Integrating dynamic modeling and continuous cultivation to enhance recombinant β-glucosidase production in Komagataella phaffii.

Cultivation under hypothermia has emerged as an interesting strategy for enhancing recombinant protein production in Komagataella phaffii, yet predictive approaches capable of integrating the effects of temperature and cell growth rate for rational bioprocess optimization remain limited. In this work, we investigated the influence of cultivation temperature on recombinant β-glucosidase (BGL1) production in the hybrid K. phaffii strain S467 by combining bioprocess characterization with dynamic mathematical modeling. Batch cultures performed at 20, 25 and 30 °C showed that hypothermia increased

Optimizing Enteral Nutrition During and After Therapeutic Hypothermia: An Evidence-Based Practice Initiative.

BACKGROUND: Feeding practices during therapeutic hypothermia (TH) vary widely across neonatal intensive care units, and many centers delay enteral nutrition because of concerns about gastrointestinal intolerance and necrotizing enterocolitis. Current evidence suggests that early, clinically guided enteral feeding may be safe and beneficial for neonates with hypoxic-ischemic encephalopathy. An evidence-based feeding guideline was implemented to promote enteral nutrition during and after TH. PURPOSE: To evaluate whether initiating minimal enteral feeding in clinically stable neonates undergoing

Implanted and degradable hydrogel evaporative cooler for regional and repeated mild hypothermia therapy of traumatic brain injury.

Implantable, regional and repeated mild hypothermia represents a promising therapeutic strategy for traumatic brain injury. Here, we develop a soft, degradable hydrogel microchannel evaporative cooler capable of removing heat through water evaporation in microchannels and achieving focused regional cooling within a short time. Fabricated from degradable hydrogels and using water as the coolant, the cooler shows biocompatibility and tissue adhesion. In vivo intracranial experiments with the cooler adhered on the rat dura mater enabled precise temperature regulation during repeated regional hypo

Myxoedema Coma With Type 2 Respiratory Failure and Hypothermia in an Oedematous Patient: A Case Report.

Myxoedema coma is a rare, life-threatening endocrine emergency representing the most severe form of hypothyroidism. We present the case of a South Asian woman in her early 70s admitted with generalised oedema, chest pain, general malaise, and reduced appetite. Initially treated as decompensated heart failure, she deteriorated with hypothermia, hypotension, bradypnea, and reduced consciousness, requiring intensive care admission and intubation. Thyroid function testing revealed profound hypothyroidism (TSH 67 mU/L, T4 <2 pmol/L), confirming myxoedema coma. She received intravenous liothyronine

Enteral Feeding During Therapeutic Hypothermia for Hypoxic-Ischemic Encephalopathy in Neonates: A Multicentre Study from Italy and Belgium.

Background: Enteral feeding during therapeutic hypothermia (TH) for hypoxic-ischemic encephalopathy (HIE) remains heterogeneous across centres because of concerns regarding feeding intolerance and the risk of necrotizing enterocolitis (NEC). Evidence regarding optimal feeding initiation and advancement remains limited. Methods: We performed a secondary analysis of a multicentre retrospective cohort of infants with HIE treated with TH at two tertiary neonatal intensive care units in Italy and Belgium between 2011 and 2024. We included survivors with complete nutritional data, aware that this re

Unplanned out-of-hospital deliveries: Insights from 3473 births of a 13-year prospective cohort study in France.

INTRODUCTION: Although poorly researched, unplanned out-of-hospital deliveries are considered to carry a high risk of maternal and neonatal complications, with the primary suspected risk factor being the home-to-hospital travel time. The objective of this study was to describe the characteristics of unplanned out-of-hospital deliveries and to identify factors associated with maternal and neonatal complications. MATERIAL AND METHODS: Data were collected from the French unplanned out-of-hospital deliveries registry (79 of 402 mobile intensive care units). Analysis included 3473 deliveries betwe

Recent advances in medical hemostatic materials: Mechanisms, evaluation, and clinical perspectives.

Uncontrolled hemorrhage remains a major cause of preventable death in trauma and surgery, yet the performance of hemostatic materials varies markedly with wound geometry, blood flow, coagulation status, and tissue sensitivity. This review critically examines natural polymer-based, synthetic polymer-based, inorganic, and composite hemostats by linking material structure to six principal mechanisms: fluid absorption and surface adsorption, coagulation activation, bioactive-factor delivery, photothermal assistance, adhesive sealing, and microstructural regulation. Beyond cataloging materials, we

Generalized Megaesophagus Concurrent With Toxocariasis in a Cat.

Feline megaesophagus is a rare disorder characterized by esophageal hypomotility and dilation, resulting in regurgitation, weight loss, and occasionally death. This report describes the clinical and paraclinical findings of generalized megaesophagus with a possible contribution of toxocariasis in a kitten. A 2-month-old female cat was referred with chief complaints of lethargy, hyporexia, dyspnea, and vomiting/regurgitation. Physical examination of the kitten revealed bradycardia, bradypnea, hypothermia, dehydration, and lung crackles. The most notable laboratory results were leukopenia with s

Changes in Core Temperature Following Administration of FC5, a Blood-Brain Carrier Fused with Neurotensin in Mice, Rats and Non-Human Primates.

Development of therapies and pharmacological management of central nervous system diseases is significantly hampered by the presence of the highly selective blood-brain barrier (BBB). Receptor-mediated transcytosis (RMT) has proven to be a robust mechanism for delivering a wide variety of therapeutic modalities across the BBB. The present study describes the use of a neurotensin-based pharmacodynamic model to evaluate the brain penetrating capacity of FC5, a well-characterized RMT shuttle. Here we produce fusion molecules of FC5 with human Fc and neurotensin and show that intact but not C-term

Moderate therapeutic hypothermia with mechanical left ventricular unloading on non-infarct area in an acute myocardial infarction model.

Patients with cardiogenic shock due to acute myocardial infarction (AMI) treated with ECPELLA support have a poor prognosis. Impaired left ventricular (LV) wall motion is observed in both infarcted and non-infarcted areas. Therapeutic hypothermia and mechanical LV unloading have different myocardial protective effects. This study aimed to evaluate myocardial protective effects of moderate therapeutic hypothermia combined with mechanical LV unloading in non-infarcted areas using a porcine AMI model. The coronary artery was ligated, and an extracorporeal ventricular assist device was driven unde

From dam to neonate: microbial, metabolic, and functional pathways shaping early-life development in dogs.

Within minutes, the canine neonate transitions from placental support to respiration, thermoregulation, glucose homeostasis, feeding, and microbial colonization. These events are often considered separately, yet they form a connected developmental process that begins in the dam and continues through birth and lactation. This narrative review integrates evidence on maternal, microbial, metabolic, and functional determinants of early-life development in dogs. During gestation, maternal nutrition, metabolism, endocrine adaptation, and the maternal microbiota can influence fetal growth and maturat

High Altitude Hypothermia Reversal in the Field: A Case from Northern Pakistan, Karakoram Range.

Severe accidental hypothermia is a well-recognized risk in high altitude environments, where sudden weather changes, prolonged exposure to unstable temperatures, and limited medical access can rapidly become life threatening. In remote areas such as the Karakoram Range of northern Pakistan, improvised rewarming techniques may be the only viable option until evacuation is possible. We report a case of a 32-y-old healthy male trekker who was found unresponsive by his companions at ∼4200 m in the Karakoram Range with a recorded axillary temperature of 27°C, measured using a portable low-reading t

Ginsenoside Ra1 alleviates endothelial barrier dysfunction and multi-organ injury in experimental sepsis via NMMHC IIA inhibition.

BACKGROUND AND PURPOSE: Non-muscle myosin heavy chain IIA (NMMHC IIA) has emerged as a significant therapeutic target in vascular diseases and sepsis. However, effective interventions targeting NMMHC IIA in sepsis-induced endothelial dysfunction remain unelucidated. Here, we have assessed ginsenoside Ra1 (Ra1) as a potent NMMHC IIA inhibitor and evaluate its efficacy and underlying mechanisms in sepsis. EXPERIMENTAL APPROACH: We used the caecal ligation and puncture model of polymicrobial sepsis in mice. Survival rate, body temperature, pathological changes, vascular permeability and plasma b
