Ocular and cutaneous melanomas arose in new inbred lines of transgenic mice having an integrated recombinant gene comprised of the tyrosinase promoter, expressed in pigment cells, and the simian virus 40 early-region transforming sequences. The tumors were hypomelanotic and were histopathologically similar to corresponding human melanomas. Eye melanomas often originated at a young age, chiefly from the retinal pigment epithelium, also from the choroid, and rarely from the ciliary body. The eye tumors grew aggressively, were highly invasive, and metastasized to local and distant sites. The earl

A mouse tyrosinase minigene, mg-Tyrs-J, in which genomic 5' noncoding flanking sequence was fused to a mouse tyrosinase cDNA, was introduced into fertilized eggs of BALB/c albino mice. Six transformed mice that exhibited brown agouti hair with some variations in the intensity of pigmentation were obtained. By crossing the founder mice with BALB/c albino mice, transgenic lines and sublines were established. Each subline expressed a characteristic phenotype with their respective band patterns in Southern blot analysis. This indicates that the character is expressed by transgenes integrated at a

While using transgenic mice to study the regulation of alpha-fetoprotein (AFP) it was noted that two different alpha-fetoprotein-chloramphenicol acetyl transferase (CAT) transgenes resulted in the appearance of craniofacial anomalies in 11% of the offspring derived from crosses between transgenic mice and nontransgenic mates. A total of 13 fetuses exhibited abnormalities; two are described in detail. Ninety-two percent of the affected fetuses had some form of mandibular abnormality while zygomatic and ossicular defects appeared in more than 40% of the specimens. Aglossia and aberrant musculatu

Transgenic mice expressing the cell receptor for poliovirus have been generated and are susceptible to poliovirus infection. TgPVR mice have been used to answer questions about the pathogenesis of poliovirus infection. Despite the widespread pattern of PVR expression, poliovirus infection in TgPVR mice is restricted to only a few sites, indicating that poliovirus tropism is not controlled solely by the ability of cells to bind virus. After intramuscular inoculation, poliovirus travels to the spinal cord by axonal transport. This route of entry into the central nervous system may play a role in

A useful approach to establishing the biologic actions, in vivo, of granulocyte-macrophage colony-stimulating factor (GM-CSF) is to assess the consequences of long-term elevation of the factor in transgenic mice. Two lines of transgenic GM-CSF mice were analyzed. The major abnormality exhibited was an elevation in the number of macrophages, eosinophils, and polymorphs in the peritoneal and pleural cavities. Disease states exhibited by the lines were dependent on the insertion site of the GM-CSF gene. These disease states seem best explained on the basis of CSF-mediated macrophage activation an

Although autoreactive T cells are thought to play a prominent role in autoimmune disease in MRL-lpr/lpr mice, it has been difficult to directly determine if autoreactive T cells escape from the thymus and react with self-antigens in the periphery. Defective expression of the Fas apoptosis antigen in MRL-lpr/lpr mice results from the insertion of the ETn retrotransposon. The fas defect can be partially corrected in CD2-fas transgenic mice in which the expression of fas is corrected in T cells. To identify a possible defect in clonal deletion or clonal anergy induction of auto-specific T cells,

Persistent hepatitis B virus (HBV) infection is epidemiologically closely associated with the development of human hepatocellular carcinoma (HCC). A molecular mechanism that would explain the strong epidemiological link of persistent HBV infection with HCCs is still open to debate. Among HBV carriers, HCC develops only in patients with chronic liver disease, such as chronic active hepatitis, liver fibrosis, or liver cirrhosis. In other words, hepatitis-related proliferative changes, which are mainly sustained by repeating cycles of cell death and regeneration either immunologically or through

Muscle mass is regulated by a wide range of hormonal and nutritional signals, such as insulin and IGF. Tuberous sclerosis complex (TSC) is an inherited hamartoma disease with tumor growth in numerous organs. TSC is caused by mutation in either TSC1 or TSC2 tumor suppressor genes that negatively regulate insulin-induced S6K activation and cell growth. Here we report that expression of human TSC1 (hTSC1) in mouse skeletal muscle leads to reduction of muscle mass. Expression of hTSC1 stabilizes endogenous TSC2 and leads to inhibition of the mTOR signaling. The hTSC1-mTSC2 hetero-complex and its d

PURPOSE: To produce transgenic mice that express the SV40 T-antigen oncogene specifically in photoreceptor cells, giving rise to retinoblastoma tumors of photoreceptor cell origin; to characterize the mice with regard to transgene expression and pathology and to characterize the resulting tumors histologically.
METHODS: Transgenic mice were generated that express T-antigen under the control of the murine interstitial retinol binding protein promoter.
RESULTS: All mice produced developed either ocular or intracranial tumors, or both, at an early age. One line of mice was generated, and all mi

A distinctive feature of gonadal maturation in mammals is the movement to an extraabdominal location. Testicular descent is a complex, multistage process whereby the embryonic gonads migrate from their initial abdominal position to the scrotum. Failure in this process results in cryptorchidism, a frequent congenital birth defect in humans. We report here a new mouse transgenic insertional mutation, cryptorchidism with white spotting (crsp). Males homozygous for crsp exhibit a high intraabdominal position of the testes, associated with complete sterility. Heterozygous males have a wild-type phe

Melanosis was found to various extents in a wide array of tissues of all 23 autopsied mice whose transgene consisted of the tyrosinase promoter fused to the simian virus 40 early-region oncogenic sequences. Pigmentation in a given animal was attributable to any or all of the following; an increase in numbers of some normally pigmented cells of neural crest origin (a result compatible with early stages of transformation); elicitation of melanin synthesis in some cells that normally have little melanin, or none at all (the latter possibly signaling metaplasia); unusual intercellular transfer of

In order to study the oncogenesis of melanocytes, transgenic mouse lines were established that express a mutated human Ha-ras (TPras) gene in pigment producing cells. The ras transgenic mice exhibit an altered phenotype, including melanocytic hyperplasia and a muted agouti coat, indicative of hyperproliferative melanocytes. These mice and their wild-type littermates have been subjected to a variety of carcinogenesis protocols, including 7, 12-dimethylbenz-[a]anthracene (DMBA), 12-O-tetradecanoylphorbol-13-acetate (TPA) and UV radiation exposure. Topical DMBA treatment of TPras mice resulted in

Although infestations by a number of Demodex mite species have been described in mice, the occurrence of Demodex musculi infestation was last reported by Hirst in 1917. This communication describes the occurrence of D. musculi infestation in two lines of transgenic mice and their F1-hybrid offspring. We first found the Demodex mite in mouse hair samples collected during efficacy screenings in an ongoing ectoparasite treatment trial for the fur mite Radfordia affinis. An investigation was undertaken to determine the extent of the Demodex infestation within the facility and the original source o

OBJECTIVE: Transgenic (Tg) mice expressing chimeras of mouse and human prion proteins (PrPs) have shorter incubation periods for Creutzfeldt-Jakob disease (CJD) prions than mice expressing full-length human PrP. Increasing the sequence similarity of the chimeric PrP to mouse PrP, by reverting human residues to mouse, resulted in a Tg line, denoted Tg22372, which was susceptible to sporadic (s) CJD prions in approximately 110 days.
METHODS: Mice expressing chimeric mouse/human PrP transgenes were produced. The mice were inoculated intracerebrally with extracts prepared from the brains of patie

When present in excess amounts during fetal life, androgens can impair female development by inducing masculinization. On way to modify fetal steroid concentration is by altering the expression of hydroxysteroid (17beta) dehydrogenases (HSD17Bs). Human HSD17B1 converts weak estrogen estrone to estradiol, and with lower catalytic efficiency, weak androgen androstenedione to testosterone. We have recently shown that over-expression of human HSD17B1 in transgenic mice results in masculinized phenotype in female mice. In the present study, we further show that in addition to the Müllerian ducts, H

Maternal diabetes during pregnancy is responsible for the occurrence of diabetic embryopathy, a spectrum of birth defects that includes heart abnormalities, neural tube defects, and caudal dysgenesis syndromes. Here, we report that mice transgenic for the homeodomain transcription factor Isl-1 develop profound caudal growth defects that resemble human sacral/caudal agenesis. Isl-1 is normally expressed in the pancreas and is required for pancreas development and endocrine cell differentiation. Aberrant regulation of this pancreatic transcription factor causes increased mesodermal cell death, a

Recent experiments have shown that the microinjected kappa-chain gene of transgenic mice is expressed in a tissue-specific fashion only in B lymphocytes. The next step was to determine whether, within the B-lymphocyte lineage, the kappa-chain gene was expressed in a normal developmental fashion. Normally, only mu heavy(H)-chain genes, and not kappa-chain genes, are expressed in pre-B cells. To obtain cloned cell lines derived from early cells of the B-cell lineage, we transformed bone marrow cells from kappa-transgenic mice with Abelson murine leukaemia virus (A-MuLV) and tested the resultant

Calcitonin gene-related peptide (CGRP) plays an important role in several physiological processes such as vasodilation, cardiovascular homeostasis and transmission of pain. Here we report the generation of a transgenic mouse overexpressing αCGRP and the impact of this on baseline physiological responses. αCGRP transgenic mice displayed significantly increased αCGRP mRNA levels in the kidney, heart and hippocampus. To assess cardiovascular physiology, we measured arterial pressure using a tail cuff system. Heart rate, systolic pressure, mean arterial pressure and diastolic pressure were signifi
