Effect of heat acclimation training frequency on the physiological adaptations of Thoroughbred horses.

Heat acclimation enhances thermoregulation and cardiovascular function. Although daily training protocols are typically recommended for humans, optimal training protocols for Thoroughbred horses remain unclear. Here, we compared the effects of two heat acclimation protocols, consecutive and intermittent, in Thoroughbred horses. In a randomized crossover study, eight trained Thoroughbred horses completed either a consecutive (CONS: 9 consecutive days) or an intermittent (INT: 3 days/wk for 3 wk) heat acclimation protocol, comprising 30 min of exercise in hot conditions (WBGT 30°C). Incremental

A differential single-cell transcriptome atlas of left-sided and right-sided colorectal cancer.

Left-sided colorectal cancer (LCRC) and right-sided colorectal cancer (RCRC) show marked differences in prognosis and therapeutic response, yet the underlying molecular mechanisms remain poorly understood. We hypothesize that these clinical differences reflect distinct cell-type-specific transcriptional programs and microenvironmental interactions that can be resolved at single-cell resolution. To test this hypothesis, this study analyzed single-cell transcriptomic data from 16 colorectal cancer (CRC) patients (8 LCRC and 8 RCRC), revealing molecular differences between LCRC and RCRC in tumor

Combined single-cell transcriptome and immune repertoire analysis reveals hepatic and renal immune injury by heat stroke.

Heat stroke (HS) is the most severe heat-related emergency, and its pathophysiology remains largely unknown, especially for exertional HS (EHS), which affects younger populations, athletes, and manual workers. Herein, we performed single-cell-transcriptomics, T cell receptor sequencing, and flow cytometry of PBMCs from 9 healthy control participants, 9 patients with heat exhaustion, and 9 patients with EHS to explore complex immunological responses associated with HS pathobiology. We showcased that granzyme-positive T cells and CD56dim NK cells with high cytotoxicity features and IL-1B+NLRP3+

Simulated heat waves aggravate the immune response to sublethal acetamiprid exposure in the honey bee (Apis mellifera).

Simultaneous exposure to thermal stress and agrochemicals represents a realistic yet understudied toxicological scenario for pollinators. Honey bees (Apis mellifera) increasingly face intense heat waves while foraging in pesticide-treated landscapes. Understanding the cumulative impact of these stressors is therefore critical for predicting pollinator health. We investigated the combined effects of simulated heat waves (exposed to 42 °C for two hours over three consecutive days) and chronic sublethal exposure to the neonicotinoid acetamiprid on honey bee immunity and physiology. By subjecting

SPOCK1 acts as a negative regulator of ferroptosis in non-small cell lung cancer.

Non-small cell lung cancer (NSCLC) is a leading cause of cancer-related morbidity and mortality worldwide. Therefore, there is an urgent need to discover precise diagnostic markers and novel therapeutic strategies. A genome-wide association study (GWAS) was conducted using the Kunshan Elderly Cohort to identify genetic susceptibility factors underlying NSCLC. GWAS results identified that seven single-nucleotide polymorphisms (SNPs) were associated with NSCLC risk, including SPOCK1 (SPARC/osteonectin, CWCV, and Kazal-like domains proteoglycan (1), which emerged as a key candidate oncogene drivi

Time-dependent hepatic immune-metabolic reprogramming in Channa argus under thermal stress: An integrated transcriptomic, biochemical and histological analysis.

Water temperature is a pivotal environmental factor governing fish physiology and health, and disease outbreaks in aquaculture are frequently constrained to specific thermal ranges. In Channa argus, rhabdovirus infections have been reported within a relatively narrow temperature window, yet the underlying host physiological responses remain poorly understood. In this study, transcriptomic, biochemical, and histological approaches were integrated to examine the effects of short-term and prolonged heat stress on host responses. Transcriptomic analysis reveals duration-dependent transcriptional p

Diagnosis and management of heat stroke and other heat-related illness in the emergency department.

Heat-related illness encompasses a spectrum of conditions ranging from heat cramps and heat exhaustion to life-threatening heat stroke, all resulting from impaired thermoregulation under excessive environmental heat load. The incidence and severity of heat-related illnesses are increasing, underscoring the need for evidence-based emergency management strategies. Accurate diagnosis requires differentiating heat-related illness from conditions that may mimic hyperthermia. Timely recognition and rapid cooling are the cornerstones of management. This review summarizes current evidence on the diagn

Coping with the heat: Thermoregulation and welfare of the plains vizcacha under acute thermal challenge (Rodentia: Chinchillidae).

Environmental temperature is a key determinant of animal welfare, and rising global temperatures increase the risk of heat stress. While thermal tolerance is critical for predicting wildlife vulnerability to climate change, physiological responses in South American wild rodents remain understudied. Here, we assessed heat stress responses in the plains vizcacha (Lagostomus maximus), a semi-fossorial rodent with key ecological functions, to establish upper critical Temperature-Humidity Index (THI) thresholds. Within an observed THI range of 18.5 to 30.7, non-invasive longitudinal monitoring and

Icy Hot: A Case of Unexpected Heat Illness.

Finish-line collapse is a unique phenomenon among athletes and includes a broad working differential. We explore the case of a young, healthy runner who collapsed at the end of a half marathon in the fall from exertional heat illness. This illness covers a wide range of severity from the mild symptoms of heat cramps to heat exhaustion and the extreme of life-threatening heat stroke. Development of these conditions is often associated with patient-specific acquired or congenital vulnerability factors such as extremes of age, fitness, obesity, medication use (eg, antipsychotics, anticholinergics

DNA Repair Exhaustion Activates Orthocaspase-Mediated Cell Death and Promote Inclusive Fitness of Cyanobacterial Population Under Abiotic Stress.

Orthocaspases execute regulated cell death (RCD) in cyanobacteria, yet their post-stress activation and physiological relevance remain elusive. Here, we investigated this using heterologous expression of orthocaspases in Synechococcus elongatus PCC 7942. Orthocaspase-expressing (OE) strains, SynOC2_GFP and SynOC6_GFP, were generated using GFP-fused orthocaspases, AnaOC2 and AnaOC6. Catalytically inactive C/A variants were used to generate mutant-expressing (ME) controls, SynC140A_GFP and SynC134A_GFP. Under non-stress conditions, OE and ME strains accumulated orthocaspases without losing viabi
