DDIAS shields vulnerable single-stranded DNA to direct neurogenesis and protect cancer cells.

Papers in Molecular Cell and Cell by Xue et al.1 and Tsukuda2 et al., respectively, reveal that DDIAS serves as an essential single-stranded DNA (ssDNA)-binding effector of the CIP2A-TOPBP1 complex that safeguards genome integrity by shielding post-replicative ssDNA lesions during mitosis, with implications for human cancer and neurodevelopment.

Taking the heat: Breaking the temperature limits of eukaryotic life.

What's the highest temperature at which complex life can grow? It's an old question. Here, Rappaport and colleagues describe a thermophilic amoeba that breaks the previous record, which was set at ∼60°C, and suggest the beginnings of an answer to what innovations enable this organism to take the heat.

Hepatic FoxO1 loss leads to pancreatic β cell deficiency via liver-derived C18/22/24 ceramides.

Preserving pancreatic β cells is requisite for antihyperglycemic strategy. Hepatic FoxO1 represents a promising therapeutic target. However, the systemic consequences on β cell homeostasis upon hepatic FoxO1 loss and the underlying mechanism remain largely unexplored. Using liver-specific Foxo1 knockdown mouse and transgenic zebrafish models, we uncover that hepatic FoxO1 loss leads to β cell deficiency via triggering apoptosis. Lipidomic and pharmacological analyses identify that hepatic FoxO1 loss increases the liver-derived C18/22/24 ceramides, which contribute to apoptosis by inhibiting th

Diet and Risk of Cardiovascular Disease.

Diet ranks among the most modifiable determinants of cardiovascular disease risk, yet the evidence base is vast, conflicting, and burdened by observational confounding. This narrative review synthesizes evidence on controversial dietary components and established heart-healthy dietary patterns. Moderate egg intake is acceptable within a high-quality dietary pattern; preparation method meaningfully modifies risk. Habitual moderate coffee consumption is neutral to modestly cardioprotective; unfiltered preparations elevate low-density lipoprotein cholesterol (LDL-C) through diterpene retention. N

Fluid amyloid-β (Aβ) biomarkers reflect early β-sheet-rich Aβ deposition during the preclinical stage in Alzheimer's disease model 5XFAD mice.

Early detection of disease progression using clinically-relevant biomarkers in animal models is important for mechanistic studies and for developing therapeutics in neurodegenerative diseases including Alzheimer's disease (AD). The preclinical stage of AD, when amyloid-β (Aβ) starts to accumulate before cognitive decline, provides a critical window for disease modification. In humans, decreases in cerebrospinal fluid (CSF) Aβ42 and the Aβ42/Aβ40 ratio in preclinical AD are considered to reflect the preferential sequestration of aggregation-prone Aβ42 into β-sheet-rich deposition in the brain,

Molecular mechanism of the LncRNA MALAT1/NF-κB signaling pathway in mediating vascular endothelial injury under glucose and lipid metabolism disorders.

BACKGROUND: Disordered glucose and lipid metabolism exacerbates endothelial injury, promoting atherosclerosis (AS). This study aimed to elucidate the role of the lncRNA MALAT1 in endothelial injury under high glucose and palmitic acid (HGPA) conditions via in vivo and in vitro experiments. METHODS: Bioinformatics identified MALAT1 as a key regulator. Functional assays, RIPpull, and Western blotting assessed its role in inflammation, apoptosis, and ferroptosis. Animal and clinical studies (100 AS patients vs. 100 controls) validated the findings. RESULTS: MALAT1 was upregulated by HGPA, activ

Widespread molecular evidence of Trypanosoma evansi among asymptomatic dogs in Iran.

Despite the widespread distribution of Trypanosoma evansi in camels and horses in Iran, only a few case reports of canine infection are available. We investigated the prevalence of trypanosomosis in 423 apparently healthy sheltered stray and owned dogs from five provinces representing different climatic regions of Iran (Hamedan, Kermanshah, Mazandaran, Yazd, and Khuzestan). Blood samples were examined microscopically and analyzed by PCR targeting the RoTat 1.2 variant surface glycoprotein (VSG) gene of T. evansi. RoTat-PCR detected T. evansi DNA in 97 dogs (22.9%), with provincial prevalences

The bile acid chenodeoxycholic acid attenuates renal fibrosis in mouse precision-cut kidney slices.

Renal fibrosis plays a central role in the progression of chronic kidney disease (CKD). Previous studies have reported that the bile acid receptors farnesoid X receptor (FXR) and Takeda G protein-coupled receptor 5 (TGR5) can protect against fibrosis development. However, these studies primarily relied on animal models or cell monocultures. In animal models, systemic influences can obscure kidney-specific effects, whereas cell monocultures lack the complex multicellular interactions that characterize kidney fibrosis. Here, we investigated whether the antifibrotic effects of FXR and TGR5 activa

Geometric morphometric analysis of cardiac shape changes associated with myxomatous mitral valve disease in Cavalier King Charles Spaniels.

Myxomatous mitral valve disease (MMVD) is a progressive cardiac disorder in Cavalier King Charles Spaniels (CKCS), characterized not only by cardiac enlargement but also by complex morphological remodeling of the cardiac silhouette. This study aimed to evaluate stage-dependent configurational changes in heart shape across the American College of Veterinary Internal Medicine (ACVIM) stages (A-D) using two-dimensional geometric morphometric analysis of thoracic radiographs. A total of 54 CKCS dogs (Stage A: n = 9, Stage B1: n = 11, Stage B2: n = 10, Stage C: n = 12, Stage D: n = 12) that underwe

Gut-Liver Axis and Multi-Omics Analysis Reveal the Protective Effect and Mechanism of Luteolin against Dibutyl Phthalate Induced liver injury.

Dibutyl phthalate (DBP), a widely prevalent environmental endocrine disruptor, has raised significant public health concerns regarding its hepatotoxicity. Luteolin (Lut) possesses anti-inflammatory, antioxidant, and multi-target regulatory properties, but the mechanisms underlying its protection against DBP-induced liver injury have not been fully elucidated. Here, we employed a systematic strategy...to investigate these effects and their core mechanisms. Network analysis predicted the PI3K-AKT and TNF signaling pathways as core pathways involved in both DBP-induced hepatotoxicity and Lut inte

Stepwise multi-scale reconstruction of cell spatial organization from single-cell RNA sequencing data with Cell2space.

Elucidating the spatial organization of cells is fundamental to understanding tissue architecture and function, yet tissue dissociation during single-cell RNA sequencing (scRNA-seq) strips cells of their native architectural context. It is essential to integrate scRNA-seq data with spatial transcriptomics (ST) references to infer the spatial information of individual cells. Despite the development of various tools, achieving precise and biologically relevant spatial reconstructions remains challenging. Here, we present Cell2space, a deep learning framework that integrates scRNA-seq data with S

The Progression of Prion Diseases Is Not Altered in the Absence of TMEM106B.

Transmembrane protein 106B (TMEM106B) is a type II transmembrane glycoprotein localized in late endosomes and lysosomes. This protein has been attributed to relevant roles in disease risk and progression of multiple neurodegenerative disorders. In experimental models, TMEM106B has been linked with the prion-like spread of misfolded tau proteins responsible for Alzheimer's disease and other tauopathies. The role of this protein in the spread of other misfolded proteins, like prions, has not been explored. Here, we tested whether the removal of TMEM106B altered the progression of prion diseases
