OBJECTIVE: Lymph nodes within the parotid gland (PG) are known as 1) an origin of Warthin's tumors and 2) a metastatic focus of nearby cancers. This study aimed to investigate factors that provide the specific features.
METHODS: Using histological sections from 28 human fetuses at approximately 15-31 weeks, we examined developing acini, ducts and lymph vessels (LVs) in the PG and compared the results with the other exocrine glands.
RESULTS: In the PG at 15-16 weeks, developing acini were embedded in a meshwork of LVs that corresponds to a major lymphatic route from the posterolateral head to

Striated duct adenoma (SDA) is a rare benign salivary gland neoplasm with limited cytologic characterization. We report a case of SDA with cytologic, histologic, immunohistochemical, and molecular correlation to further define its morphologic spectrum and diagnostic considerations. The patient was a 62-year-old male with a submandibular gland mass. Cytologically, the specimen demonstrated loose clusters of monomorphic epithelial cells ranging from cuboidal to rare elongated or slightly spindled morphology, with moderate amounts of variable oncocytic cytoplasm. Nuclei were ovoid and normochroma

Salivary gland (SG) hypofunction significantly affects the life quality of patients with hyposalivation. Current treatment options for these conditions are limited and often result in undesirable side effects. To address this issue, we've developed a developmentally inspired strategy to derive salivary gland epithelial progenitor (SGEP) cells from human pluripotent stem cells (PSC). Then we employ SGEP cells derived from human PSC with a SOX10::GFP reporter to engineer three-dimensional lumenized SG organoids, with well-defined lumens and secretory function, reminiscent of human salivary gland

OBJECTIVES: IgG4-related disease (IgG4-RD) involves elevated serum IgG4 and several autoantigens, but the role of different antibody isotypes in pathogenesis remains unclear. This study systematically characterized isotype‑specific autoantibody responses against representative autoantigens and evaluated their clinical and immunological correlations.
METHODS: We enrolled 61 patients with IgG4‑RD, 31 with systemic lupus erythematosus (SLE), 11 with Sjögren's syndrome (SS), and 30 healthy controls (HC). Serum levels of isotype‑specific autoantibodies were measured using in‑house ELISA. Correlati

Benign submandibular gland tumors are usually resected through a transcervical incision, which provides direct exposure but leaves visible cervical scarring. Remote-access endoscopic techniques improve scar concealment but can be limited by two-dimensional visualization, assistant-dependent camera control, and rigid instrument collision in the narrow submandibular gland space. This article presents a standardized video protocol for robot-assisted submandibular gland excision via a gasless trans-hairline approach for benign tumors. The procedure begins with postauricular trans-hairline incision

RATIONALE: NUT carcinoma (NC) is a rare and highly aggressive malignancy defined by rearrangements of the NUTM1gene. Primary NC of the submandibular gland is exceptionally rare, with nonspecific imaging features that often lead to misdiagnosis as common salivary gland tumors and delayed treatment.
PATIENT CONCERNS: A 32-year-old man presented with a painless mass in the left submandibular region that rapidly enlarged from 2 cm to approximately 4 cm within 1 month, accompanied by the development of ipsilateral pharyngeal and periauricular pain.
DIAGNOSES: Preoperative multimodal imaging-compu

Excessive sugar intake contributes to noncommunicable diseases, including dental caries. According to the Developmental Origins of Health and Disease (DOHaD) theory, early-life nutrition influences organ development and future disease susceptibility. This study examined the impact of high-sucrose intake during the growth period on salivary gland development in rats and its potential influence on dental caries after growth. Male Wistar rats were divided into two groups: a high-sucrose group (HS), receiving a 15% sucrose solution, and a control group (C), receiving distilled water. Measurements

BACKGROUND: 131I therapy is a targeted therapy for patients with differentiated thyroid cancer (DTC) after surgery, reducing the risk of tumor recurrence and metastasis. Radioactive iodine can accumulate in the salivary glands, causing radiation damage. Dynamic imaging of the salivary glands can objectively assess the morphology and function of the salivary glands, making it a simple and non-invasive diagnostic method. The purpose of this study was to quantitatively analyze salivary gland function changes in patients with DTC undergoing postoperative 131I therapy using salivary gland dynamic i

BACKGROUND: Submandibular sialolithiasis is a common condition that can cause significant discomfort and complications if left untreated. Treatment strategies depend largely on the size and location of the sialolith, with various surgical and nonsurgical techniques available.
CASE PRESENTATION: This report describes the successful management of a large sialolith in the submandibular gland via sialadenectomy using an extraoral approach. The case highlights the importance of a comprehensive diagnostic and therapeutic strategy for managing large sialoliths and demonstrates the efficacy of sialad

Pediatric submandibular sialolithiasis with a salivary cutaneous fistula is exceptionally rare. A 15-year-old boy presented with a 4-month history of a persistent right submandibular inflammatory lesion with a central opening and hypergranulation despite previous incision and drainage. Contrast-enhanced computed tomography demonstrated a 10.8 mm distal Wharton duct stone, a 34 mm inflammatory lesion extending toward the skin, and marked atrophy of the right submandibular gland. Because of persistent inflammation, fistula formation, and severe glandular atrophy, the fistulous tract and affected

BACKGROUND: Large-cell neuroendocrine carcinoma (LCNEC) is an aggressive malignancy that commonly arises in the lungs. Primary salivary gland involvement is exceptional, presenting a major diagnostic challenge in differentiating it from metastatic pulmonary disease.
CASE PRESENTATION: A 62-year-old man presented with a right submandibular mass that had been stable for 16 years before rapidly expanding. Imaging revealed a massive 11 cm necrotic submandibular tumor with disseminated pulmonary and hepatic metastases. Following partial debulking, pathology revealed a high-grade neuroendocrine tum

OBJECTIVES: Immunoglobulin G4-related disease (IgG4-RD) is a systemic, immune-mediated disease-causing inflammation and fibrosis in multiple organs, often associated with elevated serum IgG4 concentrations. Whether IgG4-RD is associated with malignancy remains unclear. We investigated whether there is an association between IgG4-RD and incident malignancy.
METHODS: We performed a single-centre, retrospective cohort study of patients who fulfilled the American College of Rheumatology/European Alliance of Associations for Rheumatology IgG4-RD Classification Criteria. Malignancy history was coll

Sensory neurons have been increasingly recognized as vital contributors to deep tissue function. However, how these specialized neurons contribute to salivary gland function remains largely undefined. Here, we uncover a role for trigeminal somatosensory afferents in salivary gland perception and function using in situ -based classification, in vivo calcium imaging, behavioral assays, and targeted ablation. Retrograde labeling from the submandibular gland complex revealed substantial direct innervation from trigeminal neurons. Further categorization confirmed that Trpv1 + sensory neurons provid

Sjögren's disease (SD) is an autoimmune condition characterized by chronic inflammation and impaired secretory function of the salivary glands (SG). The proresolving lipid mediator AT-RvD1 restores SG function in SD-like mice; however, its tissue pharmacokinetics, exposure-response relationships, and human dose predictions remain poorly understood. Therefore, this exploratory study characterizes AT-RvD1 tissue pharmacokinetics, develops an SG-focused physiologically based pharmacokinetic (PBPK) model, explores the relationship between SG exposure and secretory response, and predicts human expo

Palisading adenocarcinoma is a recently recognized, biphasic salivary gland tumor with a distinctive histomorphological pattern. Its low rate of local recurrence and absence of documented metastatic potential make accurate recognition clinically relevant. We report two cases of palisading adenocarcinoma arising in the sublingual gland of female patients aged 39 and 54 years. Both tumors were well-circumscribed and encapsulated, and showed solid growth composed of trabecular and islet-like structures with prominent peripheral palisading, admixed with ductal elements, with focal necrosis observe

Eosinophilic sialodochitis (ES) is an increasingly recognized cause of recurrent sialadenitis but likely remains underdiagnosed as it is commonly considered to be nonspecific chronic sialadenitis. ES is likely increasing in prevalence in association with the growing number of allergic diseases and should be considered in patients presenting with multiple episodes of salivary gland pain and swelling refractory to conventional treatment. We highlight this difficulty in diagnosis with a report of ES established following Wharton duct remnant swelling after submandibular gland (SMG) resection for

Metastasis of renal cell carcinoma (RCC) to the head and neck region is rare, accounting for approximately 8%-14% of all RCC metastases. The thyroid gland is the most common site of metastasis in this region. However, RCC is known for its ability to metastasize to unusual anatomical sites, including the salivary glands. Approximately 0.1% of metastatic salivary gland neoplasms originate from renal malignancies, and the parotid gland is the most frequently involved site. Metastases to the submandibular and sublingual glands are considerably less common. In this report, we present a 62-year-old
