Broader autism phenotype, the default mode network, and brain entropy: A network-level hypothesis.

Despite well-documented disparities by sexual and gender minority (SGM) status in suicide attempt and mortality rates, few studies have investigated the lived experiences that contribute to SGM people's disproportionate risk of suicide. Having a history of at least one near-fatal (or medically serious) suicide attempt serves as a proxy for suicide mortality, but no known study has involved SGM people who have made such an attempt. Ideation-to-action theories of suicide posit that individuals acquire the capability for suicide through repeated exposure to painful and provocative events - namely

FLT3-directed BiTE molecules vs CAR T cells in AML: costimulatory signals mitigate T-cell exhaustion.

T-cell-based immunotherapies have revolutionized treatment paradigms in B-cell malignancies, yet their translation to acute myeloid leukemia (AML) has been hindered by a scarcity of tumor-restricted antigens and the risk of on-target off-leukemia toxicity. FLT3 has emerged as a promising therapeutic target with limited expression in healthy hematopoietic tissues. Here, we performed a head-to-head preclinical comparison of an FMS-like tyrosine kinase 3 (FLT3)-directed bispecific T-cell engager (BiTE) molecule and second-generation FLT3-specific chimeric antigen receptor (CAR) T cells. Both appr

Physics-Based Models of Extraction Kinetics in Solvent-Swollen Polymers: Using Non-Exhaustive Extractions to Estimate Total Extractable Quantities.

Leachables from polymeric medical devices can migrate into the body, potentially impacting patient health. Physics-based mass-transport models can estimate patient exposure but require knowledge of the initial leachable amount, M 0 $$ {M}_0 $$ . Although M 0 $$ {M}_0 $$ can be determined through exhaustive extraction testing, this may be impractical for some solutes due to kinetic or thermodynamic limitations. We developed a free-volume model to estimate M 0 $$ {M}_0 $$ from non-exhaustive extractions, accounting for solvent-swelling effects on solute diffusivity, D $$ D $$ , in

Revitalizing T cells: breakthroughs and challenges in overcoming T cell exhaustion.

T cell exhaustion is a prevalent phenomenon in chronic infections and tumor microenvironments, severely compromising the effectiveness of antitumor and antiviral immunity. In recent years, there has been significant progress in understanding the underlying mechanisms of T cell exhaustion, including external factors and intrinsic cellular changes that drive this dysfunctional state. Key external factors such as persistent antigen exposure, immune checkpoint signaling, and the cytokine milieu, as well as intrinsic changes such as altered metabolic processes, epigenetic modifications, and transcr

The Impact of High-Intensity Exercise in Normobaric Hypoxia on Right Ventricular Function in Trained and Untrained Men-An Echocardiographic Study.

Acute exposure to hypoxia affects the cardiovascular system, especially pulmonary circulation and right heart hemodynamics. However, the impact of normobaric hypoxia on the right heart chambers during exercise is still not clear. This study examined whether a single bout of high-intensity exercise to voluntary exhaustion under acute moderate normobaric hypoxia (~3000 m a.s.l.; FiO2 = 14.4%) induces significant changes in right ventricular (RV) and right atrial (RA) dimensions or RV systolic function compared to normoxia in trained and untrained men. RPA exhaustion activates SLFN11 to eliminate cells with heightened replication stress.

SLFN11 is epigenetically silenced and confers chemoresistance in half of all cancers. In response to replication stress, SLFN11 triggers translation shutdown and p53-independent apoptosis, but how DNA damage activates SLFN11 remains unclear. Here through CRISPR-based screens we implicate SLFN11 as the critical determinant of cisplatin sensitivity in cells lacking primase-polymerase (PrimPol)-mediated repriming. SLFN11 and the downstream integrated stress response uniquely promote cisplatin-driven apoptosis in PrimPol-deficient cells. We demonstrate that replication protein A (RPA) exhaustion a

Combined metallomics and metabolomics reveal impact of metal homeostasis on biological pathways in C. elegans.

Trace metals and metal homeostasis play an essential role in cell metabolism, and an imbalance in this careful balance leads to pathological changes. In neurodegenerative diseases such as Parkinson's disease, trace metals and metal species are increasingly recognized as key factors in disease progression in mechanisms such as ferroptosis. In this study, we combined modern analytical techniques like Fourier transform ion cyclotron resonance mass spectrometry (FT-ICR-MS) with metal-free high-pressure liquid chromatography inductively coupled plasma mass spectrometry (HPLC-ICP-MS) to analyze meta

The ubiquitin ligase KLHL6 drives resistance to CD8+ T cell dysfunction.

The multifaceted dysfunction of tumour-infiltrating T cells, including exhaustion and mitochondrial dysfunction, remains a major obstacle in cancer immunotherapy1-6. Transcriptomic and epigenomic regulation of T cell dysfunction have been extensively studied7-9, but the role of proteostasis in regulating these obstacles remains less defined. Here we combined computational analyses of atlases of T cell exhaustion and mitochondrial fitness with performed targeted in vivo CRISPR screens, which identified the E3 ubiquitin ligase KLHL6 as a dual-negative regulator of both T cell exhaustion and mito

Ecotoxicological effects of cadmium on Echinometra mathaei: Oxidative stress, genotoxicity, and biochemical responses.

Cadmium (Cd), a non-essential heavy metal, poses a persistent threat to marine ecosystems. The edible sea urchin Echinometra mathaei, widely recognized as a sentinel species, was used to evaluate the effects of environmentally relevant Cd exposure. These organisms were exposed to 0, 0.01, and 0.1 mg L-1 Cd for 28 days, and gonadal tissues were analyzed for Ca content, oxidative stress biomarkers (H₂O₂, MDA, SOD, CAT), protein and carbohydrate reserves, and genotoxic endpoints (tail intensity, tail moment, tail factor). Cd accumulated progressively in gonads, coinciding with calcium depletion,

Exhausted/senescent CD4+ T cells as predictors of vaccine failure and severe coronavirus disease 2019 in solid organ transplant recipients.

Despite booster vaccinations, solid organ transplant (SOT) recipients develop suboptimal immunity and remain at risk of severe coronavirus disease 2019 (COVID-19). Chronic immunosuppression and repeated antigen exposure favor T cell exhaustion/senescence, potentially challenging the development of protective immunity. In a prospective multicenter study including 80 naïve SOT patients receiving 5 severe acute respiratory syndrome coronavirus 2 mRNA-based vaccine doses, exhausted/senescent T cell phenotypes were longitudinally profiled at distinct time points, and their impact on antigen-specifi

Overcoming Field-Amplification Decay: A Counterflow Electroextraction Strategy for Direct, Fast, and Exhaustive Enrichment of Per- and Polyfluoroalkyl Acids from Cosmetic Matrices.

Per- and polyfluoroalkyl acids (PFAAs) in cosmetics pose significant human health risks. However, analytical methods for their monitoring remain scarce due to matrix interference, as the direct, fast, and exhaustive enrichment of trace targets from complex semisolid and solid cosmetic matrices is rather challenging. To address this gap, we developed a novel counterflow-sustained field amplification nonaqueous miscible liquid-liquid electroextraction (CSFA-NMLEE) technique. The CSFA strategy dynamically sustains a strong electric field in the sample donor phase via counterflow electrolyte reple

Multilevel toxicological impacts of industrial settleable particulate matter in a freshwater crustacean, Macrobrachium rosenbergii.

As integral components of aquatic food webs, crustaceans serve as pivotal conduits for energy transfer and sensitive bioindicators of environmental quality. This study characterizes the toxicological consequences of settleable atmospheric particulate matter (SePM)-a byproduct of industrial iron-ore processing-utilizing the freshwater shrimp Macrobrachium rosenbergii as a model organism. Experimental exposure demonstrated that SePM significantly enhances the aqueous solubility of both conventional (Fe, Al, Zn, Cu, Mn) and emergent (Cr, Ni, Sr, Sn, La, Ce) metals, establishing it as a potent vec
