Effect of fenbendazole on an autoimmune mouse model.

Fenbendazole is an anthelmintic drug widely used to treat and prevent pinworm infection in laboratory rodents. Data regarding possible side effects of fenbendazole on the immune system are conflicting, potentially due to the design of treatment protocols. The purpose of the current study was to determine the effects of 2 fenbendazole therapeutic regimens (continuous for 5 wk and alternating weeks [that is, 1 wk on, 1 wk off] for 9 wk) on the development of autoimmune disease in (NZB × NZW)F1 mice. No significant differences in survival curves or weight were observed between the treatment group…

Safety of fenbendazole in common peafowl (Pavo cristatus).

The present study was undertaken to find out the safety levels of fenbendazole in common peafowl. This bird, raised on aviaries and zoos, can be severely parasitized with Ascaridia galli (enteric worms) and Syngamus trachea (gapeworm) along with other parasitic worms. Fenbendazole is a highly effective benzimidazole-class anthelmintic in animals. The objective of this work was to provide target animal safety data in young peafowl and to demonstrate reproductive safety in adult birds. During the experimental study, diets containing fenbendazole at 0, 100, 200 and 300 ppm were fed for 21 days (t…

Biologic effects of fenbendazole in rats and mice: a review.

This review summarizes findings from toxicologic, carcinogenic, immunologic, and metabolic studies on fenbendazole (FBZ). Currently, FBZ is used to treat or prevent pinworm outbreaks in laboratory rodents. Because antiparasitic treatments usually are not part of experimental designs, interactions from the medication on the outcomes of ongoing experiments are a concern. At therapeutic levels, FBZ does not alter the total content of cytochromes P450 but does induce certain hepatic cytochrome P450 isoforms, namely 1A1, 1A2, and 2B1. Although expressed constitutively at low or undetectable levels,…

Fenbendazole for treatment of Paragonimus kellicotti infection in dogs.

The effect of fenbendazole therapy was studied in 9 dogs with pulmonary paragonimiasis induced by inoculation of metacercariae (25/dog) of Paragonimus kellicotti. At 42 to 47 days after 6 dogs were inoculated, they were given fenbendazole in 2 divided doses totaling 50 mg (4 dogs) or 100 mg (2 dogs)/kg of body weight each day for 10 to 14 days. Three dogs were not treated. The passage of Paragonimus eggs in the feces ceased after 3 days at the high dosage and after 3 to 8 days at the low dosage. All dogs were euthanatized and necropsied on day 14. Live flukes were not recovered from the lungs …

Efficacy of Fenbendazole against gastrointestinal parasites of sheep.

Lambs severely parasitized by Haemonchus, Ostertagia, Trichostrongylus, Cooperia, Oesophagostomum, and Nematodirus were treated with fenbendazole (methyl 5-(phenylthio)-2-benzimidazolecarbamate) given at 3 dose levels: 3.5, 5.0, and 7.5 mg/kg of body weight. Efficacies against these genera, except Haemonchus and Nematodirus, were 100% at the 3 dose levels. Efficacies for the 3 doses against Haemonchus were 93.4, 95.3, and 99.8%, respectively, and against Nematodirus, 99.5, 99.6, and 100%. Efficacies for the doses against Trichuris were 69.1, 83.6, and 98.2%. Toxicosis due to the durg was not s…

Comparison of the larvicidal efficacies of moxidectin or a five-day regimen of fenbendazole in horses harboring cyathostomin populations resistant to the adulticidal dosage of fenbendazole.

Despite widespread acknowledgement of cyathostomin resistance to adult icidal dosages of benzimidazole (BZD) anthelmintics, many strongyle control programs continue to feature regularly scheduled larvicidal treatment with fenbendazole (FBZ). However, no studies have been conducted to evaluate the efficacy of larvicidal regimens against encysted cyathostomins in a BZD-resistant (BZD-R) population. A masked, randomized, controlled clinical study was conducted with 18 juvenile horses harboring populations of cyathostomins that were considered BZD-R on the basis of fecal egg count reduction (FECR)…

Exacerbation of acetaminophen hepatotoxicity by the anthelmentic drug fenbendazole.

Fenbendazole is a broad-spectrum anthelmintic drug widely used to prevent or treat nematode infections in laboratory rodent colonies. Potential interactions between fenbendazole and hepatotoxicants such as acetaminophen are unknown, and this was investigated in this study. Mice were fed a control diet or a diet containing fenbendazole (8-12 mg/kg/day) for 7 days prior to treatment with acetaminophen (300 mg/kg) or phosphate buffered saline. In mice fed a control diet, acetaminophen administration resulted in centrilobular hepatic necrosis and increases in serum transaminases, which were eviden…

Effects of fenbendazole on the murine humoral immune system.

Pinworms are highly contagious parasites that have been effectively treated in laboratory rodents with fenbendazole (FBZ). Whether FBZ has any detrimental side effects that may compromise experimental results is unknown. Here we asked whether the immune systems from young and aged mice are altered under FBZ treatment. We compared control and FBZ-treated groups of young (age, 2 to 4 mo) and old (age, 22 to 24 mo) BALB/cN mice. The treated mice received a total of 4 wk (alternating-week treatment regimen) of FBZ-medicated feed. Spleen and bone marrow were collected for immunologic assays, and he…

The effects of mebendazole and fenbendazole on Trichinella spiralis in mice.

Oral administration of either Mebendazole or Fenbendazole at 50 mg/kg body weight was effective in killing a 7-hr Trichinella spiralis infection in mice. Adult worms, 72 hr postingection, were unaffected by the same dose of Fenbendazole, while Mebendazole showed partial activity. Both Mebendazole and Fenbendazole were active against developing muscle larvae. Mebendazole was effective against encysted muscle larvae while 50 mg of Fenbendazole per kg per day for 7 days from 28 days postinfection failed to have any effect on this stage of the life-cycle.

Influence of subclinical nematodosis on the kinetic disposition of fenbendazole in buffaloes.

The disposition kinetics of fenbendazole was studied in buffaloes subclinically infected with gastrointestinal nematodes. There was significantly reduced uptake of the drug in infected animals compared to uninfected controls. The pH of the duodenal liquor was highly alkaline compared to the acidic pH in uninfected animals. The egg count in the faeces never became zero though the numbers were reduced to a great extent compared to pre-treatment values. The influence of the host's physiology on the reduced bioavailability of fenbendazole is discussed.

Evaluation of fenbendazole as an extended anthelmintic treatment regimen for swine.

Fenbendazole was given in the feed to swine at a cumulative dosage of 9 mg/kg of body weight over a period of 3, 6, and 12 days to compare efficacy. Four treatment groups of ten 2- to 3-month-old pigs each, with a mean of 15 kg of body weight per group, received 3 mg of fenbendazole/kg/day for 3 days, 1.5 mg/kg/day for 6 days, 0.75 mg/kg/day for 12 days, and no medication. Medicated feed was scheduled so that all treated pigs reached the last day of treatment on the same day, thus making the time between the last treatment and necropsy equal for all groups. Ascaris suum and Trichuris suis were…

[Cholestasis following use of fenbendazole as alternative cancer treatment].

BACKGROUND: Many patients use alternative cancer treatments, but evidence of their efficacy is limited. Fenbendazole is reported to cause cholangitis and liver failure in humans. Cholangitis is a very rare complication of immunotherapy. CASE PRESENTATION: A patient with cancer was admitted two weeks after immunotherapy due to a tenfold increase in cholestatic parameters. He was in good clinical condition, with normal ASAT/ALAT, bilirubin and INR. Ultrasonography findings were normal, and MRCP showed focal inflammation. No immunosuppressive treatment was initiated. We were informed that the pa…

Anthelmintic resistance of horse strongyle nematodes to fenbendazole in Lithuania.

BACKGROUND: Control of strongyle infections presents a global challenge for horse practitioners due to the development of anthelmintic resistance (AR), however comprehensive information on AR in Lithuania is still lacking. The aim of this study was to assess the current situation of fenbendazole (FBZ) AR in horses at stable level in Lithuania. RESULTS: Faecal samples from 121 horses from six stables were examined using the Mini-FLOTAC method. Of these, 89 horses met the inclusion criteria that included strongyle faecal egg counts (FEC) exceeding 200 eggs per gram (EPG). Faecal egg count reduc…
