The effect of alpha-latrotoxin on the neurosecretory PC12 cell line: studies on toxin binding and stimulation of transmitter release.

alpha-Latrotoxin of black widow spider venom was found to bind with high affinity (KA = 1.8 X 10(9)M-1) to specific sites present in discrete number (approximately 6300/cell, approximately 12/micron2) at the surface membrane of PC12 cells. This binding correlated with (and therefore, probably caused) the secretory response produced by the toxin. Binding was enhanced (approximately 2-fold) in the presence of mM concentrations of various divalent cations (Ca2+, Mn2+ and Co2+) while Ba2+ and Sr2+ had a smaller effect and Mg2+ was inactive. Hypertonicity, concanavalin A and trypsin pretreatment of

Anaphylaxis after Hymenoptera stings in three patients with urticaria pigmentosa.

Three patients with urticaria pigmentosa are reported who developed symptoms of anaphylaxis after Hymenoptera stings. Serum IgE antibodies to various Hymenoptera venoms could not be detected in any of the patients. Skin tests were completely negative in one patient, and borderline reactions with honeybee and yellow jacket venom, respectively, were found in the other two. Peripheral blood leukocytes of these latter two patients did not release significant amounts of histamine after exposure to the respective venoms. In patients with mastocytosis, anaphylaxis after insect stings may not be IgE-m

A mammal toxin derived from the venom of a chactoid scorpion.

1. It has been shown that the low toxicity to mammals (LD50 of about 200 mg per kg mice body weight) of the chactoid scorpion venom Scorpio maurus palmatus (Scorpionidae) is due to a single low molecular weight basic protein. 2. This compound was purified by the aid of gel filtration and ion exchange column chromatography, possessed about 80% of the mice lethality of the crude venom with an increase of about 60 fold in its specific toxicity. 3. It is composed of 32 amino acids (mol. wt = 3478) and devoid of isoleucine, leucine, phenylalanine, histidine and tryptophan. 4. The unique amino acid

delta-Philanthotoxin, a semi-irreversible blocker of ion-channels.

1. The digger wasp, Philanthus triangulum, which preys on honeybees, produces a paralysing venom possessing a wide variety of activities. 2. In insects, the venom has a central as well as a peripheral effect; the latter effect consists of a presynaptic as well as a postsynaptic block of the skeletal neuromuscular transmission. 3. The presynaptic block is probably caused by an inhibition of the re-uptake of the transmitter. The postsynaptic effect probably consists of a block of open ion channels. 4. The venom contains at least four active toxins called alpha-, beta-, gamma- and delta-philantho

Sensitivity to hymenoptera in adult males.

The authors studied the sera of 86 selected adult male office workers for IgE antibodies to yellow jacket and honey bee venoms and to environmental inhalants and foods. Structured histories provide information about the experiences of these men with the Hymenoptera and other immunological information. Three men (3%) gave a history of a local or general reaction to the Hymenoptera but no antibodies were detected. Ten men (12%) had IgE antibodies to yellow jacket or honey bee venoms but none gave a history of a reaction. Nine of these men were atopic. The authors speculate that IgE antibodies to

Isolation of a pure toxic polypeptide from the venom of the spider Phoneutria nigriventer and its neurophysiological activity on an insect femur preparation.

The venom of the South American spider Phoneutria nigriventer has been separated into eleven fractions by gel filtration. The neurophysiological activity of each fraction was tested by perfusion of a locust femur preparation. Fractions which gave a neurophysiological response on this perfusion were fractionated further by ion-exchange chromatography. The purity of each sub-fraction was monitored by isoelectrofocusing on polyacrylamide gels and isoelectric points determined. From one of the polypeptide-containing fractions, a pure, almost neutral polypeptide was isolated and shown to have a mol

Secondary structure of Bombyx mori and Dictyostelium discoideum 5S rRNA from S1 nuclease and cobra venom ribonuclease susceptibility, and computer assisted analysis.

The 5S rRNAs from Bombyx mori and Dictyostelium discoideum were end-labeled with [32-P] at either the 5' or 3' end and sequenced using enzymatic digestion. The secondary structure of these molecules was studied using the single-strand specific S1 nuclease and the base-pair specific cobra venom ribonuclease. Computer analysis of these results was performed and was used to generate a consensus secondary structure for each molecule. A comparison of these results with those of other workers is presented.

Allergens in hymenoptera venom. IX. Species specificity to Polistes (paper wasp) venoms.

Sera from 62 patients allergic to Polistes venom were used to study the allergenic relationships among four individual species of Polistes by both direct and inhibition RAST. Both direct and inhibition results showed that the four species' venoms were closely related. The species within the same subgenus were more closely related than those in different subgenera. The Polistes species were more closely related to each other than various yellow jacket (Vespula) species are to each other. The commercial Polistes venom mix appears to contain all of the relevant allergens.

Actions of insect toxin and other toxins derived from the venom of the scorpion Androctonus australis on isolated giant axons of the cockroach (Periplaneta americana).

1. Insect toxin, mammal toxins I and II and crustacean toxin were obtained from the venom of the scorpion Androctonus australis. Their effects on the isolated giant axon of the cockroach Periplaneta americana were investigated by current-clamp and voltage-clamp techniques. 2. In current-clamp conditions, mammal toxins and crustacean toxin (1.3-13 microM) induced a large prolongation of the falling phase of the evoked action potentials. Insect toxin (0.13-3.3 microM) induced a progressive slow depolarization of the membrane potential and repetitive firing of action potentials. No changes in the

Allergy in beekeepers.

Beekeepers represent a high-risk group for allergic disorders. They are affected by beestings but only a very limited fraction of the population develops severe anaphylactic symptoms. On the other hand several beekeepers have slight anaphylactic symptoms when they start working. Immunologically they are like "spontaneously desensitized subjects". They often have bee-venom specific IgE and all of them have very high titers of bee-venom specific IgG. Beekeepers may present an atopic sensitization to bee-hive dust and develop asthma and rhinitis. Finally, they may present a contact dermatitis to

Peripheral catecholamine release by alpha-latrotoxin in the rat.

Intraarterial injection of alpha-latrotoxin (alpha LTx), the major toxin of the black widow spider (Latrodectus mactans tredecimguttatus) venom, into carotid catheterized rats, induced prompt and marked rises in plasma adrenaline and noradrenaline concentrations, indicating that the toxin stimulates catecholamine release from both the adrenal medulla and sympathetic nerve terminals. Pretreatment with the ganglionic blocker chlorisondamine greatly reduced the plasma adrenaline response to alpha LTx but had almost no effect on the noradrenaline response, indicating that alpha LTx-stimulation of

Freeze-fracture study of cardiotoxin action on axonal membrane and axonal membrane lipid vesicles.

Freeze-fracture electron microscopy was used to follow morphological changes induced by Naja mossambica mossambica venom cardiotoxins on crab axonal membranes and thier lipids. It was shown that the extent of morphological changes depended drastically on the purity of cardiotoxin preparations and on their nature. Highly purified cardiotoxin induced mainly fusion of membrane or lipid vesicles. The extent of fusion and other morphological changes depended on the nature of cardiotoxin used: VII4 cardiotoxin induced only fusion while VII1 led to further modifications of membranes and liposomes. Comparisons of the neurotoxic activity of the venom of several species of funnel web spiders (Atrax).

A bioassay has been developed to allow comparison of the neurotoxic activity of the venom of individuals of the four most common species of funnel web spiders (Atrax species). In the absence of purified Atrax neurotoxin, the stimulatory action of the venom on skeletal muscle from the cane toad, Bufo marinus, was compared with that of known concentrations of acetylcholine. Several experiments to validate the assay were performed, then venom samples from both male and female spiders were assayed. It was found that individual female spiders of all Atrax species normally secreted several times as

Naturally occurring inhibitors of the venom of funnel web spiders (Atrax species).

It has been shown that the blood plasma of rats and many other vertebrate species, but not man, contains a factor that inhibits the toxic actions of the venom of funnel web spiders (Atrax) on isolated toad voluntary muscle and rat lung preparations and also on anaesthetized whole rats. This factor was found to be effective against all obvious symptoms produced by the venom of both sexes of A. infensus and of the females of the other three common species of Atrax, particularly when administration of the factor to the test preparation preceded that of the venom.

Electrophysiological evidence for the existence of separate receptor mechanisms mediating the action of 5-hydroxytryptamine.

The transepithelial potential recorded across the salivary gland of the blowfly Calliphora responded to 5-hydroxytryptamine (5-HT) with a complex depolarising response characterised by oscillatory activity at low doses. A rapid screening procedure for characterizing the electrophysiological response of the gland to 5-HT and its analogues was developed. It consisted of recording the potential during the gradual addition of the agent in the form of a ramp. 5-HT gave a characteristic electrophysiological signature which was then compared with that of other analogues. Tryptamine, bufotenine and di

Sequence and specificity of two antibacterial proteins involved in insect immunity.

Immune responses have been described for many different insect species. However, it is generally acknowledged that immune systems must therefore differ from those of vertebrates. An effective humoral immune response has been found in pupae of the cecropia moth, Hyalophora cecropia. The expression of this multicomponent system requires de novo synthesis of RNA and proteins and its broad antibacterial activity is due to at least three independent mechanisms, the most well known of which is the insect lysozyme. However, this enzyme is bactericidal for only a limited number of Gram-positive bacter

Bites and stings of uncommon arthropods. 1. Spiders.

In addition to the Hymenoptera, less common arthropods, such as spiders and reduviids, can inflict injuries of medical importance. Identification of the source of the bite or sting may be difficult but is essential to proper treatment. Part 1 of this article describes recognition and treatment of the bites of various spiders; part 2, beginning on page 107, considers bites and stings of other arthropods.

On the specificity of 125-I-alpha-bungarotoxin binding to axonal membranes.

125I-alpha-Bungarotoxin (alpha-BGT) was used to characterize the binding sites for cholinergic ligands in lobster walking leg nerve membranes. The toxin binding component has been visualized histochemically on the external surfaces of intact axons and isolated axonal membrane fragments. Binding of alpha-BGT to nerve membrane preparations was demonstrated to be saturable and highly reversible (KappD congruent to 1.7 +/- 0.32 X 10(-7) M; Bmax congruent to 249 +/- 46 pmol/mg protein) at pH 7.8, 10 mM-Tris buffer. Binding showed a marked sensitivity to ionic strength that was attributable to the c

Purification and pharmacological properties of eight sea anemone toxins from Anemonia sulcata, Anthopleura xanthogrammica, Stoichactis giganteus, and Actinodendron plumosum.

Eight different polypeptide toxins from sea anemones of four different origins (Anemonia sulcata, Anthopleura xanthogrammica, Stoichactis giganteus, and Actinodendron plumosum) have been studied. Three of these toxins are new; the purification procedure for the five other ones has been improved. Sea anemone toxins were assayed (i) for their toxicity to crabs and mice, (ii) for their affinity for the specific sea anemone toxin receptor situated on the Na+ channels of rat brain synaptosomes, and (iii) for their capacity to increase, in synergy with veratridine, the rate of 22Na+ entry into neuro

Potential-dependent effects of sea anemone toxins and scorpion venom on crayfish giant axon.

Effects of two kinds of sea anemone toxin (Parasicyonis actinostoloides and Anemonia sulcata) and scorpion venom (Leiurus quinquestriatus) on crayfish giant axons were examined electrophysiologically. All toxins acted on the axon in a similar manner to prolong the falling phase of the action potential. In all cases the development of toxicity was reduced when the nerve membrane was depolarized by a current injection. However, the ranges of membrane potential where the significant reduction in toxicity took place were different for each toxin. The action of Parasicyonis toxin was also suppresse
