Nutrigenomics in postgenomic era.

The last years modified major our conceptions about nutrition. These revolutionary changes were produced by implementation of new techniques of functional genomics. The nutrigenomics and nutrigenomics provide powerful approaches to unravel the complex relationships between bioactive molecules, genetic polymorphisms and biological system and can give rise to personalized nutrition and dietary recommendations. In monogenic diseases (phenylketonuria, galactosemia, lactose intolerance etc.) diet influence phenotypic expression and nutrigenomics will improve the prevention or treatment by the early…

Nutrigenomics and inflammatory bowel diseases.

The field of nutrigenomics recognizes gene-diet interactions, with regard to both the impact of genetic variation on nutrient requirements, and conversely nutrient regulation of the expression of genes. Crohn's disease and ulcerative colitis are inflammatory bowel diseases for which twin studies reveal genetic susceptibility that is impacted by diet and environment. Apparently contradictory data on the role of diet in inflammatory bowel disease would be entirely explainable if genetic variability determined dietary requirements and intolerances. Considering Crohn's disease, we recognize three …

Uninformed consent in nutrigenomic research.

Genetic testing for personalizing diet and wellness programs is performed without extensive counseling that informs about the potential implications of knowing one's genotype status. Genetic counseling seems redundant for genes that impact the effect of diet on biomarkers such as cholesterol and blood pressure, but the same genes may have pleiotropic effects that cannot be ignored. A well-known example is the APOE gene, which is implicated in cholesterol regulation and is a major risk factor for Alzheimer's disease. Not fully informing participants about the major pleiotropic effects of genes

Microbiota and nutrigenomics: beneficial allies or hidden threats.

Nutrigenomics explores how nutrients interact with the genome to influence metabolic and physiological functions through microbial and molecular mechanisms. Nutrients and bioactive food compounds act as biochemical modulators that regulate gene expression and immune responses, sustaining physiological stability. On the other hand, the gut microbiota create a balanced, complex ecosystem that is vital for maintaining host physiology and metabolic balance. The type and composition of diet directly shape gut microbial communities and populations, which, in turn, influence host gene expression. Dis…

Nutrigenomics and Precision Nutrition in Inflammatory Bowel Disease.

Inflammatory bowel disease (IBD) represents a growing public health challenge worldwide, and its prevalence is increasing year by year. A growing body of evidence suggests that diet plays a key role in shaping the gut microbiota, whose dysbiosis is a significant component of IBD pathogenesis. In response to the limitations of traditional, one-size-fits-all dietary recommendations, the concept of precision nutrition is emerging, which involves tailoring nutrition to a patient's clinical, biological, and genetic characteristics. Of particular importance in this context is nutrigenomics, the scie…

[Nutrigenomics. Scientific basis, status and perspectives of application].

Nutrigenomics investigates the interaction between nutrition and the genome, thereby combining nutritional research with functional genomics. Its aims are (1) to correlate heterogeneous effects of nutrients with sequence variations in the genome and (2) to investigate the effects of nutrients and other food components on gene expression at a genome-wide scale (mRNA profiling), and on patterns of metabolite alterations in serum (metabolite profiling). The field will provide important information as to the biological effects of food components, and to the functional consequences of genetic varia…

Vitamin B2 and B3 nutrigenomics reveals a therapy for NAXD disease.

Vitamins are essential metabolites that must be obtained from external sources. In modern times, they have become widely available, leading to their ad hoc consumption. We developed a nutritional genomics framework to systematically identify monogenic diseases responsive to micronutrient modulation. Genome-wide CRISPR screens under varying vitamin B2 and B3 levels revealed dozens of candidate disease genes amenable to rescue by individual vitamins. In the vitamin B3 screen, NAD(P)HX dehydratase (NAXD) was the top hit; this enzyme repairs an aberrant, hydrated form of NADH (6-hydroxy-1,4,5,6-te

Nutrigenomics of taste - impact on food preferences and food production.

Food preferences are influenced by a number of factors such as personal experiences, cultural adaptations and perceived health benefits. Taste, however, is the most important determinant of how much a food is liked or disliked. Based on the response to bitter-tasting compounds such as phenylthiocarbamide (PTC) or 6-n-propylthiouracil (PROP), individuals can be classified as supertasters, tasters or nontasters. Sensitivity to bitter-tasting compounds is a genetic trait that has been recognized for more than 70 years. Genetic differences in bitter taste perception may account for individual diff

Genetic predispositions and nutritional strategies: a review of nutrigenomics in Parkinson's disease.

Parkinson's disease (PD) is a neurodegenerative disease that manifests motor and non-motor symptoms due to the progressive loss of dopaminergic neurons in the substantia nigra. While its pathogenesis is multifactorial, with genetic as well as environmental components, more and more evidence suggests that nutrition and diet play an important role in regulating both the development and progression of the disease through crosstalk with genetics, a term known as nutrigenomics. This review discusses genetics and nutrition in PD, emphasizing putative mechanisms underlying neurodegeneration, includin…

Personalized dietary management of advanced prostate cancer using nutrigenomics: a case report.

While there are emerging reports in the scientific literature on potential associations between cholesterol/lipids and prostate cancer, information on the dietary management of these cancer patients is currently lacking. We report on a 57-year-old white Australian male diagnosed with advanced prostate cancer who had personalized dietary management in preparation for and following his medical treatment: radiation and radical prostatectomy. Dietary recommendations were based on his blood results and nutrigenomic tests which showed a history of and genetic predisposition to dyslipidemia. Nutritio

Nutrigenomics and Epigenetics in the Dietary Management of Inflammatory Bowel Diseases.

Inflammatory bowel diseases (IBD), including Crohn's disease (CD) and ulcerative colitis (UC), are chronic diseases with complex aetiology involving genetic, immunological, and environmental factors and intestinal microbiota disorders. Mutations in genes such as NOD2, ATG16L1, IRGM, TLR4, and IL23R disrupt the functioning of the intestinal barrier and the immune response, increasing susceptibility to chronic inflammation. Recent studies indicate that interactions between diet, gene expression, and epigenetic mechanisms play a key role in modulating the course of IBD, e.g., DNA methylation, his

Insights into garlic (Allium Sativum)'s nutrigenomics-associated fly-repellent potency in cattle.

Despite effective control of flies using synthetic pesticides, fly resistance and environmental contamination have led to the inadequacy of this strategy. The use of integrated pest management approaches has since been advocated in contemporary research to sustainably control fly populations. Recent studies have found garlic (Allium Sativum) and its derivative bioactive compounds to possess insect-repellent attributes among other key health and production enhancing properties. This highlights the potential of garlic as a botanical pesticide to control flies in cattle. Moreover, the ability of

Nutrigenomics of fat-soluble vitamins and micronutrients in hepatocyte lipotoxicity and MASLD.

Metabolic dysfunction-associated steatotic liver disease (MASLD) is a highly prevalent liver disorder deriving from the chronic exposure to hepatocyte steatosis and its lipotoxicity effects, as mitochondrial dysfunction, overactivation of stress response and inflammatory genes, and cell death signaling. Genomics and post-genomic disciplines are now offering unprecedent opportunities in disease mechanisms characterization, precision diagnostics, and treatment. These disciplines include nutrigenomics, i.e. the use of genomics techniques to study the health effects of the interaction between diet…
