Host-Associated Differentiation in a Parasitoid Wasp Connected to Venom-Related but Not Chemosensory Genes.

The mechanisms driving the megadiversity of parasitic wasps remain a central question in evolutionary ecology. To investigate the genomic architecture underlying host-associated differentiation in parasitic wasps, we generated a de novo genome assembly and performed whole-genome re-sequencing of 17 populations of the parasitoid wasp Asecodes parviclava (Eulophidae) across a latitudinal gradient and multiple Galerucella host species. Despite panmixia in host populations, we identified three genetically differentiated wasp lineages, with the most profound divergence separating Galerucella tenell

Two new arthropod toxins purified from the venom of the Mexican scorpion Centruroides nigrescens.

The venom of the slender black scorpion Centruroides nigrescens, from the State of Guerrero México, was fractionated by Sephadex G-50 gel filtration, followed by carboxymethyl cellulose ion separation and high-performance liquid chromatography, permitting to obtain two new peptides toxic to arthropods. The estimate LD50 of the soluble venom for the insect Acheta domesticus was 11.1 μg/g body weight, and for the arthropod Armadillidium vulgare was 61.1 μg body weight . The amino acid sequence of toxin Cnige1 (toxic to A. domesticus and A. vulgare) contains 65 amino acids, with a molecular mass

The honey bee stinger: An unusual pump with collapsible double pistons.

Most stinging animals deliver venom by contracting muscles around a venom reservoir, but honey bees employ a fundamentally different strategy. The stingers of worker bees have the unique ability to autotomize, separating from the body, to continue to sting independently. Despite more than a century of anatomical descriptions, the mechanics of venom pumping have remained unresolved due to the challenges of observing them in action. We recorded high-speed videos of active, autotomized stingers, combined with micro computed tomography-based three-dimensional reconstruction to characterize the bio

The new vrdg183 toxin from the Colombian spider Pamphobeteus verdolaga inhibits L-type Ca2+ currents through an allosteric mechanism.

BACKGROUND: Spider venoms are enriched in toxins that modulate mammalian voltage-gated ion channels (VGIC) with pharmacological potential. However, the inventory of toxins of American spiders with effects on voltage-gated Ca2+ channels has lagged the number of toxins interacting with other channels. Here, we aimed at identifying new Ca2+ channel-modulating toxins of the Colombian spider Pamphobeteus verdolaga. METHODS: The sequences of five novel short, positively charged, disulfide-bridged peptides, and one more short, non-bridged control peptide, were selected from the transcriptomic databa

Microbiota discovered in scorpion venom.

With low nutrient availability and presence of numerous antimicrobial peptides, animal venoms have been traditionally considered to be harsh sterile environments that lack bacteria. Contrary to this assumption, recent studies of animal venom and venom-producing tissues have revealed the presence of diverse microbial communities, warranting further studies of potential microbiota in other venomous animals. In this study we used 16S rRNA amplicon sequencing to elucidate whether scorpion venom contained bacteria, to characterize the bacterial communities, and determine if venom microbiomes differ

Megalomyrmex milenae Transcriptome Reveals a Complex Venom Cocktail.

Megalomyrmex ant species have a rich natural history that provides an interesting backdrop to understanding how venom has been shaped by evolution. However, like many other species in the tribe Solenopsidini, alkaloid investigations have dominated, limiting our understanding of the diversity of venom components. Here we use transcriptomics to qualify and quantify the proteins and peptides within Megalomyrmex milenae, a species of ant native to the Panamanian rainforest along the Panama Canal. RNA transcripts associated with and over-expressed in the venom gland allow the description of putativ

Tachinid Endoparasitoid Flies do not Rely on Domesticated Viruses, Unlike Multiple Hymenopteran Lineages.

Viral domestication, the co-option of integrated viral genes for host functions, has been repeatedly documented in endoparasitoid Hymenoptera. Whether this phenomenon extends to other parasitoid insects remains to be assessed. Here, we tested this hypothesis in tachinid flies, the largest group of non-hymenopteran parasitoids. To this end, we investigated patterns of viral endogenization in 52 genomes, including 37 newly sequenced species. Similarly to hymenopteran endoparasitoids, tachinid genomes were found to display numerous endogenous viral elements (EVEs), primarily derived from insect-i

From venom peptides to neurotherapeutics: BmK defensins and short-chain peptides as modulators of ion channels.

Scorpions, having inhabited the Earth long before the emergence of humans, represent an ancient lineage of arthropods. While often regarded with fear due to their potential to induce severe pain or fatal envenomation, scorpion venoms constitute complex cocktails of bioactive molecules known as toxins. Notably, these toxic components have been repurposed in medical research as valuable sources for therapeutic development. In traditional Chinese medicine (TCM), the venom of Buthus martensii Karsch (BmK), commonly referred to as the Chinese scorpion, has been historically employed in the treatmen

The venom gland transcriptome of Tityus paraguayensis reveals a diverse array of bioactive molecules from the Brazilian Cerrado.

Scorpions are arthropods with venom glands at their telson that produce chemicals such as peptides and proteins. These compounds may have pharmacological effects, including antimicrobial, ion channel modulating, and antihypertensive activities. Our study aims to examine the transcripts from the venom glands of Tityus paraguayensis, focusing on identifying and annotating the genes expressed in these glands. A transcript encoding a potassium channel-modulating peptide was selected for 3D structural modeling, phylogenetic analysis, and interaction assessment. Initially, the scorpions' telsons wer

The Genetic Architecture of Venom Resistance: A Novel Approach to Target Identification and Coevolutionary Discovery.

All species evolve under selective pressures that emerge from their interactions, often antagonistic, with other species. Phenotypes mediating species interactions manifest as the combined products of the genomes of interacting species; understanding the evolutionary processes acting in one lineage therefore cannot be attained without bridging the genomes of interacting species. Venoms have arisen independently more than 100 times in animals and serve diverse roles in species interactions, including predation and defense. Each venom is evolutionarily entwined with reciprocal phenotypes, such a

Immunogenicity of two representative American consensus scorpion neurotoxins from the genera Tityus and Centruroides.

Two consensus scorpion toxins derived from venoms of the genera Centruroides (NATx, North America), and Tityus (SATx, South America) were rationally designed and recombinantly expressed in Escherichia coli Origami. Both recombinant proteins were produced as inclusion bodies and subsequently purified and refolded in vitro to obtain biologically active isoforms. The expression yields were approximately 1 mg/L for rNATx and 0.5 mg/L for rSATx. Structural characterization by circular dichroism spectroscopy revealed that rNATx and rSATx exhibited folding patterns typical of scorpion β-toxins. The p

Self-toxicity and tolerance mechanisms of honeybee venom in honeybees.

Honeybee venom (apitoxin) is a potent mixture of biologically active peptides and enzymes, primarily evolved as a defence against insect predators - including other bees. Recent evidence suggests that honeybees also employ components of apitoxin for both external and internal defence against parasites and microbial infections. Consequently, they are predicted to exhibit a remarkable resistance to their own venom, which they frequently encounter within colonies. To investigate this phenomenon, we examined the physiological responses of honeybee workers and drones to envenomation. Individuals we

Caste-Specific Functional Variation in the Venom of the Army Ant Eciton hamatum.

In ants, division of labour is often associated with caste-specific adaptations. In this study, we examined the morphology of the venom apparatus (venom reservoir volume and sting length) and the composition and activity of the venom across castes (soldier, submajor and media/minor) of the army ant Eciton hamatum to investigate how caste differentiation influences venom traits. Morphometric analyses revealed that soldiers have larger venom reservoirs and longer stings, consistent with their specialised role in defending the colony against vertebrates. Integrative venom profiling using transcri

Venom proteome of the monophagous spider Ammoxenus amphalodes: a potential source of termite-specific toxins.

Venoms are complex chemical arsenals that have evolved across various animal lineages for predation, defence, and other ecological functions. In predatory species, venom composition is often shaped by diet, potentially leading to unique toxin profiles. Spiders are the most diverse group of terrestrial venomous predators, yet the venoms of most species, including highly specialised stenophagous species with narrow trophic niches, remain poorly understood. In this study, we investigated the venom of Ammoxenus amphalodes, a spider that preys exclusively on termites, to explore its molecular compo

Entomopathogenic Nematode Steinernema carpocapsae Venom Proteins Disrupt Developmental Physiology and Reproduction of Spodoptera frugiperda (Lepidoptera: Noctuidae).

The use of Steinernema carpocapsae infective juveniles as biological control agents is a long-standing practice, yet the oral impact of their secreted venom proteins on crop pests remains largely unknown. We evaluated the oral toxicity of S. carpocapsae venom proteins against Spodoptera frugiperda using artificial diet assays. Ingestion caused significant dose-dependent toxicity in early-instar larvae, resulting in mortality and a prolonged developmental duration. Carry-over effects were profound; treated pupae were smaller and malformed, with only 19% of larvae fed on 1000 ng g-1 venom protei

Comparative peptidomics of four wasp venoms reveals extensive peptide diversity, proteolytic patterns, and predicted bioactivities.

Wasp venoms possess complex compositions and diverse bioactivities, making them potential pharmacological sources. In this study, venoms from four wasp species (Vespa mandarinia, V. velutina, V. basalis, and Provespa barthelemyi) were collected by electrical stimulation and analyzed using liquid chromatography-tandem mass spectrometry. A total of 681 peptides were identified, nearly 90% of which had not been previously reported. Comparative analyses revealed pronounced species-specific signatures at both peptide and peptide family levels. Sequence-based analyses indicated that peptide release

AnvRV virus in the parasitoid wasp Anagyrus vladimiri: localization, effect on gene expression, and prevalence.

UNLABELLED: Insect-virus associations have been studied extensively in the context of pathogenic viruses transmitted by insects, whereas research on nonpathogenic viruses remains relatively scarce. Recently, we discovered three nonpathogenic RNA viruses in the parasitoid wasp Anagyrus vladimiri: AnvRV, AnvDV, and AnvIfV. Here, using transmission electron microscopy, we detected AnvRV in the wasp oocytes and in a distinct group of cells in the ovaries, which we termed "satellite cells," but not in the venom gland or venom reservoir, indicating that AnvRV is transmitted transovarially. Next, we

Multiple genetic lineages of the invasive brown widow spider (Araneae: Theridiidae) from inland areas in the Kanto region, Japan.

The brown widow spider, Latrodectus geometricus Koch, 1841, is a medically important invasive species that possesses neurotoxic venom capable of causing latrodectism in humans. This species has recently been found in inland areas of the Kanto region in Japan, raising concerns about its ongoing range expansion and associated public health risks. Here, we analyzed partial sequences of mitochondrial cytochrome c oxidase subunit I (cox1) and 16S ribosomal DNA from 99 individuals (3 males and 96 females) collected from multiple sites in Tokyo and Kanagawa prefectures, evaluating their phylogenetic
