Immunological dynamics in orthotopic compared with subcutaneous murine models of HPV-positive oropharyngeal cancer.

The necessity of reliable preclinical models for evaluating the efficacy of novel therapeutic strategies is imperative. Nevertheless, the degree to which tumor-bearing murine models represent the immunological characteristics of human papillomavirus (HPV)-positive oropharyngeal squamous cell carcinoma (OPSCC) has largely been unexplored. By utilizing single-cell RNA sequencing technology, our research elucidated that subcutaneous (SC) murine models more accurately reflect the early immunogenic phase of human HPV-positive OPSCC, marked by a stage-dependent increase in effector T-cell infiltrati

Cellular schwannoma of the base of tongue: A case report.

Schwannomas are solid, well-encapsulated masses that develop eccentrically toward the nerve from that they originate. They typically affect the head and neck and involve Schwann cells surrounding autonomic, peripheral, and cranial nerves. Intraoral schwannoma is most affected by the tongue, with 54% male preponderance. The most common neural tumors in the oral cavity include neurofibroma, schwannoma, and malignant peripheral nerve sheath tumor (MPNST). CD56 is more specific in schwannomas than neurofibromas, with CD56 expressed strongly in 90% of schwannomas and negative in 86% of neurofibroma

Extraskeletal chondroma of the tongue in a two-year-old quarter horse colt: a case report.

Extraskeletal chondromas (ESCs) are rare benign tumours composed of mature cartilage that typically develop in soft tissues. Their occurrence in horses is extremely uncommon and, to the authors' knowledge, no previous cases of lingual ESCs in equines have been reported. This report describes a two-year-old Quarter Horse colt presented with progressive dysphagia. Clinical examination revealed a solitary, multilobulated, firm-elastic mass located at the dorsal aboral portion (base) of the tongue. Diagnostic imaging ruled out bone involvement. The mass was surgically excised under general anaesth

Corticosteroid injection for pyogenic granuloma of the tongue in a rare resembling ulcerative malignancy: a case report.

BACKGROUND: Pyogenic granuloma (PG) is a benign vasoproliferative lesion that usually occurs in the skin And mucosa. The clinical manifestations of PG are rapid growth, pedicled or broad base of a single red smooth papule, easily bleeding with minor lesions. Most of the oral lesions occur at the gingiva, And the extra-gingival lesions is less common, and the ulcerative type is even rarer. We present a case of ulcerative lesion located at the lateral margin of the tongue with a peripheral bulge for more than 5 years. It was initially suspected to be tongue cancer and was confirmed to be a PG by

Cutaneous basaloid squamous cell carcinoma in a captive blue-fronted amazon (Amazona aestiva) in Paraguay: gross description, histological and immunohistochemical findings.

Squamous cell carcinoma (SCC) is a malignant neoplasm that usually arises from the integument, and is reported uncommonly in pet birds. Basaloid SCC is a variant from the prototypic squamous cell carcinoma and is a biologically high grade tumor that shows a sharp predilection for the base of the tongue, the hypopharynx and the supraglottic larynx, manifesting deep and lateral invasion. We present the case of an Amazon aestiva from Paraguay with a basaloid SCC in the skin, of 8 months of evolution. Histologically, the findings were compatible with poorly differentiated carcinoma, and immunohist

Plumbagin remodels the tumor immune microenvironment to overcome cisplatin resistance in tongue squamous cell carcinoma via KLF2 activation and ZWINT/NF-κB inhibition.

Tongue squamous cell carcinoma (TSCC) is a therapeutic challenge due to frequent cisplatin resistance and an immunosuppressive tumor immune microenvironment (TIME). Effective strategies must address both barriers. We evaluated the antitumor effects of plumbagin (PLB) in cisplatin-sensitive and -resistant TSCC cells, as well as in xenograft, patient-derived xenograft, and humanized mouse models. PLB downregulated ZWINT and inhibited NF-κB signaling, thereby restoring cisplatin sensitivity in resistant cells. In vivo, PLB significantly suppressed tumor growth and enhanced cisplatin efficacy. Patterns of Cervical Metastases and Implications for Neck Management in Minor Salivary Gland Cancer Treatment.

OBJECTIVES: Minor salivary gland cancer (MiSGC) is a rare and heterogeneous disease, with different degrees of tumor aggressiveness. The scarce data in the literature show that the presence of lymph node metastases (LNMs) negatively impacts the prognosis. Furthermore, there is a lack of information about the patterns and extension of LNMs in MiSGC. Therefore, it is necessary to identify the clinical and pathological features associated with LNMs in MiSGC to guide the cervical dissection intent of those patients. This study's endpoints were the identification of clinicopathological features ass

Fenestrating Adenocarcinoma: A Novel Salivary Gland Cancer With EWSR1::BEND2 Fusion and Predilection for the Pharynx.

Most salivary gland tumors are found to have genetic fusions driving their oncogenesis, with an explosion of such findings in recent years. They typically represent previously identified and well-characterized tumors and the fusions are generally not necessary for diagnosis but rather serve as diagnostic arbitrators in challenging cases. That said, occasional unexpected molecular findings are broadening our understanding of these common tumors and have led to the emergence of new entities, some of which were "hiding in plain sight" and are quite common, like secretory carcinoma. In contrast, o

Very Rapid Eradication of a Large Squamous-Cell Carcinoma of the Head and Neck Treated With First-line Combination Chemotherapy, a Low-methionine Diet, and Oral Recombinant Methioninase.

BACKGROUND/AIM: Treatment options for squamous-cell carcinoma of the head and neck (HNSCC) include chemotherapy, surgery, radiation, and their combinations. However, these treatments have a risk of severe side-effects, including disfigurement and functional impairment. Therefore, treatment of locally-advanced or metastatic HNSCC is often challenging, and prognosis remains poor. The present case report describes the rapid eradication of a large neck HNSCC treated with first-line combination chemotherapy, a low-methionine diet, and oral recombinant methioninase (o-rMETase). Application of machine learning to create a prediction model for the need of aggressive nutritional intervention during head & neck cancer radiotherapy.

BACKGROUND: Curative-intent radiotherapy (RT) or chemoradiotherapy (CRT) for head and neck squamous cell carcinoma (HNSCC) frequently leads to mucositis, dysphagia, and odynophagia, resulting in nutritional compromise, weight loss, and the need for feeding tube (FT) insertion. This study aimed to develop and internally validate an artificial neural network (ANN)-based machine learning model to predict patients at risk of requiring reactive FT insertion, >10 % weight loss, or grade ≥3 dysphagia during or after treatment. METHODS: This prospective, single-institutional observational study inclu

Thyroid cancer with internal jugular vein tumor embolism and skull base invasion causing polyneuropathy.

We report a rare case of thyroid cancer with tumor embolism of the internal jugular vein (IJV) that progressed cranially into the skull base and caused multiple cranial neuropathies. An 80-year-old woman presented with a progressively enlarging right cervical mass, which had been neglected for 10 years. Ultrasonography and magnetic resonance imaging revealed a cystic cervical lesion with a tumor embolism within the IJV extending to the jugular foramen and hypoglossal and facial canals. Fine-needle aspiration cytology yielded inconclusive results on two occasions, likely because of cystic degen
