Breast cancer is the most prevalent malignancy in women, with over 70% of cases being estrogen receptor α (ERα)-positive. Photodynamic therapy (PDT) is spatially precise yet limited by tumor-selective photosensitizers. Herein, we report the first co-crystal structure of 2,2'-dihydroxytriphenylamine bound to ERα, which provided a structural blueprint for designing EP5, an innovative ERα-targeted photodegrader. EP5 exhibits high binding affinity toward ERα (KD = 56.7 nM), enabling its selective accumulation in ERα-positive breast cancer cells. Upon white-light irradiation, EP5 triggers selective

Polo-like kinase 4 (PLK4) is a serine/threonine kinase that has an essential role in regulating centriole duplication and centrosome biogenesis during cell-cycle progression and a potential therapeutic target in various human cancers. Structure-guided lead optimization of an aminopyrazole scaffold at three key vectors led to the identification of EXEL-1855, which demonstrated a significant improvement in cellular potency, increased selectivity over related-kinases and an improved pharmacokinetic profile. Targeting the non-conserved Met91 residue in the PLK4 lower hinge region and optimizing th

Nanoscale proteolysis-targeting chimeras (nano-PROTACs) offer modular platforms for targeted protein degradcine-rich α-2-glycoprotein 1 (LRG1). i16αCF3-LipoTAC showed greater degradation potend endocytosis, and promoted endosomal escape. In 4T1 tumor models, i16αCF3-LipoTAC prolonged circulation, increased tumor accumulation and target depletion, and improved antitumor efficacy. These findings support fluorination tuning as a strategy to enhance nano-PROTAC trafficking and degradation efficiency.Nanoscale proteolysis-targeting chimeras (nano-PROTACs) offer modular platforms for targeted protei

Despite strong antimicrobial activity, high haemolytic/cytotoxic properties, associated with its non-cell-selective membrane-lytic property and its large size prohibits considering bee-venom, α-helical, 26-residue, melittin as a lead molecule for antimicrobial-drug development. Herein, we have explored the design of shorter non-haemolytic/non-cytotoxic antimicrobial peptides with low hydrophobic/amphipathic property and β-sheet secondary structure by using a 5-mer carboxy-terminus melittin segment, "WIKRK", which contains hydrophobic and polar-cationic residues at its two ends. Three peptides,

Nonhistone acetylation is a key epigenetic mechanism in osteoarthritis (OA), but its specific role in cartilage degeneration remains unclear. Here, we integrated transcriptomics, proteomics, and acetyl-proteomics to systematically analyze molecular changes in rat OA cartilage. Multiomics coenrichment identified the ECM-receptor interaction pathway as the core target, reprogrammed by increased acetylation of Fn1, Tnc, and Itgb1. Using 113 machine-learning strategies, we identified 11 core diagnostic genes, with Wisp3 showing the highest efficacy validated in independent data sets. In vitro expe

Helicobacter pylori (H. pylori) induces gastric precancerous lesions (GPL), but their underlying biomarkers remain poorly characterized. To establish a GPL model, BALB/c mice were inoculated with H. pylori via gavage. Immunofluorescence confirmed successful modeling by detecting H. pylori distribution and aberrant expression of precancerous lesion markers. Serum-based label-free proteomics and bioinformatic analyses were performed to screen key proteins. Data are available via ProteomeXchange (PXD076808). Western blot and immunofluorescence were used to verify key proteins and markers of DNA d

Climate change is causing shifts in the phenology of taxa across the globe, with phenological shifts detected in species, including but not limited to, plants, insects, and birds. However, changes in the phenology of caterpillar activity have been relatively understudied, despite the importance of caterpillar activity for conservation, plant and wildlife ecology, forest management practices, and agriculture. To improve understanding of the potential impacts of climate change on caterpillar phenology, we modeled shifts in the onset date of observable activity, termination date of observable act

Dense active materials must continuously relieve internal mechanical stress to remain structurally stable as they are driven far from equilibrium. In epithelial tissues, this relief occurs through cell division, where, according to Hertwig's century-old rule, cells divide along their long axis. We show that as epithelia densify, this geometric rule is superseded by a mechanical one: cells divide along the principal axis of anisotropic stress, independently of the isotropic stress state. Using direct force measurements and stress inference, we demonstrate that stress orientation governs divisio

Hippocampal activity provides a representation of the world around us, as well as our position within that world. However, it is not known if and to what extent the chosen navigational reference frame can influence hippocampal representations during memory-based tasks, including those focused on future activity. Here, we develop and employ two naturalistic, carefully controlled variants of the everyday memory task to model the use of egocentric and allocentric coordinates in the same arena. By recording hippocampal neural activity through miniature microscopes in male rats performing each of t

Precise gene dosage control is critical for cellular identity and development, particularly for imprinted genes, where dosage imbalances are linked to neurodevelopmental disorders. The Mest/Copg2 imprinted locus exemplifies this fine-tuned regulation: while Mest is constitutively paternally expressed, Copg2 shifts from biallelic to maternally-biased expression specifically during neural differentiation, a transition proposed to involve transcriptional interference by the long Mest isoform, MestXL, which extends into Copg2. However, the mechanisms underlying this switch and whether factors beyo

Transcytosis-mediated delivery effectively enhances tumor penetration of nanodrugs but is less efficacious in stroma-rich tumors. Cancer associated fibroblasts (CAF) can represent a physical barrier for drug delivery in tumor cells. Herein, by selectively targeting NPC1L1-cancer associated fibroblasts, surface-displayed cholesterol enhances the delivery of a platinum nanodrug (PtD-Chol) in stroma-rich tumors, restoring anti-tumor efficacy. The findings reveal that PtD-Chol, acting through the 'shared entry portal' of NPC1L1 triggers cross-multicellular transport (CMCT) within tumors, shuttling

Equitable and sustainable governance of small-scale fisheries (SSFs) is essential for achieving food security and conservation in coastal areas. Yet in increasingly industrialized oceans, competition and conflict with large-scale fisheries (LSFs) threaten the contributions of SSFs to the United Nations Sustainable Development Goals. Here we have developed a small-scale and industrial fisheries interaction index and applied it to 126 countries using 2 recent global datasets. Vulnerability to conflict is highest in countries where SSFs are vital for nutrition, but governance is weak, especially

OBJECTIVES: This in vitro study aimed to compare the effects of soy milk and coconut milk on primary molar enamel microhardness after an acidic challenge.
MATERIALS AND METHODS: Microhardness of 30 primary molar enamel blocks was measured using a Vickers tester. For the acidic challenge, they were immersed in citric acid with a pH of 3 for 25 h, and then their microhardness was measured again. The specimens were then randomly assigned to 3 groups (n = 10) for immersion in either soy milk, coconut milk, or artificial saliva (control) 3 times a day, each time for 5 min, for 7 days. Subsequently

BACKGROUND: FHL2 is an oncogenic scaffold protein that is overexpressed in lung adenocarcinoma (LUAD), making it a promising therapeutic target. SHK, a natural naphthoquinone, exhibits antitumor activity, but its molecular mechanism in LUAD, particularly its relationship with FHL2, remains unclear.
AIM OF THIS STUDY: The present study aimed to define the effects of SHK on NSCLC and identify the potential underlying molecular mechanisms.
METHODS: The anti-tumor effects of SHK were evaluated in LUAD cell lines (A549andH1299) and a patient-derived xenograft (PDX) model using CCK-8, colony forma

The apolipoprotein E ε4 allele (APOE4) is the strongest genetic risk factor for sporadic Alzheimer's disease (AD) and confers disproportionate risk in women, particularly after reproductive senescence. The brain changes preceding overt neurodegeneration in APOE4 carriers remain incompletely characterized. We conducted neuroimaging and behavioral studies in aged (18-month) female APOE4 humanized knock-in rats and APOE3 controls. Regional brain volumes across 197 areas were quantified by voxel-based morphometry (VBM); white and gray matter microstructure by diffusion tensor imaging (DTI), yieldi

The citrus red mite, Panonychus citri, has developed severe resistance to the widely used pesticide like abamectin. Understanding the inheritance, cross-resistance profile, and biochemical mechanism of pesticide resistance in P. citri is critical for developing effective resistance management strategies. Here, we established an abamectin-resistant near-isogenic strain (NIL-Aba) by backcrossing the susceptible strain (SS strain) with a field collected abamectin-resistant strain (NN-Aba strain) for eight generations, coupled with continuous abamectin selection. The NIL-Aba strain exhibited 2278.

Huanglongbing (HLB), caused by Candidatus Liberibacter asiaticus (CLas) and transmitted by Asian citrus psyllid (ACP), Diaphorina citri, poses a severe threat to global citrus production. In this study, a dual-functional nanopesticide, THX@ZIF-8/PDA, by encapsulating thiamethoxam (THX) within zeolitic imidazolate framework-8 (ZIF-8) followed by polydopamine (PDA) coating was developed, and THX loading capacity was 17.25%. THX@ZIF-8/PDA exhibited pH-responsive release, with cumulative release rates of 99.54%, 65.53%, and 59.14% at pH 5.0, 7.0, and 9.0 after 168 h, respectively. PDA modification

Plant-parasitic nematodes (PPNs) cause substantial global crop losses, yet the effectiveness of chemical nematicides is declining due to adaptive responses in nematode populations. Current understanding of nematicide resistance remains incomplete, often limited to target-site mutations and metabolic detoxification. This review addresses this gap by providing an integrated perspective on how PPNs evade nematicides across molecular, physiological, and ecological scales. We synthesize established resistance mechanisms alongside underexplored strategies, including cuticle remodeling, behavioral av

Transgenic crops expressing Bacillus thuringiensis (Bt) toxins are central to global pest management, but their long-term efficacy is threatened by the evolution of insect resistance. Elucidating the genetic basis of resistance is essential for early detection and for designing proactive resistance management strategies. In lepidopterans, cadherin (Cad) and the ATP-binding cassette transporters ABCC2 and ABCC3 function as key midgut receptors for Cry1 toxins, although their contributions vary among toxins and species. Following recent studies on Cry1Ab and Cry1Fa receptor pathways in the Asian

While naturally occurring insect-pathogenic viruses are valuable resources utilized as biocontrol agents, the covert viruses maintained in laboratory insect populations have been comparatively less explored. In this study, we report the isolation and characterization of a novel nucleopolyhedrovirus, CypoNPV, from a laboratory population of Cydia pomonella. Whole-genome sequencing revealed a genome size of 112,909 bp for CypoNPV, exhibiting close phylogenetic proximity to Cryptophlebia peltastica nucleopolyhedrovirus (CrpeNPV). Comparative life-history analyses of populations designated as norm
