Pathophysiology : the official journal of the International Society for Pathophysiology2026eng
BACKGROUND: Hypoxic-ischaemic brain injury (HI) is a major contributor to neurological deficits following stroke. Understanding what happens to the smallest functional and structural unit of the central nervous system in the face of oxygen and nutrient deprivation is essential to fully comprehend the pathogenesis of diseases and disorders associated with HI, such as ischaemic stroke.
AIM: The aim of this study was to develop a robust in vitro tool for initial screening of potential therapeutics and identification of diagnostic markers of brain hypoxic injury.
METHODS: This study details and

AIMS: Exertional heatstroke (EHS) can induce marked scrotal hyperthermia and impair male reproductive integrity. This study tested whether sustained-release menthol protects against EHS-induced testicular and genitourinary injury via transient receptor potential melastatin 8 (TRPM8) activation.
MATERIALS AND METHODS: Adult male Wistar rats received sustained-release menthol gel (30 mg·kg-1, subcutaneously) or vehicle for 2 weeks before EHS induction (36 °C, 50% relative humidity; approximately 52 min of treadmill running). Some rats received AMTB, a selective TRPM8 antagonist. Scrotal tempera

Rationale: Record-breaking heatwaves caused by greenhouse effects lead to multiple hyperthermia disorders, the most serious of which is exertional heat stroke (EHS) with the mortality reaching 60 %. Repeat exercise with heat exposure, termed heat acclimation (HA), protects against EHS by fine-tuning feedback control of body temperature (Tb), the mechanism of which is opaque. This study aimed to explore the molecular and neural circuit mechanisms of the HA training against EHS. Methods: Male C57BL/6 mice (6-8 weeks) and male TRPV1-Cre mice (6-8 weeks) were used in our experiments. The EHS model

Nitric oxide synthase (NOS) is an important mediator of cutaneous vasodilation during exercise-heat stress. We recently reported that pharmacological activation of transient receptor potential ankyrin 1 (TRPA1) channel mediates cutaneous vasodilation via NOS-dependent mechanisms under nonheat stress-resting conditions. Here, we hypothesized that TRPA1 channel activation would contribute to cutaneous vasodilation during exercise in the heat via NOS-dependent mechanisms. To assess this response, we first conducted TRPA1 channel antagonist verification substudy (10 young adults and 5 women) where

Extreme heat and traffic-related air pollution (TRAP) have been linked to worsening chronic health disorders, however, their combined effects on diabetic nephropathy (DN) are little understood. Type II diabetic mice were exposed to heat (40 °C) and NO2 (5 ppm) separately for 4 h per day over 6 weeks to investigate the synergistic effects on the progression of DN. We found that exposure to high temperature and NO2 elevated blood glucose levels and exacerbated histopathological changes. Additionally, there were increased oxidation indicators (ROS, MDA, 8-OHdG) and decreased antioxidant indicator

Hyperthermia is a promising anticancer treatment that induces heat stress, stimulating various signal transduction pathways to maintain cellular homeostasis. Mitogen-activated protein kinases (MAPKs) link various extracellular stimuli with cytoplasmic and nuclear mediators through a three-tiered cascade of kinases, including MAPKs, MAP2Ks, and MAP3Ks. In mammals, three major groups of MAPKs have been characterized: extracellular signal-regulated protein kinases (ERK), p38 MAPKs, and c-Jun NH2-terminal kinases (JNK). Each group of MAPKs is heat-activated and exhibits distinct biological functio

Most of the deep-sea pressure-resistant structures are made of high-strength materials. The pressure-resistant structures are made with large-thickness welding technology, which can easily result in stress concentration and cracks. In this paper, a method for crack identification and life evaluation of pressure-resistant structures based on the crack is proposed. On the one hand, the principle of eddy current thermal imaging detection is used to identify cracks. The experimental platform is built to detect cracks in pressure-resistant structures. The titanium alloy samples with micro-cracks ar

BACKGROUND: Maternal hyperthermia, an increasingly prevalent environmental stressor linked to global climate change, is epidemiologically associated with a higher risk of neurodevelopmental disorders, including autism and schizophrenia. However, the molecular mechanisms underlying this neurotoxicity, particularly those leading to long-term neurological deficits, remain poorly understood. This study tested the novel hypothesis that hyperthermia acts not only as an acute physiological stressor but also as a potent genotoxic agent, inducing persistent genomic alterations in neural progenitor cell

Brown adipose tissue (BAT) has shifted from being considered a transient thermogenic organ of infancy to a metabolically dynamic and multifunctional tissue throughout life. Histologically and developmentally distinct from white and beige adipocytes, BAT originates from a myogenic lineage and is characterised by a high mitochondrial density, multilocular lipid droplets, and abundant sympathetic innervation. Its defining function, non-shivering thermogenesis, is mediated by uncoupling protein 1 (UCP1) and complemented by alternative mechanisms such as futile creatine and calcium cycling. Beyond

Ryanodine receptor 1 (RYR1) is the sarcoplasmic reticulum (SR) Ca2+ release channel required for both skeletal muscle contraction and Ca2+ leak. Mutations in RYR1 cause malignant hyperthermia susceptibility (MHS) and enhanced sensitivity to heat stroke (ESHS), which can result in death due to excessive skeletal muscle thermogenesis upon exposure to volatile anesthetics or heat. Here, we investigated the molecular and physiological functions of phosphorylation of RYR1 at Ser2902 by the kinase striated muscle preferentially expressed protein (SPEG). Muscle from SPEG-deficient mice expressing RYR

BACKGROUND: As a result of climate change, dairy cows even in confinement systems are exposed to high ambient temperatures and environmental factors inducing heat stress. However, there are indications that chronic heat stress with reduced feed intake initially stimulates a systemic inflammatory response and progressively reduces the immunocompetence. This finally increases the disease susceptibility. This study aimed to elucidate the effects of chronic heat stress or equivalent reduced feed intake via pair-feeding at thermoneutrality on the daily hematological profile, leukocyte NF-κB p65 sig

In cold regions, asphalt pavements undergo severe deterioration driven by the synergistic effects of thermal contraction and dynamic heavy loading, resulting in premature failures such as thermal cracking and rutting. This study presents a comprehensive thermo-mechanical coupling analysis to address this critical issue. A sophisticated seven-degree-of-freedom (7-DOF) vehicle model was developed to simulate realistic dynamic loads, with load magnitudes quantified via MATLAB simulations. A three-dimensional (3D) finite element (FE) model was established, incorporating temperature-dependent mater

TRPV1, a member of the transient receptor potential vanilloid subfamily, mediates nociception and thermoregulation. TRPV1-targeting analgesics frequently induce hyperthermia, underscoring the need for structural insights to guide the development of safer compounds. Here, we determined the structures of rat TRPV1 bound to the clinical candidate analgesics AMG517, AMG9810, and SB366791. AMG517 and AMG9810 are deeply situated within the S3-S4 interface of the vanilloid pocket, where they interact with residues from the S3-S6 helices, as well as the S4-S5 linker. These interactions induce local de

Consuming cold fluids during heat stress can alter sweating responses; however, it remains unclear why chemical activation of cold-sensitive transient receptor potential melastatin 8 (TRPM8) with menthol only induces a cold sensation. Animal models propose that complete cold transduction with TRPM8 chemical stimulation must be combined with physical cooling to impact thermoeffector responses. To address this, 12 participants (5 females and 7 males; 22 ± 5 yr; 79.5 ± 15.0 kg; 1.8 ± 0.1 m) conducted four passive heating protocols in random order where, within trial, they consumed 3.2 mL/kg body

Noise-induced hearing loss (NIHL) is a major public health problem caused by damage to cochlear hair cells, synapses, and spiral ganglion neurons (SGNs). Since effective treatments are lacking, we investigated cellular stress responses induced by moderate and loud noise in a mouse model of cochlear synaptopathy. RNA sequencing and spatial transcriptomics revealed that noise exposure elicited a robust but transient upregulation of endoplasmic reticulum chaperones and proteasome subunits in SGNs and their supporting cells. To target this response, we administered TRC051384, a small-molecule acti

In order to complete their energetically demanding journeys, migratory birds undergo a suite of physiological changes to prepare for long-duration endurance flight, including hyperphagia, fat deposition, reliance on fat as a fuel source, and flight muscle hypertrophy. In mammalian muscle, SLN is a small regulatory protein which binds to sarco/endoplasmic reticulum Ca2+-ATPase (SERCA) and uncouples Ca2+ transport from ATP hydrolysis, increasing energy consumption, heat production, and cytosolic Ca2+ transients that signal for mitochondrial biogenesis, fatigue resistance and a shift to fatty aci

Gas turbine engines subject materials to extreme conditions. Their high temperature materials and co-developed coatings must survive combustion gas temperatures currently approaching 1800 °C, large thermal gradients, severe thermal shock, and static and fatigue inducing applied stresses, all the while operating in highly reactive, high-pressure, high-speed combustion gas flows containing significant partial pressures of water vapor, oxygen, and other reactive species for many tens of thousands of hours. We describe the design and development of a test facility for the study of materials under

Cannabidivarin (CBDV) and cannabigerol (CBG) are minor phytocannabinoids from Cannabis sativa, whose health benefits have been reported. However, studies about the impact of these cannabinoids on fundamental cellular processes in placentation are scarce. Placental development involves physiological endoplasmic reticulum (ER) stress, however when exacerbated it can lead to altered angiogenesis and pregnancy disorders, such as intrauterine growth restriction and preeclampsia. In this work, the effects of CBDV and CBG (1-10 µM) on placental extravillous trophoblasts were studied, using the in vit
