Single-cell atlas profiling revealed cellular characteristics and dynamic changes after PD-1 blockade therapy of brain metastases from laryngeal squamous cell carcinoma.

Brain metastasis (BM) in laryngeal squamous cell carcinoma (LSCC) is uncommon but prognosis is poor. Anti-PD-1 immunotherapy benefits some advanced LSCC cases, yet its efficiency is limited by tumor complexity. We analyzed paired metastatic tumor samples from before and after immunotherapy using single-cell RNA sequencing (scRNA-seq), along with a primary LSCC dataset and bulk RNA sequencing. This identified changes post-immunotherapy and revealed differences in single-cell transcriptomes among LSCC, primBM, and neoBM. Our findings show that anti-PD-1 treatment suppresses metastasis-promoting

Case discussion: The effect of extreme temperatures on an older adult.

Climate change can cause high temperatures that can affect the older adult in significant ways. Older adults may not be aware of the dangers of high temperature days and may continue with old habits such as staying in the sun to garden without sunscreen or a hat as they may have done in years past. High temperatures can cause impairment of the tone and structure of blood vessels by interfering with nitric oxide synthesis and cytokine production and can cause systemic inflammation, all of which significantly contribute to dehydration in older adults, who are known to have a decreased sense of t

Endosome-microautophagy targeting chimera (eMIATAC) for targeted proteins degradation and enhance CAR-T cell anti-tumor therapy.

Rationale: Since oncogene expression products often exhibit upregulation or abnormally activated activity, developing a technique to regulate abnormal protein levels represent a viable approach for treating tumors and protein abnormality-related diseases. Methods: We first screened out eMIATAC components with high targeted degradation efficiency and explored the mechanism by which eMIATAC induced target protein degradation, and verified the degradation efficiency of the target protein by protein imprinting and flow cytometry. Next, we recombined eMIATAC with some controllable elements to verif

Green whitening of polyester fabric using fluorescent brightener OB-1 in a mixture of water and decamethylcyclopentasiloxane media.

Fluorescent brightening agent OB-1 (OB-1) is often used in plastic goods because of its non-toxic nature, chemical stability, remarkable heat resistance, and light stability. Raw OB-1 is challenging to use in textiles using the exhaustion method. This study used a novel method using raw OB-1 powder to whiten polyester fabric in water and decamethylcyclopentasiloxane (D5). The Taguchi approach investigated the interaction between whitening process parameters such as temperature, OB-1 mass, water: D5 ratio, and treatment time with four levels. The study shows that the temperature and water: D5 r

Heat stress: a major threat to ruminant reproduction and mitigating strategies.

Stress is an external event or condition that puts pressure on a biological system. Heat stress is defined as the combination of internal and external factors acting on an animal to cause an increase in body temperature and elicit a physiological response. Heat stress is a set of conditions caused by overexposure to or overexertion at excess ambient temperature and leads to the inability of animals to dissipate enough heat to sustain homeostasis. Heat exhaustion, heat stroke, and cramps are among the symptoms. For the majority of mammalian species, including ruminants, heat stress has a negati

Changes in lactate concentration are accompanied by opposite changes in the pattern of fat oxidation: Dose-response relationship.

It is unknown whether changes in lactate concentration produced by different situations (e.g., glycogen depletion or heat) modify fat oxidation. If confirmed, we could determine a dose-response relationship between lactate and fat. The aim of this study was to determine whether changes in lactate concentration (due to glycogen depletion or heat) alter fat oxidation during exercise. 11 males and eight females performed an incremental exercise test under three situations: control, glycogen depletion, and heat. At rest, in the last minute of each step and immediately post-exhaustion, lactate was

The repeatability of a cycling exercise-heat stress test in a male population.

INTRODUCTION: There is considerable inter-individual variability in the physiological responses to environmental stressors and so to accurately assess and monitor changes in an individual's ability to cope with exercise-heat stress, a reliable protocol is required. The aim of this study was to examine the repeatability of a 90-min steady-state heat exercise bout with physiological and subjective variables, and performance during an incremental test to exhaustion post 90-min steady-state exercise. METHOD: Sixteen mixed ability males (Age: 39 ± 15yrs; Height: 176.5 ± 4.8 cm; BM: 79.7 ± 10.3 kg;

Optimizing a mouse model of exertional heat stroke to simulate multiorgan and brain injuries.

BACKGROUND: Exertional heat stroke (EHS) is a clinical entity characterized by abnormalities of the central nervous system (CNS) and is associated with multiple organ injury, some of which may be irreversible. It is valuable to establish an optimized model of EHS that is able to induce and assess damage to the CNS and multiple organs. METHODS: We induced EHS by using an environmental chamber with adjustable temperature and humidity and a mice forced running wheel. The endpoint for the EHS was defined as either exhaustion or a core temperature of 42.5°C being reached. Injury to the liver, kidn

Exertional Heatstroke Encephalopathy With Chronic Neurological Deficit.

Heat exhaustion progresses to heat stroke and then on to heatstroke encephalopathy, a serious illness. Extreme hyperthermia (over 40.5 °C), central nervous system failure, multiorgan dysfunction, and hypovolemic shock are the hallmarks of the clinical presentation of heatstroke. A 27-year-old male was presented to the causality with loss of consciousness followed by altered sensorium while working outdoors (outside temperature was 40°C), low blood pressure (70/50 mmHg), hyperthermia (41°C), tachycardia, focal seizures (1 episode), hypovolemic shock. At presentation, the Glasgow Coma Scale was

Time-course for onset and decay of physiological adaptations in endurance trained athletes undertaking prolonged heat acclimation training.

Short-term heat acclimation (HA) appears adequate for maximizing sudomotor adaptations and enhancing thermal resilience in trained athletes. However, for enhanced erythropoiesis and transfer effects to exercise capacity in cooler environments, prolonged HA appears necessary. To establish the time-course for physiological adaptations and performance effects, 20 male elite cyclists were divided into an intervention group (HEAT; n = 10) completing 5 weeks of HA (six one-hour HA-training sessions per week) and control (n = 10) tested pre and post in hot (40°C) and cool conditions (20°C). HEAT comp

Elevated core temperature in addition to mental fatigue impairs aerobic exercise capacity in highly trained athletes in the heat.

The purpose of this study was to investigate the effects of elevated core temperature by exposure to heat stress vs. heat exposure without elevated core temperature (mean skin temperature only) in addition to mental fatigue on aerobic exercise capacity in the heat. Seven highly trained athletes completed two experimental conditions: elevation in core and skin temperatures (hyperthermia: HYP), and skin temperatures (SKIN). Participants performed the AX-Continuous Performance Task and Stroop Task to induce mental fatigue during a warm water immersion at 40 °C (HYP) and a passive seated heat expo

Theoretical Design of Smart Bionic Skins with Self-Adaptive Temperature Regulation.

Thermal management presents a significant challenge in electric design, particularly in densely packed electronic systems. This study proposes a theoretical model for radiative bionic skin that emulates human skin, enabling the self-adaptive modulation of the thermal exhaustion rate to maintain homeostasis for objects covered by the skin in fluctuating thermal environments. The proposed artificial skin consists of phase change material (VO2) nanoparticles embedded in a low-loss matrix situated on a metallic substrate with a minimal thickness of several micrometers. The findings from our theore

Glutamine Supplementation: A Possible Strategy to Help Mitigate Health Risks of Heat-Related Illness.

A rise in body temperature caused by physical work, including exercise, in a hot climate can lead to heat-related illnesses such as exertional heat exhaustion and stroke. Individuals who work physically demanding occupations in hot environments are at heightened risk of heat injury. The mechanisms that contribute to heat illness resulting from physical work in the heat are complex and include dehydration, tissue ischemia and damage, oxidative stress, and inflammatory events. Therefore, it is important to develop strategies that address these mechanistic underpinnings to prevent exacerbation to

Effect of Ischemic Preconditioning on Endurance Running Performance in the Heat.

Ischemic preconditioning (IPC) is a strategy that may enhances endurance performance in thermoneutral environments. Exercising in the heat increases thermoregulatory and cardiovascular strain, decreasing endurance performance. The current study aimed to determine whether IPC administration improves endurance performance in the heat. In a randomized crossover design, 12 healthy subjects (V̇O2max: 54.4 ± 8.1 mL·kg-1·min-1) underwent either IPC administration (220 mmHg) or a sham treatment (20 mmHg), then completed a moderate-intensity 6-min running (EX1) and a high-intensity time-to-exhaustion r

Effects of Precooling on Endurance Exercise Performance in the Heat: A Systematic Review and Meta-Analysis of Randomized Controlled Trials.

An increasing number of studies have explored the effects of precooling on endurance exercise performance in the heat, yet the available results remain inconsistent. Therefore, this study aimed to investigate the effects of different precooling strategies on endurance exercise performance in the heat. A comprehensive search was conducted across PubMed, Web of Science, Cochrane, Scopus, and EBSCO database. The Cochrane risk assessment tool was employed to evaluate the methodological quality of the included studies. A meta-analysis was subsequently conducted to quantify the standardized mean dif

Cocktail strategy-based nanomedicine: A synergistic cascade of starvation, NIR-II photothermal, and gas therapy for enhanced tumor immunotherapy.

Immunotherapy has emerged as a highly promising strategy in the realm of cancer treatment, wherein immunogenic cell death (ICD) is considered a potential trigger for anti-tumor immunity by inducing adaptive immunity to dying cell antigens. This process is often accompanied by the exposure, active secretion, or passive release of a large number of damage-associated molecular patterns (DAMPs), which activate dendritic cells (DCs) and enhance their antigen-presenting capacity. Subsequently, it promotes the recruitment and activation of cytotoxic T lymphocytes, ultimately leading to tumor growth i

Self-reported exertional heat illness and risk factors among collegiate marching band artists.

Marching band (MB) artists are often part of the general student population and not required to complete a pre-participation health screening to identify predisposing medical conditions or risks for injury/illness. Anecdotally, exertional heat illnesses (EHI) are a concern for MB artists. As more athletic trainers provide MB healthcare, research is needed on EHI occurrence and MB associated EHI risk factors. We utilized an exploratory cross-sectional study design to determine EHI risk factors, including previous EHI occurrence, among collegiate MB artists. MB artists (n = 1 207; age = [19.

2024 Scholars' Research Symposium Abstract: Kindling Wellness: Creating a Student-Run Podcast Series on Medical Student Wellness.

INTRODUCTION: Research has consistently shown that the prevalence of burnout symptoms (such as emotional and physical exhaustion, cynicism, or lack of interest in schoolwork, the sense of incompetence, or the feeling that you cannot be effective) in medical students is greater than the prevalence in the general population. Students with preexisting anxiety, depression, mood disorder or other psychological distress are more vulnerable to burnout. It is estimated that at least half of U.S. medical students experience or are affected by burnout during their education. With the high rates of burno
