HLA-G promotes immune tolerance.

HLA-G is a non-classical major histocompatibility complex class I molecule that differs from the classical HLA-A, -B and -C molecules by (i) alternative splicing of mRNAs encoding for at least four membrane-bound and two soluble HLA-G isoforms, (ii) a limited polymorphism, and (iii) a tissue-restricted distribution. Studies over the past few years have elucidated the function of HLA-G demonstrating inhibition of both NK cell- and T cell-mediated cytolysis. Furthermore, aside from its expression during pregnancy, we have shown that HLA-G is also expressed in solid tumor cells (i.e. human melano

Immune Tolerance Regulation Is Critical to Immune Homeostasis.

The body's immune response plays a critical role in defending against external or foreign antigens while also preserving tolerance to self-antigens. This equilibrium, referred to as immune homeostasis, is paramount for overall health. The regulatory mechanisms governing the maintenance of this delicate immune balance are notably complex. It is currently accepted that immune tolerance is a dynamic outcome regulated by multiple factors, including central and peripheral mechanisms. Its induction or elimination plays a significant role in autoimmune diseases, organ transplantation, and cancer ther…

The development of inhibitors in haemophilia patients is one of the most serious challenges to effective treatment. The effect of factor (F) … des in haemophilic patients. In group 1, inhibitor titres decreased markedly 0.5-21 months prior to immune tolerance therapy (ITT) and remained low for a further 1-19 months. However, use of rFVIIa instead of aPCC as prophylactic treatment in group 2 patients undergoing ITT failed to show favourable results for rFVIIa. This may be due to the short half-life of rFV …

New strategies in immune tolerance induction.

Induction of tolerance in clinical organ transplantation that will obviate the use of chronic immunosuppression and preserve host immune response to other antigens remains the goal of transplant research. The thymus plays a critical role in the ability of the immune system to discriminate between self- and nonself-antigens or harmful and harmless alloantigens. We now know that multiple factors determine how the immune system responds to a self-antigen or foreign antigen. These determinants include developmental stage of the host, stage of T-cell maturity, site of antigen encounter, type and ma

Fetomaternal Immune Tolerance: Crucial Mechanisms of Tolerance for Successful Pregnancy in Humans.

For many years, the question of how the maternal immune system tolerates the foreign fetus has remained unanswered, and numerous studies hav … ells, dendritic cells, decidua cells, IDO, Class I HLA molecules, TGF-β, and IL-10, induce maternal immune tolerance in the fetus in numerous ways. The findings on fetomaternal immune tolerance have remained controversial and require further research.

Tryptophan metabolism in liver transplantation immune tolerance.

Liver transplantation is a life-saving treatment for end-stage liver disease, but long-term outcomes are limited by complications of lifelong immunosuppression. Inducing immune tolerance has become a major research priority. Tryptophan (Trp) metabolism, particularly the kynurenine pathway, is a key endogenous regulator of peripheral tolerance. This review moves beyond a simple description of metabolic routes and provides a critical, integrated analysis. We systematically compare the non-redundant immunosuppressive roles of indoleamine 2,3-dioxygenase (IDO) and tryptophan 2,3-dioxygenase (TDO2)…

Update: Immune Tolerance Induction Practice in Haemophilia.

ABSTRACT: The emergence of non-factor therapies has fundamentally changed inhibitor management in haemophilia. Historically, immune tolerance induction (ITI) was considered indispensable for eradicating factor VIII (FVIII) or IX inhibitors and restoring responsiveness to replacement therapy. However, the introduction of rebalancing therapies (TFPI inhibitors, tissue factor pathway inhibitors) and emicizumab, a bispecific antibody mimicking factor VIIIa activity, provides highly effective bleed protection regardless of inhibitor status, challenging the traditional paradigm of universal ITI. Sub…

Deciphering immune tolerance in allogeneic pig pregnancy.

Embryo transfer (ET) has transformed swine biotechnology, enabling genetic advancements and disease control. However, its success remains inconsistent, partly due to immune-mediated challenges at the maternal-fetal interface. This study explores the immunological environment of hemi-allogeneic pregnancies (via artificial insemination, AI) versus allogeneic pregnancies (via ET) in pigs during the critical implantation phase. Sows were categorized into groups based on pregnancy type and fetal counts, reflecting varying outcomes. Endometrial immune cell populations, including T lymphocytes, regul

Endometrial Microbiota and Immune Tolerance in Pregnancy.

Recent studies have demonstrated that the uterus has its own microbiota. However, there is no consensus on endometrial microbiota composition, thus its role in the healthy uterine environment is still a frontier topic. Endometrial receptivity is key to embryo implantation, and in this specific context immunological tolerance against fetal antigens and the tightly regulated expression of inflammatory mediators are fundamental. According to recent evidence, endometrial microbiota may interact in a very dynamic way with the immune system during the peri-conceptional stage and later during pregnan

Insm1 regulates mTEC development and immune tolerance.

The expression of self-antigens in medullary thymic epithelial cells (mTECs) is essential for the establishment of immune tolerance, but the regulatory network that controls the generation and maintenance of the multitude of cell populations expressing self-antigens is poorly understood. Here, we show that Insm1, a zinc finger protein with known functions in neuroendocrine and neuronal cells, is broadly coexpressed with an autoimmune regulator (Aire) in mTECs. Insm1 expression is undetectable in most mimetic cell populations derived from mTECs but persists in neuroendocrine mimetic cells. Muta…

TETs Link Hydrogen Sulfide to Immune Tolerance.

Demethylation of the Foxp3 locus maintains gene expression and Treg cell stability. Yang et al. (2015) show that the gasotransmitter hydrogen sulfide co-operates with growth factor TGF-β and interleukin-2 to activate Tet-mediated DNA demethylation of Foxp3 to promote immune tolerance.

[The young hemophiliac, inhibitors and immune tolerance].

The occurrence of inhibitor in a hemophilic patient is the greatest therapeutic complication in 1999. The inhibitor incidence is higher in hemophilia A (20-30%) than in hemophilia B (3%). At the moment, the best management is immune tolerance induction. This consists in frequent infusion of antihemophilic factor (every day or every other day). The risk of inhibitor development is higher in a young child than in an adult. Consequently, a venous access device is essential for this treatment although not devoid of complications in the young boy. Moreover, the probability of inhibitor disappearanc…
