Histone acetyltransferases and N-terminal acetyltransferases orchestrate development and metamorphosis in the yellow fever mosquito, Aedes aegypti.

Mosquitoes, especially Aedes aegypti, transmit major human diseases such as dengue, Zika, and chikungunya. Epigenetic regulation, including histone acetylation, plays a key role in controlling Ae. aegypti development. However, the specific roles of histone acetyltransferases (HATs) and N-terminal acetyltransferases (NATs) in mosquito development remain poorly understood. In this study, we investigated 25 HAT/NAT genes functions by knocking them down via feeding Ae. aegypti larvae with dsRNA nanoformulations. Gene expression analysis confirmed that target gene expression was reduced by more tha…

Histological localization of acetyltransferases in human tissue.

The localization of human arylamine acetyltransferases (NAT) transcripts was performed by non-isotopic in situ hybridization, utilizing a combination of six NAT1 or NAT2 specific antisense oligonucleotide probes, in order to identify those tissues and organs that might be susceptible to the carcinogenic effects of aromatic amines. The intratissue differences in the level of NAT mRNA were observed: the most abundant NAT2 transcripts were found in hepatocytes, while NAT1 ones dominated in the urothelium and in the colon epithelial cells. The specific NAT1 and NAT2 mRNAs were present also in the …

Expression of N-acetyltransferases in periodontal granulation tissue.

Smoking is a major risk of periodontal diseases. At the site of first contact, the gingiva is exposed to aromatic amines and polycyclic hydrocarbons. These are metabolized by the N-acetyltransferases (NAT), leading to local detoxification and/or activation reactions contributing to the risk of periodontal destruction in smokers. The purpose of this study was to detect the expression of N-acetyltransferase isoenzymes NAT1 and NAT2 in periodontal granulation tissue. In 24 specimens obtained from periodontitis patients or control subjects, mRNA encoding for NAT1 and NAT2 was detected by RT-PCR, a…

How Histone Acetyltransferases Shape Plant Photomorphogenesis and UV Response.

Higher plants have developed complex mechanisms to adapt to fluctuating environmental conditions with light playing a vital role in photosynthesis and influencing various developmental processes, including photomorphogenesis. Exposure to ultraviolet (UV) radiation can cause cellular damage, necessitating effective DNA repair mechanisms. Histone acetyltransferases (HATs) play a crucial role in regulating chromatin structure and gene expression, thereby contributing to the repair mechanisms. HATs facilitate chromatin relaxation, enabling transcriptional activation necessary for plant development…

Assays of sialate-O-acetyltransferases and sialate-O-acetylesterases.

The O-acetylation of sialic acids is one of the most frequent modifications of these monosaccharides and modulates many cell biological and pathological events. Sialic acid-specific O-acetyltransferases and O-acetylesterases are responsible for the metabolism of esterified sialic acids. Assays were developed for the analysis of the activities and specificities of these enzymes. The methods had to be varied in dependence on the substrate assayed, the kind of biological source, and the state of enzyme purity. With the new techniques the primary site of O-acetyl incorporation at C-7, catalyzed by…

Bacterial GCN5-Related N-Acetyltransferases: From Resistance to Regulation.

The GCN5-related N-acetyltransferases family (GNAT) is an important family of proteins that includes more than 100000 members among eukaryotes and prokaryotes. Acetylation appears as a major regulatory post-translational modification and is as widespread as phosphorylation. N-Acetyltransferases transfer an acetyl group from acetyl-CoA to a large array of substrates, from small molecules such as aminoglycoside antibiotics to macromolecules. Acetylation of proteins can occur at two different positions, either at the amino-terminal end (αN-acetylation) or at the ε-amino group (εN-acetylation) of …

N-acetyltransferases: pharmacogenetics and clinical consequences of polymorphic drug metabolism.

Since the discovery of polymorphic N-acetylation of drugs nearly 40 years ago, great progress has been made in understanding the molecular genetics of acetylation as well as the clinical consequences of being a rapid or slow acetylator. Inborn errors (several different alleles) at the NAT2 locus are responsible for the traditional acetylator polymorphism. Studies have revealed variant alleles at the NAT1 locus as well. The consequences of pharmacogenetic variation in these enzymes include (i) altered kinetics of specific drug substrates; (ii) drug-drug interactions resulting from altered kinet

Histone H3 thiol reactivity and acetyltransferases in chicken erythrocyte nuclei.

Chicken erythrocyte nuclei previously incubated separately with two novel mercury compounds (N-chloromercuribenzoyl)-biocytin and bis(p-(chloromercuribenzoyl))-[3H]lysine diamide) were digested with micrococcal nuclease and the digest products fractionated according to their solubility in 0.15 M NaCl and molecular size. The identity and quantitation of the chromatin fractions and proteins containing covalently bound mercury were determined by Western blotting, autoradiography, and scintillation counting. The most highly acetylated species of histone H3 in the 0.15 M NaCl-soluble polynucleosome

N-acetyltransferases as markers for asthma and allergic/atopic disorders.

An increasing prevalence of asthma noted worldwide has stimulated research on the phenotypic complexity resulting from interaction between the genetic and environmental components. Particularly, an increase in the prevalence of allergic rhinitis and asthma in industrialized countries indicate the importance of pulmonary metabolism of environmental xenobiotics. The arylamine N-acetyltransferases (NATs) are a unique family of enzymes that are involved in the biotransformation and detoxification of hydrazine and arylamine drugs/xenobiotics and could have a major role to play in atopy/asthma patho…

Antifungal resistance: from classical mechanisms to the expanding role of acetyltransferases.

Antifungal resistance represents a growing global health challenge, driven by limited therapeutic options, inadequate use of both clinical and environmental drugs, and the remarkable adaptive capacity of fungal pathogens. While resistance has classically been attributed to genetic alterations affecting drug targets and efflux systems, accumulating evidence indicates that regulatory mechanisms play a critical role in shaping antifungal susceptibility. This review highlights that acetyltransferases constitute important and active components of antifungal resistance, operating through multiple in…

Neurodevelopmental functions and activities of the KAT3 class of lysine acetyltransferases.

The human lysine acetyltransferases KAT3A (CREBBP) and KAT3B (EP300) are essential enzymes in gene regulation in the nucleus. Their ubiquitous expression in metazoan cell types controls cell proliferation and differentiation during development. This comprehensive review delves into the biological roles of KAT3A and KAT3B in neurodevelopment, shedding light on how alterations in their regulation or activity can potentially contribute to a spectrum of neurodegenerative diseases (e.g., Huntington's and Alzheimer's). We explore the pathophysiological implications of KAT3 function loss in these dis…

Classification and phylogeny for the annotation of novel eukaryotic GNAT acetyltransferases.

The enzymes of the GCN5-related N-acetyltransferase (GNAT) superfamily count more than 870 000 members through all kingdoms of life and share the same structural fold. GNAT enzymes transfer an acyl moiety from acyl coenzyme A to a wide range of substrates including aminoglycosides, serotonin, glucosamine-6-phosphate, protein N-termini and lysine residues of histones and other proteins. The GNAT subtype of protein N-terminal acetyltransferases (NATs) alone targets a majority of all eukaryotic proteins stressing the omnipresence of the GNAT enzymes. Despite the highly conserved GNAT fold, sequen…

MOZ and MORF acetyltransferases: Molecular interaction, animal development and human disease.

Lysine residues are subject to many forms of covalent modification and one such modification is acetylation of the ε-amino group. Initially identified on histone proteins in the 1960s, lysine acetylation is now considered as an important form of post-translational modification that rivals phosphorylation. However, only about a dozen of human lysine acetyltransferases have been identified. Among them are MOZ (monocytic leukemia zinc finger protein; a.k.a. MYST3 and KAT6A) and its paralog MORF (a.k.a. MYST4 and KAT6B). Although there is a distantly related protein in Drosophila and sea urchin, t…

Nα-acetyltransferase NAA50 mediates plant immunity independent of the Nα-acetyltransferase A complex.

In humans and plants, 40% of the proteome is cotranslationally acetylated at the N-terminus by a single Nα-acetyltransferase (Nat) termed NatA. The core NatA complex is comprised of the catalytic subunit Nα-acetyltransferase 10 (NAA10) and the ribosome-anchoring subunit NAA15. The regulatory subunit Huntingtin Yeast Partner K (HYPK) and the acetyltransferase NAA50 join this complex in humans. Even though both are conserved in Arabidopsis (Arabidopsis thaliana), only AtHYPK is known to interact with AtNatA. Here we uncover the AtNAA50 interactome and provide evidence for the association of AtNA

Structural plasticity drives convergent evolution and functional diversification of GNAT polyamine acetyltransferases.

Gcn5-related N-acetyltransferases (GNATs) are considered a "megafamily" that ranks among the most structurally and sequentially diverse superfamilies in the CATH (Class, Architecture, Topology, Homology) database. In this vast superfamily, several types of protein functions have been explored throughout evolution, yet the evolutionary pathways that led to such diversity remain poorly understood. To investigate these concepts further, we selected a functionally distinct GNAT subgroup called polyamine N-acetyltransferases (PAATs). These enzymes acetylate polyamines that are crucial for cellular …

A computational study exploring echinoderm-derived compounds for inhibition of aminoglycoside acetyltransferases.

Aminoglycoside acetyltransferases (AACs) catalyze the acetylation of aminoglycoside antibiotics, playing a major role in bacterial resistance and posing a serious threat to global healthcare. Despite growing interest in natural products, echinoderm-derived metabolites remain largely unexplored as AAC inhibitors. This study presents a comprehensive in-silico investigation into the potential of these marine compounds to inhibit four key AAC enzymes: Aminoglycoside 2'-N-acetyltransferase, AAC(3)-Ib, AAC(6')-Im, and AAC(3)-Iva. A virtual screening of 1600 echinoderm metabolites was performed using…

Molecular and Functional Characterization of N-Acetyltransferases in Common Marmosets and Pigs.

Arylamine N-acetyltransferases (NATs) are drug-metabolizing enzymes that are essential for the metabolism of endogenous substrates and xenobiotics. The molecular characteristics of NATs have been extensively investigated in humans but remain to be investigated in common marmosets and pigs, animal species that are often used in drug metabolism studies. In this study, marmoset NAT1 and pig NAT1 cDNAs were isolated from liver samples and were characterized by molecular analyses and drug-metabolism assays. These NAT genes were intronless and formed gene clusters with one other NAT gene in the geno…

Properties of clock-controlled and constitutive N-acetyltransferases from chick pineal cells.

The pineal gland synthesizes its hormone melatonin (O-methyl-N-acetylserotonin) from serotonin. Acetyl-CoA: serotonin N-acetyltransferase (SNAT), the enzyme that catalyzes the committed step in this biosynthesis, is largely restricted to the pineal gland and is regulated by adrenergic and circadian mechanisms. Another enzyme, acetyl-CoA: arylamine N-acetyltransferase (ANAT), having an apparently similar activity, is also present in the pineal. This enzyme, however, is not rhythmically regulated. SNAT activity of cultured chick pineal cells was obtained without ANAT after ammonium sulfate preci

Encoding of Promiscuity in an Aminoglycoside Acetyltransferase.

Aminoglycoside antibiotics are a large family of antibiotics that can be divided into two distinct classes on the basis of the substitution pattern of the central deoxystreptamine ring. Although aminoglycosides are chemically, structurally, and topologically diverse, some aminoglycoside-modifying enzymes (AGMEs) are able to inactivate as many as 15 aminoglycosides from the two main classes, the kanamycin- and neomycin-based antibiotics. Here, we present the crystal structure of a promiscuous AGME, aminoglycoside- N3-acetyltransferase-IIIb (AAC-IIIb), in the apo form, in binary drug (sisomicin,

Arylalkylamine N-acetyltransferase: "the Timezyme".

Arylalkylamine N-acetyltransferase controls daily changes in melatonin production by the pineal gland and thereby plays a unique role in biological timing in vertebrates. Arylalkylamine N-acetyltransferase is also expressed in the retina, where it may play other roles in addition to signaling, including neurotransmission and detoxification. Large changes in activity reflect cyclic 3',5'-adenosine monophosphate-dependent phosphorylation of arylalkylamine N-acetyltransferase, leading to formation of a regulatory complex with 14-3-3 proteins. This activates the enzyme and prevents proteosomal pro
