BACKGROUND: Trifluoperazine is an inexpensive accessible 'high potency' antipsychotic drug, widely used to treat schizophrenia or related psychoses.
OBJECTIVES: To estimate the effects of trifluoperazine compared with placebo and other drugs.
SEARCH STRATEGY: Searches of the Cochrane Schizophrenia Group's register of trials (March 2002), supplemented with hand searching, reference searching, personal communication and contact with industry.
SELECTION CRITERIA: All clinical randomised trials involving people with schizophrenia and comparing trifluoperazine with any other treatment.

A 19-year-old woman, recently discharged from the hospital and being treated for schizophrenia, presented with an unusual reaction to trifluoperazine. She complained of nausea and vomiting and experienced bilateral swelling of the tongue. Symptoms subsided when the medication was discontinued. Although dystonic reactions to high doses of phenothiazines are not uncommon, we postulate that this case represented an unusual allergic reaction to the medication.

Trifluoperazine (TFP), a phenothiazine, is a commonly used antipsychotic drug whose therapeutic effects are attributed to its central anti-adrenergic and anti-dopaminergic actions. This preliminary note describes the preventive action of trifluoperazine against cholesterol-induced atherosclerosis in rabbits. Although this drug had no significant effect on the elevated levels of serum lipids induced by the atherogenic diet, it completely inhibited the initiation of atherosclerotic lesions in rabbits fed atherogenic diets. Based on these findings, we propose that calmodulin and protein kinase 'C' may play a key role in the development of the atherosclerotic process.

Effect of trifluoperazine (TFP) (2.5 X 10(-7) M and 2.5 X 10(-6)M)--an inhibitor of calmodulin, on Langendorff-perfused rabbit hearts was examined. In the normoxic hearts TFP increased coronary flow and reduced force of contraction and oxygen consumption. The cytogenetic effects of trifluoperazine have been studied in male Swiss albino mice using the micronucleus test, chromosomal analysis in germ cells and a sperm morphology assay.
