A harvester ant venom: chemistry and pharmacology.

The mammalian toxicity of the potently algogenic venom of the ant Pogonomyrmex badius is greater than that reported for any other insect venom. This enzyme-rich venom contains high concentrations of phospholipase A2 and B, hyaluronidase, acid phosphatase, lipase, and esterases. This hemolytic secretion from the poison gland products unusual symptoms in mammals and appears to have been evolved as a deterrent for vertebrate predators.

Pseudolymphoma of skin induced by oriental hornet (Vespa orientalis) venom ultrastructural study.

A single subcutaneous injection of Oriental hornet venom into black mice led to the development of a subcutaneous nodule which showed no change during the period of 1 year. Study of the ultrastructure revealed an accumulation of lymphocytes and lymphoblasts and a few plasma cells with formation of Russel bodies. In addition, a few melanin-containing macrophages and macrophages filled with cellular debris were found. This reaction is compared to other pseudolymphomatous reactions in the dermis.

Release of neurotransmitters and depletion of synaptic vesicles in cerebral cortex slices by alpha-latrotoxin from black widow spider venom.

The effect of alpha-latrotoxin on cerebral cortex slices was studied by both biochemical and morphological methods. This toxin greatly stimulates the release of preloaded gamma-amino[3H]butyric acid from cortex slices. The response increases linearly with dose. The release is not dependent on the presence of extracellular Ca2+, and therefore it is not mediated by the release of other transmitters from other types of neurons. In contrast, no significant increase in the release of a nontransmitter substance alpha-amino[14C]isobutyric acid is observed. Since previously we have shown that alpha-la

The use of the radioallergosorbent test in the diagnosis of Hymenoptera anaphylaxis.

IgE antibody levels to Hymenoptera (honey bee, hornet, wasp or yellow jacket) venom or venom sac were measured in the serum of ninety-six patients with a history of immediate hypersensitivity reactions to these insect stings. Normal levels of IgE antibody were found in fourty-four of these patients tested. The severity of the systemic reaction was similar in those patients with normal and elevated IgE antibody levels, and the interval from the systemic reaction to antibody determination was also similar. In twelve patients with a definite reaction to honey bee venom, the IgE antibody was eleva

Near-fatal and fatal anaphylactic reactions to insect sting.

Two patients experienced anaphylactic reaction to insect stings, with residual encephalopathy. One patient had anaphylaxis despite hyposensitization with whole-body extracts, thus supporting recent reports that reliable protection cannot be achieved unless pure venom is used. The other patient had pathologic confirmation of anoxic encephalopathy, thus supporting the concept that encephalopathy with anaphylaxis is secondary to associated circulatory collapse rather than a primary allergic response of brain disease.

Survey of 2144 cases of red-back spider bites: Australia and New Zealand, 1963--1976.

An analysis has been made of 2144 consecutive cases of latrodectism (envenomation by the red-back spider, Latrodectus mactans hasselti) reported to the Commonwealth Serum Laboratories. In the last eight years, notifications have averaged 240 cases per annum. Bites, usually on the extremities (74%), occurred most frequently in the summer months, and in the afternoon or evening. Most victims (79%) were aged between 18 and 50 years and 64.4% of them were males. Males are still often bitten on the genitals or buttocks (9.7% of cases). Local pain, redness and swelling were the most common symptoms,

Harvester ant sensitivity: in vitro and in vivo studies using whole body extracts and venom.

Harvester ant stings by Pogonomyrmex maricopa (Pm) or Pogonomyrmex rugosus (Pr) resulted in serious reactions in 8 patients, 4 with generalized reactions and 1 with large local reactions. Exposure to one species in the genus Pogonomyrmex (P) appeared to cross-sensitize ant-sensitive patients to other species in the same genus as evidenced by skin testing and leukocyte histamine release, but these patients were less sensitive to extracts from other stinging Hymenoptera, including bee, wasp, yellow jacket, hornet, and Formica ant. Pr ant venom was obtained by electrical stimulation of live ants

Allergens in bee venom II. Two new high molecular weight allergenic specificities.

Two new allergenic specificites were detected in honeybee venom and the two corresponding protein substances isolated by gel filtration, immunoadsorption, and ion exchange chromatography. The first of these, allergen B, has a molecular weight ranging from 49,000 to more than 200,00 d and can be recognized by rabbit and guinea pig antisera as well as by human reaginic sera using the radioallergosorbent test (RAST). Allergen B gives a single line in immunodiffusion distinct from hyaluronidase, phospholipase A, melittin, and the other high molecular weight substances described and gives a single

Allergens in bee venom. III. Identification of allergen B of bee venom as an acid phosphatase.

Allergen B previously isolated from honeybee venom and shown to be a mildly acidic protein consisting of polymers of a chain of 49,000 d is shown to have acid phosphatase activity. Allergen B is homogeneous by several criteria. No acid phosphatase, alkaline phosphatase, or esterase activity was found in any other allergen or fraction of bee venom. Acid phosphatase activity was also found in yellow jacket venom and extracts of venom sacs from bumblebees and paper wasps.

Ants and bees: whole body extracts vs venom.

Ants, bees, and other stinging hymenoptera can induce serious anaphylactic reactions in susceptible individuals. Certainly, hymenoptera venom contains potent antigens that have been useful in testing and treating hymenoptera-sensitive patients who fail to acquire protection from whole body extract therapy. However, before whole body extracts are forever abandoned, their antigen content and stability should be re-examined by means of modern immunologic methods.

[Effect of sodium deprivation on the isolated guinea pig ileum in a physiological medium or in the presence of scorpion venom].

We concluded from our results that contractures which occurred when NaCl is replaced by other substitutes could be due to increased Ca entry. This ionic disturbance does not produce any increase of the resting acetylcholine output such as that described for the innervated longitudinal muscle obtained from the ileum. In this environment, venom from the Scorpion (Androctonus australis) is shown to have no effect.

Stimulation of sodium and calcium uptake by scorpion toxin in chick embryo heart cells.

Scorpion toxins, the basic miniproteins of scorpion venom, stimulated the passive uptake of Na+ and Ca2+ in chick embryo heart cells. Half-maximum stimulation was obtained for 20-30 nM Na+ and 40-50 nM Ca2+. Scorpion toxin-activated Na+ and Ca2+ uptakes were fully inhibited by tetrodotoxin, a specific inhibitor of the action potential Na+ ionophore in excitable membranes. Half-maximum inhibition was obtained with the same concentration of tetrodotoxin (10 nM) for both Na+ and Ca2+. Scorpion toxin-stimulated Ca2+ uptake was dependent on extracellular Na+ concentration and was not inhibited by C

Imported fire ant hypersensitivity. Studies of human reactions to fire ant venom.

It is now apparent that venom and venom components of the Hymenoptera superfamilies of Apida (honeybee) and Vespida (wasps, yellow jackets, and hornets) are becoming increasingly important in the diagnosis and treatment of hypersensitivity reactions. Stings from fire ants (superfamily Formicidae, family Myrmicinae) have also been recognized as causes of systemic reactions in man. Fire ant venom is unique in its composition, consisting mainly of alkaloids in aqueous suspension with only trace amounts of protein. This study compares the skin reactivity of fire ant venom and synthesized alkaloid

Studies of the antigenicity and allergenicity of phospholipase A2 of bee venom.

The antigenic and allergenic properties of phospholipase A2 (PLA2) and whole bee venom were compared by measuring the IgG and IgE antibody responses in animals and man. Precipitating antibodies raised in rabbits and reaginic and other antibodies raised in mice reacted about equally with both bee venom and PLA. The majority of human sera containing bee venom-specific IgE also contained PLA-specific IgE, although in somewhat lower titers. Similarly, most human sera with significant amounts of total antibodies reacting with bee venom also had antibodies reacting with PLA. Histamine and SRS-a rele

A new method for labelling saxitoxin and its binding to non-myelinated fibres of the rabbit vagus, lobster walking leg, and garfish olfactory nerves.

1. A new method of labelling saxitoxin (STX) is described, based on transfer of tritium from tritiated water to the toxin. 2. The radiochemical purity of the labelled toxin has been directly determined, rather than being based on indirect biochemical means, as in previous experiments with Wilzbach-labelled STX and TTX. 3. The specific activity of the labelled toxin, 66 d..m.f-mole-1, corresponds with one tritium atom per molecule STX, an improvement of about 300-fold over other means of labelling TTX and STX. 4. The binding of this toxin to rabbit, lobster and garfish olfactory nerve fibres ha

Experimental studies on the aetiology of acute scorpion pancreatitis.

This study examined the action of the venom of the scorpion Tityus trintatis on the pancreas and gastrointestinal tract in anaesthetized dogs, on the isolated extracorporeal haemoperfused canine pancreas and on the isolated canine sphincter of Oddi. The venom induces exocrine secretion in both the isolated and intact pancreas and causes contraction of the isolated sphincter of Oddi. These results are discussed in relation to the pathogenesis of acute scorpion pancreatitis and possibly of some other forms of acute pancreatitis.

Allergens in bee venom. I. Separation and identification of the major allergens.

Honeybee venom was separated into seven fractions by gel filtration on Sephadex G-75. Allergenic activities of these fractions were assessed by the paper disc radioallergosorbent test (RAST) with a panel of sera from 24 individuals who had systemic reactions to bee stings, 7 who had large local reactions, and 10 control subjects who had reactions of 5 cm or less following bee stings. Three fractions were identified by enzyme or direct hemolytic activity. Twenty-nine of 31 sera from patients having either systemic or large local reactions to bee stings were positive when radioallergosorbent tes

An enzyme mechanism in explanation of pain in angina pectoris.

The effect of catecholamines, represented by epinephrine and norepinephrine, on the activity of phospholipase A2 from bee venom was studied. It was shown that the hydrolysis of l-alpha-lecithin to lysolecithin and a fatty acid was considerably activated by preincubation of the lecithin with the biogenic amine. On the other hand, addition of nitroglycerin or propranolol to the enzyme solution considerably curtailed activation by the catecholamines. [Biological significance of animal toxins].

Animal toxins are not to be understood as substances of random occurrence; they are of great importance in ecology. The venom-secreting organs have developed in a wide variety of ways and serve either as an offensive weapon or for protection against enemies. Accordingly, we may distinguish between actively venemous and passively venemous animals. There are other animals that are toxic rather by chance and due to extraneous factors; these we may call poisonous animals. Research into animal toxins is important from both the chemical and the biological points of view. In addition to basic knowled
