Comparison of Anorectic Potencies of Type A Trichothecenes T-2 Toxin, HT-2 Toxin, Diacetoxyscirpenol, and Neosolaniol.

Trichothecene mycotoxins are common contaminants in cereal grains and negatively impact human and animal health. Although anorexia is a common hallmark of type B trichothecenes-induced toxicity, less is known about the anorectic potencies of type A trichothecenes. The purpose of this study was to compare the anorectic potencies of four type A trichothecenes (T-2 toxin (T-2), HT-2 toxin (HT-2), diacetoxyscirpenol (DAS), and neosolaniol (NEO)) in mice. Following oral exposure to T-2, HT-2, DAS, and NEO, the no observed adverse effect levels (NOAELs) and lowest observed adverse effect levels (LOA

T-2 toxin as an emetic factor in moldy corn.

Extracts of Fusarium poae (NRRL 3287) grown either on sterile corn at 8 C or in Richards solution at room temperature were shown to have emetic activity in pigeons at nonlethal concentration under conditions of oral and intravenous administration. The causative agent was found to be T-2 toxin (3-hydroxy-4,15-diacetoxy-8-[3-methylbutyryloxy]-12,13-epoxy-Delta(9)-trichothecene). Oral and intravenous mean toxic dose values for this compound were found to be 0.72 and 0.15 mg/kg, respectively, as compared with an oral mean lethal dose of 2.75 mg/kg. The fact that T-2 toxin causes emesis at nonletha

T-2 Toxin Neurotoxicity: Molecular Mechanisms and Emerging Chemoprotective Strategies.

Mycotoxins can contaminate food and raw food materials and are a threat to animal and human health. T-2 toxin is the most toxic secondary metabolite mainly produced by Fusarium species among trichothecenes. T-2 toxin exposure can induce multiple toxic effects, including hepatotoxicity, nephrotoxicity, immunotoxicity, gastrointestinal toxicity, and reproductive toxicity. Recent studies have reported that T-2 toxin can cross the blood-brain barrier and trigger neurotoxicity. In this review, we summarized the neurotoxic effects caused by T-2 toxin exposure and the underlying molecular mechanisms.

Combined Inhibitory Effects of Citrinin, Ochratoxin-A, and T-2 Toxin on Aquaporin-2.

Aquaporins form a large family of transmembrane protein channel that facilitates selective and fast water transport across the cell membrane. The inhibition of aquaporin channels leads to many water-related diseases such as nephrogenic diabetes insipidus, edema, cardiac arrest, and stroke. Herein, we report the molecular mechanism of mycotoxins (citrinin, ochratoxin-A, and T-2 mycotoxin) inhibition of aquaporin-2 (AQP2) and arginine vasopressin receptor 2. Molecular docking, molecular dynamics simulations, quantum chemical calculations, residue conservation-coupling analysis, sequence alignmen

The role of gut microbiota in anorexia induced by T-2 toxin.

T-2 toxin is one of trichothecene mycotoxins, which can impair appetite and decrease food intake. However, the specific mechanisms for T-2 toxin-induced anorexia are not fully clarified. Multiple research results had shown that gut microbiota have a significant effect on appetite regulation. Hence, this study purposed to explore the potential interactions of the gut microbiota and appetite regulate factors in anorexia induced by T-2 toxin. The study divided the mice into control group (CG, 0 mg/kg BW T-2 toxin) and T-2 toxin-treated group (TG, 1 mg/kg BW T-2 toxin), which oral gavage for 4 wee

p38 mediates T-2 toxin-induced Leydig cell testosterone synthesis disorder.

T-2 toxin is an unavoidable food and feed contaminant that seriously threatens human and animal health. Exposure to T-2 toxin can cause testosterone synthesis disorder in male animals, but the molecular mechanism is still not completely clear. The MAPK pathway participates in the regulation of testosterone synthesis by Leydig cells, but it is unclear whether the MAPK pathway participates in T-2 toxin-induced testosterone synthesis disorders. In this research, testosterone synthesis capacity, testosterone synthase expression and MAPK pathway activation were examined in male mice and TM3 cells e

Protective Effect of Selenomethionine on T-2 Toxin-Induced Rabbit Immunotoxicity.

T-2 toxin is a trichothecene mycotoxin produced by fusarium species, which is mainly prevalent in grain and livestock feed. One of the main effects of this toxin is immunodepression. Previous studies have shown that T-2 toxin can cause damage to immune organs and impaired immune function in animals. However, selenomethionine (SeMet) as an organic selenium source can not only promote the growth and development of the body but also effectively improve the body's immune function. In this study, rabbits were exposed to 0.4-mg/kg T-2 toxin, and abnormal blood routine indicators were found in the ra

Reduced expression of α2 integrin is involved in T-2 toxin-induced matrix degradation in C28/I2 cells and cartilages from rats administrated with T-2 toxin.

T-2 toxin is a mycotoxin demonstrating several harmful effects on chondrocyte and cartilage functions. In the present study, we investigated the toxic effects of T-2 toxin on cartilage matrix degradation and evaluated the involvement of α2 integrin in T-2 toxin-induced matrix damage. In C28/I2 cells, T-2 toxin decreased cell viability in a dose-dependent manner. Regarding matrix degradation, T-2 toxin decreased type II collagen and increased matrix metalloproteinase 13 (MMP-13) expression. Moreover, T-2 toxin significantly decreased the expression of α2 integrin in C28/I2 cells, indicating imp

Emetic responses to T-2 toxin, HT-2 toxin and emetine correspond to plasma elevations of peptide YY3-36 and 5-hydroxytryptamine.

Trichothecene mycotoxins are a family of potent translational inhibitors that are associated with foodborne outbreaks of human and animal gastroenteritis in which vomiting is a clinical hallmark. Deoxynivalenol (DON, vomitoxin) and other Type B trichothecenes have been previously demonstrated to cause emesis in the mink (Neovison vison), and this response has been directly linked to secretion of both the satiety hormone peptide YY3-36 (PYY3-36) and neurotransmitter 5-hydroxytryptamine (5-HT). Here, we characterized the emetic responses in the mink to T-2 toxin (T-2) and HT-2 toxin (HT-2), two

Neurotoxic mechanisms of mycotoxins: Focus on aflatoxin B1 and T-2 toxin.

Aflatoxin B1 (AFB1) and T-2 toxin are commonly found in animal feed and stored grain, posing a serious threat to human and animal health. Mycotoxins can penetrate brain tissue by compromising the blood-brain barrier, triggering oxidative stress and neuroinflammation, and leading to oxidative damage and apoptosis of brain cells. The potential neurotoxic mechanisms of AFB1 and T-2 toxin were discussed by summarizing the relevant research reports from the past ten years. AFB1 and T-2 toxin cause neuronal damage in the cerebral cortex and hippocampus, leading to synaptic transmission dysfunction,

T-2 toxin causes dysfunction of Sertoli cells by inducing oxidative stress.

T-2 toxin is an inevitable mycotoxin in food products and feeds. It is a proven toxicant impairing the male reproductive system. However, previous studies have concentrated on the toxic effect of T-2 toxin on Leydig cells, with little attention on the Sertoli cell cytotoxicity. Therefore, this study aimed to establish the toxic mechanism of T-2 toxin on Sertoli cells. The Sertoli cell line (TM4 cell) was cultured and exposed to different concentrations of T-2 toxin with/without N-acetyl-L-cysteine (NAC) for 24 h. A CCK-8 assay then measured the cell viability. In addition, the expression of TM
