Non-photochemical quenching speeds up with increasing temperatures.

Photosynthesis constantly adapts to fluctuating light and temperature. Under high light, non-photochemical quenching (NPQ) protects the photosynthetic machinery by dissipating excess excitation energy as heat. With rising global temperatures and more frequent heat events, understanding how photosynthesis responds to combined light and temperature stress is critical. In vascular plants, NPQ is regulated by the protonation of Photosystem II subunit S (PsbS) and the enzymatic formation of zeaxanthin, but it remains unclear how temperature influences the complete NPQ response and the roles of thes

Combined single-cell transcriptome and immune repertoire analysis reveals hepatic and renal immune injury by heat stroke.

Heat stroke (HS) is the most severe heat-related emergency, and its pathophysiology remains largely unknown, especially for exertional HS (EHS), which affects younger populations, athletes, and manual workers. Herein, we performed single-cell-transcriptomics, T cell receptor sequencing, and flow cytometry of PBMCs from 9 healthy control participants, 9 patients with heat exhaustion, and 9 patients with EHS to explore complex immunological responses associated with HS pathobiology. We showcased that granzyme-positive T cells and CD56dim NK cells with high cytotoxicity features and IL-1B+NLRP3+

Targeted Disruption of the Outer Membrane of Gram-Negative Bacteria with Enediyne-Labeled Light-Responsive Au Nanorods.

Today a significant number of Gram-negative bacteria have evolved to inactivate traditional antibiotics like penicillin and continue to develop resistance against next-generation antimicrobial drugs at a shocking rate. Taking inspiration from actinobacteria (order Actinomycetales) that make reactive enediynes with a (Z)-1,5-diyn-3-ene core to arm themselves, we report the photoactivation of the enediyne precursor octa-4-en-2,6-diyne-1,8-diamine (EDDA) to kill DH5α cells. To trigger Bergman cyclization of EDDA to make diradicals, we used gold nanorods (AuNRs) of chosen dimensions to act as a li

Behavioral thermoregulation in relation to humidity in wild Japanese macaques (Macaca fuscata yakui): the significance of semi-shade.

Recent climate change is increasing thermal stress, highlighting the importance of understanding how wild animals behaviorally buffer these conditions. Although microhabitat use is known to vary with temperature and to function as behavioral thermoregulation, the effect of humidity on this process (i.e., the effect of humidity on temperature-dependent microhabitat use) remains poorly understood. Here, I aimed to examine microhabitat use in wild Japanese macaques (Macaca fuscata yakui) on Yakushima, Japan, across sunlight categories (sun, semi-shade, shade), explicitly accounting for both tempe

Simulated heat waves aggravate the immune response to sublethal acetamiprid exposure in the honey bee (Apis mellifera).

Simultaneous exposure to thermal stress and agrochemicals represents a realistic yet understudied toxicological scenario for pollinators. Honey bees (Apis mellifera) increasingly face intense heat waves while foraging in pesticide-treated landscapes. Understanding the cumulative impact of these stressors is therefore critical for predicting pollinator health. We investigated the combined effects of simulated heat waves (exposed to 42 °C for two hours over three consecutive days) and chronic sublethal exposure to the neonicotinoid acetamiprid on honey bee immunity and physiology. By subjecting

Organelle contact reorganization drives calcium-dependent autophagy under proteostatic stress.

Disruption of proteostasis is a defining feature of cancer and other chronic diseases. The AAA+ ATPase VCP/p97 (valosin containing protein) is a key regulator of proteostasis by disassembling ubiquitinated substrates for degradation. VCP overexpression supports cancer cell survival and correlates with poor prognosis, promoting the development of VCP inhibitors as anti-cancer agents. However, the molecular basis for cancer-selective vulnerability of VCP inhibition remains unclear. Here, we demonstrate that allosteric VCP inhibition triggers cell-type specific macroautophagy/autophagy through dy

Enhancing Thermal Stability of Perovskite Film for durable Solar Cells.

The practical application of metal halide PSCs (PSCs) is badly impeded by their long-term stability concerns, with the issues primarily rooted in the perovskite film. In real-service environments, PSCs endure multiple external factors, light, heat, humidity, and oxygen. Among them, thermal instability represents a particularly severe and fundamental challenge, since heat acts not only as a direct degradation driver but also as a potent accelerator of other degradation pathways. Herein, this review discusses the thermal degradation mechanisms of perovskite from the perspectives of phase transit

An Innovative Bioengineering Approach to Investigate the Response of Melanin-Rich Cells to Intense Pulsed Light (IPL).

Light-based therapies are becoming increasingly = more mainstream, not only within the medical science space, but also within the fields of cosmetic dermatology and personal grooming. Intense Pulsed Light (IPL) harnesses the ability of the natural chromophore-melanin to absorb light energy, which is translated into heat energy and consequently results in targeted thermolysis of cells rich in melanin. This mechanistic pathway lends itself to a wide range of applications, including long-term hair removal, skin rejuvenation, the treatment of unwanted pigmentation, and the treatment of ophthalmic

Progress in Metal-Based Nanomaterials for Photothermal Therapy in the Second Near-Infrared Window.

Photothermal therapy (PTT), characterized by its non-invasiveness, minimal side effects, and potent anti-tumor efficacy, represents a promising alternative to conventional cancer treatments. PTT in the near-infrared-II (NIR-II) window (1000-1700 nm) obtains significant interest due to its lower energy dissipation, deeper penetration, and higher power safety windows than its counterparts conducted in other optical bands. Among those materials capable of coupling light and heat within the NIR-II window, metal-based nanomaterials bear a strong photothermal conversion capability that establishes t

SPOCK1 acts as a negative regulator of ferroptosis in non-small cell lung cancer.

Non-small cell lung cancer (NSCLC) is a leading cause of cancer-related morbidity and mortality worldwide. Therefore, there is an urgent need to discover precise diagnostic markers and novel therapeutic strategies. A genome-wide association study (GWAS) was conducted using the Kunshan Elderly Cohort to identify genetic susceptibility factors underlying NSCLC. GWAS results identified that seven single-nucleotide polymorphisms (SNPs) were associated with NSCLC risk, including SPOCK1 (SPARC/osteonectin, CWCV, and Kazal-like domains proteoglycan (1), which emerged as a key candidate oncogene drivi

Advances in IR-780-derived theranostics: A comparative analysis of free and nanoencapsulated forms.

IR-780 is a lipophilic heptamethine cyanine dye that exhibits strong absorption and fluorescence emission in the near-infrared (NIR) region (700-900 nm). Similar to other cyanine dyes, IR-780 is emerging as a promising fluorophore for fluorescence-guided surgery (FGS). In addition, due to its high molar extinction coefficient, IR-780 can generate reactive oxygen species (ROS), enable efficient conversion of light into heat, and impart additional phototherapeutic capabilities. Consequently, IR-780 represents a promising "all-in-one" single molecule for FGS and phototherapy applications. However

Time-dependent hepatic immune-metabolic reprogramming in Channa argus under thermal stress: An integrated transcriptomic, biochemical and histological analysis.

Water temperature is a pivotal environmental factor governing fish physiology and health, and disease outbreaks in aquaculture are frequently constrained to specific thermal ranges. In Channa argus, rhabdovirus infections have been reported within a relatively narrow temperature window, yet the underlying host physiological responses remain poorly understood. In this study, transcriptomic, biochemical, and histological approaches were integrated to examine the effects of short-term and prolonged heat stress on host responses. Transcriptomic analysis reveals duration-dependent transcriptional p

Increased sensitivity of sublethally injured Escherichia coli O157:H7 to environmental factors induced by dielectric barrier discharge plasma.

The sensitivity of sublethally injured Escherichia coli O157:H7 caused by dielectric barrier discharge (DBD) plasma to diverse environmental factors was investigated in the work. The sublethally injured E. coli cells were achieved by DBD plasma treatment at 18 W for 40 s. The cultivability of injured cells decreased progressively with increasing NaCl concentration in trypticase soy agar (TSA). Although both normal and injured cells were sensitive to mild heat, the latter exhibited increased sensitivity. Incubation at pH 3.0 and 4.0 for 40 min had no effect on the viable counts of normal cells.
