Rheum undulatum Linné Exerts Neuroprotective Effects Against Aβ Oligomer-Induced Neuronal Damage by Modulating p53-Mediated Signaling.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by cognitive decline and synaptic dysfunction. Accumulating evidence indicates that β-amyloid oligomers (AβO), the most neurotoxic form of β-amyloid, play a critical role in the pathogenesis of AD by inducing synaptic loss, neuronal death, and neuroinflammation. Therefore, targeting AβO-driven pathological processes through multi-target therapeutic strategies represents a promising approach to treating AD. This study aimed to investigate the neuroprotective effects of Rheum undulatum Linné rhizome (RUL) and the

Multimodal nanoengineered scaffolds as regenerative microenvironment orchestrators: coupling mechanical, bioelectrical, immunological, and spatiotemporal therapeutic cues.

Nanoengineered scaffolds are evolving from passive structural supports into instructive platforms capable of regulating the regenerative microenvironment. This narrative review examines how mechanical, bioelectrical, immunological, and spatiotemporal therapeutic cues are engineered and integrated within multimodal scaffolds. A literature search was performed using PubMed, Web of Science, Scopus, and Google Scholar databases for publications from January 2015 to March 2026, with earlier studies included when mechanistically essential. Mechanically instructive architectures regulate cellular org

MEGH-DDI: MolT5-driven edge-aware graph learning with Hop2Token representation for drug-drug interaction prediction.

Drug-drug interactions (DDIs) are critical in pharmacology, as synergistic or antagonistic effects between co-administered drugs can enhance therapeutic outcomes or cause toxicity, posing major challenges for drug safety assessment and new drug development. Current molecular representation methods often fail to capture dynamic substructure relationships or semantic interactions among functional groups. To overcome these limitations, this study proposes MEGH-DDI, a multimodal framework that integrates complementary molecular representation learning with graph-structure dynamic perception. The f

A "Progenitor Cell Ark" hydrogel system for intervertebral disc regeneration via mitochondrial targeting and mitophagy enhancement.

Intervertebral disc degeneration (IVDD), driven by progenitor cell loss and oxidative stress, remains a major clinical challenge with limited effective regenerative therapies. Here, we present a "Progenitor Cell Ark," a triple-layered hydrogel microsphere system designed for synergistic cell delivery and microenvironment remediation. The Ark comprises an outer nucleus pulposus progenitor cell (NPPC) layer for cell replenishment, a middle GelMA/HAMA scaffold for support, and an inner SS31-modified ultrasmall Prussian blue nanozyme (SS31-UPN) layer that specifically targets mitochondria to scave

HiNet3D: Integrating patient hierarchical aggregation network and molecular 3D-aware for medication recommendation.

Accurate medication recommendation from electronic health records (EHR) is crucial for clinical decision support. However, prior methods often treat EHR as a flat visit sequence and encode drugs with 2D molecular graphs, which limits capturing inadequate patient representation and drug molecular semantics. Therefore, we propose a framework named HiNet3D for the recommendation task. Specifically, HiNet3D builds a patient history graph by representing each visit as a node and performs cross-visit pairwise aggregation to form a layer of virtual nodes, enabling rich cross-visit context modeling to

Recycling of polycarbonate from optical lens manufacturing waste via solvent-targeted recovery and precipitation: A process feasibility and sustainability assessment.

Polycarbonate (PC) is widely used in optical lenses, and lens manufacturing generates a heterogeneous waste stream known as swarf that contains PC, polyurethanes, thermoset resins, coatings, and process debris and is currently unrecyclable. This preliminary feasibility study evaluates swarf as a potential feedstock for PC recycling via solvent-targeted recovery and precipitation (STRAP™). Solvent selection was guided by COSMO-RS modeling to identify solvents that dissolve PC under mild conditions (65-120 °C, ambient pressure) while minimizing urethane-resin dissolution. Among the solvents test

A GelMA hydrogel co-loaded with ursolic acid, ferulic acid and naringin facilitates dentin regeneration by providing a multi-component odontogenic microenvironment.

Dental caries is a prevalent bacterial infectious disease, yet current clinical treatments remain limited because conventional restorative materials provide only mechanical filling and fail to induce dental tissue regeneration. Herein, we developed a composite gelatin methacryloyl (GelMA) hydrogel system (Gel/UFN) co-loaded with three active components from traditional Chinese medicine, including ursolic acid (UA), ferulic acid (FA) and naringin (Nar), to remodel the carious microenvironment and promote dentin remineralization and vascularized pulp-dentin-like tissue formation. Gel/UFN showed

Phenylboronate hydrogel for administration route tailored dual-drug delivery to anterior and posterior ocular segments.

Frequent multidrug eyedrop instillation for ocular perioperative prophylaxis (e.g., cataract surgery) suffers from rapid nasolacrimal clearance, limited corneal penetration, and poor access to the posterior segment. To address these limitations, we engineered an interface-adaptive phenylboronate ester hydrogel for rapid in situ gelation and the co-entrapment of two frequently instilled drugs (moxifloxacin and dexamethasone) in their free or liposomal forms at physiological pH. This platform utilizes chemical crosslinking between hyaluronic acid-conjugated 2-aminophenylboronic acid and the diol

Powder Handling and Frozen Texture of a Reconstitutable Strawberry-Oat Sorbet Premix After Sucrose Replacement.

A dry sorbet premix must remain manageable as a powder and still develop an appropriate structure after reconstitution and freezing. This study compared a sucrose-containing control with a strawberry-oat premix in which sucrose was replaced by maltodextrin, inulin, and a commercial high-intensity sweetener blend. Three independently prepared batches were evaluated in the dry state and after reconstitution and dynamic freezing. The sucrose-replaced powder had a higher angle of repose (44° vs. 39°) and a lower compaction force (2.65 vs. 7.65 N); both powders wetted in approximately 4 s. It also

Alkali Heat-Treated Quinoa Protein-Tannic Acid Nanoparticles Stabilized Pickering Emulsions: Stability, Dihydroquercetin Delivery and in Vitro Digestion Properties.

Dihydroquercetin, a naturally occurring flavonoid, has limited applications in food systems because of its poor aqueous solubility and low bioaccessibility. A food-grade delivery system was developed to improve the stability and bioaccessibility of dihydroquercetin. Alkali heat treatment was applied to modify the conformational structure and enhance the functional properties of quinoa protein. Covalent quinoa protein-tannic acid nanoparticles were prepared under alkaline conditions and used to stabilize Pickering emulsions for dihydroquercetin encapsulation. The nanoparticles were comprehensiv

Phospholipid codes at virus-host membrane interfaces: mechanisms and therapeutic opportunities in animal viral diseases.

Animal viruses depend on host membranes at every stage of infection, yet membrane lipid states are often treated as background rather than as causal determinants of virus-host interactions. This Review develops a phospholipid-code framework in which headgroup identity, acyl-chain chemistry, leaflet topology, and lipid flux define infection-stage-specific membrane states. Evidence from Newcastle disease virus, infectious bronchitis virus, avian influenza virus, porcine reproductive and respiratory syndrome virus, African swine fever virus, Senecavirus A, Japanese encephalitis virus, and porcine

Effects of straw return combined with different amendments on soil enzyme activities and bacterial community structure in acidic purple soil.

Soil microorganisms are fundamental drivers of nutrient cycling and ecosystem functioning in agricultural soils, but their responses to management practices in acidic agroecosystems remain insufficiently understood. Acidification-induced changes in soil environmental conditions can constrain bacterial activity and disrupt carbon and nutrient transformation processes. This study investigated how straw return combined with different amendments affects soil enzyme activities and bacterial community structure in acidic purple soil under a 2-year rice-rapeseed rotation system. Six management treatm

Chitosan Nanoparticles Alleviate Salinity-Induced Growth Inhibition in in Vitro Shoot Cultures of Tobacco (Nicotiana tabacum L.).

Salinity stress is a major environmental constraint that limits plant growth and productivity by disrupting ionic balance, inducing osmotic stress, and promoting oxidative damage. Chitosan (Ch), a naturally derived biopolymer, enhances plant tolerance to abiotic stresses, but its effectiveness may be limited by poor solubility and reduced interaction with plant tissues. Chitosan nanoparticles (ChNP) may overcome these limitations through improved bioavailability and cellular interaction. This study evaluated the effectiveness of Ch and ChNP in alleviating salinity stress in in vitro shoot cult

Waterlogging tolerance in pigeonpea: current understanding of physiological responses, genetic resources, and genomic prospects.

Waterlogging is an escalating threat to pigeonpea (Cajanus cajan L. Millsp.) production in tropical and subtropical regions, where increasingly erratic monsoon rainfall and poorly drained Vertisols frequently expose crops to root-zone hypoxia during germination and early vegetative growth. Waterlogging in pigeonpea has historically been associated with substantial production losses in affected areas; however, the magnitude of these losses depends on stress duration, developmental stage, genotype, environmental conditions, and post-stress recovery. Oxygen deprivation disrupts root respiration,

Structure-Guided Development of Selective HDAC4 and HDAC7 Inhibitors That Modulate Th17 Cell Transcription.

Class IIa histone deacetylases (HDACs) are attractive therapeutic targets due to their specialized regulatory functions and restricted expression patterns. Recent studies identified HDAC4 and HDAC7 as critical regulators of Th17-cell differentiation, acting through distinct transcriptional mechanisms to promote Th17 lineage commitment. Building on these findings, we designed, synthesized, and characterized a series of selective HDAC4/7 inhibitors derived from the Class IIa scaffold TMP269. Structure-guided optimization of the metal-proximal and hydrophobic-binding regions generated analogs wit

Optimization of enzymatic hydrolysis for generating potential multifunctional peptides from goat milk: Bioactivity evaluation, structural characterization and molecular interaction analysis.

Food-derived bioactive peptides have attracted increasing interest for functional food development. In this study, goat milk proteins were enzymatically hydrolyzed to generate peptides with inhibitory activities against α-glucosidase (α-G), dipeptidyl peptidase-IV (DPP-IV), and xanthine oxidase (XO). Protease screening and response surface methodology were applied to optimize the composite hydrolysis process. The optimized goat milk hydrolysate exhibited inhibitory activities of 73.05 ± 1.38%, 88.63 ± 2.76%, and 80.69 ± 1.65% against α-G, DPP-IV, and XO, respectively. The active peptide fracti

Diverse oxidosqualene cyclases for triterpenoid biosynthesis.

Triterpenoids, a significant group of secondary metabolites, possess diverse skeletons and excellent pharmacological activities and are widely applied in cosmetics, medicine, and biopesticides. Traditional plant extraction and chemical synthesis cannot meet the needs of commercial production, and heterologous biosynthesis of triterpenoids has become a promising strategy. Oxidosqualene cyclases (OSCs) are the key enzymes that determine triterpenoid biosynthesis. However, most OSCs exhibit low heterologous expression, low catalytic activity, and promiscuous catalytic products, which impedes the

PXL: a nucleic acid-binding module of the promyelocytic leukemia protein.

The promyelocytic leukemia protein (PML) is a stress-response factor that assembles into PML nuclear bodies, dynamic subnuclear compartments involved in tumor suppression and antiviral defense. The most abundant isoform, PML-1, has been linked to transcriptional regulation, genome stability, and antiviral responses, yet the molecular basis of these functions remains unclear and is generally attributed to proteins associated with PML nuclear bodies. Here, we report that PML-1 itself harbors a previously structurally uncharacterized nucleic acid-binding domain. The domain, termed PXL, is encoded

Multi-enzyme cascade reactions synthesize diverse structurally defined glycoside conjugates.

Protein glycosylation serves essential biological functions. While glycosyltransferases catalyze protein glycosylation, their practical applications are limited by the dependence on high-cost nucleotide sugar donors. In this work, we bypass these nucleotide sugar intermediates and develop a coupled biocatalytic system with monosaccharide donors, allowing economical synthesis of structurally defined glycoconjugates.
