An anatomical evaluation of the submental artery in a select South African population: a cadaveric study.

BACKGROUND: Understanding the anatomical variation of the submental artery (SA) is crucial for dissection of the submental region. Failure to recognize variations in this artery can lead to undesirable outcomes when using the SA island flap. This study aimed to evaluate the anatomical characteristics of the SA in a select donor cohort housed at our institution. MATERIALS AND METHODS: This cohort consisted of 19 adult human donor cadavers (n = 38; age range: 52-99 years) used for anatomy education and research. The submental regions were dissected to document the anatomical parameters of the S

Altered mitochondrial ultrastructure in salivary epithelial cells of patients with Sjögren's disease is associated with mitochondrial DNA release and increased activation of pattern recognition receptors: potential use of tofacitinib as therapy.

OBJECTIVES: This study aimed to analyse structural and functional mitochondrial alterations, the release of mitochondrial DNA (mtDNA), and the activation of inflammatory signalling pathways that can be reversed by tofacitinib in the salivary glands (SG) of patients with Sjögren's disease (SjD). METHODS: SG from patients with SjD and controls, as well as from mice with SjD treated with or without tofacitinib, were analysed. We determined the mitochondrial ultrastructure, the presence of mtDNA in the cytosol, and the levels and localisation of pattern recognition receptors (PRRs) that recognise

Gland-preserving surgery versus total gland excision for benign submandibular tumors: A systematic review and meta-analysis.

BACKGROUND: Preservation of gland function is gaining importance in the surgical management of benign submandibular gland (SMG) tumors, yet comparative evidence between gland-preserving and total excision approaches remains limited. This study aimed to compare functional, oncologic and cosmetic outcomes between submandibular gland-preserving surgery (SGPS) and total submandibular gland excision (SGTE) for benign SMG tumors. METHODS: A systematic review and meta-analysis were conducted using studies retrieved from PubMed, Embase, and the Cochrane Library. The primary outcome was postoperative

Submandibular gland oncocytoma: Imaging features and histopathological correlation of a rare benign salivary gland tumor.

Submandibular gland oncocytoma is a rare benign epithelial tumor accounting for less than 1% of all salivary gland neoplasms. It is composed of oncocytes-large polygonal epithelial cells with abundant eosinophilic granular cytoplasm resulting from mitochondrial accumulation-and typically affects adults between the fifth and seventh decades of life, without a clear gender predilection. Radiation exposure has been reported as a possible predisposing factor. Clinically, oncocytomas usually present as painless, slow-growing masses. Imaging modalities, particularly ultrasound and contrast-enhanced

Organ-specific local cytokine release syndrome after anti-CD19 CAR-T therapy with salivary gland involvement: a case report, literature review, and a diagnostic alert for tocilizumab-associated cervical swelling.

Background. Cytokine release syndrome (CRS) is the most frequent toxicity after chimeric antigen receptor T-cell (CAR-T) therapy and typically presents as a systemic inflammatory syndrome. In recent years, a localized form of CRS (L-CRS), most commonly involving the craniocervical region, has been increasingly recognized. L-CRS may occur with or without overt local tumor involvement and can progress to airway impairment, requiring prompt recognition and treatment. Case presentation. We report two cases of patients with refractory diffuse large B-cell lymphoma (DLBCL) treated with anti-CD19 CAR

Partial Submandibular Gland Resection in Deep Neck Surgery: An Assessment of Safety Using the LigaSure Exact Dissector.

BACKGROUND: Optimal cervicomandibular contour often necessitates management of subplatysmal structures, including the submandibular glands (SMGs). Partial SMG excision may enhance neck definition but remains controversial because of concerns regarding bleeding and other potential complications. The LigaSure Exact Dissector (Medtronic, Minneapolis, MN) is a bipolar vessel-sealing device that has been adopted by some surgeons for SMG management during aesthetic neck surgery. OBJECTIVES: To evaluate the short-term safety and postoperative complication profile of partial SMG excision using the Li

Redefining Neck Rejuvenation: The Novel Hyoid-to-Mastoid Crevasse Neo-Ligament and OnderKaak Angle in Deep Plane Neck Lift.

BACKGROUND: Neck rejuvenation has evolved from superficial skin tightening to deeper structural modifications targeting the platysma, submental and subplatysmal fat, and the submandibular gland (SMG). Significant controversy surrounds the surgical maneuvers necessary to achieve the ideal neck. OBJECTIVES: The objective is to introduce the Hyoid-to-Mastoid Crevasse (HMC) neo-ligament for neck rejuvenation that elevates the SMG via a suture secured from the hyoid periosteum to the mastoid crevasse. Moreover, we introduce the OnderKaak (OK) angle, representing the region overlying the submandibu

Differential metabolic responses to hydrogen peroxide-induced oxidative stress in parotid and submandibular gland acinar cell lines.

This study explored how oxidative stress induced by hydrogen peroxide alters metabolite levels in submandibular and parotid salivary gland acinar cells. We investigated the impact of oxidative stress on the metabolite profiles of two salivary gland cell lines and examined differences in response. Rat-derived immortalized acinar epithelial cell lines from the parotid and submandibular salivary glands were exposed to 50 μM and 150 μM hydrogen peroxide for 24 h. Metabolite levels were then analyzed using liquid chromatography mass spectrometry, and succinate levels were independently assessed usi

Postoperative radiotherapy versus postoperative radiochemotherapy after surgery of salivary gland cancer: a systematic review and meta-analysis.

There is a lack of prospective clinical trials on the therapy of salivary gland cancer (SGC) due the rarity and large variety of histological subtypes. This study aimed to compare overall survival (OS) after primary surgery of SGC followed by postoperative radiotherapy (PORT) versus radiochemotherapy (PORCT) in curative intent. A systematic review and meta-analysis were performed on studies reporting OS after the two postoperative therapy modalities using hazard ratios (HRs) or allowing to calculate the HRs from the data. These studies were identified from PubMed, Web of Science, and Cochrane

Neonatal Submandibular Sialadenitis Without Abscess Formation: A Rare Cause of Neck Swelling.

Acute neonatal sialadenitis is a rare condition, most commonly involving the parotid gland, whereas isolated submandibular gland involvement is uncommon and may pose diagnostic challenges. We report the case of a two-week-old term male infant who presented with right submandibular swelling in the absence of fever, systemic signs of infection, or abscess formation. Laboratory evaluation revealed leukocytosis with elevated inflammatory markers, and ultrasound imaging confirmed acute submandibular sialadenitis. The infant responded well to intravenous antibiotics without the need for surgical int

The Impact of Alternate-Day Fasting on the Salivary Gland Ductal Compartments and the Differentiation Potential of Keratin 5+ Salivary Gland Progenitor Cells in an Induced Mouse Model of Sjögren's-like Hyposalivation.

Intermittent fasting confers protection in diverse diseases through various mechanisms, including the clearance of senescent and pathogenic cells, modulation of tissue inflammation and enhancement of stem/progenitor cell niche and functionality. Our previous study demonstrated the beneficial impact of alternate-day fasting (ADF) on xerostomia and sialadenitis, along with an improvement in salivary gland ductal compartments, where salivary gland progenitor cells reside, in non-obese diabetic mice, a spontaneous model of Sjögren's syndrome (SS). In the present study, we induced SS-associated hyp

Rapid SDS/trypsin decellularization of rat submandibular gland yields an ECM scaffold supporting salivary gland tissue engineering.

INTRODUCTION: Salivary gland hypofunction caused by radiation, Sjögren's syndrome, or other insults leads to xerostomia and currently lacks effective regenerative treatments. Decellularized organ-specific extracellular matrix (ECM) scaffolds offer a promising strategy toward gland restoration by recapitulating the native microenvironment. METHODS: In this study, we developed a rapid decellularization protocol for rat submandibular glands (SMGs) to create a biomimetic ECM scaffold for salivary gland tissue engineering. Following freeze-thaw pretreatment, detergent and enzyme incubations totale
