Tumor-reactive TCRs within exhausted TILs reveal cancer type-specific immune landscapes in renal cell carcinoma.

Clear cell renal cell carcinoma (ccRCC) presents a unique immunological paradox: abundant CD8+ tumor-infiltrating lymphocytes (TILs) correlate with poor prognosis. To clarify their functional status and therapeutic potential, we performed single-cell transcriptomic profiling of TILs from 15 patients with ccRCC and functionally validated dominant T cell receptor (TCR) clonotypes using autologous tumor-derived organoids. Single-cell RNA sequencing revealed dynamic shifts in T cell composition, with effector and progenitor-exhausted CD8+ T cells declining and terminally exhausted CD8+ and regulat

Size-Transformable Supramolecular Nanoprodrugs Enable Redox Imbalance Amplification and Cholesterol Modulation to Boost Multidimensional Tumor Immunotherapy.

Low immunogenicity and highly immunosuppressive tumor microenvironment (TME) present major challenges in immunotherapy, as they restrict T lymphocyte infiltration and activation. Although ferroptosis induction has emerged as a promising approach for enhancing tumor immunogenicity and promoting T cell recruitment, its efficacy is frequently compromised by cholesterol-driven T cell functional exhaustion. To address these limitations, we engineered tumor-targeting, TME-responsive, and size-switchable supramolecular nanoprodrugs that enable multidimensional immune activation. Nanoprodrugs orchestr

The micronucleus assay in exfoliated buccal cells induced by lifestyle factors: an overview.

We describe the impact of lifestyle related factors on the formation of micronuclei (MN) in buccal cells. It is known that consumption of alcohol and of other drugs including tobacco and other plants cause damage of the genetic material and leads to adverse health effects. Overall, 235 articles were evaluated in total. Tobacco smoking, i.e., consumption of cigarettes (75 publications), water pipes (11 publications) and crack smoking (5 publications) caused mainly positive effects, while negative results were obtained with pure marijuana (2 publications). Chewing habits (smokeless tobacco (27 p

How do physical activity and social media use predict the study wellbeing profile of comprehensive school students?

Students' study wellbeing plays a crucial role in protecting against social and academic challenges, both within and outside school. However, little is known about how the extent of physical activity and social media use affects study wellbeing. To explore this, we identified profiles of study engagement and burnout among Finnish primary school students (n = 345, age 11), and lower secondary school students (n = 447, age 14). Using latent profile analysis, we identified five study wellbeing profiles: three showing a negative association between engagement and burnout-engaged, burned-out, and a

Drug-controlled CAR T cells through the regulation of cell-cell interactions.

Chimeric antigen receptor (CAR) T cell therapy is constrained by on-target, off-tumor toxicities and cellular exhaustion because of chronic antigen exposure. CARs incorporating small-molecule controlled on- and off-switches can enhance both safety and therapeutic efficacy but their design is limited by the scarcity of nonimmunogenic protein elements responsive to nonimmunosuppressive, clinically approved drugs with favorable pharmacodynamics. Here we combine rational design and library-based optimization of a protein-protein interaction (PPI) of human origin to develop venetoclax-controlled dr

Lymphocyte Suppression and Exhaustion, Conventional, and Accelerated.

The essential mammalian immune system lymphocytes include T cells, B cells, and natural killer cells. Any impairments of their functionalities can have severe consequences, since these lymphocytes each contribute as an interactive team in responding to pathogen infections or cancers. Such impairments include lymphocyte exhaustion, such as T cell, B cell, or natural killer cell exhaustion, or lymphocyte suppression impairing one or more of these cells. Lymphocyte exhaustion can have any intensity from mild to severe, having a severity scale worsened by various exposures and time periods of cons

Examining the effects of traumatic event exposure and somatic symptoms on emotional exhaustion: The mediating role of social support and self-esteem.

In the current study we examined factors contributing to emotional exhaustion, examining traumatic event exposure and somatic symptoms as explaining individual differences in emotional exhaustion, directly and indirectly through their effects on social support and self-esteem. Four hundred-and-ninety-seven respondents from Israel's general population were examined using self-report measures, and mediation analyses were conducted to examine the expected associations. The relationships between traumatic event exposure, somatic symptoms, and emotional exhaustion were mediated by self-esteem, wher

Generation of CCR4/CD7 Bispecific CAR-T Cells Resistant to Fratricide and Exhaustion.

The use of chimeric antigen receptor (CAR) T-cell therapy for T-cell malignancies is limited by fratricide, antigen-escape, lack of functional endurance and adverse events such as multilineage cytopenia. To address these limitations, we developed a simple single-step CD7-depletion process followed by transduction with a lentiviral vector encoding a CCR4/CD7 bispecific CAR. CD7-negative (CD7N) CCR4/CD7 CAR-T cells could expand without experiencing fratricide, unlike the bulk CCR4/CD7 CAR-T cells. The CD7N CAR-T exhibited robust cytotoxicity against malignant T-cell lines with heterogeneous CD7

Prevalence of Emotional Exhaustion Among Respiratory Therapists.

BACKGROUND: Respiratory therapists (RTs) face high burnout risk, with rates as high as 79% reported during the COVID-19 pandemic. We hypothesized that the rate of burnout has decreased since the COVID-19 pandemic. METHODS: We performed a multi-center, multinational study between January 6 and February 1, 2025, evaluating the prevalence of RT burnout. Validated survey instruments were used to measure emotional exhaustion (burnout), incivility, intent to leave, and leadership. RESULTS: We received 1,033 responses (34% response rate). Seven hundred nineteen (70%) respondents were burned out, wi

Hyperinsulinemia drives glomerular podocyte injury and albuminuria via a self-perpetuating GSK3β-IRS1 insulin desensitization circuit.

Epidemiological evidence indicates that hyperinsulinemia is an independent risk factor for albuminuria and renal impairment, yet its molecular basis remains unclear. In prediabetic db/db mice, hyperinsulinemia coincided with albuminuria, podocyte injury, and impaired glomerular insulin signaling, characterized by insulin receptor exhaustion and hypoactivity of the IRS1/PI3K/Akt insulin signaling. This led to diminished inhibitory phosphorylation of GSK3βS9 in podocytes, denoting GSK3β hyperactivity. Notably, GSK3β co-localized and physically interacted with IRS1 in glomerular podocytes, phosph

NR2F6 deletion revives CAR-T cell function and induces antigen-agnostic immune memory in solid tumors.

CAR-T cell therapy is effective in hematologic malignancies but remains challenging in solid tumors owing to antigen heterogeneity and tumor microenvironment-induced exhaustion. Here, gene editing of the nuclear receptor NR2F6 restores CAR-T cell functionality, sustaining a TCF1⁺ progenitor-exhausted phenotype, enhancing metabolic fitness, and preserving cytotoxic potency under chronic antigen exposure. In immunocompetent models, Nr2f6-deficient CAR-T cells suppress solid tumor growth and induce robust, polyclonal host antitumor responses that persist after CAR-T clearance, as demonstrated by

Immune Exhaustion in Chronic Infection and Cancer: Signaling Pathways and Therapeutic Interventions.

Immune exhaustion is a state of sustained lymphocyte dysfunction that occurs following chronic antigenic stimulation and constitutes a shared hallmark of chronic infection and cancer. Beyond being a passive consequence of persistent antigen exposure, it actively drives tumor progression by fostering immunosuppressive microenvironments. Pathogens that evade immune detection to establish chronic infection can directly induce immune exhaustion through sustained inflammatory signaling, thereby crippling cytotoxic T cell-mediated tumor surveillance. This impairment facilitates both de novo tumorige

Development and psychometric validation of the media-influenced healthcare violence perception scale in Türkiye.

BACKGROUND: Violence against healthcare professionals is a growing public health concern with significant implications for workforce sustainability, work-life integration, quality of care, and patient safety. Although the prevalence and consequences of violence in healthcare are well documented, the ways in which media exposure shapes public perceptions, emotional responses, and the psychosocial well-being of healthcare workers remain insufficiently measured. This study aimed to develop and psychometrically validate the Media-Influenced Healthcare Violence Perception Scale (MHVPS). METHODS: An

Mechanistic basis and therapeutic modulation of T cell fitness to enhance CAR-T cell efficacy in hematological malignancies.

T cell fitness has emerged as a critical determinant of the efficacy and persistence of Chimeric Antigen Receptor (CAR)-T cell therapy. Defined by the capacity of T cells to proliferate, resist exhaustion, persist in vivo, and exert sustained effector functions, T cell fitness reflects the integration of a dynamic network of intrinsic and extrinsic regulatory mechanisms. In this review, we present a comprehensive synthesis of the molecular and cellular foundations underlying T cell fitness, emphasizing the influence of differentiation trajectories, signaling pathways, metabolic reprogramming,

NCOA2 promotes the return of hematopoietic stem cells to quiescence after irradiation stress by regulating FOXO3a-dependent mitophagy.

Hematopoietic stem cells (HSCs) are responsible for replenishing blood cells under stress conditions through increasing proliferation and differentiation. After the hematopoietic function has reconstructed, HSCs must re-enter a quiescent state to avoid their depletion, whereas the underlying mechanisms remain to be elucidated. Here, we show that the translocation of nuclear receptor coactivator 2 (NCOA2) into the nucleus is gradually increased in HSCs during the hematopoietic recovery phase after sub-lethal dose irradiation (IR). Although deletion of NCOA2 only slightly affects the steady stat

Balancing Intrinsic and Extrinsic Factors in CD8+ T Cell Therapy.

Persistent Ag exposure in tumors, chronic infections, and autoimmune diseases progressively drive CD8+ T cell dysfunction-a process known as T cell exhaustion. During this process, progenitor exhausted CD8+ T (Tpex) cells represent an early subset with stem cell-like properties and serve as key mediators of immune checkpoint blockade responses. Despite their longevity and proliferative capacity, Tpex cells display limited cytotoxicity. Upon sustained TCR stimulation, they differentiate into exhausted CD8+ T (Tex) cells that express high levels of granzyme B and contribute critically to tumor e
