Toxicological response of silver catfish (Rhamdia quelen) after acute exposure to a commercial insecticide containing thiamethoxam.

This study assessed the hematological, enzymatic and osmoregulatory responses of silver catfish (Rhamdia quelen) exposed to sublethal concentrations (1.125 and 3.750 µg/L) of a commercial thiamethoxam-containing insecticide used on rice crops. Groups of 6 fish per tank (in triplicate, n = 3, total 54 fish) were exposed for up to 96 h to different concentrations of the compound. After this period, fish were placed in clean water for 48 h. Two fish from each tank (6 per treatment) that had been exposed to the insecticide for 24 h were anesthetized with eugenol and blood was collected to evaluate

Toxicological impacts of herbicide paraquat dichloride on histological profile (gills, liver, and kidney) of freshwater fish Channa punctatus (Bloch).

Paraquat dichloride is a broad-spectrum herbicide used worldwide. It is very fast acting and used to kill a wide range of grasses and broad-leaved weeds. Paraquat dichloride gets run off to aquatic water bodies, and its presence has been reported by various researchers, where its effect is certain on aquatic organisms. Fish are vulnerable to aquatic pollutants as they are in direct contact with their environment. Therefore, our study was designed to evaluate the effects of herbicide paraquat dichloride on histology of vital organs (gills, liver, and kidney) of the fresh water fish Channa punct

Use of embedded Chelex chelating resin and sediment toxicity bioassays with medaka embryos to determine the bioavailability and toxicity of lead-contaminated sediment.

The aquatic sediment acts as a reservoir for multiple sources of pollutants including toxic metals. Most analytical methods used for estimating the bioavailability of sediment heavy metals have not been biologically validated by correlation with an aquatic organism's response. A reliable whole-sediment contacting toxicity assay using vertebrate species is lacking and the exposure routes for sediment metals are unclear. This study established a novel bio-analytical approach involving the Chelex-100 resin detection system and sediment toxicity assessment with embryo-larval stages of medaka fish

Effects of dechlorane plus on intestinal barrier function and intestinal microbiota of Cyprinus carpio L.

Dechlorane Plus (DP) is a typical polychlorinated flame retardant that has been emerged in chemical products. Due to its accumulation and amplification effect, the toxicity of DP has become a widespread environmental safety issue. However, whether DP can affect the intestinal tract of teleost fish remains largely unclear. To understand its effects on the intestinal barrier, morphological characteristics and intestinal microbiome of common carp, different concentrations (30, 60 and 120 μg/L) of DP were exposed to common carps for 4 weeks. The results indicated that DP evidently shortened the in

Sublethal effects of carbendazim in Jenynsia multidentata detected by a battery of molecular, biochemical and genetic biomarkers.

The fungicide carbendazim (CBM) has been applied all around the world but its potential adverse effects other than its recognized activity as endocrine disruptor in non target organisms have been scarcely studied. The aims of this work were (1) to use a battery of biomarkers that can reflect potential negative effects such as oxidative stress, genotoxicity, neurotoxicity or altered immune response; and (2) to examine biomarkers of detoxification by analyzing the gene expression of cytochrome P4501A1 (CYP1A1) and the multi-xenobiotic resistance protein P-glycoprotein (P-gp) in the freshwater fi

Bisphenol A and 17α-ethinylestradiol-induced transgenerational gene expression differences in the brain-pituitary-testis axis of medaka, Oryzias latipes†.

Endocrine disrupting chemicals (EDCs), such as bisphenol A (BPA) and 17α-ethinylestradiol (EE2), can have far reaching health effects, including transgenerational abnormalities in offspring that never directly contacted either chemical. We previously reported reduced fertilization rates and embryo survival at F2 and F3 generations caused by 7-day embryonic exposure (F0) to 100 μg/L BPA or 0.05 μg/L EE2 in medaka. Crossbreeding of fish in F2 generation indicated subfertility in males. To further understand the mechanisms underlying BPA or EE2-induced adult onset and transgenerational reproducti

Inflammatory lncRNA AK039862 regulates paraquat-inhibited proliferation and migration of microglial and neuronal cells through the Pafah1b1/Foxa1 pathway in co-culture environments.

Emerging evidences having suggested that particular lncRNAs have a potential effect on PD progression through provoking damage and inflammatory responses of microglia/ dopaminergic cells. In addition, paraquat can be accumulated in human body through various approaches and have an increased risk for Parkinson's disease. However, the specific role and mechanism of lncRNA related to neurotoxic in the progression of PD is unclear. In our study, a mouse PD model was established induced by the intraperitoneal injection of paraquat (5 mg/kg and 10 mg/kg) every three days (10 times). We determined di

Comparative Determination of Cytotoxicity of Sub-10 nm Copper Nanoparticles to Prokaryotic and Eukaryotic Systems.

Copper nanoparticles demonstrate antibacterial activity, but their toxicity to eukaryotic systems is less understood. Here, we carried out a comparative study to determine the biocompatibility and cytotoxicity of sub-10 nm copper nanoparticles to a variety of biological systems, including prokaryotic cells (Escherichia coli), yeast, mammalian cell lines (HEK293T, PC12), and zebrafish embryos. We determined the bearing threshold for the cell-death-inducing concentration of copper nanoparticles by probing cell growth, viability, as well as embryological features. To exclude the partial toxicity

Effects on Ultrastructure, Composition and Specific Surface Area of the Gills of Odontesthes bonariensis Under Subchronic Glyphosate-Based Herbicide Exposure.

Gills represent one of the major sites of gas exchange of fish, consequently they are in continuous close contact with the aquatic environment and its pollutants. In the present study the effects on gills of pejerrey fish, Odontesthes bonariensis, under glyphosate-based herbicide subchronic exposure were analyzed. Adult animals were exposed to sublethal concentrations of a glyphosate-based commercial formulation (1 and 10 PMG mg L-1, PMG: glyphosate active ingredient) for 15 days, while control group was maintained in rearing water. Ultrastructural changes in gills were observed by scanning el

Effects of crude oil vapors on the cardiovascular flow of embryonic Gulf killifish.

Direct contact with toxicants in crude oil during embryogenesis causes cardiovascular defects, but the effects of exposure to airborne volatile organic compounds released from spilled oil are not well understood. The effects of crude oil-derived airborne toxicants on peripheral blood flow were examined in Gulf killifish (Fundulus grandis) since this model completes embryogenesis in the air. Particle image velocimetry was used to measure in vivo blood flow in intersegmental arteries of control and oil-exposed embryos. Significant effects in oil-exposed embryos included increased pulse rate, red

Contact micropredation may play a more important role than exotoxicity does in the lethal effects of Karlodinium australe blooms: Evidence from laboratory bioassays.

Multiple dinoflagellate species from the genus Karlodinium have been well known to form massive and toxic blooms that consequently cause fish kills in many coastal waters around the world. Karlodinium australe is a mixotrophic and potentially ichthyotoxic species associated with fish kills. Here, we investigated phagotrophy of K. australe (isolate KaJb05) established from a bloom event in the West Johor Strait, Malaysia, using several prey species (phytoplankton, zooplankton, and larval fish). The results showed that K. australe ingested relatively small prey cells of co-occurring microalgae b

Wastewater remediation of heavy metals and pesticides using rice straw and/or zeolite as bioadsorbents and assessment of treated wastewater reuse in the culture of Nile tilapia (Oreochromis niloticus).

The remediation of wastewater (WW) is a promising solution for limited water sources. This study aimed to evaluate rice straw (RS) and zeolite (Z) as bioadsorbents for the removal of pollutants, including heavy metals (HMs) (cadmium [Cd], nickel [Ni], and lead [Pb]) and malathion (PC), from WW and to assess the suitability of reusing remediated WW in fish rearing units. A total of 11 treatment groups with 3 replicates each were designed with different combinations of RS and/or Z for the treatment of real WW contaminated with HMs and malathion, where the WW remained in contact with the adsorben

Characterizing transcriptomic responses of southern flounder (Paralichthys lethostigma) chronically exposed to Deepwater Horizon oiled sediments.

To obtain a deeper understanding of the transcriptomic responses to oil in southern flounder (Paralichthys lethostigma), we performed quantitative PCR and RNA sequencing on liver and gill tissue after a chronic exposure (35 days) to Deepwater Horizon crude oiled sediment and after a 30-day recovery period. We wanted to understand which specific genes are differentially expressed in liver and gill tissues directly after oiled sediment exposure and with the addition of a recovery period. Furthermore, we wanted to examine specific enriched pathways in these two tissues to determine the impact of

Exposure to Aerosolized Algal Toxins in South Florida Increases Short- and Long-Term Health Risk in Drosophila Model of Aging.

Harmful algal blooms (HABs) are a rising health and environmental concern in the United States, particularly in South Florida. Skin contact and the ingestion of contaminated water or fish and other seafood have been proven to have severe toxicity to humans in some cases. However, the impact of aerosolized HAB toxins is poorly understood. In particular, knowledge regarding either the immediate or long-term effects of exposure to aerosolized cyanotoxins produced by freshwater blue-green algae does not exist. The aim of this study was to probe the toxicity of aerosolized cyanobacterial blooms usi

Tolerance and dose-response assessment of subchronic dietary ethoxyquin exposure in Atlantic salmon (Salmo salar L.).

Ethoxyquin (EQ; 6-Ethoxy-2,2,4-trimethyl-1,2-dihydroquinoline) has been used as an antioxidant in feed components for pets, livestock and aquaculture. However, possible risks of EQ used in aquafeed for fish health have not yet been characterized. The present study investigated the toxicity and dose-response of subchronic dietary EQ exposure at doses ranging from 41 to 9666 mg EQ/kg feed in Atlantic salmon (Salmo salar L.). Feed at concentrations higher than 1173 mg EQ/kg were rejected by the fish, resulting in reduced feed intake and growth performance. No mortality was observed in fish expose

Bio-interactions and risks of engineered nanoparticles.

Nano technological research offered uncountable opportunities for engineered nanoparticles (ENPs) in the field of biomedical, pharmaceutical, agricultural, cosmetics, textiles, automobiles and electronic industry. Large scale commercial production and use of nanoparticles with smaller size and characteristic physico-chemical properties enhance the possibility of amenable toxicity to the environment. Primary important species of the ecosystem like bacteria, algae, fishes and plants are at high risk with nanoparticle (NP) toxicity. ENP distributed in air, water and soil can directly affect the l

Predicted sediment toxicity downstream of combined sewer overflows corresponds with effects measured in two sediment contact bioassays.

The present study investigated the sediment toxicity along 100 m transects downstream from three different combined sewer overflows (CSOs) using a coupled substance risk assessment and bioassay approach. As a first step, a risk assessment according to the consensus based sediment quality guideline was performed to estimate the probability for an incidence of toxicity to benthic organisms in freshwater sediments using their pollutant concentrations of metals, PAHs and PCBs. Secondly, two sediment contact assays (SCAs) using the nematode Caenorhabditis elegans and embryos of the fish Danio rerio

Effects of Neonicotinoid Insecticides on Physiology and Reproductive Characteristics of Captive Female and Fawn White-tailed Deer.

Over the past decade, abnormalities have been documented in white-tailed deer (Odocoileus virginianus) in west-central Montana. Hypotheses proposed to explain these anomalies included contact with endocrine disrupting pesticides, such as imidacloprid. We evaluated the effects of imidacloprid experimentally at the South Dakota State University Wildlife and Fisheries Captive Facility where adult white-tailed deer females and their fawns were administered aqueous imidacloprid (an untreated control, 1,500 ng/L, 3,000 ng/L, and 15,000 ng/L).
