Real-world evidence on gadoteridol safety: A 20-year FAERS database study revealing critical adverse reaction patterns.

Gadoteridol, a gadolinium-based contrast agent used for magnetic resonance imaging, has been used in clinical settings since its approval in the United Kingdom in 1992. Although its efficacy in enhancing imaging results is well established, investigating its associated adverse reactions remains critically important. This study aimed to comprehensively assess the safety profile of gadoteridol-related adverse drug events (ADEs), as reported in the FDA Adverse Event Reporting System (FAERS) database. Adverse event reports on gadoteridol spanning from the first quarter of 2004 to the fourth quarte

Assessing the changes in outcome measurements in randomized controlled trials of regional anesthesia interventions: a registry-publication comparison study.

BACKGROUND: Regional anesthesia (RA) is increasingly used due to its safety and effectiveness in reducing postoperative pain and opioid use. As in other areas of clinical research, transparent and consistent outcome reporting is important for interpreting trial evidence. This study assessed the extent and nature of temporal changes to prespecified outcomes in interventional RA trials registered on ClinicalTrials.gov after the implementation of the Final Rule and the Food and Drug Administration Amendments Act 801. METHODS: We conducted a registry-based cross-sectional analysis of RA trials in

Public support for novel carbon removal hinges on procedural and distributive fairness.

Scientifically valid data on the safety of drug use during pregnancy are a significant public health need. Data are rarely available on the fetal effects of in utero exposure in human pregnancies, particularly when a drug is first marketed. Data from animal reproductive toxicology studies, which function as a screen for potential human teratogenicity, are usually all that is available in a product's labelling. For practising clinicians, translating known animal risks into an accurate assessment of teratogenic risks in their patients is very difficult, if not impossible. Without human data on t

US postmarketed drug removals: Analysis of discontinued and withdrawn drugs.

Drugs removed from the US market after approval can be either discontinued by the manufacturer or withdrawn by the US Food and Drug Administration (FDA). However, a systematic overview is currently lacking. This minireview provides a comprehensive analysis of removed drugs, identifies patterns regarding drug classes, indications, adverse effects, and scientific interest, and compares US removals with the European Union and FDA boxed warnings. A total of 166 drugs were removed between 1955 and 2026, including 88 discontinued and 78 withdrawn drugs. Removed drugs represented a wide range of ther

Development and Testing of Waterpipe Tobacco Text and Pictorial Warnings Among Young Adults in the United States.

INTRODUCTION: Waterpipe tobacco smoking (WTS) is associated with health harms, yet the United States requires only a single text warning. METHODS: We developed 30 text warnings, across 12 health themes, based on scientific evidence and expert input. Text warnings were tested in an online survey among a national convenience sample of US young adults (18-29, Study 1). We then selected six text warnings and paired each with three images, creating 18 pictorial warnings that were tested in an online survey among a second national convenience sample of US young adults (18-29, Study 2). RESULTS: St

Postmarket Drug Safety-Related Actions Before and After the US Food and Drug Administration Amendments Act.

IMPORTANCE: The 2007 US Food and Drug Administration (FDA) Amendments Act (FDAAA) expanded its safety-related regulatory authorities, including enhanced postmarketing safety surveillance and new clinical study requirements. However, whether FDAAA has been associated with differences in the frequency and timing of postmarket safety-related actions remains poorly understood. OBJECTIVES: To assess whether FDAAA was associated with differences in time to first FDA postmarket drug safety-related action, and to assess whether therapeutic and regulatory characteristics were associated with differenc

Endocrine advantages of PD-1/PD-L1 therapy: Comparative analysis of FAERS-JADER.

With the extensive clinical application of immune checkpoint inhibitors (ICIs), immune-related adverse events (irAEs) associated with these agents have increasingly garnered significant attention. Unlike other irAEs, endocrine irAEs are mostly irreversible, with variable and nonspecific symptoms, which poses challenges for clinicians in diagnosis. As a result, this study leveraged the U.S. Food and Drug Administration Adverse Event Reporting System (FAERS) and the Japanese Adverse Drug Event Report (JADER) pharmacovigilance databases to conduct an in-depth investigation into adverse events ind

Mid-infrared spectroscopy combined with chemometrics for the detection of ciprofloxacin and Bronopol® residues in milk.

The presence of antibiotic residues in milk poses a critical challenge to public health and food safety due to the risks of adverse reactions, disruptions in intestinal microbiota, and especially the spread of antimicrobial resistance. Although conventional detection methods such as chromatography are effective, they have limitations including high cost, lengthy analysis time, and the need for specialized labor. This study investigated the application of mid-infrared Fourier-transform infrared spectroscopy (FTIR-MIR), combined with chemometrics, as a rapid and effective alternative to detect c
