Role of p38 MAPK in the cytokine reprogramming of a human leukemic cell line with a drug-resistant phenotype.

BACKGROUND: Tumor cells have the capacity to develop coordinated resistance mechanisms that promote their survival and progression through the acquisition of multidrug resistance (MDR). This phenotype is the consequence of global cellular changes caused by antineoplastic drugs and may include modifications in the secretome, which influence the antitumoral immune response and the fate of resistant cells. RESULTS: In this study, we determined the cytokine secretion profile of human leukemic cells sensitive and resistant to several antineoplastic drugs. We also analyzed the p38 MAPK signaling pat

Efficacy and safety of Kangaroo mother care vs. conventional care during hospitalization for preterm and/or low birth weight infants: a meta-analysis with trial sequential analysis of randomized controlled trials.

BACKGROUND: Kangaroo mother care (KMC) plays a significant role in reducing mortality among preterm or low birth weight (LBW) infants. However, comprehensive evidence regarding various clinical outcomes during hospitalization for neonates receiving KMC remains limited. This meta-analysis focuses on hospital-based KMC and aims to evaluate its efficacy and safety for neonates during their hospital stay. METHODS: A systematic search of the literature was undertaken across major databases, including PubMed, Web of Science, Embase, and the Cochrane Library, to identify randomized controlled trials

Non-invasive convective head cooling during stroke thrombectomy: A prospective multi-center feasibility trial.

INTRODUCTION: Non-invasive convective head cooling is a promising putative neuroprotective therapy for ischemic stroke patients as it may portably, non-invasively, and selectively cool the ischemic penumbra. We aimed to investigate the feasibility of utilizing non-invasive convective head cooling in ischemic stroke patients before and during endovascular thrombectomy (EVT). PATIENTS AND METHODS: We conducted a multi-center, prospective, non-randomized, open-label trial at two comprehensive stroke centers in ischemic stroke patients where EVT was planned. Patients were assessed for eligibility

Temperature at admission and mortality in older adults with infection: Limited prognostic value in non-sepsis cases.

BACKGROUND: Temperature abnormalities are established prognostic markers in sepsis, but their predictive value in older adults with infection without sepsis remains unclear. OBJECTIVE: To examine the association between arrival temperature and 28-day all-cause mortality among emergency department patients aged ≥65 years with infection, stratified by sepsis status. METHODS: We performed a post-hoc analysis of a prospective cohort from a Danish emergency department. Sepsis was defined as infection plus an acute ≥2 point increase in the Sequential Organ Failure Assessment (SOFA) score. Propensi

Design and Validation of an Arduino-Based, Cost-Effective Heated Anesthesia Induction Chamber for Mice.

Rodents are highly susceptible to peri-anesthetic hypothermia because of their small body size, large surface area to volume/mass ratio, and anesthetic-induced thermoregulatory depression. Early heat loss during induction is rapid and can be difficult to reverse, emphasizing the need for simple, reliable warming solutions during anesthetic induction. We designed and validated a low-cost, Arduino-controlled heated induction chamber for mice and evaluated its safety and efficacy in preventing early hypothermia. The device uses proportional integral derivative control to regulate a heated chamber

Impact of Thermal Processing on Cashew Ana o 3: Multifaceted Insights into Structural Changes, IgE Binding Capacity, and In Vivo Allergenicity.

Reducing cashew nut allergenicity is critical for ensuring sustainable utilization of nut resources and safeguarding the health of individuals with cashew allergies. Here, we investigated the effects of different thermal processing, including water bath (WB), oil bath (OB), and microwave (MW) processing, on the structure and allergenicity of the major allergen Ana o 3 in cashew nuts. Results showed that WB and MW caused minor structural changes with negligible effects on IgE binding. OB treatment significantly denatured Ana o 3, primarily through the loss of its small subunit, which resulted i

Target temperature management in acute ischemic stroke.

Acute ischemic stroke (AIS) is an acute neurological deficit that results from focal cerebral ischemia associated with permanent brain infarction, and is a leading cause of death and disability worldwide. Considerable attention has been paid to reducing mortality and improving the prognosis of patients with AIS. Targeted temperature management (TTM), including hypothermia therapy, normothermia control, and febrile intervention, has been widely investigated in laboratory and preclinical studies and has provided substantial protection for neurological function. The effect of TTM on neurological

Enhanced metabolic benefits of dietary methionine restriction in cold resistant hybrid UCP1-deficient mice.

Dietary methionine restriction (MR) improves metabolic health in various animals. Beneficial effects on mice include reduced obesity and improved metabolism associated with the appearance of multilocular uncoupling protein 1 (UCP1) positive adipocytes in white fat. This suggests that MR-induced UCP1 thermogenesis contributes significantly to metabolic health. To test this, we evaluated MR effects using unique UCP1-deficient mice on a hybrid (F1) background that is resistant to developing hypothermia upon acute cold exposure. F1.Ucp1HET and F1.Ucp1KO mice were fed high fat control diet (HF-CD)

Operating-room temperature and warming protocols as predictors of postoperative hypothermia in ovarian cancer surgery: A retrospective cohort study.

Perioperative hypothermia is a common but preventable complication associated with increased morbidity and delayed recovery in surgical oncology patients. However, limited evidence exists regarding its prevalence, predictors, and consequences in women undergoing cytoreductive surgery for ovarian cancer. This retrospective cohort study included 245 patients who underwent primary or interval cytoreductive surgery for epithelial ovarian cancer at a tertiary care center between 2014 and 2022. Patients were stratified into normothermia (≥36.0°C) and hypothermia (<36.0°C) groups based on core temper

Current status and influencing factors of anxiety in patients with malignant tumors after anesthesia recovery.

BACKGROUND: General anesthesia is essential for maintaining adequate sedation, minimizing intraoperative stimuli, and ensuring smooth surgery completion. However, when faced with invasive procedures and anesthetic risks, patients often exhibit nonspecific physiological and psychological responses. AIM: To investigate the occurrence of agitation and the prevalence of anxiety during anesthesia recovery in patients with malignant tumors (MTs) and to analyze their influencing factors, providing a theoretical basis for clinical management. METHODS: An analysis was performed on 168 patients who un

Drug repurposing reveals Fmoc-l-leucine as an adipose thermogenesis activator in cold-exposed mice by targeting PPARγ/UCP1.

Cold-induced injuries, such as hypothermia and frostbite, pose significant health risks and have limited therapeutic options. Drug repurposing using integrated bioinformatics approaches offers an effective strategy for identifying potential agents. In this study, we aimed to identify thermogenic compounds through drug repurposing and validate their efficacy in a cold-exposure animal model. Gene expression signatures from brown and inguinal white adipose tissues from cold-exposed mice (5 °C, 7 days) were obtained from transcriptomic profiles and compared with drug-induced gene expression signat

Consequences of heterothermy during development on the physiology and growth of Antarctic storm-petrel chicks.

The ability to endure a wide range of body temperatures, called heterothermy, offers many species worldwide the possibility to survive times of malnourishment by lowering metabolic rate and body temperature. During development, facultative hypothermia and therefore lowered body temperatures might have disadvantages. Our study aimed at identifying potential trade-offs between the use of facultative hypothermia and investment into growth, immunity, and nutritional physiology in chicks of an Antarctic seabird, the Wilson's Storm-petrel (Oceanites oceanicus). To answer these questions, we used the

The Sigma1 receptor antagonist CM304 potentiates the antinociceptive effects of the cannabinoid receptor agonist delta9-tetrahydrocannabinol in rats and mice.

BACKGROUND: Mu opioid receptor (MOR) agonists are the primary medication class used to treat moderate to severe pain; however, the ongoing opioid overdose crisis underscores the need for alternatives to MOR agonists for pain treatment. Delta9-tetrahydrocannabinol (THC), a cannabinoid CB1 receptor (CB1R) agonist, and sigma1 receptor (σ1R) antagonists elicit antinociceptive effects albeit less effectively than MOR agonists. Co-administration of σ1R antagonists with THC may offer a safer alternative for pain management. METHODS: Antinociception (warm water tail withdrawal in mice and hot plate as

Knowledge, attitude, and practice toward hypothermia in severe trauma patients among healthcare professionals in emergency departments and trauma centers in Shanghai: a cross-sectional study.

BACKGROUND: Hypothermia is a critical component of the trauma triad of death and has been shown to increase morbidity and mortality among severely injured patients. Early recognition and effective temperature management are essential to improving outcomes, yet clinical knowledge and implementation remain inconsistent. Understanding healthcare professionals' preparedness for hypothermia management is therefore of great importance. To examine the knowledge, attitudes, and practices (KAP) of medical staff in emergency departments and trauma centers in Shanghai regarding the management of hypother

Implementing and evaluating a low-carbon, high-quality perioperative patient warming pathway.

BACKGROUND: Intraoperative hypothermia can lead to adverse clinical outcomes and avoidable financial and environmental costs. Environmentally preferable warming practices have been identified, including using reusable resistive blankets, extending the life cycle of forced air warming (FAW) garments and minimising flannel blanket use. This study integrates existing environmental data with best practices and quality improvement methodology to develop an optimised patient warming pathway (OPWP). This pathway was adapted to our local context, implemented and evaluated. METHODS: The OPWP was devel

Patient blood management and surgery: Impact of new surgical techniques on transfusion needs.

BACKGROUND AND OBJECTIVES: Patient blood management (PBM) is a multidisciplinary approach aimed at reducing the use of blood products. It considers the patient's own blood as a valuable resource to preserve and seeks to avoid the routine use of transfusions to treat anaemia. PBM is primarily based on interventions implemented by anaesthesiologists, including preoperative anaemia correction, targeted administration of coagulation factors and tranexamic acid and strict adherence to transfusion protocols. However, the role of surgical approaches in this context deserves attention. Indeed, surgica
