2+-Prkca pathway.

BACKGROUND: Tensile force is a key regulator for condylar cartilage remodeling in the temporomandibular joint (TMJ), and this biomechanical characteristic underlies the mechanisms of mandibular growth modification in orthodontic practice. Cartilage stem/progenitor cells (CSPCs) in the superficial layer of condylar cartilage play an essential part in the development and remodeling of condylar cartilage. However, the regulatory role of tensile force on condylar CSPCs remains unclear. This study aimed to investigate the impact of tensile loading on condylar CSPCs and explore the molecular mechani

Land use and land cover changes influence the rate of accidents involving venomous animals.

Direct human-nature interactions can have positive and negative impacts on people's health and well-being, and there is an increasing trend of some negative direct human-nature interactions in many parts of the world. Every year, hundreds of thousands of people suffer accidents with venomous animals around the world, including fatal accidents. However, the drivers of the increase in the number of accidents remain unclear. In this study, we evaluated whether land use and land cover changes affect the rate of accidents involving venomous animals in Brazil. We recorded the rate of accidents per c

A Vision for VenomsBase: An Integrated Knowledgebase for the Study of Venoms and Their Applications.

Venoms are complex bioactive mixtures that have independently evolved across diverse animal lineages, including snails, insects, sea anemones, spiders, scorpions, and snakes. Despite the growing interest in venom research, data is fragmented across disparate databases which lack standardization and interoperability. A vision for the proposed VenomsBase platform presented here seeks to address these challenges by using the best practices approach in creating a centralized, open-access platform adhering to FAIR principles (Findable, Accessible, Interoperable, and Reproducible). VenomsBase will u

Mechanical stretch promotes sustained proliferation and inflammation in developing human airway smooth muscle.

Preterm infants frequently require respiratory support, including continuous positive airway pressure (CPAP), that imposes mechanical stretch on highly compliant perinatal airways. How this excess stress impacts airway development and function is not completely understood. Using human fetal airway smooth muscle (fASM), a key cell type in airway contractility and remodeling, as a model, we investigated the effects of stretch, focusing on the role of mechanosensitive ion channels Piezo1 and Piezo2. We found that CPAP-like static stretch did not alter Piezo1 and Piezo2 protein expression per se a

Offensive and Defensive Exploitation of Ants by Termitophagous Spiders (Araneae: Zodariidae).

Myrmecophagy is one of the most common types of dietary specialization among predators. It can include exploitation of ants, termites, or both. Although ants and termites share a few traits, they are distantly related and possess different defensive mechanisms. Therefore, adaptations to ants and termites should differ, especially in arthropod predators of similar body size as their prey. We investigate offensive and defensive adaptations in Zodariidae spider genus Diores, reported to feed on termites. The ancestral state reconstruction of the diet favored termitophagy for the genus, but metaba

The Adjuvant Effect of Hyperbaric Oxygenation for Loxosceles rufescens Bite: A Case Series.

BACKGROUND: The venom of Loxoscelesrufescens (L.r.), also known as the violin and/or brown spider, contains a wide variety of proteins and can induce a complex, intense, and uncontrolled inflammatory response, hemolysis, thrombocytopenia, dermo-necrosis, and renal failure. Studies have postulated the efficacy of hyperbaric oxygen therapy (HBOT) for Loxosceles bites. However, data describing the use and beneficial effects of HBO are, to date, relatively scarce. Only a few cases of Loxosceles bites in Northern Italy have been documented, and there is no laboratory test available for the diagnosi

The Dissociation of Latrophilin Fragments by Perfluorooctanoic Acid (PFOA) Inhibits LTXN4C-Induced Neurotransmitter Release.

α-Latrotoxin stimulates neurotransmitter release by binding to a presynaptic receptor and then forming ion-permeable membrane pores and/or stimulating the receptor, latrophilin-1, or Adhesion G-protein-coupled receptor type L1 (ADGRL1). To avoid pore formation, we use the mutant α-latrotoxin (LTXN4C), which does not form pores and only acts through ADGRL1. ADGRL1 is cleaved into an N-terminal fragment (NTF) and a C-terminal fragment (CTF), which behave as independent cell-surface proteins, reassociating upon binding LTXN4C. We investigated the role of the NTF-CTF association in LTXN4C action,

Cyanine 5-huwentoxin-IV as a novel imaging probe to detect hNav1.7 channel overexpressed in non-small cell lung cancer.

Currently, tools for detecting cancer cells belonging to the non-small cell lung cancer (NSCLC) are lacking. As overexpression of human voltage-gated sodium channel subtype 1.7 (hNav1.7) channels was identified as a promising marker in NSCLC, we investigated whether huwentoxin-IV (HwTx-IV), known to interact preferentially with this sodium channel subtype at nanomolar concentrations, may serve as a practical tool for detecting NSCLC. To achieve this goal, we coupled HwTx-IV to the cyanine 5 fluorophore and evaluated the potential of cyanine 5-HwTx-IV (Cy5-HwTx-IV) as an imaging agent to improv
