Robotic Totally Intracorporeal Bilateral Boari Flap Reconstruction for Complex Bilateral Ureteral Strictures: A Step-by-Step Technique.

PURPOSE: Boari flap reconstruction is effective for unilateral ureteral strictures (1-5), but its use for bilateral disease is rarely reported. Robotic surgery offers enhanced visualization and dexterity for complex ureteral reconstruction (4, 6-9). We describe robotic totally intracorporeal bilateral Boari flap (BBF) reconstruction for bilateral mid- and distal ureteral strictures (BMDUS). MATERIALS AND METHODS: A 42-year-old woman with BMDUS underwent BBF reconstruction. The left ureter was dissected, ligated, and transected proximal to the stricture, then trimmed to healthy tissue until a

Perineal Urethrostomy After Urethral Stricture: Step-by-Step Surgical Technique.

PURPOSE: Urethral stricture is a chronic narrowing of the urethral lumen caused by inflammation and iatrogenic interventions, including previous surgery and prostate cancer treatment (1). Treatment options include urethral dilation, internal urethrotomy, urethroplasty, and perineal urethrostomy. Perineal urethrostomy creates a permanent urethral meatus and is increasingly considered a primary treatment option for complex cases or patients with significant comorbidities (2, 3). It is associated with high patient satisfaction, preserved continence, and favorable voiding outcomes (4). This video

(Immuno-)Histological Analysis of Ewing Sarcoma.

The diagnosis of Ewing sarcoma requires integrating information from numerous techniques, some of which are very sophisticated. However, the first steps of the diagnostic process are crucial to achieve the maximum possible diagnostic performance. In this chapter, we will review how to handle the diagnostic specimen from its collection, how to prepare it for diagnosis, how to make a complete pathology report, and provide guidance for the reasonable use of immunohistochemical techniques in this malignancy.

Ewing Sarcoma PDX Models.

Ewing sarcoma (EWS) is a rare malignant pediatric tumor, and patient-derived xenografts (PDXs) could represent a possibility to increase the number of available models to study this disease. Compared to cell-derived xenografts (CDX), PDXs are reported to recapitulate better tumor microenvironment, heterogeneity, genetic, and epigenetic features. They are considered reliable models due to their superior predictive value when comparing preclinical efficacy and treatment response in patients. In this chapter, we extensively describe a method for generating EWS PDX models, along with their validat

Foundation artificial intelligence models for health recognition by use of face photographs (FAHR-Face): a model development and validation study.

BACKGROUND: Facial appearance offers a non-invasive window into health. We built Foundation Artificial Intelligence for Health Recognition-Face (FAHR-Face), a foundation model trained on more than 40 million facial photographs and fine-tuned it for two distinct tasks: biological age estimation (FAHR-FaceAge) and survival prediction (FAHR-FaceSurvival). METHODS: FAHR-FaceAge underwent two-stage, age-balanced fine-tuning on ten public image datasets; FAHR-FaceSurvival was fine-tuned on 34 363 photographs of patients with cancer. Model robustness (cosmetic surgery, make-up, pose, and lighting) a

Tumor Growth Analysis of Ewing Sarcoma Cell Lines Using Subcutaneous Xenografts in Mice.

Subcutaneous murine xenograft models are one of the most commonly used in vivo experimental methods in the cancer research field. Due to the lack of syngenic animal models for Ewing sarcoma, subcutaneous murine xenograft models currently offer the simplest way to investigate antineoplastic effects of therapeutics or biological functions of target genes in vivo. In this chapter, we describe in detail a widely used procedure for subcutaneous injection in mice, focusing on the specific characteristics of Ewing sarcoma cell lines.

Orthotopic Implants in Mice.

By directly implanting patient tumor cells into mice in a relevant location, we can mimic both the biology and tumor microenvironment of the original tumor. Here, we describe the process of generating an orthotopic patient-derived xenograft model by injecting a single cell suspension of Ewing sarcoma cells into the femur of a recipient mouse.

Metastasis Assessment in Ewing Sarcoma Using Orthotopic Xenografts.

Orthotopic models are based on the implantation of tumor cells directly into the organ of origin, which allows interaction between the cells and the surrounding host tissues. Here, we describe a modified version of an orthotopic model that closely recapitulates the steps required for metastasis development in Ewing sarcoma: tumor cells are injected into the calf muscles of the mouse, and once the tumor reaches a certain volume, the muscles containing the tumor are surgically resected. This procedure involves a nonaggressive surgery of the muscle, which allows for the survival of the mouse duri

Analysis of Colony Formation Capacity in Ewing Sarcoma.

Colony formation assays enable an assessment of the cells' ability to grow in colonies under in vitro conditions. Following the seeding and an incubation period, the cell colonies are stained using a crystal violet staining solution, and the resulting colonies are imaged and assessed. This method can be leveraged to investigate the effect of genetic modifications or the application of different drug compounds on colony growth. Here, we describe a protocol applied to Ewing sarcoma cell lines, as well as possible modifications.

Conjugation Assay for Antimicrobial Resistance Plasmids in Salmonella enterica.

The conjugation assay is an essential technique for investigating horizontal gene transfer and assessing the mobility of antimicrobial resistance plasmids in Enterobacteriaceae. Here we describe a commonly used method for performing conjugation experiments to quantify resistance plasmid transfer in Salmonella enterica under laboratory conditions. It outlines the required materials, experimental conditions, and verification of transconjugants. The method is applicable to most plasmid incompatibility groups within Enterobacteriaceae and can be adapted to other bacterial species.

Preprocessing Large-Scale Proteomics Data.

Recent advances in mass spectrometry instrumentation and computational analysis have established label-free data-independent acquisition (DIA) as a powerful tool in quantitative proteomics, offering depth, throughput, and reproducibility. These developments now enable large-scale projects with hundreds or even thousands of samples, opening new applications in systems biology and biomedicine. However, large-scale datasets require special consideration during data preprocessing. Here, we present a practical guide for preprocessing large-scale DIA datasets, illustrated using a public dataset. We

Using Zebrafish Larvae as a Xenotransplantation Model to Study Ewing Sarcoma.

Tumor models allowing for the in vivo investigation of molecular mechanisms driving tumor progression and metastasis are important to develop novel strategies for cancer treatment. Unfortunately, for Ewing sarcoma, no adequate genetic animal models are currently available, although there is progress in zebrafish. Nevertheless, mouse xenotransplantation models are the state-of-the-art to model Ewing sarcoma in vivo. Here, we describe an alternative Ewing sarcoma xenotransplantation model in embryonic and larval zebrafish. This zebrafish model offers live imaging and easy compound testing opport

Artifacts and Practical Issues in Atomic Force Microscopy.

As with any other microscopic technique, in atomic force microscopy (AFM), artifacts and problems can arise. Some of these happen due to improper use of the microscope by the operator, and some are due to particular characteristics of the sample. Some occur depending on the type of instrument or from probe damage. Some of them are artifacts inherent in the technique. Knowledge of these issues is important for correct data acquisition and interpretation, and in many cases, training in AFM is inadequate. In this chapter, we show examples of common artifacts in AFM and describe, where possible, h

Effects of Antimicrobial Peptides on Bacterial Cells.

In recent decades, antimicrobial peptides (AMPs) have emerged as some of the most extensively studied alternatives to conventional antibiotics. To gain deeper insights into their mechanisms of action and development, AFM has played a crucial role as an imaging and force spectroscopic tool. Here, we describe the methods used for examining the effects of AMPs on bacteria using imaging and force spectroscopy techniques. These approaches have proven essential for elucidating their mechanisms of action and identifying potential outcomes.
