Research progress in intratumoral microbiota: unlocking the potential of cancer immunotherapy.

In recent years, intratumoral microbiota (ITM), a crucial component of the tumor microenvironment, have received increasing attention because of their role in cancer immunotherapy. ITM not only influence tumor initiation and progression but also modulate local and systemic immune responses, thereby affecting the efficacy of immune checkpoint inhibitors and other immunotherapeutic approaches. This review summarizes the compositional characteristics of ITM, their immunoregulatory mechanisms, and their effects on tumor metastasis and responsiveness to immunotherapy. Particular emphasis is placed

The microbiome-metabolite-vaccine axis: context-dependent modulators of immunogenicity and durability.

Vaccine responses vary markedly between individuals, yet age, genetics, prior antigen exposure, comorbidity, nutrition and vaccine platform explain only part of this heterogeneity. The intestinal microbiome is a candidate modulator through microbial metabolites and parallel non-metabolic signals. Human studies link microbial features to vaccine outcomes, but taxa rarely replicate across geography, age and platform. Small randomised antibiotic studies identify effects of treatment assignment, not necessarily microbiome mediation; infant microbial rescue has been demonstrated in mice rather than

Effects of artificial milk on growth performance and intestinal barrier function in suckling piglets: a review.

With the continuous improvement of genetic selection in modern swine production, litter size has steadily increased. However, due to the limited capacity of sows to enhance milk production, some piglets experience insufficient milk intake and restricted growth during the lactation period. As an important supplement to sow milk, artificial milk has been widely used in both production practice and scientific research. This review summarizes the effects of artificial milk on the growth performance and intestinal health of suckling piglets. It analyzes how insufficient milk yield and the nutrition

Machine learning-assisted fluorescent sensor array based on siloxane-derived porous polymers for high-throughput discrimination of multiple antibiotics in milk.

Both simultaneous monitoring of multiple antibiotic residues for public health security and accurate single-analyte quantification for regulatory compliance are critically important, though the former remains challenging due to structural diversity among analytes. Herein, we constructed a four-channel fluorescent sensor array based on siloxane-derived porous polymers for discriminative detection of 12 antibiotics spanning four categories (chloramphenicols, nitrofurans, nitroimidazoles, and tetracyclines), and further achieved discrimination of 6 antibiotics in real milk samples at lower concen

Between Likes and Ethics: Benefits and Challenges of Social Media Use in Medical Practice in a Pre- and Post-intervention Study.

BACKGROUND: Social media use among medical students is nearly universal, yet it often occurs without adequate ethical-legal literacy, particularly considering the update introduced by Resolution No. 2,336/2023 of the Brazilian Federal Council of Medicine (Conselho Federal de Medicina, CFM). This study assessed medical students' knowledge and perceptions of the benefits, risks, and ethical limits of social media use before and after an educational intervention based on practical scenarios grounded in that resolution. METHODOLOGY: This was a quasi-experimental study with a paired pre- and post-

Selection on PER2 Phosphorylation Regulation Underlies Atypical Mammalian Circadian Rhythms.

Circadian rhythms are fundamental biological processes that enhance survival and reproductive fitness by coordinating physiology and behavior with predictable environmental cycles. However, in some species, circadian rhythms are weakened, irregular, or even undetectable, likely reflecting evolutionary responses to extreme environments or specialized ecological niches. Nevertheless, the genetic basis of these atypical phenotypes remains incompletely understood. Here, we investigated the molecular evolution of 20 PER2-associated genes across 71 representative mammalian species. We identified fou

From Byproduct to Platform: Can Kraft Lignin Outcompete Lignosulfonates?

Lignosulfonates (LS) have long set the standard of dispersants for construction, agrochemical, oilfield, and animal feed industries, due to their amphiphilic structurearomatic domains linked with sulfonated functionalities, which result in efficient and cost-effective colloidal stabilization. Nevertheless, LS production is intrinsically limited by the falloff in sulfite pulping, which is today only a minor percentage of the world's pulp manufacturing. On the other hand, kraft lignin (KL) is produced at multimillion-tonne levels annually and is now being acknowledged as a renewable aromatic po

Pangenome variation analysis uncovers the impacts of SNP-independent structural variations in modern European cattle breeding.

While pangenome studies emphasize structural variations (SVs), their functional significance beyond single-nucleotide polymorphisms (SNPs) in cattle remains unexplored. We established a comprehensive variation map for 1379 European cattle from nine breeds using a pangenome graph. Although SV and SNP distributions were similar genome-wide, 19.3% of SVs exhibited low linkage disequilibrium (r² < 0.2) with nearby SNPs. These non-SNP-tagged SVs are preferentially localized to chromosome ends and promoters, and play crucial roles in artificial selection for production traits. Integrated analyses of

Host-specific and deterministic microbiome assembly in major coleopteran stored-grain pests.

Dual DNA- and RNA-based sequencing profiled the microbiomes of five major coleopteran stored-grain pests and their wheat flour substrate. The microbiomes differed significantly among hosts and from flour, yet 11 core bacterial and 9 core fungal taxa were shared across hosts. RNA data revealed host-specific active microbiota. Deterministic processes dominated community assembly, exceeding 90% in Rhyzopertha dominica and Oryzaephilus surinamensis. Homogeneous selection identified eight conserved bacterial clades. Although microbiomes were strongly host-specific, they showed no significant correl

BACKGROUND: Artificial Intelligence (AI) is becoming increasingly significant in veterinary medicine, particularly in answering pet owners' medical questions. AI's integration could reduce veterinary professionals' workload while improving access to medical advice. However, the reliability and quality of AI-generated responses compared to veterinarians' remain unexplored. The objective was to determine whether AI-generated responses were perceived as comparable to veterinarian-generated responses. METHODS: This cross-sectional study analysed the answers to a sample of pet owners' medical ques

Developmental plasticity in response to early-life conditions affects male sexual traits in guppies.

Early environmental conditions play an important role in animal development. Lack of sufficient food early in life can lead to impaired growth and long-term fitness consequences. To avoid that, many animal species compensate during a period of low food availability by increasing their growth rate when food becomes abundant again, ie compensatory growth. However, compensatory growth during development may divert resources away from the ontogeny of reproductive and somatic functions, imposing hidden costs that are only apparent later in life. For ectotherms, these trade-offs are expected to be e

Harnessing microbial metabolism and nutrition to target the gut-mammary axis for better livestock production and health.

The gut-mammary axis orchestrates bidirectional crosstalk linking intestinal homeostasis and mammary function through integrated neural, endocrine, and immune pathways. Dysregulation driven by gut microbiota dysbiosis or metabolic dysfunction compromises lactation performance and induces mastitis via pathogen translocation and epigenetic reprogramming. This review systematically discusses the molecular mechanisms underlying microbial metabolite-mediated mammary development and lactation efficiency, evaluates the impact of gut-derived pathogens on mammary barrier integrity and inflammatory casc

Oleoylethanolamide regulates intestinal stem cell activity and villus size via PPARα signaling pathway.

The size of intestinal villi plays a critical role in determining digestive and absorptive functions, directly affecting nutrient digestibility and feed efficiency. Therefore, elucidating the key mechanisms regulating villus development is of significant importance. This study compares intestinal development in Shaziling pigs and Yorkshire pigs, which exhibit distinct lipid metabolism characteristics, revealing significant enrichment of different fatty acid metabolic pathways and the peroxisome proliferator-activated receptors (PPAR) signaling pathway. LC-MS metabolomics analysis further explo

Imaging Microtubule Dynamics In Vivo in Drosophila Sensory Dendrites.

Drosophila larval class IV sensory neurons are located just beneath the cuticle and are characterized by highly branched dendritic arbors that expand isotropically in two dimensions as the larvae develop. The growth of these arbors is driven by stochastic processes involving growth, shrinkage, and pausing of the dendritic terminal tips. To test the hypothesis that tip dynamics are driven by microtubule dynamics, protocols have been developed for imaging fluorescently labeled microtubules within dendrites. The method enables visualization of microtubule dynamics in fine terminal dendrites of cl

Genetic, Phenotypic, and Quantitative Analysis of Myoblast Fusion in Drosophila Embryos.

The fusion of mononucleated myoblasts into multinucleated syncytia is a fundamental process in skeletal muscle development. Drosophila melanogaster is a powerful, genetically tractable model for rapidly and efficiently assessing the function of candidate genes involved in this process. Here, we describe a comprehensive protocol with step-by-step instructions for analyzing the myoblast fusion phenotype in Drosophila embryos. This method includes mutant line selection for candidate gene, embryo collection and fixation, whole-mount immunostaining, developmental staging, confocal imaging, and phen
