Elevated activity of the mesolimbic dopamine system promotes feeding during pregnancy in mice.

The pregnancy period is accompanied by increased feeding behavior to accommodate the elevated energy demands associated with fetal growth and development. However, the underlying neural circuitry and molecular mechanisms mediating increased feeding during pregnancy are largely unknown. Here, we utilized a combination of fiber photometry, chemogenetics, and mouse behavioral assays to characterize altered feeding behavior during pregnancy in mice. We uncover that pregnancy increases the average activity of the mesolimbic dopamine system during feeding behavior in mice. VTA dopamine neurons promo

Risk of bone fractures in patients with prostate cancer treated with maximal androgen blockade therapy: a systematic literature review and meta-analysis.

BACKGROUND: Addition of an androgen receptor pathway inhibitor (ARPI) to androgen deprivation therapy (ADT) (ADT + ARPI, i.e., maximal androgen blockade, MAB) improves survival outcomes compared to ADT monotherapy in patients with prostate cancer (PC). It is known that ADT increases the risk of fractures in patients with PC, but it is unclear if this risk is higher with MAB. The aim of this study is to conduct a systematic review and meta-analysis to determine if MAB increases the incidence of fractures compared to ADT alone, and if the incidence of fractures was influenced by the type of ARPI

Genomic and Clinical Impact of Smoking on Therapeutic Outcomes in Prostate Cancer: A Public Databases Analysis.

BACKGROUND: In prostate cancer (PCa), smoking history is associated with more aggressive clinicopathological features, reduced efficacy of androgen deprivation therapy, and poorer overall survival (OS). However, its impact on survival outcomes in patients receiving taxane chemotherapy, poly ADP-ribose polymerase inhibitor (PARPi), or immune checkpoint inhibitor (ICI) therapy for PCa remains unclear. We compared the genetic profiles of smokers and non-smokers with PCa in several public databases to explore the potential influence of smoking on treatment outcomes. METHODS: We retrospectively co

Estimating the determinants of health literacy for policy prioritisation: a local level case study in Newham, London.

This study investigates the determinants of health literacy in Newham, London, using a statistical modelling approach. Health literacy is defined using literacy, numeracy, and Information and Communication Technology (ICT) literacy from the ONS Skills for Life (SfL) survey 2011. The research combines survey, the Newham Residents Survey (NRS), and the UK 2021 Census data. Multilevel regression with post-stratification (MRP) and average treatment effects (ATE) are used to quantify the strength of association of various factors on health literacy at the local level. The study identifies several si

[How can the suitability of a geriatric patient for uro-oncological treatment be determined?].

BACKGROUND: Geriatric patients undergoing uro-oncological treatments are particularly susceptible to complications in many organ systems. Prior to starting treatment, a careful risk-benefit analysis must be performed, taking into consideration the patient's remaining life expectancy and existing functional deficits. These should be weighed against the side effect profile of the planned treatment. OBJECTIVE: After an introduction of the three-step geriatric assessment to identify geriatric patients, systematically identify functional deficits, and evaluate them in depth, the example of androge

Differential impacts of the COVID-19 pandemic on sociodemographic groups: A mathematical model framework.

Deprivation and age can both drive disparities in infectious disease transmission and outcomes; however, few models capture their combined effects. We developed a deterministic ordinary differential equation model stratified by age and deprivation decile coupled with time-dependent testing proportion to examine how mixing patterns shape inequalities in disease burden, using COVID-19 in England as a case study. The framework allows three mixing scenarios-diagonal, preferred, and proportionate, and we simulated the epidemic with movement restrictions to reflect lockdown measures. We assessed the

Promoting mental health among at-risk adolescents in Malaysia (MyHeRo): study protocol for a cluster randomised controlled trial to evaluate the effectiveness and cost-effectiveness of a school-based intervention compared with study skills condition for adolescents identified as at risk for anxiety and depression.

BACKGROUND: In Malaysia, adolescent anxiety and depression are increasing faster than ever, and rates of suicidal behaviour are rising especially among those living in deprived communities. However, Malaysia's mental health system is currently constrained by limited workforce capacity, affecting the delivery of effective interventions. The overall aim of this trial is to use a school-based intervention to promote mental health among at-risk adolescents from low-income communities in Malaysia. Our primary aim is to evaluate the effectiveness and cost-effectiveness of a school-based intervention

Patient, facility, and community determinants of timely follow up after abnormal screening mammograms in Appalachia: A multilevel analysis.

PURPOSE: Examine the extent that community and organizational characteristics influence mammogram recall and biopsy rates in Appalachia. METHODS: We collected facility survey and Medicare claims (2016-2018) data on 191 FDA-registered breast imaging organizations serving 2.65 million women in 5 Appalachian states (KY, OH, PA, VA, and WV). Primary outcomes were recall rates and time to recall and biopsy. Generalized linear mixed-effects models (GLMM) (patient level with facility random intercepts) and facility-level structural equation models were used to evaluate multilevel predictors of recal

The Diverse Effect of HDAC Inhibitors: Sodium Butyrate and Givinostat on Microglia Polarization After Hypoxia-Ischemia In Vitro.

Microglia play a key role in the development of neuroinflammation induced by cerebral ischemia. On the other hand, these cells participate in neurorepair processes. This dual role of microglia stems from the ability to shift their phenotype from pro-inflammatory M1 to protective M2. Histone deacetylase inhibitors (HDACis) are a group of agents that exhibit neuroprotective effects in some models of ischemia, among others, by modulation of signaling pathways that regulate microglial activation. This study aimed to examine the effect of HDACis-sodium butyrate and Givinostat-on polarization of mic

Evaluating the effectiveness and acceptability of free door-to-door transport to increase the uptake of breast screening appointments in Yorkshire: a cluster randomised GP feasibility trial (DOORSTEP protocol).

INTRODUCTION: Breast screening uptake remains low in parts of the UK, partly due to barriers including limited transport access. Offering free transport to screening appointments may help address this and improve uptake. This general practitioner (GP) cluster-randomised feasibility trial will assess whether offering free door-to-door transport alongside routine screening invitations increases attendance. METHODS AND ANALYSIS: Eight general practices in Yorkshire will be randomised to either the intervention (routine invitation plus information about booking free door-to-door transport) or con

Akkermansia muciniphila Ameliorates Chronic Sleep Deprivation-Induced Glucose Intolerance and Inflammatory Cytokine Activation.

OBJECTIVE: Emerging evidence indicates that Akkermansia muciniphila (A. muciniphila or AKK) regulates host glucose metabolism through multiple pathways. In this study, we examined the therapeutic effects of A. muciniphila on chronic sleep deprivation (CSD)-induced glucose dysregulation and the underlying mechanisms. METHODS: A modified multiplatform water environment method was used to generate a mouse model of CSD. The mice were divided into three groups: the control (CON) group (ad libitum sleep), the CSD group (subjected to sleep deprivation), and the CSD+AKK group (CSD mice were supplemen

Mitochondrial responses to anoxia-reoxygenation exposure in crucian carp (Carassius carassius).

The crucian carp (Carassius carassius) is one of the most anoxia-tolerant vertebrates. While physiological underpinnings of its ability to withstand O2 deprivation are well studied, the ability to tolerate the return to normoxia is still enigmatic. Such reoxygenation is associated with detrimental oxidation damage in other organisms, where mitochondria play a central role in the damaging effects. This leads to the question whether mitochondrial adaptations play a central role in the anoxia and reoxygenation tolerance of crucian carp. We here address whether mitochondria from crucian carp circu

Metabolite profiling of the effect of prenatal stimuli across postnatal treatments in the liver.

Hepatic molecular mechanisms can be modulated by pro- and anti-inflammatory signals associated with infections and nutritional changes that can, in turn, affect the endocrine system. The sex-specific interplay between stimuli on hepatic pathways was studied using a biomedical model. The liver metabolome of pigs exposed to a prenatal immune activation from maternal infection was compared to that of matching female and male controls. Within prenatal treatment and sex group, the postnatal treatments were synthetic inflammatory factor, feeding deprivation (fasting), or saline. Liquid chromatograph

Glucose Deprivation of Tumor Cells via Selective Nutrient Delivery: A Potential Therapy for Metastatic Breast Cancer.

BACKGROUND/AIM: Metastatic breast cancer remains a major clinical challenge despite the availability of various chemotherapeutic agents. Current metabolic inhibitors have limitations, prompting the need for innovative strategies that selectively target tumor cells while sparing normal tissues. This study aimed to propose a novel approach focused on depriving tumor cells of glucose while ensuring nutrient delivery to normal cells. MATERIALS AND METHODS: A comprehensive literature review was conducted using PubMed and Google Scholar. The proposed strategy involved focusing on studies that deple

Circadian feeding promotion by Ninjin'yoeito counteracts frailty in aged mice.

Frailty in aging is a major health challenge, requiring solution. Older people with frailty often exhibit malnutrition and dysregulated feeding. Feeding behavior displays circadian rhythm, while aging and frailty involve rhythm disorders, suggesting possible role of circadian feeding in frailty and treatment. Herbal medicine, Ninjin'yoeito (NYT), reportedly ameliorates frail symptoms. The present study explored impacts of NYT on circadian feeding and psychological/physical functions in aged mice. Here, we report that oral NYT independent of administration timing increases food intake specifica

More efficient iron enrichment in heterotrophic Auxenochlorella protothecoides by nitrogen source manipulation.

Microalgae are increasingly explored as sustainable carriers for micronutrient delivery in food, feed, and agriculture, particularly for iron enrichment. Among them, Auxenochlorella protothecoides is a promising strain owing to its ability to grow heterotrophically to high cell densities using food industry side streams and organic carbon and nitrogen sources. However, under such conditions, direct iron supplementation in the cultivation medium is largely ineffective because ferric ions are poorly soluble at near-neutral pH and readily form complexes with organic matter, limiting cellular upta

Mice from lines selectively bred for innately high activity levels have altered behavioral and energetic responses to repeated sleep deprivation.

Sleep deprivation negatively impacts both physical and psychological health in both humans and animal models. Exercise, on the other hand, can have beneficial effects on various aspects of physical and mental health. However, little is known about the ways in which sleep deprivation and exercise may interact, especially for exceptionally high levels of exercise. We studied High Runner (HR) mice from a long-term artificial selection experiment to investigate how genetically high exercise level could impact the response to sleep deprivation. A total of 192 adult mice from four replicate HR and f

Health service contacts for mental health and substance use on release from prison: a retrospective population-based data linkage study.

BACKGROUND: Mental health and substance use problems among people released from prison contribute substantially to premature mortality and emergency services demand. Understanding of mental health and substance use-related health service contacts prior to these severe and costly outcomes is limited. We assessed mental health and substance use-related contact with multiple services, comparing rates of contact among people released from prison to a matched general population sample who had not recently been in prison. OBJECTIVES: To compare rates of health service contacts for mental health and

Effect of timing of morning blood draws on sleep quantity and quality in hospitalized patients.

BACKGROUND: Sleep deprivation significantly impacts millions of hospitalized patients and is a critical factor in their recovery process. One major contributor to sleep disturbances is early morning blood draws. OBJECTIVE: This study investigated the effect of delaying early morning blood draws by 2 h on both sleep quality and quantity. METHODS: In this quasi-experimental study, we recruited adult medical patients admitted to two similar acute medical units. Patients in one unit were assigned to have their routing morning blood draws at the usual time of 4:00 a.m., while those in the other u
