Chemically Mediated Artificial Electron Transport Chain.

Electron transport chains (ETCs) are ubiquitous in nearly all living systems. Replicating the complexity and control inherent in these multicomponent systems using ensembles of small molecules opens up promising avenues for molecular therapeutics, catalyst design, and the development of innovative energy conversion and storage systems. Here, we present a noncovalent, multistep artificial electron transport chains comprising cyclo[8]pyrrole (1), a meso-aryl hexaphyrin(1.0.1.0.1.0) (naphthorosarin 2), and the small molecules I2 and trifluoroacetic acid (TFA). Specifically, we show that 1) electr…

The induction of microsomal electron transport enzymes.

Liver endoplasmic reticulum contains as NADPH-dependent electron transport complex where the family of hemeproteins, termed cytochrome P-450, serve as catalysts for the oxidation of a variety of different organic chemicals. The content and inventory of the types of cytochrome P-450 is readily modified following in vivo treatment of animals with 'inducing agents' such as barbiturates, steroids and polycyclic hydrocarbons. Recent studies have applied the methods of molecular biology to evaluate changes in the transcription and translation of genomic information occurring concomitant with the ini…

Impairment of mitochondrial electron transport by tetrachloroethylene.

Impairment of electron transport was studied in rat liver sonicated submitochondrial particles. The most susceptible portion was located from NADH dehydrogenase to coenzyme Q. Tetrachloroethylene increased the Michaelis constant (Km) and decreased the maximum velocity (Vmax) of cytochrome c reduction by NADH-cytochrome c reductase. The results suggest that tetrachloroethylene does not represent a specific inhibitor of the particular carrier but decreased the electron flow at the susceptible portion in the mitochondrial inner membrane.

The electron transport chain of Rhizobium trifolii.

The presence of a branched respiratory chain in Rhizobium trifolii grown in batch culture on a defined medium was demonstrated. The branches of the respiratory chain differ in sensitivity to inhibition by cyanide and in affinity for oxygen. The branching point is localized at cytochrome b-555. Electron flow from cytochrome b-555 via cytochrome c-549 and cytochrome a-597 towards oxygen has a relatively low affinity for oxygen and is sensitive to inhibition by cyanide. The second terminal oxidase is cytochrome b-561, which is directly linked to cytochrome b-555. This pathway has a higher affinit

Electron transport chain defects in heart failure.

In recent years, the possibility that disorders of cardiac metabolism play a role in the mechanisms that lead to ventricular dilatation and dysfunction in heart failure has attracted much attention. Electron transport chain is constituted by a series of multimeric protein complexes, located in the inner mitochondrial membranes, whose genes are distributed over both nuclear and mitochondrial DNA. Its normal function is essential to provide the energy for cardiac function. Many studies have described abnormalities in mitochondrial DNA genes encoding for electron transport chain (ETC) in dilated …

Tau-induced mitochondrial reverse electron transport drives neurodegeneration.

Hyperphosphorylation and aggregation of tau are pathological hallmarks of tauopathies. Mitochondrial dysfunction is also a common feature of tauopathies. The mechanistic link between tau abnormalities and mitochondrial dysfunction and its relationship to the physiological function of tau, however, is unclear. Here, we demonstrate that tau regulates mitochondrial reverse electron transport (RET), which produces excess reactive oxygen species (ROS), reduces the NAD+/NADH ratio, and is activated by aging or stress. In flies, mice, and human induced pluripotent stem cell (hiPSC)-derived neurons, t…

Paraclostridium sp. AKS46 vesicles facilitate extracellular electron transport.

AIMS: Paraclostridium sp. AKS46 was shown to have high exoelectrogenic activity. The current study investigated whether membrane vesicles (M … produces extracellular vesicles packed with electron-rich molecules and thus, these vesicles act as electron transporters. The work highlights a vesicle-mediated mechanism for energy harvesting from waste degradation in microbial fuel cells.

Targeting the Electron Transport System for Enhanced Longevity.

Damage to mitochondrial DNA (mtDNA) results in defective electron transport system (ETS) complexes, initiating a cycle of impaired oxidative phosphorylation (OXPHOS), increased reactive oxygen species (ROS) production, and chronic low-grade inflammation (inflammaging). This culminates in energy failure, cellular senescence, and progressive tissue degeneration. Rapamycin and metformin are the most extensively studied longevity drugs. Rapamycin inhibits mTORC1, promoting mitophagy, enhancing mitochondrial biogenesis, and reducing inflammation. Metformin partially inhibits Complex I, lowering rev…

Enhancing ROS-Inducing Nanozyme through Intraparticle Electron Transport.

Iron oxide nanoparticles (IONPs) have garnered significant attention as a promising platform for reactive oxygen species (ROS)-dependent dis … iron (Fe0 and Fe2+ ), which accelerates the recycling of surface Fe3+ to Fe2+ through intraparticle electron transport. Additionally, ultrasmall wüstite nanoparticles are generated by tuning the thermodecomposition-based nanocrystal synthesis, resulting in a Fenton reaction rate 5.3 times higher than that of ferumoxytol, an FDA-approved IONP. Compared with ferumoxyt …

Electron-Transfer Gated Ion Transport in Carbon Nanopipets.

Coating the inner wall of a quartz nanopipet with a thin layer of carbon yields a nanopore with tunable surface charge and chemical state for resistive-pulse and rectification sensing. Herein we report the experimental study and modeling of the electron-transfer gated ion transport processes in carbon nanopipets. The potential of the unbiased carbon layer can be tuned by adding very low (sub-nM) concentrations of redox species to the solution via bipolar electrochemistry. The potential of the carbon layer determines the electrical double-layer structure that, in turn, affects the ionic transpo

Plasma membrane electron transport in frog blood vessels.

In an attempt to see if frog blood vessels possess a plasma membrane electron transport system, the postcaval vein and aorta isolated from Rana tigrina were tested for their ability to reduce ferricyanide, methylene blue, and 2,6-dichloroindophenol. While the dyes remained unchanged, ferricyanide was reduced to ferrocyanide. This reduction was resistant to inhibition by cyanide and azide. Heptane extraction or formalin fixation of the tissues markedly reduced the capability to reduce ferricyanide. Denuded aortas retained only 30% of the activity of intact tissue. Our results indicate that the …

Electron-electrolyte coupling in AC transport through nanofluidic channels.

The transport properties of nanofluidic channels are usually studied under constant (DC) voltage or pressure driving. However, the frequency response under sinusoidal (AC) drivings offers rich insights into the time-dependent transport mechanisms. Inspired by recent electrochemical approaches, we investigate the couplings between ionic and electronic transport under AC driving. We show that conduction electrons of the channel walls participate in ionic current via capacitive electrochemical coupling, defining a critical frequency and length scale where electron-dominated conductivity emerges.
