Source of the nonlinear dependence of bacteriophage SP82 transfection on deoxyribonucleic acid concentration.

Extracts of competent cells of Bacillus subtilis exhibited nuclease activity on radioactively labeled cell deoxyribonucleic acid (DNA). The activity was not decreased when Mg was omitted from the reaction mixture but was decreased to zero by addition of ethylenediaminetetraacetic acid. Of the other metals tested, only Ca increased nuclease activity more than Mg. Addition of 1.5 x 10(-3)m Ca to transfection mixtures increased the nonlinearity of the relationship between number of transfectants and DNA concentration. The inferred role of the Ca-activated nuclease was checked by showing that the

-aminobutyric acid as a required germinant for mutant spores of Bacillus megaterium.

Germinated spores of Bacillus megaterium QM B1551 were irradiated with ultraviolet light, and spore-forming survivors were screened for germination requirements. Spore strains which failed to germinate in a variety of defined solutions germinative for spores of the parent strain were obtained. Mutant spores germinated readily in solutions containing yeast extract or one of numerous complex preparations. gamma-Aminobutyric acid, obtained from yeast extract by column chromatography, was shown to be required for germination by the mutant spores. gamma-Aminobutyric acid and l-alanine at final conc

Urea-mercaptoethanol-soluble protein from spores of Bacillus thuringiensis and other species.

Treatment with urea-mercaptoethanol of purified spores of Bacillus thuringiensis, other Bacillus species, and Clostridium roseum solubilizes a protein fraction between 5 and 12% of the dry weight of the spores. This fraction behaves identically to the crystal protein of B. thuringiensis on acrylamide-gel electrophoresis. The protein from all of the Bacillus species shows partial homology with crystal protein, using the Ouchterlony immunodiffusion technique. A further fraction, similar in amount, can be removed from spores of B. thuringiensis by the addition of sodium lauryl sulfate to the urea

An electrokinetic model of transduction in the semicircular canal.

Transduction in the semicircular canal was studied by focusing an infrared beam on either side of exposed ampullae from the posterior canals of Rana pipiens. The direction of fluid movement resulting from a stimulus was inferred by observing the polarity of the change in afferent impulse mean rate relative to the spontaneous value. On the basis of the accepted functional polarization of this receptor, the results indicate that fluid moved toward the warmer side of the ampulla. Convection and thermal reception were shown to be unlikely explanations for these results. Morover, cupular displaceme

The separation of beta-glucanases produced by Cytophaga johnsonii and their role in the lysis of yeast cell walls.

1. When Cytophaga johnsonii was grown in the presence of suitable inducers the culture fluid was capable of lysing thiol-treated yeast cell walls in vitro. 2. Autoclaved or alkali-extracted cells, isolated cell walls and glucan preparations made from them were effective inducers, but living yeast cells or cells killed by minimal heat treatment were not. 3. Chromatographic fractionation of lytic culture fluids showed the presence of two types of endo-beta-(1-->3)-glucanase and several beta-(1-->6)-glucanases; the latter may be induced separately by growing the myxo-bacterium in the presence of

Bacterial attachment to a specific wound site as an essential stage in tumor initiation by Agrobacterium tumefaciens.

The number of tumors initiated by Agrobacterium tumefaciens strain B6 on primary pinto bean leaves was decreased when cells of an avirulent strain (IIBNV6) were included in the inoculum. With sufficient B6 cells to initiate ca. 50% of the maximal number of tumors per leaf, inhibition was detected at a 1:1 ratio of B6 to IIBNV6 cells and increased linearly with the logarithm of the number of IIBNV6. Varying the number of B6 in the presence of a constant number of IIBNV6 or varying the number of both, while maintaining a constant ratio of B6 to IIBNV6, showed that the inhibition was a function o

The action of light on culture media.

Failure of growth of staphylococci on nutrient agar plates was shown to be due to previous exposure to sunlight in an exceptionally well lit laboratory. Such plates inhibited the growth of Staph. aureus, Staph. albus, some Proteus species and Providence, but not that of various other bacteria. The affected species are found to be more susceptible to the action of hydrogen eroxide added to the medium. The effect was produced in plates of five different nutrient agars, but not in two others which contained casein. Plates awaiting inoculation should be protected from light; a white fabric blind w

Adeno-associated satellite virus interference with the replication of its helper adenovirus.

Adeno-associated satellite virus type 4 interferes with the replication of its helper adenovirus. No interferon-like soluble substance could be detected in satellite-infected cultures and other DNA- and RNA-containing viruses were not inhibited by coinfection with satellite virus under conditions which reduced adenovirus yields by more than 90% in monkey cells. Altering the concentration of adenovirus in the presence of constant amounts of satellite resulted in a constant degree of interference over a wide range of adenovirus inocula and suggested that adenovirus concentration was not a signif

Conductance changes produced by light in rod outer segments.

1. Changes in the admittance of rod outer segments produced by illumination with brief flashes were studied by two methods: one, in which maintained changes in real and imaginary parts of admittance were observed in the frequency range 15 c/s-60 kc/s; the other, in which the time course of change in absolute value of admittance (Delta|Y|) was observed at frequencies of 100 kc/s-1.0 Mc/s.2. The response to light absorbed by rhodopsin was resolved into components. One of these components was a transient increase in conductance which arose from a rapid degradation into heat of the light energy. A

Stellar and 0ther high-temperature molecules.

Optical spectroscopy of stellar molecules trapped at 4 degrees K is particularly valuable when the data can be used to complement the corresponding gas data. The ground state is then directly established by measurement of the absorption spectrum at the low temperature, since all transitions beginning from excited states are eliminated. Isotopic substitution establishes the (0,0) bands of transitions to excited electronic states, and when these data are combined with infrared and fluorescence measurements at 4 degrees K, most of the vibrational properties of the ground and excited states can be

Pesticins. 3. Expression of coagulase and mechanism of fibrinolysis.

Mutational loss of pesticin I, a bacteriocin-like substance produced by Pasteurella pestis, is known to result in concomitant loss of a coagulase and fibrinolytic factor. No relationship was detected between pesticinogeny and other tested properties either associated with virulence or peculiar to P. pestis. Pesticin I was distinguished from the coagulase and fibrinolytic activities on the basis of anatomical distribution, behavior during gel filtration, and sensitivity to heat. Coagulase and the fibrinolytic factor were not differentiated by these criteria. Spontaneous suppressor mutations cau

Behavior of carbon dioxide and other volatiles on Mars.

We have found that a rather simple thermal model of the Martian surface, in combination with current observations of the atmospheric composition, points strongly toward the conclusion that the polar caps of Mars consist almost entirely of frozen CO(2). This study was based upon the following principal assumptions. 1) Carbon dioxide is a major constituent of the Martian atmosphere. 2) The blanketing effect of the atmosphere is small, and due principally to the absorption band of CO(2) near 15 microns. 3) Lateral and convective heat transfer by the atmosphere is negligible. 4) The far-infrared e

Brenner, Don J. (University of Washington, Seattle), and Neal B. Groman. Inhibition of coliphage reproduction after superinfection of induced lysogens. J. Bacteriol. 92:1727-1734. 1966.-Purified preparations of phages lambda and lambda112 inhibited lysis, phage reproduction, and endolysin synthesis by ultraviolet-induced strains of K-12 (lambda) and K-12 (lambda112). Structural and partial functional integrity of phage was required for inhibition, and the role of phage deoxyribonucleic acid (DNA) in inhibition was demonstrated. Characterization of a factor formed in the course of adenovirus infection of tissue cultures causing detachment of cells from glass.

Infectious tissue culture fluids of the majority of serotypes of adenovirus at low dilutions detach HeLa or KB cells from glass surfaces within a few hours after inoculation. A reproducible method for testing cell detachment was devised. The factor present in infectious tissue culture fluids and responsible for cell detachment is trypsin-sensitive and non-dialyzable; it is smaller and more resistant to the effect of heat or ultraviolet light than the infectious virus particle. Cell detachment activity was found to be temperature-dependent, and the cell-detaching titer of infectious tissue cult

Inactivation and reactivation of B. megatherium phage.

Preparation of Reversibly Inactivated (R.I.) Phage.- If B. megatherium phage (of any type, or in any stage of purification) is suspended in dilute salt solutions at pH 5-6, it is completely inactivated; i.e., it does not form plaques, or give rise to more phage when mixed with a sensitive organism (Northrop, 1954). The inactivation occurs when the phage is added to the dilute salt solution. If a suspension of the inactive phage in pH 7 peptone is titrated to pH 5 and allowed to stand, the activity gradually returns. The inactivation is therefore reversible. Properties of R.I. Phage.- The R.I.

The Complex Behaviour of High-Frequency Currents in Simple Circuits.

The fact that standing wave phenomena exist along transmission lines and loops conducting high-frequency electrical energy is responsible for effects of which therapeutic use can be made.A. Power measurements are made possible because parallel transmission lines behave as power transformers of which the ratio varies with the length of these lines. In a generator designed by the G.E.C. the dimensions of the lines are such that after a preliminary estimation of the impedance of the load in the treatment field, the sensitivity of the meter can be adjusted so that the meter subsequently registers
