Dnmt3b and Dnmt3l knockdown reduces blastocyst development in early mouse embryos.

A one-cell embryo called a zygote develops into a blastocyst through several successive cell divisions and lineage specification, this process is called early embryo development. Both embryonic genome activation (EGA) and the first lineage specification during early embryonic development depend on tightly coordinated epigenomic organization. Regulation of the epigenome is primarily governed by DNA methylation mediated through DNA methyltransferase (Dnmt) enzymes. Dnmt1 is responsible for the maintenance of methylation during cellular division, while Dnmt3a/Dnmt3b enzymes play a role in the est

Dose-dependent effects of selenium (Se)-enriched yeast containing nano-scale Se particles on Se bioavailability, rumen fermentation, hematological profile, and growth performance in female Thin-Tail sheep.

BACKGROUND AND AIM: Selenium (Se) deficiency remains a significant nutritional challenge in sheep production due to the limited bioavailability of inorganic Se sources and the low Se availability in many soils and forages. Se-enriched yeast containing nano-scale Se particles may provide a more bioavailable organic Se source while also influencing rumen fermentation and animal physiology. This study evaluated the dose-dependent effects of Se-enriched yeast containing nano-scale Se particles on Se bioavailability, rumen fermentation, hematological profile, and growth performance in female Thin-T

Breed- and age-associated variation in hematological parameters and Toll-like receptor 2 gene expression in peripheral blood mononuclear cells of commercial and purebred laying hens during the laying cycle.

BACKGROUND AND AIM: Hematological parameters and innate immune responses are influenced by genetic background and physiological status, making them valuable indicators of health and immune competence in laying hens. However, comparative information on breed- and age-related variation in hematological characteristics and Toll-like receptor 2 (TLR2) gene expression in PBMCs during the laying cycle remains limited. This study evaluated the effects of breed and age on hematological traits and TLR2 gene expression in commercial Lohmann Brown (LB) and purebred White Leghorn (WL) laying hens to impro

Plasmonic property of gold nanoparticles to predict nanozyme activity: towards the colorimetric detection of manganese(ii).

Because of its remarkable versatility and indispensable properties, the need for manganese for different applications has grown tremendously. The ever-growing need and the beneficiation of manganese ore, however, have increased the emission of manganese into the environment, such as into water sources, negatively affecting human and animal health. This necessitates a simple method for the analysis of manganese in water resources. The intrinsic peroxidase-like activity of gold nanoparticles (AuNPs) is considered for the detection of Mn2+. The peroxidase-like activity of AuNPs using 3,3',5,5'-te

A complex of MAST1 and 14-3-3η regulates Tau phosphorylation in the developing cortex.

The MAST family of serine/threonine kinases has been implicated in a spectrum of human neurodevelopmental disorders. However, little is known about their biological function or regulation. Seeking to fill these gaps in our knowledge, we have identified upstream and downstream partners of MAST1. 14-3-3η, a neuronal 14-3-3 paralog, specifically interacts with MAST1 at two regulatory serines, S90 and S161. p21-activated kinase (PAK), a neuronal regulator of the actin cytoskeleton, phosphorylates MAST1 to regulate its interaction with 14-3-3η. Arginine Kinase 1 supports energy homeostasis in Drosophila flight muscle development.

In Drosophila, Arginine kinase 1 (Argk1) is involved in maintaining ATP homeostasis during bursts of activity in tissues with high and variable rates of energy turnover such as muscle. However, its role beyond stress conditions is less understood. Argk1 is the sole phosphagen kinase with dynamic expression throughout flight muscle development. Here, we show that at least one of the Argk1 isoforms localizes to mitochondria in the developing myofibers, and its function is also necessary for proper flight muscle development. Depleting Argk1 specifically in the muscles lead to low ATP level and NA

Evolution, Mechanisms, and Therapeutic Implications of Mobile Tetracycline Destructases.

Tetracycline destructases (TDases) pose an emerging global threat by enzymatically inactivating all generations of tetracycline (Tet) antibiotics, including last-resort agents such as tigecycline. Despite their recent identification, TDases have rapidly disseminated worldwide, largely driven by mobile genetic elements and environmental reservoirs. This review synthesizes current knowledge on TDase genomics, structural and catalytic mechanisms, ecological niches, and clinical impacts. We detail the mechanistic distinctions between type 1 and type 2 TDases, emphasizing their divergent structural

Glycerol Phenylbutyrate Treatment in a Pediatric Cohort of SLC6A1-Related Neurodevelopmental Disorder.

BACKGROUND: The SLC6A1 gene encodes the gamma-aminobutyric acid transporter 1 protein, which facilitates the reuptake of gamma-aminobutyric acid from the synaptic cleft in inhibitory synapses back into presynaptic neurons and glial cells. SLC6A1 haploinsufficiency leads to neurodevelopmental impairment including epilepsy, movement disorders, intellectual disability, and autism spectrum disorder. SLC6A1-related neurodevelopmental disorder (SLC6A1-NDD) is a rare condition with autosomal dominant inheritance for which there is no cure. Acting as a chemical chaperone, glycerol phenylbutyrate, used

Prepubertal ovariectomy alters dorsomedial striatum indirect pathway neuron excitability and explore/exploit balance in female mice.

Decision-making circuits are modulated across life stages (e.g. juvenile, adolescent, or adult)-as well as on the shorter timescale of reproductive cycles in females-to meet changing environmental and physiological demands. Ovarian hormone signaling may contribute to this flexibility by regulating neural circuits involved in adaptive decision making. Here, we examined how prepubertal ovariectomy (pOVX) influences adult performance in an odor-guided multiple-choice reversal task. During reversal learning, pOVX females exhibited a distinct pattern of errors compared to sham-operated controls. Re

The Influence of the Ozonation Process on the Quality Parameters and Physicochemical Stability of Horse Meat.

The aim of this study was to demonstrate the effect of gaseous ozonation at a concentration of 10 ppm on the quality of horse meat. Samples of the longissimus thoracis (LT) muscle were exposed to ozone for 1, 5, and 15 min and then stored at 4 °C for 6 and 48 h. Ozonation caused a deterioration in the water-holding capacity of muscle tissue. Significant changes in the color of horse meat were observed, as longer exposure to ozone intensified oxidative processes in the meat. The volatile compound profile after 48 h indicated significant lipid degradation and an increase in aldehydes, typical of

Paraventricular nucleus of the hypothalamus activation and forebrain neuroinflammation in salt-loaded rats.

Salt loading (SL) in rats for 7 days raises mean arterial pressure (MAP) and induces astrogliosis in the paraventricular nucleus of the hypothalamus (PVN). However, the exact role of the PVN in elevating MAP and sympathetic nerve activity (SNA), as well as its effects on astrocytes in forebrain osmosensitive areas, remain unclear. Male Holtzman and Wistar rats (300-320 g) received only 2% NaCl solution (SL) to drink for 7 days, while controls drank water (WT). Daily fluid and food intake, body weight, urinary volume, and MAP were recorded. On day 7, blood and cerebrospinal fluid (CSF) were col

Nontoxic mRNA vehicles based on electrically neutral self-assembling peptides.

The cationic molecules and low-molecular-weight amphiphiles commonly used in nonviral gene delivery vehicles often induce adverse nonspecific effects, notably cytotoxicity and detergent-like unfavourable membrane activity. To address these limitations, here we report the development of a nontoxic mRNA-condensing agent based on electrically neutral self-assembling peptides (ENSPs). Driven by cooperative hydrogen bonding and hydrophobic interactions, ENSPs coassemble with mRNA into discrete nanotube-shaped complexes. ENSP-based mRNA vehicles are almost nontoxic to mammalian cells and exhibit neg

Transcription elongation mechanisms that control gene expression in erythropoiesis.

The regulation of RNA transcript abundance relies on the control of two key early steps in gene expression: transcription initiation and productive elongation. Although DNA-directed RNA polymerase provides the intrinsic catalytic activity required for RNA synthesis, its ability to effectively produce a transcript relies on accurate recruitment to specific DNA sites to initiate RNA polymerization, followed by release from promoter-proximal pausing into a productive elongation machine. These steps are governed by accessory factors that not only facilitate the assembly of a functional pre-initiat

The rate and spectrum of germline mutations in chicken from a commercial pedigree line.

BACKGROUND: Pedigree-based studies of germline mutations provide a direct understanding of the origin and consequences of genetic variation, and allow quantification of how mutations affect functional variation. Here, we use whole-genome sequencing data from 194 chicken (Gallus gallus) trios from a commercial pedigree line to estimate the germline mutation rate and spectrum, and to characterize patterns of mutational variation across the genome and genes, aiming to rank protein coding genes by their expected contributions to mutational variance (Vm). RESULTS: We estimated a mutation rate of µ
