Nanohydrogel Composite Vaccine Capable of Bypassing the Blood-Brain Barrier and Targeting Tumors for Glioblastoma Immunotherapy via Intranasal Immunization.

Background: Glioblastoma (GBM) is a primary malignant tumor of the central nervous system and has a high lethal rate despite therapeutic advances. Although immunotherapies have achieved great success in solid tumors, the highly immunosuppressive tumor microenvironment and the blood-brain barrier (BBB) obstruction hinder the development of immunotherapies for GBM. In this study, we innovatively developed a nanohydrogel composite vaccine (nanoCOM-GEL) for GBM immunotherapy via intranasal immunization. Methods: The nanoCOM-GEL used GelMA as the hydrogel matrix and was co-formulated with antigenic

Accounting for variable detection functions in temporal abundance modeling via transfer learning.

Relative abundance, measured as the number of animals caught per unit of sampling effort (catch per unit effort, CPUE), is commonly used to monitor fish and wildlife populations, largely because sampling methods are cost-effective to implement. Modeling relative abundance from CPUE data, however, requires the assumption that the detection probability is constant across sampling events. This assumption is likely not valid, as the probability of detection often varies as a function of several factors, including the characteristics of individual animals and environmental conditions at the time of

Fabrication and Application of Injectable Bpei/Graphene Oxide/Silver Nanoparticle Hydrogel for Wound Healing.

Acute full‑thickness skin wounds represent a significant clinical challenge due to tissue loss, risk of bacterial contamination, and the need for rapid and effective regeneration. Injectable thermosensitive hydrogels incorporating antibacterial nanomaterials represent a promising strategy for advanced wound care. In this study, a multifunctional hydrogel composed of Graphene oxide (GO), branched polyethyleneimine (BPEI), and silver nanoparticles (AgNPs) dispersed in Pluronic F127 was developed and evaluated for wound healing applications. The GO-BPEI-AgNP nanocomposite was synthesized via amid

Luteinizing hormone promotes functional remodeling of immature porcine Leydig cells associated with DNA methylation changes.

Luteinizing hormone (LH) is a central regulator of testicular development, while the epigenetic mechanisms underlying LH-mediated regulation of immature Leydig cell (iLC) development and function warrant further investigation. In the present study, graded administration of a gonadotropin-releasing hormone agonist (GnRH-a) established differential serum LH levels in Shaziling boars. Reduced LH levels were associated with delayed testicular development and increased body weight, body length, and chest circumference. In vitro, LH treatment promoted G1-to-S phase progression and cell proliferation

Targeting NLRP3 with GDC-2394 ameliorates airway inflammation and smooth muscle hyperplasia in experimental asthma by suppressing the IL-6/JAK2/STAT3 axis.

Uncontrolled childhood asthma contributes to chronic airway inflammation and remodeling, primarily characterized by bronchial smooth muscle hyperplasia. Although the NLR family pyrin domain-containing 3 (NLRP3) inflammasome regulates airway inflammation, its role in abnormal airway smooth muscle cell (ASMC) proliferation remains poorly understood. Here, we evaluated the selective NLRP3 inhibitor GDC-2394 in a house dust mite (HDM)-induced murine model of asthma with a vehicle control group, focusing on the NLRP3-IL-6/JAK2/STAT3 signaling axis. HDM-challenged mice displayed increased airway hyp

Limited Antibodies to Henipaviruses Detected Among Nigerian Horses Using Pseudotyped Virus Neutralization Tests.

The Hendra virus (HeV; Henipavirus hendraense) and Nipah virus (NiV; Henipavirus nipahense), members of the genus Henipavirus, are high-consequence pathogens that have caused multiple zoonotic outbreaks with high mortality. Frequently, pigs or horses have acted as bridging hosts between bats, the reservoir hosts, and people. We used pseudotyped viruses with the fusion (F) and glycoprotein (G) envelope proteins of the NiV and Ghana virus (GhV; Henipavirus ghanaense) in neutralization tests (PVNTs) to screen serum samples from 138 apparently healthy horses in northwest Nigeria. No antibodies to

Sarcolipin is a Conformation-Dependent Regulator of the Sarcoplasmic Reticulum Calcium Pump SERCA.

Muscle relaxation is enabled by the sarco-endoplasmic reticulum Ca2+-ATPase (SERCA), which removes calcium from the cytosol and returns it to the lumen of the sarcoplasmic reticulum. During transport, SERCA adopts a variety of conformational states which differ in their structure and affinity for substrates. The transition between these states is mediated by calcium and ATP binding and the formation of an aspartyl-phosphate intermediate, which enables the transport of calcium across the membrane. SERCA function is highly regulated because of the importance of calcium in processes such as muscl

Latitudinal variation in seasonal migration of Eurasian curlews Numenius a. arquata breeding in Great Britain: breeding failure advances departure timing in southern populations.

BACKGROUND: Migration plays a crucial role in the annual cycle of migratory birds. The Eurasian curlew Numenius arquata arquata, a species of high conservation concern in Great Britain, exhibits a wide range of migratory behaviours across its western distribution, from resident and short-distance movements to long-distance migrations along the East Atlantic Flyway. However, migratory behaviours of British-breeding curlews remain poorly studied. In particular, the ecological and life-history factors underlying variation in migration, such as timing and duration, remain unclear in this declining

The Centriole Biogenesis Cycle: Temporal Mechanisms, Autonomy and Coupling to the Cell Cycle and Other Cellular Clocks.

Centrioles are cytoskeletal organelles whose duplication must occur with high temporal precision to ensure faithful cell division, ciliogenesis and tissue organization. Traditionally, the centriole biogenesis cycle has been viewed as a surrogate to the nuclear division cycle, particularly to oscillations in cyclin-dependent kinase (CDK) activity. In this chapter, we revisit classic experimental observations demonstrating that centriole duplication can proceed independently of DNA replication, transcription and CDK oscillations, thereby revealing an intrinsic autonomy in this cycle. We then dis

Hematological Profiles and Oxidant-Antioxidant Status in Native Turkish White Geese: A Preliminary Comparison Between Young and Adult Geese of Both Sexes.

Hematological parameters and indicators of oxidant-antioxidant balance could provide useful information about the physiological characteristics of poultry. However, information on these parameters in native Turkish white geese of different ages and sexes remains limited. This study evaluated 40 clinically healthy native Turkish white geese aged 16 or 98 weeks (10 females and 10 males per group). Hematology was assessed manually; plasma total antioxidant status (TAS), total oxidant status (TOS), and oxidative stress index (OSI) were measured with commercial kits. Two-way ANOVA was followed by B

Evaluating larvicide performance against Culicoides sonorensis in semi-field conditions.

BACKGROUND: Culicoides biting midges are small flies that are considered a nuisance pest that also can serve as vectors for a number of human and veterinary-important pathogens. This study evaluated the efficacy of larvicide products against C. sonorensis populations under controlled semi-field conditions and investigated how environmental factors influence the degradation and persistence of the larvicide active ingredients (AIs) in an artificial larval habitat at three study locations (Lubbock, TX; Manhattan, KS; and Slidell, LA). RESULTS: All tested larvicide products resulted in larval mor

Synthetic Cell-Based Approaches to Dissect Autonomous Cellular Oscillators: Recent Applications on the Cell Cycle.

Cell-free extracts have proven to be a tremendous tool for studying cell cycle regulation. This chapter reviews recent advances that expand upon this platform by incorporating synthetic cell-like compartments to encapsulate functional cell cycle regulators. These synthetic approaches allow for a programmable reconstitution of self-sustained autonomous oscillators, including that of mitosis, exhibiting a diverse range of biochemical and physical properties. The high throughput and manipulability of this system provide a comprehensive tool to systematically analyze the single-cell dynamics of th

Inclusion of Curcumin in the Diet of Prepartum Cows and Its Effects on Animal Health and Production.

This study aimed to evaluate whether adding curcumin to prepartum cows' diets positively affects metabolism, immune response, milk and colostrum production and quality. Forty Holstein cows were divided into two homogeneous groups, with 20 animals per treatment, balanced according to milk production (kg/day) during the previous lactation and parity. The experiment was conducted in a continuous trial, with each groups of animals randomly assigned to one of the treatments: (a) addition of curcumin to the diet (152 mg/day) in the prepartum period; (b) without additive, used as control. The study w

Integrative copy-number and dependency mapping in canine high-grade nodal lymphoma identifies candidates for targeted therapy.

High-grade nodal lymphomas (HGNL) in dogs are aggressive malignancies and important comparative models for human non-Hodgkin lymphoma; however, their copy-number architecture remains incompletely defined. This study applied high-density SNP arrays optimized for FFPE DNA to 90 canine HGNLs (77 B-cell [BCL], 13 T-cell [TCL]) to map genome-wide copy-number alterations (CNAs) and recurrent copy-number altered regions (CNARs), and to integrate them with candidate gene drivers and potential therapeutic targets. Across immunophenotypes, CNAs were dominated by gains on CFA13, syntenic to human 8q24 an

Impact of Climate Change on Farm Animals: Physiological Mechanisms, Health and Productivity, and Integrated Adaptation Strategies.

Climate change is accelerating the frequency of extreme thermal environments, threatening animal welfare, global herd productivity, and agricultural economics. Heat stress-occurring when the ambient temperature-humidity index (THI) surpasses species-specific thermoneutral thresholds-initiates physiological heat-loss cascades (panting, sweating, and peripheral vasodilation), neuroendocrine disruptions (depressed thyroid hormones, elevated glucocorticoids, and insulin dysregulation), mitochondrial oxidative stress, and gut-barrier breakdown. These systemic disruptions drive substantial performan

A Seven Amino Acid Long Region in the Cardiac Cav1.2, Encompassing Ser1487, Is Essential for Mice Embryonic Development and "Fight or Flight" Responses.

The cardiac calcium channel Cav1.2 is essential for embryonic development, cardiac excitation-contraction coupling, and the "Fight or Flight" response. Regulation of Cav1.2 by protein kinase A (PKA) phosphorylation is critical to this process, though the underlying mechanism remains contentious. We previously identified serine 1458 (S1458) in the proximal C-terminus of the human Cav1.2 as essential for PKA-mediated regulation in vitro. To investigate its functional role in vivo, we generated three mouse models targeting S1487, the mouse equivalent of human S1458. Comprehensive phenotyping and

METTL3/IGF2BP1 Axis Orchestrates m6A-Dependent NCAM1 Preservation to Combat Age-Related Cognitive Decline.

Age-related decline in learning and memory functions poses significant challenges in an aging society, with epigenetic dysregulation emerging as a key contributor to cognitive deterioration. As the most prevalent internal RNA modification, N6-methyladenosine (m6A) dynamically orchestrates neural transcriptome plasticity through its "writers," "erasers," and "readers," yet its role in aging-associated cognitive impairment remains underexplored. This study employs an integrated epitranscriptomic approach to investigate m6A-mediated regulation in hippocampal aging processes. Through comparative m
