Thymocytes can tolerize thymocytes by clonal deletion in vitro.

Clonal deletion of thymocytes bearing TCR for self antigens is one major mechanism of T cell tolerance induction. Peptide antigen-induced deletion of thymocytes from alpha beta TCR transgenic mice has been studied using single cell suspension cultures. The results show that antigen-presenting immature CD4+CD8+ thymocytes can tolerize antigen-reactive immature thymocytes in vitro by programmed cell death (apoptosis) 6-8 h after antigen exposure. Antigen-induced apoptosis of immature thymocytes was inhibited by antibodies specific for the alpha beta TCR, CD3, CD8, and LFA-1 molecules. This impli…

Instructing Memory in CD8+ Thymocytes.

T cells can sometimes acquire properties of a memory cell without encountering foreign antigen. Miller et al. now show that development of such 'memory phenotype' (MP) CD8+ T cells starts with recognition of self-ligands in the thymus, and finishes in peripheral tissues. These cells respond rapidly during infection, and infiltrate tumors, where they express high amounts of PD-1.

Defects in thymocyte differentiation and thymocyte-stromal interactions in the trisomy 16 mouse.

We have examined fetal thymic development in the trisomy 16 (Ts16) mouse, which is considered to be a model for human trisomy 21, or Down Sy … o, but declines in vitro relative to normal thymuses. Reconstitution of depleted thymic stroma with thymocytes showed evidence of defects in both developmental compartments.

Thymocyte migration and emigration.

Hematopoietic precursors (HPCs) entering into the thymus undergo a sequential process leading to the generation of a variety of T cell subsets. This developmental odyssey unfolds in distinct stages within the thymic cortex and medulla, shaping the landscape of T cell receptor (TCR) expression and guiding thymocytes through positive and negative selection. Initially, early thymic progenitors (ETPs) take residence in the thymic cortex, where thymocytes begin to express their TCR and undergo positive selection. Subsequently, thymocytes transition to the thymic medulla, where they undergo negative…

Enhancement of chicken hemagglutinin response by thymocytes.

Antigen responsive cells, detectable with the cell transfer antibody assay, are abundant in the blood and spleen of chickens after immunizat … cells. The hemagglutinin response in the adoptive cell transfer system was enhanced when autologous thymocytes were added to the immunocyte-antigen mixture. Data from the kinetic and thymocyte supplement experiments taken together strongly indicate that the mouse erythrocyte antigen responsive cells of the chicken, like their mammalian counterpart the antige …

Age-related synthesis of glucocorticoids in thymocytes.

Glucocorticoids (GCs) are primarily synthesized in the adrenal glands but an ectopic production has also been reported in the brain, the gastrointestinal tract and in thymic epithelial cells (TEC). Here we show that thymocytes express genes encoding for all enzymes required for de novo GC synthesis and produce the hormone as demonstrated by both a GC specific reporter assay and a corticosterone specific ELISA assay. Interestingly, GC synthesis is detectable in cells from young mice (4 weeks) and thereafter increases during aging (14-22 weeks) together with an increased gene expression of the r…

Patterns of Differentially Expressed circRNAs in Human Thymocytes.

Circular RNAs (circRNAs) are suggested to play a discriminative role between some stages of thymocyte differentiation. However, differential aspects of the stage of mature single-positive thymocytes remain to be explored. The purpose of this study is to investigate the differential expression pattern of circRNAs in three different development stages of human thymocytes, including mature single-positive cells, and perform predictions in silico regarding the ability of specific circRNAs when controlling the expression of genes involved in thymocyte differentiation. We isolate human thymocytes at…

Beta-catenin expression in thymocytes accelerates thymic involution.

Age-related thymic involution in mammals is accompanied by decreased generation of naïve T cells without significant reduction in the number of peripheral T cells. This leads to inefficient immune responses and inadequate combating of infections and other challenges to the immune system in older mammals. The molecular mechanisms that underlie this phenomenon are not known. In this report we show that expression of beta-catenin in thymocytes enhances thymic involution. The effect of beta-catenin expression is seen in all the thymic sub-populations, suggesting that an age-related developmental p…

Sepsis-induced apoptosis of the thymocytes in mice.

Intraperitoneal injection of Gram-negative bacteria (Escherichia coli, Klebsiella pneumoniae, Pseudomonas aeruginosa) induces thymic atrophy in mice. The thymus weight, cell number, and viability began to decrease at 3 h, and reached their lowest level at 72 h. The thymocyte death was associated with DNA fragmentation of approximately 200 base pairs in ladder form. The kinetic study on histopathology revealed the process of thymocyte death and thymic atrophy. Flow-cytometric analysis showed that CD4+CD8+ thymocytes decreased predominantly. LPS caused thymocyte apoptosis, but only in LPS-respon…

Infection of human thymocytes by Epstein-Barr virus.

The Epstein-Barr Virus (EBV) causes infectious mononucleosis, and has been strongly associated with certain human cancers. The virus is thought to exclusively bind to B lymphocytes and epithelial cells via receptors (CR2/CD21) that also interact with fragments of the third component of complement (C3). Recent evidence, however, has challenged this belief. We have used two-color immunofluorescence analysis using biotin-conjugated EBV and streptavidin-phycoerythrin along with fluorescein-conjugated anti-T cell antibodies and demonstrated that CD1-positive, CD3-dull (immature) human thymocytes ex…

Terminal deoxynucleotidyl transferase activity in thymocytes of kittens.

Activity of terminal deoxynucleotidyl transferase (TdT) was measured in the thymus of 8- to 14-week-old kittens. Microscopic evidence of involution or other morphologic changes were not observed in formalin-fixed hematoxylin and eosin-stained thymic tissue sections. Thymic tissue from young kittens (8 weeks old) had low TdT activity, as determined by enzymatic catalysis or immunoassay procedures. Markedly higher TdT activity was measured in thymic tissue of older kittens (13 to 14 weeks old). Feline TdT catalyzed the polymerization of deoxynucleotides at a higher rate in Mn2+ than in Mg2+. The formaldehyde stability of thymocyte immunogens.

The differential effect of formaldehyde on mouse thymic and erythrocyte immunogens is studied using a simple technique. Changes are reflected in the antibody-independent properties of rabbit antisera to these treated cells. Taken as a whole all properties are quantitatively reduced in fifteen antisera but there are some notable exceptions. The five antisera with the highest lymphocytotoxic titres, having titres equal with fresh thymocyte antisera, show detectable levels of immunosuppression. This falls significantly below the level achieved with antisera to fresh thymocytes to suggest that som…

Compensatory anion currents in Kv1.3 channel-deficient thymocytes.

Kv1.3 is a voltage-gated potassium channel with roles in human T cell activation/proliferation, cell-mediated cytotoxicity, and volume regulation and is thus a target for therapeutic control of T cell responses. Kv1.3 is also present in some mouse thymocyte subsets and splenocytes, but its role in the mouse is less well understood. We report the generation and characterization of Kv1.3-deficient (Kv1.3-/-) mice. In contrast to wild-type cells, the majority of Kv1.3-/- thymocytes had no detectable voltage-dependent potassium current, although RNA and protein for several potassium channel subuni…

Long-term growth of malignant thymocytes in vitro.

We report a new methodology for the long-term growth of malignant T-lymphoblasts from patients with T-cell acute lymphoblastic leukemia (T-ALL) and T-cell lymphoblastic lymphoma (T-LL). When malignant cells were cultured in the presence of insulin-like growth factor I under hypoxic conditions, cellular proliferation occurred that resulted in the establishment of immortal cell lines from ten of 12 patient tumors. Authenticity of each cell line was verified by a direct comparison of the immunophenotype, karyotype, and immunogenotype with the patient's tumor cells. This improved method of cell cu

Differential chromatin looping regulates CD4 expression in immature thymocytes.

Runx1 binds the silencer and represses CD4 transcription in immature thymocytes. In this study, using looping chromatin immunoprecipitation and chromatin conformation capture assays, we demonstrated that interactions between Runx1 and positive elongation factor b (P-TEFb) appose the silencer and enhancer in CD4-negative thymoma cells and double-negative immature thymocytes. This chromatin loop decoys P-TEFb away from the promoter, thus preventing RNA polymerase II from elongating on the CD4 gene. In the absence of Runx1 on the silencer, P-TEFb interacts with the transcription complex, forming …

Muscarinic receptors and receptor-mediated actions on rat thymocytes.

Rat thymocytes possess a single class of saturable, high affinity binding sites for muscarinic antagonists of the benzilate type such as [3H]3-quinuclidinyl benzilate ([3H]3-QNB). The average number of receptors per cell is 3000 and the equilibrium dissociation constant of [3H]3-QNB on intact cells is 7.5 nM. In the work reported here we found that perturbation of the thymocyte membrane by addition of phytohemagglutinin (4 micrograms/ml) caused a transient increase in muscarinic antagonist binding, and hydrocortisone (100 mg/kg s.c.) treatment of rats for 2 days prior to sacrifice increased th…
