Latrodectus spp. toxins outside venom glands: past, present and future.

Latrodectus spp. are one of the highly poisonous spiders known in the world. Interestingly, the spiders not only produce and secrete toxins by their venom glands, but also have toxic components in other parts of the adult spiders, and even in the newborn spiderlings and unhatched eggs. The venom and venom toxins secreted by their venom glands have been quite extensively studied and, comparatively, the study on the toxins outside venom glands is limited. Although the toxins outside venom glands were discovered at a very early time, it is not until recently that the molecular basis and mechanism

Ecosystem services provided by spiders.

Spiders, ubiquitous and abundant predators in terrestrial ecosystems, often are the subjects of an unjust negative perception. However, these remarkable creatures stand as unsung heroes within our ecosystems, contributing a multitude of ecosystem services critical to human well-being. Here, we describe the diverse spectrum of ecosystem services offered by spiders and their potential to inspire or directly provide nature-based solutions. Provisioning services include the versatile uses of silk-like and other materials, inspiration for biomimetic technology, medicines derived from venom, hemolym

Investigating the immunomodulatory effects of honeybee venom peptide apamin in Drosophila platforms.

Apamin, an 18-amino-acid honeybee venom peptide, although traditionally recognized for its neurotoxic effects, demonstrates potent antimicrobial properties in our research when genetically expressed in Drosophila. This antimicrobial efficacy is independent of its disulfide bonds and is enhanced when the peptide is membrane-tethered. Apamin selectively inhibits pathogenic bacteria, such as Pseudomonas aeruginosa, Enterococcus faecalis, and Escherichia coli, while promoting beneficial bacteria like Lactobacillus plantarum, thereby improving the gut microbiome. This gut-localized antimicrobial ac

A rare, evolutionarily conserved venom protein benefits endoparasitism across parasitoids.

Although many venom proteins and other parasitic effectors have been identified in various specific systems of parasitoids, key elements that contribute to parasitic success across a broad range of taxa remain largely unexplored. Here, we focus on Leptopilina and conduct a large-scale, multi-omics study to explore common venom proteins for these drosophilid parasitoids. We find that this genus has undergone extensive chromosome rearrangements and a rapid turnover of venom gene repertoires between species. Interestingly, we identified a lineage-specific venom lipase, Leptopilina-specific venom

Envenomation by Brown recluse spider bite - a case series of cutaneous loxocelism.

Brown recluse spiders, scientifically known as Loxosceles reclusa, are predominantly found in the United States of America. These spiders generally tend to stay in places away from human habitation. The bite of the spider can result in cutaneous loxoscelism or systemic loxoscelism, which manifests as haemolysis and renal abnormalities. Here, we report a case series of cutaneous loxoscelism.

The predatory behavior of ants: an impressive panoply of morphological adaptations.

Ants are typically omnivorous insects with ovoid heads equipped with short mandibles, but there is great diversity in both the adaptations of their morphological and behavioral traits, as well as their dietary habits. Here, we review the variety of form and function in ant predation. Predation and scavenging were likely the plesiomorphic modes, with Cretaceous ants evolving mandibles well-suited to prey capture. In contemporary ground-dwelling species, mandible morphology can vary with adaptations to capturing particular prey as they may possess trap-jaw, snapping, or falciform mandibles chara

Cvill6 and Cvill7: Potent and Selective Peptide Blockers of Kv1.2 Ion Channel Isolated from Mexican Scorpion Centruroides villegasi.

Scorpion venoms are a rich source of peptides that modulate the activity of ion channels and can serve as a new drug for channelopathies. Cvill6 and Cvill7 are two new peptides isolated from the venom of Centruroides villegasi with MW of 4277 Da and 4287 Da and they consist of 38 and 39 amino acids, respectively, including six cysteines. Sequence alignment revealed high similarity with members of the α-KTx2 subfamily of potassium channel toxins. In electrophysiology, Cvill7 potently inhibited Kv1.2 ion channels with an IC50 of 16 pM and Kv1.3 with an IC50 of 7.2 nM. In addition, it exhibited p

Hymenoptera anaphylaxis and mast cell activation disorders.

PURPOSE OF REVIEW: Among patients with hymenoptera venom allergy (HVA), a high proportion have an underlying systemic mastocytosis. It is essential to identify them for an adequate management. RECENT FINDINGS: The clinical presentation of anaphylaxis after stinging -cardiovascular symptoms and absence of cutaneous- may point to a clonal mast cell disease (MCD). In addition, these patients usually have low mast cell burden, so highly sensitive techniques are needed to detect clonality. SUMMARY: It is important to recognize patients with HVA associated with a clonal MCD since the handling of i

Bee venom loaded chitosan nanoparticles enhances growth, immunity and resistance to vibrio parahaemolyticus in pacific white shrimp.

Despite the known anti-inflammatory, antibacterial, and antioxidant properties of bee venom (apitoxin), its application for promoting growth and health in shrimp (Litopenaeus vannamei), remains largely unexplored. This study explored the effects of bee venom (BV) encapsulated nano-chitosan (BV-CSNPs) on the health and disease resistance of white shrimp. Two hundred forty shrimp (mean weight 6.0 ± 0.02 g) were randomly assigned to four groups (n = 60 per group), each consisting of four replicate hapa (1 m³ each) stocked with 20 shrimp. Shrimp were fed either a basal diet (control) or diets adde

Impact of Naturally Composed and Size-Excluded Hymenoptera Venom Preparations on the Safety and Efficacy of Venom Immunotherapy: A Monocentric Experience.

UNLABELLED: <p>Introduction: The first-line treatment for patients with Hymenoptera venom allergy, a potentially life-threatening condition, is venom immunotherapy (VIT). However, for reasons still unclear, honeybee VIT (HBV-IT) is less effective than yellow jacket VIT (YJV-IT). METHODS: This retrospective monocentric study aimed to investigate the safety and efficacy of naturally composed Hymenoptera venom (NC-HV) in a rush protocol involving a high total venom dose and to compare the results with those previously obtained using size-excluded Hymenoptera venom (SE-HV). Data regarding the num

Immunomodulatory Effect of a Cysteine-Rich Secretory Protein from an Entomopathogenic Nematode with Sterol-Binding Activity.

The Steinernema carpocapsae nematode is known to release several excretory/secretory products (ESPs) in its venom upon contact and during the parasitic infection process of insect hosts. A recurrent family of proteins found in this nematode's venom is the CAP (cysteine-rich secretory protein/antigen 5/pathogenesis-related 1) protein, but the functional role of these proteins remains unknown. To elucidate the biological function, this study focused on characterising the secreted protein, first identified in the venom of the nematode's parasitic stage, and the sequence retrieved from transcripto

Venomics and insect prey specificity of the Central American scorpion Centruroides limbatus (Pocock, 1898) and its comparison with close species Centruroides bicolor (Pocock, 1898).

Centruroides limbatus and Centruroides bicolor are phylogenetically related species with different geographical distributions. The scarce envenomation cases in Costa Rica suggest they are probably 'mammal harmless', but their venom's effect on other animals has been poorly characterized. We aimed to describe the protein venom composition of C. limbatus and C. bicolor, in the context of their trophic habits and prey specificity. We applied a proteomics shotgun approach to analyze the composition of venom peptides of C. limbatus and compare them with those expressed by C. bicolor. We also tested

Spider venom peptides with unique fold selectively block Shaker-type potassium channels.

Natural toxins are highly effective at targeting ion channels with high selectivity and potency. To date, all identified spider venom peptide toxins that modulate voltage-gated potassium (KV) channels inhibit Shab (KV2) or Shal-related isoforms (KV4) by interacting with their voltage-sensing domains. In this study, we report novel spider-derived pore-blocking toxins that selectively target Shaker-type (KV1) channels with nanomolar potency. We isolated murinotoxins MnTx-1 and MnTx-2 from the orange baboon tarantula Pterinochilus murinus and sequenced them using a combination of Edman degradatio

Divergent clinical, inflammatory, and histopathological responses induced by Amazonian Tityus venoms: insights and limitations of current antivenom therapy.

Scorpion stings are considered a neglected condition and represent a serious health problem in many tropical countries, especially for children and the elderly. In Brazil, the yellow scorpion (Tityus serrulatus) is widely found and responsible for the majority of severe envenoming cases; however, other medically relevant species endemic to the Brazilian Amazon region, such as Tityus silvestris, Tityus metuendus and Tityus obscurus, remain underexplored. In the present study, we characterized the clinical, inflammatory and histopathological responses induced by venoms from these Amazonian speci

Impact of climate change on the distribution of Tityus scorpions (Buthidae) in the Amazon: Implications for conservation units and indigenous territories.

Climate change is altering the distribution of organisms, affecting ecosystem goods and services. These impacts can pose new challenges for public health due to shifts in the distribution of venomous animals, influencing the dynamics of envenomation incidents. The consequences of climate change are expected to be more severe in tropical regions, where the most vulnerable communities are located. In this context, the increase of scorpionism in recent decades across the vast Amazonian region of Brazil calls for an assessment of future risks. In this study, we analyze the effects of climate chang

Integrative transcriptomic and proteomic analysis reveals the toxin diversity of the spider Macrothele bannaensis.

Spider toxins are an important and abundant source of bioactive compounds and represent molecular candidates for the development of novel compounds for use in medicine and agriculture. Despite the vast diversity of spider species, only a small fraction of them have been studied in detail regarding venom chemistry. Macrothele bannaensis, a spider belonging to the family Macrotheleidae, is widely distributed in the Xishuangbanna region of Yunnan, China, and serves as an important natural enemy of agricultural pests. However, there is a lack of research on the composition of its venom. In this st
