A tumor microenvironment-activatable CuS@TF nanosystem with intrinsic multi-nanozyme properties for NIR-II photoacoustic imaging-guided synergistic PTT/CDT.

The efficacy of single-modal chemodynamic therapy (CDT) is often restricted by the antioxidant scavenging of overexpressed glutathione (GSH) within the tumor microenvironment (TME) and insufficient catalytic kinetics. Meanwhile, traditional photothermal therapy (PTT) also faces bottlenecks of a single therapeutic mechanism and low photothermal conversion efficiency, leading to unsatisfactory overall anti-tumor outcomes. To address these challenges, we constructed a smart-responsive, core-shell multifunctional nanotheranostic platform for near-infrared-II (NIR-II) photoacoustic (PA) imaging-gui

Panax ginseng suppresses endothelin-1-induced cardiomyocyte hypertrophy through inhibition of the local angiotensin II-AT1 receptor signaling axis.

Panax ginseng (PG) is a traditional herbal medicine widely used for the treatment of cardiovascular disorders in East Asia. Although PG has been reported to attenuate cardiomyocyte hypertrophy, the molecular mechanisms underlying its cardioprotective effects remain incompletely understood. In this study, we investigated whether modulation of the endothelin-1 (ET-1)-angiotensin II (AngII) signaling axis contributes to the anti-hypertrophic action of PG. Neonatal rat ventricular myocytes (NRVMs) were used to evaluate ET-1-induced hypertrophy, and pharmacological experiments using receptor antago

Biochar mitigates microplastic- and nanoplastic-induced soil degradation and plant stress: a global meta-analysis.

Microplastic (MP) and nanoplastic (NP) contamination threatens soil functioning and plant performance, yet the effectiveness of biochar in mitigating these impacts varies substantially among experimental conditions. We conducted a meta-analysis of 59 peer-reviewed studies comprising 1418 effect-size estimates to quantify biochar-mediated responses across the soil-plant continuum. Effect sizes were calculated as the natural logarithm of the response ratio (lnRR) and analyzed using multilevel random-effects models. Subgroup analyses examined the effects of growth medium, plant functional group,

Unraveling Tyrosine Hydroxylase in the Brain of a Crustacean: An Integrative Investigation Using Different Approaches.

Tyrosine hydroxylase (TH), the rate-limiting enzyme in dopamine (DA) synthesis, is conserved across most animal phyla. DA regulates motor activity, sensory processing motivation, neural homeostasis, and protection against oxidative stress. TH and DA have been described in vertebrates and in some invertebrates, including arthropods. This study investigated the anatomical location of TH and DA and characterized the biochemical and molecular structure of TH in the brain of the mangrove crab Ucides cordatus. This species is a useful environmental model for neurodegeneration studies, due to its occ

Glutamate coordinates with Lactobacillus amylovorus to attenuate intestinal damage induced by deoxynivalenol.

Deoxynivalenol (DON) is a prevalent trichothecene mycotoxin that threatens intestinal health, yet its effects on gut microbiota remain poorly understood. Here, Lactobacillus amylovorus isolated from Ningxiang pigs showed strong acid and bile tolerance but significantly impaired growth under DON exposure. Transcriptomic analysis revealed that DON disrupted amino acid metabolism in L. amylovorus, particularly glutamate metabolism. Among the amino acids tested, only glutamate supplementation restored bacterial growth under DON exposure. Futhermore, co-supplementation with glutamate and L. amylovo

The role of the mitochondrial phosphate carrier in modulating mitochondrial homeostasis and enhancing radiosensitivity in tumor cells.

BACKGROUND: The mitochondrial phosphate carrier (PiC) is a key protein in maintaining mitochondrial homeostasis, and its dysfunction is closely associated with cell fate determination. Recent studies suggest that PiC may regulate mitochondrial permeability transition pore (MPTP) opening. However, its precise role during radiotherapy and the mechanisms by which it regulates cell death remain elusive. METHODS AND RESULTS: Focusing on the clinical challenge of radioresistance in glioma, this study reveals for the first time that radiation specifically suppresses PiC expression, thereby inducing

CYP1B1 is a potential target of kaempferol in renal fibrosis: a multi‑omics and single‑cell transcriptomics study.

BACKGROUND: Renal fibrosis (RF) is a progressive and irreversible pathological process for which effective treatments are limited. This study explores the molecular targets and mechanisms through which kaempferol (Kae) ameliorates RF, with particular emphasis on cytochrome P450 1B1 (CYP1B1). METHODS: Using a combination of transcriptomic profiling, weighted gene co‑expression network analysis (WGCNA), multi‑algorithm machine learning [least absolute shrinkage and selection operator (LASSO), support vector machine‑recursive feature elimination (SVM‑RFE), and Random Forest], and single‑cell tra

The miR-6631-5p/ACYP2 Axis Orchestrates Myogenesis and Slow-Twitch Muscle Fiber Specification in Broilers.

Poultry skeletal muscle fiber composition, primarily defined by oxidative slow-twitch and glycolytic fast-twitch fibers, is a major determinant of meat quality. Yet the miRNA-mRNA regulatory mechanisms controlling fiber type specification remain largely unresolved. Here, we compared the transcriptomic profiles of pectoralis major (PM) and soleus muscle (SM) in Tianfu broilers and identified gga-miR-6631-5p as a key slow-twitch-associated microRNA (miRNA) through small RNA sequencing and weighted gene co-expression network analysis. Mechanistically, acylphosphatase 2 (ACYP2) was validated as a

Functional Diversity Responses to Temperature Vary Spatially and With Protection Status in Wintering Waterbird Assemblages.

Wetlands support disproportionately high biodiversity, including waterbirds, which underpin key ecological functions and regulatory services. This has led to efforts to protect these crucial sites, but whether these protections can maintain waterbird assemblages in the face of new threats such as climate change is poorly understood. Here, we calculated functional redundancy and three metrics of functional diversity from over 45,000 waterbird censuses, spanning four decades of rising winter temperatures for 2792 protected and unprotected sites in Europe. We modelled the effect of average winter

Swollen lysosomes grab Rabs for a CASM-free route to LRRK2 activation.

Multiple lysosomal stresses activate Parkinson's-linked LRRK2 kinase, and these involve the so-called "CASM" pathway to help recruit LRRK2 to lysosome surfaces. Clegg et al. (https://doi.org/10.1083/jcb.202511137) show that LRRK2, activated upon lysosome swelling caused by PIKfyve inhibition, instead relies solely on Rab GTPase binding for lysosome localization and activation.

Phytometabolite profiling and immunomodulatory potential of Aegle marmelos leaves in Concanavalin-A stimulated chicken splenocytes model system.

Aegle marmelos (L.) Correa is reported to possess antioxidant, anti-inflammatory and metabolic regulatory properties; however, its effects in chicken splenocytes remain insufficiently explored. This study employed an integrated approach combining phytochemical, elemental and gene-expression analyses employing metabolomic (Untargeted UHPLC-ESI-QTOF-MS/MS and GC-MS), ionomic (ICP-MS) and quantitative reverse transcription-PCR (qRT-PCR) based gene-expression analyses to characterise Aegle marmelos aqueous leaf extract (AME) and investigate its immunomodulatory effects in Concanavalin-A stimulated

Multiple sources contribute to predictive tactile attenuation in the mouse whisker system.

The brain predicts action outcomes using interacting neuronal systems that integrate sensory feedback, movement, and rewards. In the mouse whisker system, we investigated whether two presumed predictive mechanisms jointly attenuate ascending tactile signals during movement. To this end, we employed an open-loop whisker-reach task that monitored facial nucleus motor activity and presented experimental tactile consequences while measuring tactile attenuation in the barrel cortex. Experimentally manipulating sensory consequences revealed two distinct systems. Sensory gating, a previously document

Chemical Study of the Marine Sponge Myrmekioderma gyroderma: Biogenetic and Evolutionary Adaptation Shared with Terrestrial Plants?

AbstractThe Yucatan Peninsula contains a richness of organisms and invaluable knowledge beneath its marine waters. However, much of this value remains hidden and unknown; it has also been threatened by global climate change and other anthropogenic pressures. In this research, we present a chemical investigation of the marine sponge Myrmekioderma gyroderma, collected in the Yucatan Peninsula, from which we isolated the compound 13-Oxo-geranyllinalool, a derivative of geraniol previously isolated only from terrestrial plants. This chemical study is the first to investigate the secondary metaboli

Interspecific Comparative Lectin-Binding Analysis of Sea Urchin Adhesive Tube Feet.

AbstractGlycans have been consistently reported as important constituents of nonpermanent adhesives produced by aquatic animals across multiple metazoan phyla. However, lectin-binding patterns associated with these glycans may vary among taxa, potentially reflecting phylogenetic relationships or adaptations to local environmental conditions. Despite growing interest in bioadhesion, the number of species investigated remains limited, constraining broader evolutionary interpretations. Here we present a large comparative analysis of 15 sea urchin species representing four orders, seven families,

Community Composition of Sessile Invertebrates on Underwater Cultural Heritage Differs from Natural Reefs in Palau.

AbstractShips, planes, and other vessels that come to rest on the seafloor introduce metals to the marine environment. Underwater cultural heritage (UCH) sites become important habitats for sessile invertebrates, but the materials they are made from can impact the composition of species living on them. We collected and analyzed images from three sites in Palau (two shipwrecks and one plane wreck from WWII) along with directly adjacent (1-5 m away) naturally occurring coral reefs. Community composition on UCH after ∼80 years of submergence was significantly different from adjacent coral reefs,

Global oxidative stress estimation using chemiluminescence for evaluating semen quality in boars with different fertility indices.

Oxidative stress (OS) in ejaculates is a key indicator of semen quality. The aim was to estimate global OS using chemiluminescence and malondialdehyde (MDA) based methods in fresh (0h) and extended preserved (48h and 96h) semen of boars with different fertility indices. Based on the boar fertility index (BFI >10, ∼7-8 and <6), boars (n = 6) with three fertility (high, medium and low) levels were used to collect semen (n = 72). Routine semen evaluation tests along with OS estimation by MDA and chemiluminescence methods were done in fresh (0h) and extended semen at 48, 96 h of preservation. In r

Cryopreservation of chicken feather follicle-derived cells using chicken serum-based media: broadening cellular diversity in avian biobanking.

Feather follicles are dynamic and heterogeneous mini-organs containing epithelial, mesenchymal, pigmentary, and progenitor cell populations. However, cryopreservation studies have focused mainly on fibroblasts, and other follicular populations remain poorly explored. This study aimed to establish favorable cryopreservation conditions for cells isolated from growing feathers of Gallus gallus domesticus using chicken serum. Primary cultures were assessed by morphology, differentiation-associated staining, and reverse transcription quantitative polymerase chain reaction (RT-qPCR). Cells were cryo

Photobiomodulation and the Wnt/β-catenin pathway in apexogenesis: Rationale and evidence gaps.

Apexogenesis is a vital pulp therapy that preserves vascularized pulp in immature teeth and relies on dental pulp stem cells (DPSCs) and stem cells from the apical papilla (SCAPs) for continued root growth and apical closure. Wnt/β-catenin signaling has an established role in odontoblast differentiation, reparative dentinogenesis, and root development, although normal root development requires regulated rather than simply increased Wnt activity. Photobiomodulation therapy (PBMT) is proposed to act through mitochondrial photoreception, and in vitro studies report increased expression of odontog

Movement regimens drive region-specific humeral adaptation: whole-bone insights from an experimental animal model.

Repetitive movements impose distinct loading regimens on long bones, potentially eliciting predictable osteogenic responses and morphological change. Cortical responses to such movements are seldom investigated along whole diaphyses as opposed to selected locations. Morphometric mapping is a technique that is well-suited for comprehensively examining whole diaphyses. To assess responses to different movement regimens, we evaluated humeri of thirty-five female C57BL/6J mice raised from weeks 4 to 20 post-birth in one of three scenarios: linear-dominated (n = 12), turning-dominated (n = 12), and
