[The formation of tooth germs in teratomas].

The teeth buds which are observed in some teratomas ought to provide a useful system for the study of the development of organ structures in these tumours. They exhibit well defined characteristics in space and time. In contrast with normal development tooth bud formation in teratomas display malformations and other observable characteristics which are primarily the consequence of defects in coordination of activities among cells. Cyto-differentiation appears to be normal and the defective organogenesis must represent a breakdown of the principles which underlie the organization of structures.

[Autotransplantation of human tooth germs].

The Author presents three cases of tooth germ transplantation which were periodically checked by x-rays to show the progressive root development. The clinical result obtained were quite satisfactory. The situations requiring the transplantations in the 3 cases were all rather unusual: in one case the tooth germ was inside a large follicular cyst; in the other two the tooth germs were mesiodistally rotated 90 degrees which made eruption impossible. The cases described differ from cases frequently found in literature in that the transplanted tooth germs were in the initial stage of development; …

Tooth germ transposition: report of cases.

Six patients with transposed tooth germ before root formation are reported in cases with cleft lip or palate. A boy with complete bilateral cleft lip and palate had a maxillary permanent canine displaced posteriorly in the vomer. A girl with cleft palate had an upwards and distally transmigrated maxillary canine. Two girls with cleft palate had a transposed maxillary canine with the first premolar. One patient had a maxillary second premolar migrated between the first and second permanent molar, and another had the germ of a supernumerary maxillary premolar between the first and second permane…

Tooth Germ Development After Autotransplantation of an Immature Permanent Tooth.

Hypodontia is commonly associated with cleft lip and/or palate and can be managed by autotransplantation of teeth. In this case report, autotransplantation of the immature mandibular right second premolar to replace the congenitally missing maxillary right second premolar was performed successfully in an 11-year-old Chinese male. An ectopic tooth resembling a premolar tooth was subsequently noted to be developing palatal to the root apex of the transplanted tooth one month after surgery. The development of a dental tooth germ at the site of the autotransplantation post-surgery is a possibility…

Sonographic depiction of fetal tooth germs.

Recently, sonography of the fetal face has gained increasing importance in prenatal diagnosis. It is not yet clear whether sonographic depiction of fetal tooth germs would have an influence on the prenatal diagnosis of ectodermal dysplasia syndromes. During routine malformation screening, horizontal sections of fetal jaws were visualized and examined for tooth germs in 124 pregnant women following sonographic 'facing'. Histological jaw sections of fetuses that had died in utero at various gestational ages were produced in order to examine the degree of correspondence between the sonographic an…

Mitochondrial granule distribution in tooth germ cells.

Incisor and molar tooth germs of albino rats sacrificed at the eighteenth and twentieth days in utero and one to seven days after birth were studied with light and electron microscopy. Observations of the various stages of tooth development in molars established that intramitochondrial granules in odontoblasts were comparable to the intramitochondrial granules of other hard tissue cells. These electron-dense deposits appeared in mitochondria in an appreciable number only when odontoblasts become engaged in dentin mineralization. When dentin mineralization was advanced the odontoblast mitochond…

Application of cryopreservation to tooth germ transplantation for root development and tooth eruption.

We cryopreserved mouse tooth germs with widely open cervical margins of the enamel organ to overcome difficulties in cryoprotectant permeation and tested their efficacy by transplanting them into recipient mice. The upper right first molar germs of 8-day-old donor mice were extracted and categorized into the following four groups according to cryopreservation time: no cryopreservation, 1 week, 1 month, and 3 months. The donor tooth germs were transplanted into the upper right first molar germ sockets of the 8-day-old recipient mice. The upper left first molars of the recipient mice were used a…

Tooth replantation in germ-free and conventional rats.

The purpose of the present study was to evaluate the influence of bacterial infection on the pulpal and periodontal tissues in replanted teeth using germ-free and conventional rats. Forty maxillary and mandibular first molars from ten 6-week-old germ-free male Wistar rats were used. The animals and all materials were maintained in a germ-free environment inside vinyl isolators throughout the experimental periods. Twenty conventional male Wistar rats served as controls. The first molars were intentionally replanted immediately after extraction. At 3 days, and 1, 2, 4 and 8 weeks after replantat

Identification of human tooth germ ectomesenchymal stem cells.

Cranial neural crest cells (CNCCs) contribute to most cranio-maxillofacial mesenchymal tissues, including the dental ectomesenchyme. However … of SLITRK6+ cells enriched in the odontogenic ectomesenchyme. These cells were detected in prenatal tooth germs and postnatal dental progenitor niches, including the apical papilla and dental follicle, but were rare in mature dental tissues. Functionally, SLITRK6+ cells exhibited enhanced clonogenic and odontogenic capacities and supported dentin-like matrix …

[Pharmacological study of tooth formation and mineralization using a tooth germ cultivation system].

In this study, we introduce a cultivation method for mandibular first molars from mouse embryos. We also investigate the in vitro effects of several drugs on tooth formation. Histological observations demonstrated that the tooth germ dissected from 17-day-old embryonic mice and cultivated for 7 days under the conditions described here showed clear mineralization in the dentin and enamel. Alkaline phosphatase (ALP) activity and calcium content in the tooth germ increased developmentally during 7-day cultivation. The increase of calcium was preceded by that of ALP in the germ. The removal of ALP…

Macrophages Promote Vascular Maturation to Support Tooth Germ Morphogenesis.

Macrophages are increasingly recognized as key regulators of organogenesis, yet their contributions to tooth germ development remain poorly understood. Here, we integrated analysis of publicly available single-cell RNA sequencing datasets with multiplex immunohistochemistry to investigate the presence, distribution, and activation states of macrophages during tooth development. Macrophages were enriched in the dental follicle and dental papilla, expanded through local proliferation, and displayed a predominant tissue-resident macrophage (TRM)-like and M2-like phenotype. Spatial and transcripto…

TLR signalling can modify the mineralization of tooth germ.

OBJECTIVE: The aim of this work is to investigate the possible role of Toll-like receptor 4 (TLR4) during the development of mouse tooth germ. TLR4 is well known to inhibit mineralization and cause inflammation in mature odontoblasts and dental pulp cells. However, unlike these pathological functions of TLR4, little is known about the developmental function(s) of TLR4 during tooth development. MATERIALS AND METHODS: TLR4 expression was studied via Western blot in developing lower mouse incisors from E13.5 to E18.5. To generate functional data about the effects of TLR4, a specific agonist (LPS…

Development of rotation of mandibular premolar tooth germs in the dog.

The development of premolar tooth germs in dogs, particularly their rotation and crowding, was examined on radiographs, dissected specimens of tooth germs, serial sections stained by Masson-Goldner's technique and reconstructed models. The materials used were the mandibles of 32 puppies from birth to 6 months, and the species was a highly pedigreed Shiba Inu which has no crowding of the premolars in the mature stage. The results obtained are as follows. (1) Rotations and crowdings of premolars, similar to those in brachygnathus dogs and wolves raised in captivity, were observed during ontogene…
